Biocompatibility and Mineralization Activity of Three Calcium Silicate-Based Root Canal Sealers Compared to Conventional Resin-Based Sealer in Human Dental Pulp Stem Cells

The purpose of this study was to compare the cytotoxic effects and mineralization activity of three calcium silicate-based root canal sealers to those of a conventional resin-based sealer. Experiments were performed using human dental pulp stem cells grown in a monolayer culture. The root canal sealers tested in this study were EndoSequence BC Sealer (Brasseler), BioRoot RCS (Septodont), Endoseal MTA (Maruchi), and AH Plus (Dentsply DeTrey). Experimental disks 6 mm in diameter and 3 mm in height were made and stored in a 100% humidity chamber at 37 °C for 72 h to achieve setting. The cytotoxicity of various root canal sealers was evaluated using a methyl-thiazoldiphenyl-tetrazolium (MTT) assay. To evaluate cell migration ability, a scratch wound healing method was used, and images of the scratch area were taken using a phase-contrast microscope. Cell morphology was evaluated by a scanning electron microscope after direct exposure for 72 h to each sealer disk. In the cell viability assay, there were no significant differences between the EndoSequence BC, BioRoot RCS, Endoseal MTA, and control groups in any experimental period (p > 0.05). In the cell migration assay, there were no significant differences between the EndoSequence BC, Endoseal MTA, and control groups in any experimental period (p > 0.05). BioRoot RCS exhibited slower cell migration relative to EndoSequence BC and Endoseal MTA for up to 72 h (p < 0.05). Conversely, it showed a similar wound healing percentage at 96 h (p > 0.05). In an evaluation of cell morphology, cells in direct contact with EndoSequence BC, BioRoot RCS, and Endoseal MTA disks showed superior spreading compared to those in contact with the AH Plus disk. In an Alizarin red staining assay, EndoSequence BC, BioRoot RCS, and Endoseal MTA showed a significant increase in mineralized nodule formation compared to the AH Plus group (p < 0.05). In conclusion, all calcium silicate-based root canal sealers tested in this study showed good biological properties and mineralization activity compared to conventional resin-based sealer.

Accuracy of age estimation and assessment of the 18-year threshold based on second and third molar maturity in Koreans and Japanese

This study aimed to validate Lee’s age estimation method and assess the 18-year threshold in Korean and Japanese populations. We evaluated the maxillary and mandibular second (M2) and third molars (M3) in 2657 orthopantomograms of the Korean and Japanese populations aged 15–23 years (19.47±2.62 years for Koreans, 19.31±2.60 years for Japanese), using Demirjian’s criteria. Dental age was estimated, and correlations between chronological and dental ages were analyzed. Classification performance was calculated based on the 18-year threshold. The relationship between developmental stage and chronologic age was analyzed using multiple linear regression. Our results revealed that Lee’s method was appropriate for estimation in the Korean population. When the Lee’s method was applied to the Japanese population, a lower value of correlation coefficients between estimated and chronological age, and lower specificity were observed. Population differences were observed predominantly in the stages of root development (stages F and G) of M2s and M3s in both jaws and more frequently in females than in males. In the multiple linear regression between developmental stage and chronological age, lower values of adjusted r 2 were observed in the Japanese population than in the Koreans. In conclusion, the Lee’s method derived from the Korean population data might be unsuitable for Japanese juveniles and adolescents. To support the findings of this study, future studies with samples from multiple institutions should be conducted. Future studies with larger sample sizes are also warranted to improve the accuracy of dental age estimation and confirm the developmental pattern of teeth in the Japanese population.

Predicting early endodontic treatment failure following primary root canal treatment

Background
Understanding when and why endodontic treatments fail could help clinicians make prognoses and thus improve treatment outcomes. This study was aimed to assess potential predictors of early endodontic treatment failure. We explored factors contributing to the failure of initial root canal treatment were explored, with a specific emphasis on evaluating the influence of the time elapsed since the initial treatment.
Methods
This retrospective cohort study enrolled 1262 patients who sought endodontic treatment at our department and 175 patients were included for analysis. Potential causes of endodontic treatment failure were investigated, such as inadequate obturation quality, inadequate coronal status, the presence of additional untreated canals, anatomical complexity, instrument separation, iatrogenic perforation, cracks, and endodontic-periodontal lesions. The patients were divided into “short-term” and “long-term” groups depending on the time that had passed since the initial treatment (i.e., < 5 and > 10 years, respectively). The causes of failure in the short-term and long-term group were analyzed and compared using logistic regression analyses. Subgroup analysis was performed according to the number of years since the initial treatment in the short-term group to further investigate the association between the time and cause of failure (i.e., < 1, 2, 3, and 4 years, respectively).
Results
Untreated additional canals were present in 21.7% of all cases, and in 36.9 and 6.4% of cases in the short-term and long-term groups, respectively. Multivariable analysis showed that the presence of untreated additional canals was significantly associated with short-term compared to long-term failure. Untreated additional canals were also associated with endodontic failure within 1, 2, 3, and 4 years.
Conclusions
The presence of untreated additional canals was a predictor of endodontic failure within 5 years following initial root canal treatment. To optimize long-term prognosis, it is important to detect and treat all root canals during the initial treatment.

