VEGF/VEGFR-Targeted Therapy and Immunotherapy in Non-small Cell Lung Cancer: Targeting the Tumor Microenvironment

Non-small cell lung cancer (NSCLC) is the leading cause of death by cancer worldwide. Despite developments in therapeutic approaches for the past few decades, the 5-year survival rate of patients with NSCLC remains low. NSCLC tumor is a complex, heterogeneous microenvironment, comprising blood vessels, cancer cells, immune cells, and stroma cells. Vascular endothelial growth factors (VEGFs) are a major mediator to induce tumor microvasculature and are associated with the progression, recurrence, and metastasis of NSCLC. Current treatment medicines targeting VEGF/VEGF receptor (VEGFR) pathway, including neutralizing antibodies to VEGF or VEGFR and receptor tyrosine kinase inhibitors, have shown good treatment efficacy in patients with NSCLC. VEGF is not only an important angiogenic factor but also an immunomodulator of tumor microenvironment (TME). VEGFs can suppress antigen presentation, stimulate activity of regulatory T (Treg) cells, and tumor-associated macrophages, which in turn promote an immune suppressive microenvironment in NSCLC. The present review focuses on the angiogenic and non-angiogenic functions of VEGF in NSCLC, especially the interaction between VEGF and the cellular components of the TME. Caspase-1 affects chronic restraint stress-induced depression-like behaviors by modifying GABAergic dysfunction in the hippocampus

Major depression disorder (MDD) is one of the most common psychiatric disorders and one of the leading causes of disability in worldwide. Both inflammation and GABAergic dysfunction have been implicated in the pathophysiology of MDD. Caspase-1, a classic inflammatory caspase, regulates AMPARs-mediated glutamatergic neurotransmission. However, the role of caspase-1 in chronic stress-induced GABAergic dysfunction remains largely unknown. In this study, we found that serum and hippocampal caspase-1-IL-1β levels increased significantly in chronic restraint stress (CRS) mice, and a significant negative correlation occurred between levels of caspase-1 and depression-like behaviors. Furthermore, CRS significantly decreased GAD67 mRNA levels and GABAergic neurotransmission accompanied by the reduction of GABA concentration, reduced the amplitude and frequency of mIPSCs inhibitory postsynaptic currents (mIPSCs) and the decreased surface expression of GABAARs γ2 subunit in the hippocampus. Genetic deficiency of caspase-1 not only blocked CRS-induced depression-like behaviors, but also alleviated CRS-induced impairments in GABAergic neurotransmission. Finally, reexpression of caspase-1 in the hippocampus of Caspase-1−/− mice increased susceptibility to stress-induced anxiety- and depression-like behaviors through inhibiting GAD67 expression and GABAARs-mediated synaptic transmission. Our study suggests that CRS dysregulates GABAergic neurotransmission via increasing the levels of caspase-1-mediated neuroinflammation in the hippocampus, ultimately leading to depression-like behaviors. This work illustrates that targeting caspase-1 may provide potential therapeutic benefits to stress-related GABAergic dysfunction in the pathogenesis of MDD.

An Improved Global and Local Fusion Path-Planning Algorithm for Mobile Robots

Path planning is a core technology for mobile robots. However, existing state-of-the-art methods suffer from issues such as excessive path redundancy, too many turning points, and poor environmental adaptability. To address these challenges, this paper proposes a novel global and local fusion path-planning algorithm. For global path planning, we reduce path redundancy and excessive turning points by designing a new heuristic function and constructing an improved path generation method. For local path planning, we propose an environment-aware dynamic parameter adjustment strategy, incorporating deviation and avoidance dynamic obstacle evaluation factors, thus addressing issues of local optima and timely avoidance of dynamic obstacles. Finally, we fuse those global and local path-planning improvements to form our fusion path-planning algorithm, which can enhance the robot’s adaptability to complex scenarios while reducing path redundancy and turning points. Simulation experiments demonstrate that the improved fusion path-planning algorithm not only effectively addresses existing issues but also operates with higher efficiency.

