Factor VIII gene transfer with a single intravenous infusion of valoctocogene roxaparvovec (AAV5-hFVIII-SQ) has demonstrated clinical benefits lasting 5 years to date in people with severe hemophilia A. Molecular mechanisms underlying sustained AAV5-hFVIII-SQ-derived FVIII expression have not been studied in humans. In a substudy of the phase 1/2 clinical trial (

), liver biopsy samples were collected 2.6–4.1 years after gene transfer from five participants. Primary objectives were to examine effects on liver histopathology, determine the transduction pattern and percentage of hepatocytes transduced with AAV5-hFVIII-SQ genomes, characterize and quantify episomal forms of vector DNA and quantify transgene expression (hFVIII-SQ RNA and hFVIII-SQ protein). Histopathology revealed no dysplasia, architectural distortion, fibrosis or chronic inflammation, and no endoplasmic reticulum stress was detected in hepatocytes expressing hFVIII-SQ protein. Hepatocytes stained positive for vector genomes, showing a trend for more cells transduced with higher doses. Molecular analysis demonstrated the presence of full-length, inverted terminal repeat-fused, circular episomal genomes, which are associated with long-term expression. Interindividual differences in transgene expression were noted despite similar successful transduction, possibly influenced by host-mediated post-transduction mechanisms of vector transcription, hFVIII-SQ protein translation and secretion. Overall, these results demonstrate persistent episomal vector structures following AAV5-hFVIII-SQ administration and begin to elucidate potential mechanisms mediating interindividual variability.
The analysis of liver biopsy samples after AAV gene therapy for hemophilia A reveals normal histology and long-term persistence of the episomal vector, and identifies potential factors contributing to interindividual variability of transgene expression.

Magnetite (Fe3O4) have been thoroughly investigated as microwave absorbing material due to its excellent electromagnetic properties (permittivity and permeability) and favorable saturation magnetization. However, large density and impedance mismatch are some of the limiting factors that hinder its microwave absorption performance (MAP). Herein, Fe3O4 nanoparticles prepared by facile co-precipitation method have been coated with citric acid and embedded in a polyvinylidene fluoride (PVDF) matrix. The coated Fe3O4 nanoparticles were characterized by X-ray diffraction spectrometer (XRD), field emission scanning electron microscopy (FESEM), Fourier transform infrared spectroscopy (FTIR), and vibrating sample magnetometer (VSM). COMSOL Multiphysics based on the finite element method was used to simulate the rectangular waveguide at X-band and Ku-band frequency range in three-dimensional geometry. The citric acid coated Fe3O4/PVDF composite with 40 wt.% filler loading displayed good microwave absorption ability over the studied frequency range (8.2–18 GHz). A minimum reflection loss of −47.3 dB occurs at 17.9 GHz with 2.5 mm absorber thickness. The composite of citric acid coated Fe3O4 and PVDF was thus verified as a potential absorptive material with improved MAP. These enhanced absorption coefficients can be ascribed to favorable impedance match and moderate attenuation.

Listening is an important tool of learning as it involves devoting time and attention to the acquisition of knowledge competence on an academic subject in education. Most students regard listening as a complex interactive process which involves a dynamic process of construction of meaning. As a result of this, students at the secondary school level exhibit poor listening habit and their performance has not been encouraging in English Language. Hence, this paper sought the analysis of listening comprehension performance of senior secondary school students at the discriminative, informative and critical levels of listening. The study was a descriptive survey research. The population for the study consisted of all the senior secondary schools in Abeokuta, Nigeria. Random sampling technique was used to select 315 SSII students which served as the sample for this study. The instrument used for the study was a Listening Comprehension Test. Percentage and ANOVA were used to answer the research questions raised and analyse the hypotheses generated at 0.05 level of significance. Findings from the study showed that the general level of listening comprehension performance of senior secondary school students in Abeokuta was high. There was a significant difference in the senior secondary school students' performance in listening at the discriminative, informative and critical levels on account of subject combination. It was concluded that the general level of listening comprehension of secondary school students was high. Based on the findings, the researcher recommended that greater premium should be placed on the teaching of listening comprehension in public schools. Curriculum planners and policy makers are to readdress the issue of listening skills in secondary schools.

