The Mechanism of Ferroptosis and Applications in Tumor Treatment
Iron-dependent ferroptosis is a new form of cell death in recent years, which is driven by lipid peroxidation. The lethal lipid accumulation caused by glutathione depletion or inactivation of glutathione peroxidase 4 (GPX4) is characteristic of the ferroptosis process. In recent years, with the in-depth study of ferroptosis, various types of diseases have been reported to be related to ferroptosis. In other words, ferroptosis, which has attracted widespread attention in the fields of biochemistry, oncology, and especially materials science, can undoubtedly provide a new way for patients. This review introduces the relevant mechanisms of ferroptosis, the relationship between ferroptosis and various cancers, as well as the application of ferroptosis in tumor treatment. We also sorted out the genes and drugs that regulate ferroptosis. Moreover, small molecule compound-induced ferroptosis has a strong inhibitory effect on tumor growth in a drug-resistant environment, which can enhance the sensitivity of chemotherapeutic drugs, suggesting that ferroptosis is very important in the treatment of tumor drug resistance, but the details are still unclear. How to use ferroptosis to fight cancer, and how to prevent drug-resistant tumor cells have become the focus and direction of research. At the end of the article, some existing problems related to ferroptosis are summarized for future research.
A hybrid algorithm for clinical decision support in precision medicine based on machine learning
Purpose The objective of the manuscript is to propose a hybrid algorithm combining the improved BM25 algorithm, k-means clustering, and BioBert model to better determine biomedical articles utilizing the PubMed database so, the number of retrieved biomedical articles whose content contains much similar information regarding a query of a specific disease could grow larger. Design/methodology/approach In the paper, a two-stage information retrieval method is proposed to conduct an improved Text-Rank algorithm. The first stage consists of employing the improved BM25 algorithm to assign scores to biomedical articles in the database and identify the 1000 publications with the highest scores. The second stage is composed of employing a method called a cluster-based abstract extraction to reduce the number of article abstracts to match the input constraints of the BioBert model, and then the BioBert-based document similarity matching method is utilized to obtain the most similar search outcomes between the document and the retrieved morphemes. To realize reproducibility, the written code is made available on https://github.com/zzc1991/TREC_Precision_Medicine_Track . Findings The experimental study is conducted based on the data sets of TREC2017 and TREC2018 to train the proposed model and the data of TREC2019 is used as a validation set confirming the effectiveness and practicability of the proposed algorithm that would be implemented for clinical decision support in precision medicine with a generalizability feature. Originality/value This research integrates multiple machine learning and text processing methods to devise a hybrid method applicable to domains of specific medical literature retrieval. The proposed algorithm provides a 3% increase of P@10 than that of the state-of-the-art algorithm in TREC 2019.
Cultural Routes as Cultural Tourism Products for Heritage Conservation and Regional Development: A Systematic Review
Cultural routes are a composite set of heritage sites that refer to historical routes of human communication. As key products of cultural tourism, they provide visitors with rich cultural experiences across regions. We systematically review reports and studies related to the tourism development of 38 cultural route cases worldwide, with a special focus on their distribution, typology, planning patterns, and tools for cultural tourism. We summarized eight tools and found some differences in how often these eight tools are used by the different types of routes and different planning patterns for route tourism. This study also developed an evaluation system based on the conservation principles of cultural routes to determine how different tourism tools affect the conservation and development of historical regions. Although tourism decision-makers have made numerous efforts to protect and develop cultural routes, there are still many problems and challenges in the process of tourism development along cultural routes. We conclude the paper by making recommendations for decision-makers and researchers concerning future route tourism planning and study.
Study on the Wear Mechanism of a Diamond AFM Tip During Scribing of a Single-Crystal Silicon
To elucidate the wear mechanisms of diamond AFM tips during nanoscale scribing of single-crystal silicon, this study combines controlled experiments with atomistic molecular dynamics (MD) simulations. Scribing tests were conducted under systematically varied bias current, scribing speed, and scribing distance. Tip morphology evolution was quantitatively characterized. Concurrently, a three-dimensional MD model reproduced probe–silicon interactions to analyze bond breaking, atomic detachment, and structural transformation at the atomic scale. The results show that increasing current, speed, and distance significantly accelerate tip blunting. Simulations reveal a progressive transition in deformation behavior from elastic response to atomic attrition, plastic damage, brittle cracking, and catastrophic fracture as indentation depth increases, and cluster analysis establishes a quantitative correlation between process parameters and wear severity. This integrated experimental simulation framework provides mechanistic insight into diamond tip degradation and offers quantitative guidance for improving probe durability and process reliability in AFM-based nanofabrication.
