Flame retardants (FRs) are used in a variety of common items from furniture to carpet to electronics to reduce flammability and combustion, but the potential toxicity of these compounds is raising health concerns globally. Organophosphate FRs (OPFRs) are becoming more prevalent as older, brominated FRs are phased out, but the toxicity of these compounds, and the FR mixtures that contain them, is poorly understood. Work in a variety of in vitro model systems has suggested that FRs may induce metabolic reprogramming such that bone density is compromised at the expense of increasing adiposity. To address this hypothesis, the present studies maternally exposed Wistar rat dams orally across gestation and lactation to 1000 µg daily of the FR mixture Firemaster 550 (FM 550) which contains a mixture of brominated FRs and OPFRs. At six months of age, the offspring of both sexes were examined for evidence of compromised bone composition. Bone density, composition, and marrow were all significantly affected, but only in males. The fact that the phenotype was observed months after exposure suggests that FM 550 altered some fundamental aspect of mesenchymal stem cell reprogramming. The severity of the phenotype and the human-relevance of the dose employed, affirm this is an adverse outcome meriting further exploration.

Alzheimer’s disease (AD) is a stealthy and progressive neurological disorder that is a leading cause of dementia in the global elderly population, imposing a significant burden on both the elderly and society. Currently, the condition is treated with medications that alleviate symptoms. Nonetheless, these drugs may not consistently produce the desired results and can cause serious side effects. Hence, there is a vigorous pursuit of alternative options to enhance the quality of life for patients. Ginkgo biloba (GB), an herb with historical use in traditional medicine, contains bioactive compounds such as terpenoids (Ginkgolides A, B, and C), polyphenols, organic acids, and flavonoids (quercetin, kaempferol, and isorhamnetin). These compounds are associated with anti-inflammatory, antioxidant, and neuroprotective properties, making them valuable for cognitive health. A systematic search across three databases using specific keywords—GB in AD and dementia—yielded 1702 documents, leading to the selection of 15 clinical trials for synthesis. In eleven studies, GB extract/EGb 761® was shown to improve cognitive function, neuropsychiatric symptoms, and functional abilities in both dementia types. In four studies, however, there were no significant differences between the GB-treated and placebo groups. Significant improvements were observed in scores obtained from the Mini-Mental State Examination (MMSE), Short Cognitive Performance Test (SKT), and Neuropsychiatric Inventory (NPI). While the majority of synthesized clinical trials show that Ginkgo biloba has promising potential for the treatment of these conditions, more research is needed to determine optimal dosages, effective delivery methods, and appropriate pharmaceutical formulations. Furthermore, a thorough assessment of adverse effects, exploration of long-term use implications, and investigation into potential drug interactions are critical aspects that must be carefully evaluated in future studies.

A phase I dose-escalation trial of 2-deoxy-d-glucose alone or combined with docetaxel in patients with advanced solid tumors

Purpose
This phase I trial was initiated to evaluate the safety, pharmacokinetics (PK) and maximum tolerated dose (MTD) of the glycolytic inhibitor, 2-deoxy-
d
-glucose (2DG) in combination with docetaxel, in patients with advanced solid tumors.
Methods
A modified accelerated titration design was used. 2DG was administered orally once daily for 7 days every other week starting at a dose of 2 mg/kg and docetaxel was administered intravenously at 30 mg/m
2
for 3 of every 4 weeks beginning on day 1 of week 2. Following the completion of dose escalation, cohorts of patients were then treated with 2DG for 21 days or every day of each 4-week cycle for up to 12 cycles.
Results
Thirty-four patients were enrolled: 21 on every other week, 6 on a 21 of 28-day cycle and 7 on the continuous 2DG dosing schedule. There were no dose-limiting toxicities which met the MTD criteria. The most common adverse events were fatigue, sweating, dizziness and nausea mimicking the hypoglycemic symptoms expected from 2DG administration. Therefore, 63 mg/kg was selected as the clinically tolerable dose. The most significant adverse effects noted at 63–88 mg/kg doses were reversible hyperglycemia (100 %), gastrointestinal bleeding (6 %) and reversible grade 3 QTc prolongation (22 %). Eleven patients (32 %) had stable disease, 1 patient (3 %) partial response and 22 patients (66 %) progressive disease as their best response. There was no PK interaction between 2DG and docetaxel.
Conclusion
The recommended dose of 2DG in combination with weekly docetaxel is 63 mg/kg/day with tolerable adverse effects.

The treatment of rheumatoid arthritis remains a challenge as available therapies still entail the risk of deleterious off-target effects. The present study describes hyaluronic acid-conjugated pH-sensitive liposomes as an effective drug delivery-targeting strategy to synovial cells.
Therapeutic, cytotoxic and targeting potential of developed liposomes were studied
using macrophages and fibroblasts cell lines.
Results suggest an enhanced cellular uptake of conjugated liposomes, mainly mediated by caveolae- and clathrin-dependent endocytosis.
release studies demonstrated that prednisolone was preferentially released under acidic conditions mimicking intracellular endosomal compartments. Overall, results revealed that conjugated pH-sensitive liposomes are a promising nanoapproach for the targeted delivery of prednisolone within inflamed synovial cells for rheumatoid arthritis treatment.

