In this study, we provide crystallographic evidence, along with some binding assay data, that three clinically approved anti hepatitis C virus drugs and two other drug-like compounds covalently bind to the M
pro
Cys145 catalytic residue in the active site. Also, molecular docking studies can provide additional insight for the design of new antiviral inhibitors for SARS-CoV-2 using these drugs as lead compounds. One might consider derivatives of these lead compounds with higher affinity to the M
pro
as potential COVID-19 therapeutics for further testing and possibly clinical trials.

AMX – the highly automated macromolecular crystallography (17‐ID‐1) beamline at the NSLS‐II

The highly automated macromolecular crystallography beamline AMX/17‐ID‐1 is an undulator‐based high‐intensity (>5 × 1012 photons s−1), micro‐focus (7 µm × 5 µm), low‐divergence (1 mrad × 0.35 mrad) energy‐tunable (5–18 keV) beamline at the NSLS‐II, Brookhaven National Laboratory, Upton, NY, USA. It is one of the three life science beamlines constructed by the NIH under the ABBIX project and it shares sector 17‐ID with the FMX beamline, the frontier micro‐focus macromolecular crystallography beamline. AMX saw first light in March 2016 and started general user operation in February 2017. At AMX, emphasis has been placed on high throughput, high capacity, and automation to enable data collection from the most challenging projects using an intense micro‐focus beam. Here, the current state and capabilities of the beamline are reported, and the different macromolecular crystallography experiments that are routinely performed at AMX/17‐ID‐1 as well as some plans for the near future are presented. AMX (17‐ID‐1) is the highly automated macromolecular crystallography beamline at the NSLS‐II. Photon delivery system, beamline instrumentation, high‐performance computing environment and suites of applications are described for beamline scientists and users. AMX's primary mission is to support routine structure determination from the most challenging projects.

IntroductionThere is an urgent need to reduce the burden of non-communicable diseases (NCDs), particularly in low-and middle-income countries, where the greatest burden lies. Yet, there is little research concerning the specific issues involved in scaling up NCD interventions targeting low-resource settings. We propose to examine this gap in up to 27 collaborative projects, which were funded by the Global Alliance for Chronic Diseases (GACD) 2019 Scale Up Call, reflecting a total funding investment of approximately US$50 million. These projects represent diverse countries, contexts and adopt varied approaches and study designs to scale-up complex, evidence-based interventions to improve hypertension and diabetes outcomes. A systematic inquiry of these projects will provide necessary scientific insights into the enablers and challenges in the scale up of complex NCD interventions.Methods and analysisWe will apply systems thinking (a holistic approach to analyse the inter-relationship between constituent parts of scaleup interventions and the context in which the interventions are implemented) and adopt a longitudinal mixed-methods study design to explore the planning and early implementation phases of scale up projects. Data will be gathered at three time periods, namely, at planning (TP), initiation of implementation (T0) and 1-year postinitiation (T1). We will extract project-related data from secondary documents at TP and conduct multistakeholder qualitative interviews to gather data at T0 and T1. We will undertake descriptive statistical analysis of TP data and analyse T0 and T1 data using inductive thematic coding. The data extraction tool and interview guides were developed based on a literature review of scale-up frameworks.Ethics and disseminationThe current protocol was approved by the Monash University Human Research Ethics Committee (HREC number 23482). Informed consent will be obtained from all participants. The study findings will be disseminated through peer-reviewed publications and more broadly through the GACD network.

Experiencias de aprendizaje en las Redes Integradas de Salud en zona rural

Las experiencias de los gestores salubristas estuvo referida a los aprendizajes adquiridos en el proceso de implementación de las Redes Integradas de Salud (RIS) en la provincia de Huamanga. El objetivo del trabajo fue conocer las experiencias de aprendizaje de los salubristas y contribuir al desarrollo integral de salud en la zona rural. El enfoque de estudio fue cualitativo bajo un diseño fenomenológico, obteniéndose la información mediante la entrevista semiestructurada a gestores de la organización. Se aplicó un análisis inductivo a través de la percepción de profesionales multidisciplinarios salubristas en el proceso de implementación como una actividad de innovación personal, la oportunidad de fortalecer sus aptitudes, conocimientos y transferir intercambios culturales mejorando las actitudes de nuevos valores profesionales.