Disheveled3 enhanced EMT and cancer stem-like cells properties via Wnt/β-catenin/c-Myc/SOX2 pathway in colorectal cancer
Background Epithelial-to-mesenchymal transition (EMT) and cancer stem-like cells (CSLCs) play crucial role in tumor metastasis and drug-resistance. Disheveled3 (DVL3) is involved in malignant behaviors of cancer. However, the role and potential mechanism of DVL3 remain elusive in EMT and CSLCs of colorectal cancer (CRC). Methods UALCAN and PrognoScan databases were employed to evaluate DVL3 expression in CRC tissues and its correlation with CRC prognosis, respectively. Transwell, sphere formation and CCK8 assay were used to assess metastasis, stemness and drug sensitivity of CRC cells, respectively. Western blotting and dual luciferase assay were performed to analyze the protein expression and Wnt/β-catenin activation, respectively. Lentiviral transfection was used to construct the stable cell lines. Animal studies were performed to analyze the effect of silencing DVL3 on tumorigenicity and metastasis of CRC cells in vivo. Results DVL3 was overexpressed in CRC tissues and several CRC cell lines. DVL3 expression was also higher in CRC tissues with lymph node metastasis than tumor tissues without metastasis, and correlated with poor prognosis of CRC patients. DVL3 positively regulated the abilities of migration, invasion and EMT-like molecular changes in CRC cells. Moreover, DVL3 promoted CSLCs properties and multidrug resistance. We further identified that Wnt/β-catenin was crucial for DVL3-mediated EMT, stemness and SOX2 expression, while silencing SOX2 inhibited DVL3-mediated EMT and stemness. Furthermore, c-Myc, a direct target gene of Wnt/β-catenin, was required for SOX2 expression and strengthened EMT and stemness via SOX2 in CRC cells. Finally, knockdown of DVL3 suppressed tumorigenicity and lung metastasis of CRC cells in nude mice. Conclusion DVL3 promoted EMT and CSLCs properties of CRC via Wnt/β-catenin/c-Myc/SOX2 axis, providing a new strategy for successful CRC treatment.
Does cultural intelligence matter within cross-cultural teams in hospitality industry? Understanding the role of team dissimilarity climate
Cross-cultural teams have become integral units of international hospitality enterprises given the rapid development of globalization. How to effectively stimulate team members' performance in a cross-cultural context has thus become a growing concern. This paper aims to explore whether, how, and when cultural intelligence influences team members’ performance in cross-cultural teams. Data analysis from 440 employees nested in 100 cross-cultural teams of an international hotel chain revealed that cultural intelligence is positively associated with individual role performance via cross-cultural adjustment. In addition, the above indirect relationship was found to be stronger when a higher level of team dissimilarity climate was present. Our findings shed light on how situational factors and personal traits interact in a diverse range of work environments. The practical implications for international organizations of fully utilizing the benefits of cultural diversity in the workplace are also discussed.
YOLO-LDFI: A Lightweight Deformable Feature-Integrated Detector for SAR Ship Detection
A lightweight enhanced detection model named YOLO-LDFI is proposed in this study for ship target detection in SAR images, aiming to improve detection accuracy and deployment efficiency under complex maritime environments. Based on YOLOv11n, the model incorporates four architectural improvements in a progressive manner: linear deformable convolution (LDConv), deformable context-aware attention mechanism (DCAM), frequency-adaptive dilated convolution detection head (FAHead), and Inner-EIoU. Experiments conducted on the public SAR ship detection dataset HRSID demonstrate that the proposed model achieves an AP50 of 90.7% and an F1 score of 87.0%, with only 2.63 M parameters and a computational complexity of 6.7 GFLOPs. Ablation experiments validate the contribution of each component to improved feature alignment, reduced background interference, and more accurate target localization. Overall, the results indicate that the proposed model offers a reasonable trade-off between detection performance and computational efficiency in SAR ship detection tasks.
