Chlamydial Entry Involves TARP Binding of Guanine Nucleotide Exchange Factors
Chlamydia trachomatis attachment to cells induces the secretion of the elementary body-associated protein TARP (Translocated Actin Recruiting Protein). TARP crosses the plasma membrane where it is immediately phosphorylated at tyrosine residues by unknown host kinases. The Rac GTPase is also activated, resulting in WAVE2 and Arp2/3-dependent recruitment of actin to the sites of chlamydia attachment. We show that TARP participates directly in chlamydial invasion activating the Rac-dependent signaling cascade to recruit actin. TARP functions by binding two distinct Rac guanine nucleotide exchange factors (GEFs), Sos1 and Vav2, in a phosphotyrosine-dependent manner. The tyrosine phosphorylation profile of the sequence YEPISTENIYESI within TARP, as well as the transient activation of the phosphatidylinositol 3-kinase (PI3-K), appears to determine which GEF is utilized to activate Rac. The first and second tyrosine residues, when phosphorylated, are utilized by the Sos1/Abi1/Eps8 and Vav2, respectively, with the latter requiring the lipid phosphatidylinositol 3,4,5-triphosphate. Depletion of these critical signaling molecules by siRNA resulted in inhibition of chlamydial invasion to varying degrees, owing to a possible functional redundancy of the two pathways. Collectively, these data implicate TARP in signaling to the actin cytoskeleton remodeling machinery, demonstrating a mechanism by which C.trachomatis invades non-phagocytic cells.
A mechanism for real-time decision making and system maintenance for resource constrained robotic systems through ReFrESH
Robot operating environments and the status of robots are complex and varying, so it is practically impossible for a robotics designer to anticipate all system configurations to successfully complete a task prior to deployment. Therefore, a mechanism for dynamic decision making and configuration synthesis that copes with system fault and uncertainty is necessary. This paper implements such a mechanism within a self-adaptive framework (ReFrESH). The goal of this presented mechanism is to provide diagnosability and maintainability to manage the system performance during task execution in the presence of unexpected uncertainties. Specifically, the functionality of the proposed mechanism include: (1) detection of system performance degradation; (2) diagnosis and locate of the fault module; (3) synthesis of feasible task configurations; (4) selection of the optimal one. We illustrate the feasibility of the proposed mechanism through a visual servoing task.
Transcriptomic Analysis of Octanoic Acid Response in Drosophila sechellia Using RNA-Sequencing
The dietary specialist fruit fly Drosophila sechellia has evolved to specialize on the toxic fruit of its host plant Morinda citrifolia. Toxicity of Morinda fruit is primarily due to high levels of octanoic acid (OA). Using RNA interference (RNAi), prior work found that knockdown of Osiris family genes Osiris 6 (Osi6), Osi7, and Osi8 led to increased susceptibility to OA in adult D. melanogaster flies, likely representing genes underlying a Quantitative Trait Locus (QTL) for OA resistance in D. sechellia. While genes in this major effect locus are beginning to be revealed, prior work has shown at least five regions of the genome contribute to OA resistance. Here, we identify new candidate OA resistance genes by performing differential gene expression analysis using RNA-sequencing (RNA-seq) on control and OA-exposed D. sechellia flies. We found 104 significantly differentially expressed genes with annotated orthologs in D. melanogaster, including six Osiris gene family members, consistent with previous functional studies and gene expression analyses. Gene ontology (GO) term enrichment showed significant enrichment for cuticle development in upregulated genes and significant enrichment of immune and defense responses in downregulated genes, suggesting important aspects of the physiology of D. sechellia that may play a role in OA resistance. In addition, we identified five candidate OA resistance genes that potentially underlie QTL peaks outside of the major effect region, representing promising new candidate genes for future functional studies.
The Impact of a Peer Observation Program on Collective Efficacy for Teachers: Teacher Perceptions
The purpose of this mixed method action research was to better understand the impact of a peer observation program on teacher efficacy in a suburban high school. The study used a peer observation treatment group and a control group. Both groups took a collective efficacy scale, CE-Scale twice. The peer observation group also participated in a peer observation program with a colleague from their academic department. The research and information gathered showed that formal evaluation is most effective when it leads to feedback and reflection, teachers seek personalized professional development, a trusting relationship with an observer leads to increased collaboration, and positive teacher views of a peer observation program leads to increased collaboration and fosters student learning.
A Pilot Study on Using an Intelligent Life-like Robot as a Companion for Elderly Individuals with Dementia and Depression
This paper presents the design, development, methodology, and the results of a pilot study on using an intelligent, emotive and perceptive social robot (aka Companionbot) for improving the quality of life of elderly people with dementia and/or depression. Ryan Companionbot prototyped in this project, is a rear-projected life-like conversational robot. Ryan is equipped with features that can (1) interpret and respond to users' emotions through facial expressions and spoken language, (2) proactively engage in conversations with users, and (3) remind them about their daily life schedules (e.g. taking their medicine on time). Ryan engages users in cognitive games and reminiscence activities. We conducted a pilot study with six elderly individuals with moderate dementia and/or depression living in a senior living facility in Denver. Each individual had 24/7 access to a Ryan in his/her room for a period of 4-6 weeks. Our observations of these individuals, interviews with them and their caregivers, and analyses of their interactions during this period revealed that they established rapport with the robot and greatly valued and enjoyed having a Companionbot in their room.