Room temperature valley polarization via spin selective charge transfer

The two degenerate valleys in transition metal dichalcogenides can be used to store and process information for quantum information science and technology. A major challenge is maintaining valley polarization at room temperature where phonon-induced intervalley scattering is prominent. Here we demonstrate room temperature valley polarization in heterostructures of monolayer MoS
2
and naphthylethylammine based one-dimensional chiral lead halide perovskite. By optically exciting the heterostructures with linearly polarized light close to resonance and measuring the helicity resolved photoluminescence, we obtain a degree of polarization of up to −7% and 8% in MoS
2
/right-handed (R-(+)-) and left-handed (S-(-)-) 1-(1-naphthyl)ethylammonium lead iodide perovskite, respectively. We attribute this to spin selective charge transfer from MoS
2
to the chiral perovskites, where the perovskites act as a spin filter due to their chiral nature. Our study provides a simple, yet robust route to obtain room temperature valley polarization, paving the way for practical valleytronics devices.
Valleytronics is a portmanteau of ‘valley’ and ‘electronics’ and refers to the use of the valley degree of freedom, present in some materials, for encoding and processing information. Here, Shrestha et al demonstrate a room temperature valley polarization in heterostructures composed of molybdenum disulfide and a chiral lead halide perovskite, an important step in development of valleytronics.

Gated subspace alignment with drift compensation for parameter-efficient Class-Incremental Learning

Class-Incremental Learning (CIL) aims to enable models to continuously learn new categories while preserving existing knowledge and avoiding catastrophic forgetting. Although parameter-expansion architectures can alleviate task interference to some extent, the representations of previously learned classes often drift or degrade as the feature subspaces continuously evolve and expand, resulting in decreased recognition performance for old classes. To address this issue, we propose an efficient CIL method—Dynamic Gated Adapter for Subspace Alignment (DGASA). Based on a frozen pre-trained backbone, DGASA introduces lightweight adapters with attention-based gating for each task to construct task-specific subspaces, while dynamically fusing cross-task information via attention mechanisms. In addition, DGASA learns a linear mapping between the old and new subspaces to achieve consistent alignment of old class prototypes in the current subspace without accessing past data. Extensive experiments demonstrate that DGASA significantly improves classification accuracy and resistance to forgetting on multiple benchmark datasets, offering strong generalization and computational efficiency.

Developing natural products as potential anti-biofilm agents

Biofilm is a natural form of bacterial growth ubiquitously in environmental niches. The biofilm formation results in increased resistance to negative environmental influences including resistance to antibiotics and antimicrobial agents. Quorum sensing (QS) is cell-to-cell communication mechanism, which plays an important role in biofilm development and balances the environment when the bacteria density becomes high. Due to the prominent points of biofilms implicated in infectious disease and the spread of multi-drug resistance, it is urgent to discover new antibacterial agents that can regulate biofilm formation and development. Accumulated evidences demonstrated that natural products from plants had antimicrobial and chemo-preventive properties in modulation of biofilm formation in the last two decades. This review will summarize recent studies on the discovery of natural anti-biofilm agents from plants with clear-cut mechanisms or identified molecular addresses, as well as some herbs with unknown mechanisms or unidentified bioactive ingredients. We also focus on the progression of techniques on the extraction and identification of natural anti-biofilm substances. Besides, anti-biofilm therapeutics undergoing clinical trials are discussed. These newly discovered natural anti-biofilm agents are promising candidates which could provide novel strategies for biofilm-associated infections.