Effects of Compound Elicitors on the Biosynthesis of Triterpenoids and Activity of Defense Enzymes from Inonotus hispidus (Basidiomycetes)
Inonotus hispidus has various health-promoting activities, such as anticancer effects and immune-stimulating activity. The commercialization of valuable plant triterpenoids faces major challenges, including low abundance in natural hosts and costly downstream purification procedures. In this work, orthogonal design was used to compound methyl jasmonate (MeJA), salicylic acid (SA), oleic acid, and Cu2+, and the effects of combinations on the total triterpenes biosynthesized were studied. The optimal combination was screened out and its effect on the activity of PAL, CAT, and SOD was studied. The optimal concentration of oleic acid was 2% when MeJA was 100 mol/L, and the total triterpenoid content and mycelia production were 3.918 g and 85.17 mg/g, respectively. MeJA treatment induced oxidative stress, and at the same time increased the activity of related defense enzymes. Oleic acid is thought to regulate cell permeability by recombining cell membranes. It promotes the material exchange process between cells and the environment without affecting cell growth. When oleic acid was used in combination with MeJA, a synergistic effect on triterpene production was observed. In conclusion, our findings provide a strategy for triterpenoid enrichment of I. hispidus.
Integrated metabolic profiling and transcriptome analysis of pigment accumulation in diverse petal tissues in the lily cultivar ‘Vivian’
Background Petals are the colorful region of many ornamental plants. Quality traits of petal color directly affect the value of ornamental plants. Although the regulatory mechanism of flower color has been widely studied in many plants, that of lily flower color is still worth further exploration. Results In this study, the pigmentation regulatory network in different regions of the petal of lily cultivar ‘Vivian’ was analyzed through tissue structure, metabolites biosynthesis, and gene expression. We found that cell morphology of the petal in un-pigmented region differed from that in pigmented region. The cell morphology tends to flatten in un-pigmented region where the color is lighter. Moreover, high level anthocyanin was found in the pigmented regions by metabonomic analysis, especially cyanidin derivatives. However, flavanones were accumulated, contrast with anthocyanin in the un-pigmented regions of lily petal. To understand the relationship of these different metabolites and lily flower color, RNA-Seq was used to analyze the differentially expressed genes-related metabolite biosynthesis. Among these genes, the expression levels of several genes-related cyanidin derivatives biosynthesis were significantly different between the pigmented and un-pigmented regions, such as LvMYB5, LvMYB7, LvF3’H, LvDFR, LvANS and Lv3GT . Conclusions This data will help us to further understand the regulation network of lily petal pigmentation and create different unique color species.
Cancer therapy-related cardiovascular aging: mechanisms, monitoring, and intervention strategies
Significant improvements in cancer survival rates have been achieved through advancements in treatment and early diagnosis. However, non-cancer-related mortality among cancer survivors continues to rise each year. Cardiovascular diseases (CVDs) related to cancer therapy now rank as the second leading cause of death in survivors, sometimes surpassing the cancer itself. Among these, cardiovascular aging represents one of the most severe side effects, often leading to detrimental structural changes such as cardiac atrophy or fibrosis, which ultimately impair cardiac function and reduce survival. Preventing or treating cardiovascular aging has emerged as a promising strategy to mitigate Cancer Therapy-Related Cardiovascular Toxicity (CTR-CVT). This review offers a comprehensive analysis of the characteristics and mechanisms underlying cancer therapy-induced accelerated cardiovascular cellular senescence, outlines current monitoring and intervention strategies, and explores future research opportunities and challenges. Enhancing the understanding of Cancer Therapy-Related Cardiovascular Cellular Senescence and Cancer Therapy-Related Cardiovascular Aging (CTR-CVA) is crucial for optimizing cancer treatment, advancing medical research, and improving clinical practice, all of which are vital for preserving cardiac health and improving the quality of life of patients with cancers.