Fungal endophytes from cactus Cereus jamacaru in Brazilian tropical dry forest: a first study

Endophytic fungi live within the healthy tissues of plants and can promote host species tolerance to different environmental stresses. However, most studies have been of plants in humid environments, and there are few reports of the benefits of such associations for plants of extreme environments. Our aims were to identify endophytic fungi using morphological taxonomy, to explore richness and estimate species frequency in a cactus, C. jamacaru, in Brazilian tropical dry forest (Caatinga). We thus identified 59 taxa, corresponding to 69.7 % of the total number of isolates; the other 30.3 % were sterile mycelia. Cladosporium cladosporioides and Fusarium oxysporum were the species most commonly isolated, followed by Acremonium implicatum, Aureobasidium pullulans, Trichoderma viride, Chrysonilia sitophila, and Aspergillus flavus. Forty-seven species are recorded for the first time as endophytic fungi of cacti, and 18 others as endophytes for Brazil; this suggests that C. jamacaru harbors a highly diverse fungal community as measured by a diversity index. However, species accumulation curves suggest that our study still underestimates endophyte diversity because it does not provide an exhaustive sample. To our knowledge, this is the first report of endophytic fungi from C. jamacaru in tropical dry forests.

Vermelha Lagoon is a hypersaline shallow transitional ecosystem in the state of Rio de Janeiro (Brazil). This lagoon is located in the protected area of Massambaba, between the cities of Araruama and Saquarema (Brazil), and displays two quite uncommon particularities: it exhibits carbonate sedimentation and displays the development of Holocene stromatolites. Due to both particularities, the salt industry and property speculation have been, increasingly, generating anthropic pressures on this ecosystem. This study aims to apply a multiproxy approach to evaluate the trophic state of Vermelha Lagoon based on physicochemical parameters and geochemical data for the quantification and qualification of organic matter (OM), namely total organic carbon (TOC), total sulfur (TS), total phosphorus (TP) and biopolymeric carbon (BPC), including carbohydrates (CHO), lipids (LIP) and proteins (PTN). The CHO/TOC ratio values suggest that OM supplied to the sediment is of autochthonous origin and results, essentially, from microbial activity. The cluster analyses allowed the identification of four regions in Vermelha Lagoon. The Region I included stations located in shallow areas of the eastern sector of Vermelha lagoon affected by the impact of the artificial channel of connection with Araruama Lagoon. The Region II, under the influence of salt pans, is characterized by the highest values of BPC, namely CHO promoted by microbiological activity. The Region III include stations spread through the lagoon with high values of dissolved oxygen and lower values of TP. Stromatolites and microbial mattes growth was observed in some stations of this sector. Region IV, where the highest values of TOC and TS were found, represents depocenters of organic matter, located in general in depressed areas. Results of this work evidences that the Vermelha Lagoon is an eutrophic but alkaline and well oxygenated environment (at both water column and surface sediment) where the autotrophic activity is greater than heterotrophic one. These particular conditions make this a special and rare ecosystem.

Surface and Structural Modifications Induced by Dielectric Barrier Discharge Cold Plasma and Pulsed Light Radiation in Bean Starch Films: Implications for Bread Shelf-Life Extension

In this study, bean starch films were developed and treated with dielectric barrier discharge (DBD) cold plasma (5 min (DBD5), 10 min (DBD10), and 15 min (DBD15)) and pulsed light (PL) radiation (4 J cm−2 (PL4), 8 J cm−2 (PL8), and 12 J cm−2 (PL12)), and the effects of these treatments on the physical, barrier, mechanical, morphological, and structural properties were evaluated, as well as the practical application of the films in bread storage for 7 days. Both treatments significantly modified the film properties (p < 0.05). Film thickness decreased from 95 µm (control) to 87 µm (PL12), while solubility was reduced from 39.40% (control) to 25.32% (PL12), indicating improved water resistance. Reductions in water vapor permeability (WVP) were also observed, with a more pronounced effect for PL12 (approximately 55% reduction compared to the control). The contact angle increased from 58.30° (control) to 67.76° (PL12), indicating a moderate increase in surface hydrophobicity. The DBD cold plasma treatment increased tensile strength (up to 16.05 MPa in DBD15) and reduced elongation (44.72%), whereas PL, especially at PL8, increased flexibility (60.36%). Morphological analyses indicated increased surface roughness for DBD-treated films, while structural analyses suggested subtle changes in molecular organization rather than the formation of well-defined crystalline domains. During bread storage, the treated films, particularly PL12, were significantly more effective than the control in delaying bread staling (final firmness of 6.67 N vs. 11.82 N), reducing mass loss (5.66% vs. 12.66%), and maintaining higher water activity, thereby better preserving product quality. Overall, both treatments showed potential for tailoring film properties: DBD was more effective in enhancing mechanical strength, while PL promoted improvements in barrier properties and practical performance. Therefore, physical treatments, particularly PL, represent promising strategies to overcome intrinsic limitations of starch-based films and to develop packaging materials with potential applications in bakery product preservation.

