Dorsolateral septum somatostatin interneurons gate mobility to calibrate context-specific behavioral fear responses

Adaptive fear responses to external threats rely upon efficient relay of computations underlying contextual encoding to subcortical circuits. Brain-wide analysis of highly coactivated ensembles following contextual fear discrimination identified the dorsolateral septum (DLS) as a relay of the dentate gyrus–CA3 circuit. Retrograde monosynaptic tracing and electrophysiological whole-cell recordings demonstrated that DLS somatostatin-expressing interneurons (SST-INs) receive direct CA3 inputs. Longitudinal in vivo calcium imaging of DLS SST-INs in awake, behaving mice identified a stable population of footshock-responsive SST-INs during contextual conditioning whose activity tracked and predicted non-freezing epochs during subsequent recall in the training context but not in a similar, neutral context or open field. Optogenetic attenuation or stimulation of DLS SST-INs bidirectionally modulated conditioned fear responses and recruited proximal and distal subcortical targets. Together, these observations suggest a role for a potentially hard-wired DLS SST-IN subpopulation as arbiters of mobility that calibrate context-appropriate behavioral fear responses.Besnard et al. uncover functional heterogeneity of somatostatin interneurons (SST-INs) in the dorsolateral septum and reveal a role for a subpopulation of SST-INs as hippocampal relays that govern mobility to calibrate adaptive fear responses.

Photonic synaptic transistors with new electron trapping layer for high performance and ultra-low power consumption

Photonic synaptic transistors are being investigated for their potential applications in neuromorphic computing and artificial vision systems. Recently, a method for establishing a synaptic effect by preventing the recombination of electron–hole pairs by forming an energy barrier with a double-layer consisting of a channel and a light absorption layer has shown effective results. We report a triple-layer device created by coating a novel electron-trapping layer between the light-absorption layer and the gate-insulating layer. Compared to the conventional double-layer photonic synaptic structure, our triple-layer device significantly reduces the recombination rate, resulting in improved performance in terms of the output photocurrent and memory characteristics. Furthermore, our photonic synaptic transistor possesses excellent synaptic properties, such as paired-pulse facilitation (PPF), short-term potentiation (STP), and long-term potentiation (LTP), and demonstrates a good response to a low operating voltage of − 0.1 mV. The low power consumption experiment shows a very low energy consumption of 0.01375 fJ per spike. These findings suggest a way to improve the performance of future neuromorphic devices and artificial vision systems.

Cultivating Medical Intentionality: The Phenomenology of Diagnostic Virtuosity in East Asian Medicine

This study examines the perceptual basis of diagnostic virtuosity in East Asian medicine, combining Merleau-Ponty’s phenomenology and an ethnographic investigation of Korean medicine in South Korea. A novice, being exposed to numerous clinical transactions during apprenticeship, organizes perceptual experience that occurs between him or herself and patients. In the process, the fledgling practitioner’s body begins to set up a medically-tinged “intentionality” interconnecting his or her consciousness and medically significant qualities in patients. Diagnostic virtuosity is gained when the practitioner embodies a cultivated medical intentionality. In the process of becoming a practitioner imbued with virtuosity, this study focuses on the East Asian notion of “Image” 象 that maximizes the body’s perceptual capacity, and minimizes possible reductions by linguistic re-presentation. “Image” enables the practitioner to somatically conceptualize the core notions of East Asian medicine, such as Yin-Yang, and to use them as an embodied litmus as the practitioner’s cultivated body instinctively conjures up medical notions at clinical encounters. In line with anthropological critiques of reductionist frameworks that congeal human existential and perceptual vitality within a “scientific” explanatory model, this article attempts to provide an example of various knowing and caring practices, institutionalized external to the culture of science.

Clonal dynamics in early human embryogenesis inferred from somatic mutation

Cellular dynamics and fate decision in early human embryogenesis remain largely unknown owing to the challenges of performing studies in human embryos

. Here, we explored whole-genomes of 334 single-cell colonies and targeted deep sequences of 379 bulk tissues obtained from various anatomical locations of seven recently deceased adult human donors. Using somatic mutations as an intrinsic barcode, we reconstructed early cellular phylogenies that demonstrate (1) an endogenous mutational rate that is higher in the first cell division but decreases to approximately one per cell per cell division later in life; (2) universal unequal contribution of early cells to embryo proper, resulting from early cellular bottlenecks that stochastically set aside epiblast cells within the embryo; (3) examples of varying degrees of early clonal imbalances between tissues on the left and right sides of the body, different germ layers and specific anatomical parts and organs; (4) emergence of a few ancestral cells that will substantially contribute to adult cell pools in blood and liver; and (5) presence of mitochondrial DNA heteroplasmy in the fertilized egg. Our approach also provides insights into the age-related mutational processes and loss of sex chromosomes in normal somatic cells. In sum, this study provides a foundation for future studies to complete cellular phylogenies in human embryogenesis.
Adult human tissues from diverse sites around the body are used to reconstruct cellular phylogenies from early development, using somatic mutations as an internal barcode.

High-efficiency electrochemical thermal energy harvester using carbon nanotube aerogel sheet electrodes

Conversion of low-grade waste heat into electricity is an important energy harvesting strategy. However, abundant heat from these low-grade thermal streams cannot be harvested readily because of the absence of efficient, inexpensive devices that can convert the waste heat into electricity. Here we fabricate carbon nanotube aerogel-based thermo-electrochemical cells, which are potentially low-cost and relatively high-efficiency materials for this application. When normalized to the cell cross-sectional area, a maximum power output of 6.6 W m
−2
is obtained for a 51 °C inter-electrode temperature difference, with a Carnot-relative efficiency of 3.95%. The importance of electrode purity, engineered porosity and catalytic surfaces in enhancing the thermocell performance is demonstrated.