Valoctocogene Roxaparvovec Gene Therapy for Hemophilia A

Patients with severe hemophilia A were treated with an adenoviral construct containing coagulation factor VIII cDNA and followed for 1 to 3 years. Median factor VIII activity at 49 to 52 weeks was 24 IU per deciliter, and annualized bleeding rates decreased after treatment. Elevations in alanine aminotransferase were the most common toxic effect and were mainly controlled with glucocorticoids.

The study examined the effect of the adoption of an e-tax filing system on SMEs compliance. This was with a view to providing information on the compliance of SMEs with electronic tax filing systems in Lagos State. The study employed a descriptive survey research design, targeting a population of 91,097 SMEs. Taro Yamane’s formula was used to obtain a sample of 398 SMEs. Data were collected through well-structured questionnaires and analyzed using ordinal logit regression techniques. The study showed that factors such as performance expectancy, effort expectancy, and e-tax filing awareness have a positive significant influence on SMEs compliance in Lagos State. The study concluded that the electronic tax filing system positively impacted tax compliance of SMEs in Lagos State. The study recommended that tax authorities should enhance the performance and effort expectancy of the e-tax filing system in Lagos State by addressing technical issues, streamlining the filing process, and improving user interface design.

Reading is an indispensable tool through which a student can achieve a good academic pursuit and the teacher is a significant figure in achieving this through effective teaching. For a teacher to be effective and efficient at teaching, he needs to carefully plan, logically implement and evaluate adequately for feedback since it has been confirmed by language scholars that students are no more reading as they should. This study, however, examined the variable of teachers on their mathemagenic behaviour in reading comprehension instruction in Ogbomoso, Nigeria. The study is a descriptive survey of cross-sectional type and the population was all the senior secondary schools English language teacher, in both public and private schools, as well as urban and rural schools. Both quantitative and qualitative data were used to collect data from the sample. The research question was answered using the mean and standard deviation. Questions 2, 3, 5 and 6 with their corresponding hypotheses were tested using the t-test statistical method. One way ANOVA was used to test Research Question three together with the corresponding hypothesis at 0.5 level of significance. The study concluded that the sample did employ mathemagenic strategy in reading comprehension instructions. Also, Teacher Variables such as gender, educational qualification, teaching experience and school proprietorship did not pose any significant difference in the teachers' mathemagenic behaviour but that of the school location did. It was thereafter recommended that, teachers in rural areas should buckle up as their counterparts in the urban areas by employing the needed strategies in the reading comprehension instruction so as to bridge the gap in the performances of students in the rural and urban schools.

Reading and Expression Writing Skills of Senior Secondary School Students of Arabic in Oyo State

Reading and writing are fundamental precept to all round education. Efficiency in the skills influence's performances in language and communication, specifically student's ability to read and write in a language like Arabic which is a foreign language but quite desirable in Nigeria tend to show the degree of learners' academic progress. Unfortunately, student's performance in Nigeria secondary schools in Arabic reading and writing skills of WASCCE & NECO Examination over the years have been somehow poor. This study therefore compared students' performances in reading and expression writing skills in Arabic, Oyo state. The study is descriptive correlation method involving all the senior secondary 3 students of Arabic in the three senatorial districts of Oyo state using purposive sampling techniques to select 11 9 students from the 3 districts. Arabic reading comprehensive performance test and Arabic writing performance test was used for data collection. Validation of instrument yielded correlations value of 0.62. There is research question with corresponding hypothesis which was tested using Pearson correlation co-efficient statistic at 0.05 level of significance. The result shows that reading skills significantly influenced students expression skill in writing because cal.r (0.349) > table r (0.178). Based on the findings it was recommended that teachers of Arabic need enlighten on better approach for teaching of reading and expression in writing skill.
