Using a novel computational drug-repositioning approach (DrugPredict) to rapidly identify potent drug candidates for cancer treatment
Computation-based drug-repurposing/repositioning approaches can greatly speed up the traditional drug discovery process. To date, systematic and comprehensive computation-based approaches to identify and validate drug-repositioning candidates for epithelial ovarian cancer (EOC) have not been undertaken. Here, we present a novel drug discovery strategy that combines a computational drug-repositioning system (DrugPredict) with biological testing in cell lines in order to rapidly identify novel drug candidates for EOC. DrugPredict exploited unique repositioning opportunities rendered by a vast amount of disease genomics, phenomics, drug treatment, and genetic pathway and uniquely revealed that non-steroidal anti-inflammatories (NSAIDs) rank just as high as currently used ovarian cancer drugs. As epidemiological studies have reported decreased incidence of ovarian cancer associated with regular intake of NSAIDs, we assessed whether NSAIDs could have chemoadjuvant applications in EOC and found that (i) NSAID Indomethacin induces robust cell death in primary patient-derived platinum-sensitive and platinum- resistant ovarian cancer cells and ovarian cancer stem cells and (ii) downregulation of β-catenin is partially driving effects of Indomethacin in cisplatin-resistant cells. In summary, we demonstrate that DrugPredict represents an innovative computational drug- discovery strategy to uncover drugs that are routinely used for other indications that could be effective in treating various cancers, thus introducing a potentially rapid and cost-effective translational opportunity. As NSAIDs are already in routine use in gynecological treatment regimens and have acceptable safety profile, our results will provide with a rationale for testing NSAIDs as potential chemoadjuvants in EOC patient trials.

Thermal remodelling of Alternanthera mosaic virus virions and virus-like particles into protein spherical particles

The present work addresses the thermal remodelling of flexible plant viruses with a helical structure and virus-like particles (VLPs). Here, for the first time, the possibility of filamentous Alternanthera mosaic virus (AltMV) virions’ thermal transition into structurally modified spherical particles (SP) has been demonstrated. The work has established differences in formation conditions of SP from virions (SP V ) and VLPs (SP VLP ) that are in accordance with structural data (on AltMV virions and VLPs). SP originate from AltMV virions through an intermediate stage. However, the same intermediate stage was not detected during AltMV VLPs’ structural remodelling. According to the biochemical analysis, AltMV SP V consist of protein and do not include RNA. The structural characterisation of AltMV SP V/VLP by circular dichroism, intrinsic fluorescence spectroscopy and thioflavin T fluorescence assay has been performed. AltMV SP V/VLP adsorption properties and the availability of chemically reactive surface amino acids have been analysed. The revealed characteristics of AltMV SP V/VLP indicate that they could be applied as protein platforms for target molecules presentation and for the design of functionally active complexes.

The paradigm of emergent qualities of education management as a scientific and technological platform for sustainable development

In this paper the factors influencing sustainable development were identified. The influence of emergent qualities of education management on stable development is highlighted. The analysis of the rating of countries by the level of education, the human development index was carried out. A cognitive map-review of systematized factors of influence on the development of education in the modern socio-economic environment has been constructed. Economic-mathematical models that describe trends in indicators of the impact of emergent qualities of education management on sustainable development and testify to a fairly stable relationship between the factors influencing the stable development of education management have been constructed. It is proved that the emergent qualities of education management have a direct impact on the creation of a scientific and technological platform in Ukraine as an effective mechanism for partnership and interaction between education, science and business.

It is established that the polysaccharide glucuronoxylomannan decorated with gold nanoparticles significantly changes the spatial structuring depending on pH and temperature. A mechanism of the biopolymer conformation effect on the formation process and properties of a hybrid material is proposed. The most stable structures are formed in an alkaline medium, however, they also retain the possibility of adaptive behavior of an initial biomolecule. It has been demonstrated that biological macromolecules can be applied to form “smart” molecular hybrids with the ability to switch properties in response to external stimuli.