The integration of multiple functionalities into a single, planar, ultra-compact metasurface has presented significant opportunities for enhancing capacity and performance within compact 5G/6G communication systems. Recent advances in multifunctional metasurfaces have unveiled comprehensive wavefront manipulations utilizing phase, polarization transmission/reflection, and coding apertures. Despite these developments, there remains a critical need for multifunctional metasurfaces with expanded channel capabilities, including multiple operational frequencies, minimal crosstalk, and high-efficiency computable array factors. This study introduces a multifunctional metasurface that integrates phase- and amplitude simultaneous coding meta-atoms at dual frequencies. By altering the polarization of electromagnetic (EM) waves, it is possible to reshape the wave-fronts of reflected waves at these frequencies. The coding metasurface proficiently manipulates both

linearly polarized waves through phase and amplitude coding at dual frequencies, thereby enabling distinct functionalities such as anomalous reflection, reflection imaging, and vortex wave beam generation. Both theoretical analysis and full-wave simulation confirm the anticipated functionalities of the designed devices, paving the way for advancements in integrated communication systems with diverse functionalities.

In order to promote the transformation and development of China's economy and cope with the realistic challenges posed by the global economic recession and slowing domestic economic growth, the Fourth Session of the 12th National People's Congress adopted a strategic resolution on developing the \"new economy\" and fostering new drivers of growth. Chengdu, capital of Sichuan province, is one of the cities in China which firstly begin to carry out the layout of the new economy development. While achieving results, it also faces many troubles due to its insufficient cognition of the connotation, definition, and path of the \"new economy.\" The paper is based on a theoretical study in combination with the new economic development of Chengdu's five new paths, to analyze the concept and connotation of the \"new economy.\" It also analyzes from the perspective of higher dimensional reconstruction and new strategic path of economic development, in order to add some insights to the theorectical study and to promote better strategic value of the new economy.

The environmental repercussions of extensive carbon dioxide (CO2) emissions on the environment are crucial for policymakers and scholars. The repercussions of and connection between economic growth (ECG), tourism (TOUR), and foreign direct investment (FDI) on CO2 emission mitigation have been measured and argued from empirical and theoretical perspectives by scholars. Notwithstanding, the extant body of knowledge has failed to incorporate and investigate the function of governance in decarbonizing tourism activities and FDI from CO2 emissions to attain a healthy and quality environment in Africa. Hence, this current research investigates governance’s role in the reduction processes of CO2 emissions grounded in environmental Kuznets curve (EKC) conceptual assumptions for panel data spanning 2000 through 2020 for 27 African countries. This research utilized the Westerlund panel cointegration approach for the investigation of the cointegration of the selected variables. This study applied the Driscoll–Kraay regression approach for the long-term estimation. In addition, the dynamic ordinary least squares (DOLS) and the pooled mean group (PMG) were used for robustness checks. The findings of this research indicated that the governance (GOV) indicators employed have a statistically significant effect on the CO2 emission reduction. Besides, this study found that the appreciation of the income of the nations gives credence to the formation of the EKC theory and contributes to the decline in CO2 emissions within the selected African nations. The findings revealed that tourism, FDI, ECG, and GOV are positive and significant factors leading to increased CO2 emissions in Africa. Furthermore, the results showed that effective governance and control of FDI inflows and tourism activities can support decarbonization. These findings suggest the merits of governance in ensuring effective decarbonization policies of the environment, and policy suggestions are accordingly put forward.

Tumor Infiltrating Lymphocyte (TIL) Therapy for Solid Tumor Treatment: Progressions and Challenges

Over the past decade, immunotherapy, especially cell-based immunotherapy, has provided new strategies for cancer therapy. Recent clinical studies demonstrated that adopting cell transfer of tumor-infiltrating lymphocytes (TILs) for advanced solid tumors showed good efficacy. TIL therapy is a type of cell-based immunotherapy using the patient’s own immune cells from the microenvironment of the solid tumor to kill tumor cells. In this review, we provide a comprehensive summary of the current strategies and challenges in TIL isolation and generation. Moreover, the current clinical experience of TIL therapy is summarized and discussed, with an emphasis on lymphodepletion regimen, the use of interleukin-2, and related toxicity. Furthermore, we highlight the clinical trials where TIL therapy is used independently and in combination with other types of therapy for solid cancers. Finally, the limitations, future potential, and directions of TIL therapy for solid tumor treatment are also discussed.