Biocompatibility and Bioactivity of Set Direct Pulp Capping Materials on Human Dental Pulp Stem Cells

In this study, we assessed the biocompatibility and bioactivity of various pulp capping materials—ProRoot MTA (Dentsply Tulsa Dental Specialties), Biodentine (Septodont), TheraCal LC (Bisco), and Dycal (Dentsply Caulk)—on human dental pulp stem cells (hDPSCs). Experimental disks (diameter, 7 mm; height, 4 mm) were stored in a humified incubator at 37 °C for 48 h. Then, the pulp capping materials were tested for cytotoxic effects by methyl-thiazoldiphenyl-tetrazolium and scratch wound healing assays, and for mineralization potential by Alizarin red S (ARS) staining assay and alkaline phosphatase enzyme (ALP) activity. Cell viability and cell migration did not significantly differ between ProRoot MTA, Biodentine, and control (p > 0.05). TheraCal LC exhibited slower cell migration on days 2–4 compared to control (p < 0.05), and Dycal showed no cell migration. ALP activity was highest with Biodentine on days 10 and 14, and was lowered with TheraCal LC and Dycal (p < 0.05). In the ARS assay, hDPSCs grown in ProRoot MTA and TheraCal LC eluates showed significantly increased mineralized nodule formation on day 21 compared to Biodentine, Dycal, and control (p < 0.05). These findings indicate that ProRoot MTA, Biodentine, and TheraCal LC exhibit better biocompatibility and bioactivity than Dycal.

A Novel Method for Eliminating Glint in Water-Leaving Radiance from UAV Multispectral Imagery

Unmanned Aerial Vehicle (UAV) high-resolution remote sensing imagery has been used for unprecedented coastal environment monitoring with ground sampling distance and time intervals of a few centimeters and seconds, respectively. However, high spatial-time resolutions of UAV remote sensing data consist of unexpected signals from water surface level changes induced by wind-driven currents and waves. This leads to non-linear and non-stationary forms of sun and sky glints in the UAV sea surface image. Consequently, these surface glints interfere with the detection of water body reflections and objects, reducing the accuracy and usability of the measurements. This study employed Fast and Adaptive Multidimensional Empirical Mode Decomposition (FA-MEMD) to separate the spatial periodicity of time-continuous multispectral images of the sea surface from the original data and retain non-oscillatory signals called residual images. The residual images effectively represented the spatial-temporal radiance and flow variations in the water body by correcting the regions of surface glint. This study presents three key findings: First, homogeneous surface radiance data with surface glint removed from the raw image sequence was acquired using FA-MEMD. Second, the continuous surface glint removal effect is validated through water-leaving radiance (Lw-SBA) measurements obtained via the Skylight-Blocked Approach (SBA) method. Comparisons showed that R2 values for the data obtained from clear water before and after surface glint removal were 0.02 and 0.56 with RMSE values of 8.37 × 10−5 and 5.51 × 10−5 W·m−2·sr−1, respectively, indicating an improvement rate of 34.19%. Third, a comparative analysis with previous study methods demonstrated that our approach yielded spatially and temporally uniform homogeneous surface radiance data with less variability than traditional methods. The spatially and temporally synchronized residual images and the Lw-SBA data showed high similarity, confirming that the FA-MEMD technique effectively removed the surface glint from wave-induced roughness, enhancing the reliability of high-resolution UAV sea color observations.

Osteogenic Potential of Various Premixed Hydraulic Calcium Silicate-Based Sealers on Human Bone Marrow Stem Cells

This study aims to compare the osteogenic potential of premixed hydraulic calcium silicate-based sealers (HCSSs) with an epoxy resin-based sealer in human bone marrow-derived stem cells (hBMSCs). Three HCSSs (White Endoseal MTA, One-Fil, EndoSequence BC Sealer) were compared with AH Plus Jet, an epoxy resin-based sealer. Disk-shaped specimens were prepared using sterilized Teflon tubes and immersed in osteogenic medium to create eluates. hBMSCs were cultured in each eluate, and osteogenic potential was assessed by alkaline phosphatase (ALP) activity (n = 6), Alizarin Red-S (ARS) staining (n = 6), quantitative real-time polymerase chain reaction (qPCR) (n = 3), and Western blot analysis. Statistical analyses were conducted using SPSS (version 24.0), with significance set at p < 0.05. All experimental groups exhibited higher ALP activity than the control on day 4. ARS staining of HCSSs differed significantly from AH Plus Jet on day 14 (p < 0.05), while White Endoseal MTA exhibited the highest intensity. qPCR revealed that EndoSequence BC Sealer induced the highest SMAD1 expression on day 4, while One-Fil and EndoSequence BC Sealer significantly upregulated RUNX2 expression compared with AH Plus Jet (p < 0.05). Western blotting confirmed that EndoSequence BC Sealer induced the highest RUNX2 protein expression. Collectively, premixed HCSSs promoted superior mineralization and RUNX2 expression compared to conventional resin-based sealer in hBMSCs.