The effect of heating rate (in the range of 0.083–3.333 K s−1) on the parameters of thermal stability and crystallization kinetics of the Fe40Ni40P14B6 metallic glass has been investigated by differential scanning calorimetry and X-ray diffraction. An analytical model of glass crystallization describing the homogeneous nucleation rate, the velocity of interface-limited growth, the number of crystal volume density in crystallized samples as well as the crystallization kinetics at constant rate heating is presented. A modified procedure of estimation of thermodynamic and kinetic parameters governing the rate of crystal nucleation and growth based on the use of the measured heating rate-dependent variations of the volume fraction crystallized and the average grain size is proposed. The values of the effective diffusivity have been estimated by the Kissinger-like isoconversional method accounting the contribution of both the free energy difference and the specific interfacial nucleus-melt energy changes estimated from the structural data. It has been shown for the first time that the shapes of the non-isothermal experimental kinetic crystallization curves measured at the heating rates ≤ 1.333 K s−1 are well described by the approximate analytical equation and the values of the Avrami exponent lowering from 5.5 to 2.3 with the heating rates increasing have been estimated. The evaluated ranges of the nucleation rate (1.4 × 1016–2.5 × 1017 m−3 s−1) and growth velocity of crystals (2.05 × 10−9–3.0 × 10−7 m s−1) in Fe40Ni40P14B6 glass at temperatures from 653 to 714 K are in reasonable agreement with the experimental data. Possible variations of the established eutectic crystallization mechanism revealed by changes of the Avrami exponent with the heating rate increasing are discussed.

This article analyses the results of the proposed approach to quality assessment of the use of online courses in the system of Ukrainian Engineering Pedagogics Academy e-learning and to assess its effectiveness in solving the problem of improving the quality of education. The courses were inspected after the Spring 2020 semester, according to the following criteria: activity of the process participants; interactivity of the process when using an online course; the intensity of the process by the elements of a course; dynamics of students' educational achievements and management of the process of using an online course. This article shows that without painstaking daily systematic work regarding supporting the use of each online course, one should not hope for the effective impact of this form of studying. This should be done in every educational institution because the COVID-19 pandemic is not over and a significant part of educational institutions will spend the 2020/2021 academic year under the strict quarantine.

The Decadal-Multidecadal Changes of Extreme Air Temperature in the Black Sea Region

The decadal-multidecadal changes of extreme air temperature characteristics during winter and summer in the Black Sea region are studied in the paper. The 5th and 95th percentiles of intra-seasonal daily temperature anomalies and number of days with air temperature below the 5th and above the 95th percentile were used to describe moderate extremes. The average daily air temperature data from 13 hydrometeorological stations in 1950-2014 and from several stations in 1900-2005 were analyzed. Pacific Decadal oscillation (PDO) and the Atlantic Multidecadal oscillation (AMO) were considered as the main global processes in the ocean-atmosphere system responsible for regional climate change on decadal-multidecadal scale. The differences of extreme air temperature characteristics between positive and negative AMO and PDO phases are analyzed in the paper. It is found that the extreme winter air temperature anomalies are increased by about 2 °C during the positive PDO phase and negative AMO phase. The winter frequency of extreme low temperature is decreased by two times during the positive PDO phase and negative AMO phase. The summer changes of extreme temperature characteristics, associated with the PDO and AMO phases are insignificant.

Consideration of ecologization and standardization in the management of agricultural business operations

The article deals with the formation of financial statements of agricultural enterprises, taking into account the indicators of ecologization and standardization in their activities. It is noted that at present time there is a problem of developing innovative directions of managing the strategic development of enterprises based on the use of new forms of financial evaluation of economic activity and compliance with environmental standards. The approaches to a comprehensive assessment of the financial condition of enterprises have been defined, their characteristics and strengths and weaknesses have been outlined. During the financial estimation of the economic activity of the enterprise the set of coefficients must be taken into account, which are formed taking into consideration the indicators of the rating estimation for each parameter of ecologization and standardization. A comprehensive analysis of the financial condition of agricultural enterprises on the basis of Ukrainian and international financial reporting standards has been conducted. It is summarized that the innovative management system based on the use of environmental and unified reporting standards will ensure the improvement of strategic planning mechanisms of socio-economic policy development at the state, regional and local levels.

Interannual variability of anticyclone activity and temperature extremes in the Black sea region

Manifestations of North Atlantic oscillation (NAO) during its negative and positive phases and during different El Niño (EN) types in the anticyclone parameters and air temperature extreme thresholds in the Black sea region are studied. It is estimated quantitative characteristics of anticyclone frequency anomalies and temperature extremes associated with the NAO phases and the EN types.

The Dependencies of X-Ray Conductivity and X-Ray Luminescence of ZnSe Crystals on the Excitation Intensity

This work studies the conductivity and luminescence of ZnSe single crystals under X-ray irradiation. The experimentally derived lux-ampere characteristics of the X-ray conductivity for ZnSe crystals have a sublinear behavior within the temperature range from 8 to 420 K. The theoretical analysis of the conductivity kinetics at X-ray excitation showed that the value of maximum accumulated lightsum at deep traps does not depend on radiation intensity. However, regarding shallow and phosphorescent traps, the strength of accumulated lightsum depends on the intensity of exciting irradiation. Specifically, these shallow traps and phosphorescent traps cause the sublinear behavior of lux-ampere characteristics in the semiconductor material.