El sistema ferroviario en el Perú está siguiendo la tendencia a nivel de América Latina y el Caribe en cuanto a la especialización del uso de ferrocarriles para el transporte de carga y pasajeros, es por ello que en los últimos años se han ido desarrollando nuevos proyectos en el país en los que incluso están involucrados otros países de la región. El objetivo del presente PEA es contribuir con un sistema de transporte que contribuya a la competitividad de las materias y productos exportables en los tramos ferroviarios Sur y Sur – Oriente, así como también de generar una mejor experiencia de viaje que se adecue a las necesidades del transporte de turistas y de la población local. El transporte de carga y de pasajeros en el sector ferroviario ofrece diferentes ventajas competitivas como por ejemplo la eficiencia en el consumo de combustible en cuanto a la cantidad transportada por kilómetro al ser un medio transporte masivo, la emisión de CO2 frente a la contaminación medio ambiental que genera el transporte por carretera, la seguridad que ofrece al ser considerado como uno de los medios de transporte con menores tasas de accidentes por año, entre otros. Para lograr alcanzar estas ventajas competitivas se han propuesto cinco objetivos a largo plazo que se detallan a continuación: a) Para el 2023, FETRASA logrará la renovación del 21.62% de su infraestructura férrea, entre los tramos La Joya e Imata, b) Para el 2025, FETRASA contará con la certificación FRA III en el tramo de Matarani e Imata. Al 2019 se labora con FRA II, c) Para el 2025, FETRASA logrará el ingreso de una empresa operadora de trenes al Perú, d) Para el 2025, FETRASA logrará la reducción de accidentes provocados por el estado de la vía en un 95%, e) Para el 2026, FETRASA logrará incrementar a cinco las estaciones de transferencia con la finalidad de articular las carreteras con la vía férrea. Estos objetivos de largo plazo han sido complementados por 19 objetivos de corto plazo. Así mismo, se han desarrollado 26 estrategias y dentro de ellas tenemos 13 estrategias retenidas y 13 de contingencias.

Study habits are student behaviors on a regular basis, incorporating knowledge into the cognitive structure. The objective of this research was to characterize study habits in exact science students from the Hermilio Valdizán National University in the city of Huánuco-Peru. The research is non-experimental, descriptive and comparative with a stratified sample of probabilistic type for a population of 97 students that branched out into homogeneous non-overlapping and eligible groups with various characteristics, represented in 24 students for the academic period 2017. A test was applied from study habits to each one; including 38 dichotomous questions, classified into five dimensions. The descriptive analysis analyzed the frequency of the responses and the percentages, as well as the calculation of the median as a measure of central tendency. The comparative analysis was performed with the Mann-Whitney W non-parametric test (Wilcoxon). The most representative correlation of the research lies between class attention and general attitude (.769). The best results of the investigation explain that the most important quality for an exact science student in the development of his study habits is; place of study and general attitude.
Los hábitos de estudio son conductas en los estudiantes de forma regular, incorporando conocimientos a la estructura cognitiva. La presente investigación tuvo como objetivo caracterizar hábitos de estudio en estudiantes de ciencias exactas de la Universidad Nacional Hermilio Valdizán en la ciudad de Huánuco-Perú. La investigación es no experimental, descriptiva y comparativa con una muestra estratificada de tipo probabilística para una población de 97 estudiantes que se ramificó en grupos homogéneos no superpuestos y elegibles con diversas características, representado en 24 estudiantes para el lapso académico 2017. Se aplicó un test de hábitos de estudio a cada uno; incluyendo 38 preguntas dicotómicas, clasificadas en cinco dimensiones. El análisis descriptivo, analizó la frecuencia de las respuestas y los porcentajes, así como el cálculo de la mediana como medida de tendencia central. El análisis comparativo se realizó con la prueba no paramétrica W de Mann-Whitney (Wilcoxon). La correlación más representativa de la investigación recae entre atención en clase y actitud general (.769). Los mejores resultados de la investigación, explican que la cualidad más importante para un estudiante de ciencias exactas en el desarrollo de sus hábitos de estudios es; lugar de estudio y actitud general.