Hyperkalaemia management and related costs in chronic kidney disease patients with comorbidities in Spain

Background
Hyperkalaemia (HK) is a common electrolyte disorder in patients with chronic kidney disease (CKD) and/or treated with renin–angiotensin–aldosterone system inhibitors (RAASis). The aim of this study is to determine the severity, current management and cost of chronic HK.
Methods
We performed a retrospective cohort study of patients with chronic HK and CKD, heart failure or diabetes mellitus between 2011 and 2018. The study follow-up was 36 months.
Results
A total of 1499 patients with chronic HK were analysed: 66.2% presented with mild HK, 23.4% with moderate HK and 10.4% with severe HK. The severity was associated with CKD stage. Most patients (70.4%) were on RAASi therapies, which were frequently discontinued (discontinuation rate was 39.8, 49.8 and 51.8% in mild, moderate and severe HK, respectively). This RAASi discontinuation was similar with or without resin prescription. Overall, ion-exchange resins were prescribed to 42.5% of patients with HK and prescriptions were related to the severity of HK, being 90% for severe HK. Adherence to resin treatment was very low (36.8% in the first year and 17.5% in the third year) and potassium remained elevated in most patients with severe HK. The annual healthcare cost per patient with HK was €5929, reaching €12 705 in severe HK. Costs related to HK represent 31.9% of the annual cost per HK patient and 58.8% of the specialized care cost.
Conclusions
HK was usually managed by RAASi discontinuation and ion-exchange resin treatment. Most patients with HK were non-adherent to resins and those with severe HK remained with high potassium levels, despite bearing elevated healthcare expenditures.

Emerging Roles of Dipeptidyl Peptidase-4 Inhibitors in Delaying the Progression of Type 1 Diabetes Mellitus

Type 1 diabetes mellitus (T1DM) results from the immune cell-mediated destruction of functional pancreatic β-cells. In the presymptomatic period, T1DM is characterized by the presence of two or more autoantibodies against the islet cells in patients without glycemic decompensation. Therapeutic strategies that can modify the autoimmune process could slow the progression of T1DM. Dipeptidyl peptidase-4 (DPP-4) or CD26, a multifunctional serine protease with a dual function (regulatory protease and binding protein), can modulate inflammation and immune cell-mediated β-cell destruction. CD26 is involved in T-cell co-stimulation, migration, memory development, thymic maturation, and emigration patterns. DPP-4 degrades the peptide hormones GLP-1 and GIP. In addition to regulating glucose metabolism, DPP-4 exerts anti-apoptotic, regenerative, and proliferative effects to promote β-cell mass expansion. GLP-1 receptor signaling may regulate murine lymphocyte proliferation and maintenance of peripheral regulatory T-cells. In patients with T1DM, the serum DPP-4 activity is upregulated. Several studies have suggested that the upregulated DPP-4 activity is correlated with T1DM pathophysiology. DPP-4, which is preferentially expressed on the Th1 surface, can promote the polarization of Th1 immunity, a prerequisite for T1DM development. CD26 inhibition can suppress T-cell proliferation and Th1 cytokine production and stimulate tumor growth factor beta-1 (TGF-β1) secretion, which plays an important role in the regulation of autoimmunity in T1DM. Studies on humans or animal models of T1DM have suggested that DPP-4 inhibitors can improve β-cell function and attenuate autoimmunity in addition to decreasing insulin dependence. This review summarizes the emerging roles of DPP-4 inhibitors in potentially delaying the progression of T1DM.

Climate Change Impact on the Distribution of Forest Species in the Brazilian Amazon

Studies using ecological niche models highlight the vulnerability of forest species to climate change. This work aimed to analyze the distribution of timber species Aspidosperma desmanthum, Cariniana micranta, Clarisia racemosa, Couratari oblongifolia, and Vouchysia guianensis, which are targets of deforestation, to predict the impacts of climate change and identify areas for their conservation in the Amazon. For this purpose, 37 environmental variables were used, including climatic and edaphic factors. The models were fitted using five algorithms, and their performance was evaluated by the metrics Area Under the Curve (AUC), True Skill Statistic, and Sorensen Index. The deforestation analysis was conducted using data accumulated over a period of 14 years. The study indicated that under the most pessimistic predictions, considering continued high emissions of greenhouse gases (GHGs) from the use of fossil fuels, SSP5–8.5, potential habitat loss for the studied species was more significant. Analyses of the species show that the Western Amazon has a greater climatic suitability area for the conservation of its genetic resources. Further study of the accumulated deforestation over 14 years showed a reduction in area for all species. Therefore, in situ conservation policies and deforestation reduction are recommended for the perpetuation of the analyzed forest species.