We present a technique called white-light diffraction tomography (WDT) for imaging microscopic transparent objects such as live unlabelled cells. The approach extends diffraction tomography to white-light illumination and imaging rather than scattering plane measurements. Our experiments were performed using a conventional phase contrast microscope upgraded with a module to measure quantitative phase images. The axial dimension of the object was reconstructed by scanning the focus through the object and acquiring a stack of phase-resolved images. We reconstructed the three-dimensional structures of live, unlabelled, red blood cells and compared the results with confocal and scanning electron microscopy images. The 350 nm transverse and 900 nm axial resolution achieved reveals subcellular structures at high resolution in
Escherichia coli
cells. The results establish WDT as a means for measuring three-dimensional subcellular structures in a non-invasive and label-free manner.
The three-dimensional structures of transparent objects, such as living cells, are captured by an imaging technique that uses white-light illumination and diffraction tomography to collect a stack of phase-based images.

A 3D-CNN model with CT-based parametric response mapping for classifying COPD subjects

Chronic obstructive pulmonary disease (COPD) is a respiratory disorder involving abnormalities of lung parenchymal morphology with different severities. COPD is assessed by pulmonary-function tests and computed tomography-based approaches. We introduce a new classification method for COPD grouping based on deep learning and a parametric-response mapping (PRM) method. We extracted parenchymal functional variables of functional small airway disease percentage (fSAD%) and emphysema percentage (Emph%) with an image registration technique, being provided as input parameters of 3D convolutional neural network (CNN). The integrated 3D-CNN and PRM (3D-cPRM) achieved a classification accuracy of 89.3% and a sensitivity of 88.3% in five-fold cross-validation. The prediction accuracy of the proposed 3D-cPRM exceeded those of the 2D model and traditional 3D CNNs with the same neural network, and was comparable to that of 2D pretrained PRM models. We then applied a gradient-weighted class activation mapping (Grad-CAM) that highlights the key features in the CNN learning process. Most of the class-discriminative regions appeared in the upper and middle lobes of the lung, consistent with the regions of elevated fSAD% and Emph% in COPD subjects. The 3D-cPRM successfully represented the parenchymal abnormalities in COPD and matched the CT-based diagnosis of COPD.

Reinforcement Learning for Fail-Operational Systems with Disentangled Dual-Skill Variables

We present a novel approach to reinforcement learning (RL) specifically designed for fail-operational systems in critical safety applications. Our technique incorporates disentangled skill variables, significantly enhancing the resilience of conventional RL frameworks against mechanical failures and unforeseen environmental changes. This innovation arises from the imperative need for RL mechanisms to sustain uninterrupted and dependable operations, even in the face of abrupt malfunctions. Our research highlights the system’s ability to swiftly adjust and reformulate its strategy in response to sudden disruptions, maintaining operational integrity and ensuring the completion of tasks without compromising safety. The system’s capacity for immediate, secure reactions is vital, especially in scenarios where halting operations could escalate risks. We examine the system’s adaptability in various mechanical failure scenarios, highlighting its effectiveness in maintaining safety and functionality in unpredictable situations. Our research represents a significant advancement in the safety and performance of RL systems, paving the way for their deployment in safety-critical environments.

The broad-line region (BLR) size–luminosity relation has paramount importance for estimating the mass of black holes in active galactic nuclei (AGNs). Traditionally, the size of the Hβ BLR is often estimated from the optical continuum luminosity at 5100 Å, while the size of the Hα BLR and its correlation with the luminosity is much less constrained. As a part of the Seoul National University AGN Monitoring Project, which provides 6 yr photometric and spectroscopic monitoring data, we present our measurements of the Hα lags of high-luminosity AGNs. Combined with the measurements for 42 AGNs from the literature, we derive the size–luminosity relations of the Hα BLR against the broad Hα and 5100 Å continuum luminosities. We find the slope of the relations to be 0.61 ± 0.04 and 0.59 ± 0.04, respectively, which are consistent with the Hβ size–luminosity relation. Moreover, we find a linear relation between the 5100 Å continuum luminosity and the broad Hα luminosity across 7 orders of magnitude. Using these results, we propose a new virial mass estimator based on the Hα broad emission line, finding that the previous mass estimates based on scaling relations in the literature are overestimated by up to 0.7 dex at masses lower than 107 M ⊙.

Biomass-Derived Sustainable Electrode Material for Low-Grade Heat Harvesting

The ever-increasing energy demand and global warming caused by fossil fuels push for the exploration of sustainable and eco-friendly energy sources. Waste thermal energy has been considered as one of the promising candidates for sustainable power generation as it is abundantly available everywhere in our daily lives. Recently, thermo-electrochemical cells based on the temperature-dependent redox potential have been intensely studied for efficiently harnessing low-grade waste heat. Despite considerable progress in improving thermocell performance, no attempt was made to develop electrode materials from renewable precursors. In this work, we report the synthesis of a porous carbon electrode from mandarin peel waste through carbonization and activation processes. The influence of carbonization temperature and activating agent/carbon precursor ratio on the performance of thermocell was studied to optimize the microstructure and elemental composition of electrode materials. Due to its well-developed pore structure and nitrogen doping, the mandarin peel-derived electrodes carbonized at 800 °C delivered the maximum power density. The areal power density (P) of 193.4 mW m−2 and P/(ΔT)2 of 0.236 mW m−2 K−2 were achieved at ΔT of 28.6 K. However, KOH-activated electrodes showed no performance enhancement regardless of activating agent/carbon precursor ratio. The electrode material developed here worked well under different temperature differences, proving its feasibility in harvesting electrical energy from various types of waste heat sources.
