Transcriptomic and metabolomic profiling of drought-tolerant and susceptible sesame genotypes in response to drought stress

Background
Sesame is an important oil crop due to its high oil, antioxidant, and protein content. Drought stress is a major abiotic stress that affects sesame production as well as the quality of sesame seed. To reveal the adaptive mechanism of sesame in response to water deficient conditions, transcriptomic and metabolomics were applied in drought-tolerant (DT) and drought-susceptible (DS) sesame genotypes.
Results
Transcriptomic analysis reveals a set of core drought-responsive genes (684 up-regulated and 1346 down-regulated) in sesame that was robustly differently expressed in both genotypes. Most enriched drought-responsive genes are mainly involved in protein processing in endoplasmic reticulum, plant hormone signal transduction photosynthesis, lipid metabolism, and amino acid metabolism. Drought-susceptible genotype was more disturbed by drought stress at both transcriptional and metabolic levels, since more drought-responsive genes/metabolites were identified in DS. Drought-responsive genes associated with stress response, amino acid metabolism, and reactive oxygen species scavenging were more enriched or activated in DT. According to the partial least-squares discriminate analysis, the most important metabolites which were accumulated under drought stress in both genotypes includes ABA, amino acids, and organic acids. Especially, higher levels of ABA, proline, arginine, lysine, aromatic and branched chain amino acids, GABA, saccharopine, 2-aminoadipate, and allantoin were found in DT under stress condition. Combination of transcriptomic and metabolomic analysis highlights the important role of amino acid metabolism (especially saccharopine pathway) and ABA metabolism and signaling pathway for drought tolerance in sesame.
Conclusion
The results of the present study provide valuable information for better understanding the molecular mechanism underlying drought tolerance of sesame, and also provide useful clues for the genetic improvement of drought tolerance in sesame.

We examined whether participative leadership behavior is associated with improved work performance through a motivational process or an exchange-based process. Based on data collected from 527 employees from a Fortune 500 company, we found that the link between superiors' participative leadership behaviors and subordinates' task performance and organizational citizenship behavior toward organizations (OCBO) was mediated by psychological empowerment (motivational mediator) for managerial subordinates. Yet, for non-managerial subordinates such as supporting and front-line employees, the impact of participative leadership on task performance and OCBO was mediated by trust-in-supervisor (exchange-based mediator). Implications for theories and practices are discussed.

Genome-wide characterization and expression analysis of the HD-Zip gene family in response to drought and salinity stresses in sesame

Background
The homeodomain-leucine zipper (HD-Zip) gene family is one of the plant-specific transcription factor families, involved in plant development, growth, and in the response to diverse stresses. However, comprehensive analysis of the HD-Zip genes, especially those involved in response to drought and salinity stresses is lacking in sesame (
Sesamum indicum
L.), an important oil crop in tropical and subtropical areas.
Results
In this study, 45 HD-Zip genes were identified in sesame, and denominated as SiHDZ01-SiHDZ45. Members of SiHDZ family were classified into four groups (HD-Zip I-IV) based on the phylogenetic relationship of
Arabidopsis
HD-Zip proteins, which was further supported by the analysis of their conserved motifs and gene structures. Expression analyses of
SiHDZ
genes based on transcriptome data showed that the expression patterns of these genes were varied in different tissues. Additionally, we showed that at least 75% of the
SiHDZ
genes were differentially expressed in responses to drought and salinity treatments, and highlighted the important role of HD-Zip I and II genes in stress responses in sesame.
Conclusions
This study provides important information for functional characterization of stress-responsive HD-Zip genes and may contribute to the better understanding of the molecular basis of stress tolerance in sesame.

Identification and characterization of the bZIP transcription factor family and its expression in response to abiotic stresses in sesame

Basic leucine zipper (bZIP) gene family is one of the largest transcription factor families in plants, and members of this family play important roles in multiple biological processes such as light signaling, seed maturation, flower development as well as abiotic and biotic stress responses. Nonetheless, genome-wide comprehensive analysis of the bZIP family is lacking in the important oil crop sesame. In the present study, 63 bZIP genes distributed on 14 linkage groups were identified in sesame, and denominated as SibZIP01-SibZIP63. Besides, all members of SibZIP family were divided into nine groups based on the phylogenetic relationship of Arabidopsis bZIPs, which was further supported by the analysis of their conserved motifs and gene structures. Promoter analysis showed that all SibZIP genes harbor cis-elements related to stress responsiveness in their promoter regions. Expression analyses of SibZIP genes based on transcriptome data showed that these genes have different expression patterns in different tissues. Additionally, we showed that a majority of SibZIPs (85.71%) exhibited significant transcriptional changes in responses to abiotic stresses, including drought, waterlogging, osmotic, salt, and cold, suggesting that SibZIPs may play a cardinal role in the regulation of stress responses in sesame. Together, these results provide new insights into stress-responsive SibZIP genes and pave the way for future studies of SibZIPs-mediated abiotic stress response in sesame.

Evaluation of Multi-Source Precipitation Products in the Hinterland of the Tibetan Plateau

High-resolution precipitation products have been crucial for hydrology, meteorology, and environmental ecosystems over the Tibetan Plateau (TP). However, these products are usually subject to systematic errors, which may vary with time and topographic conditions. The study evaluated the suitability of four satellite-derived products (GPM IMERG, GSMaP, CMORPH, and PERSIANN-CDR) and four fusion precipitation products (ERA5-Land, CHIRPS, CMFD, and TPHiPr) by comparing with 22 rain gauges at a daily scale from 1 January 2014 to 31 December 2018 over the hinterland of the TP. The main findings are as follows: (1) TPHiPr and CMFD are better than the satellite-derived products, while the performance of CHIRPS is worse; (2) among the satellite-derived products, the quality of GPM IMERG is the highest on different time scales, and PERSIANN-CDR is better in the months of June to October, while GSMaP and CMORPH have poor performance; (3) the eight precipitation products have weaker detection capability for heavy precipitation events, and the quality of each product decreases with the increase in the precipitation threshold, while the rate of descent of fusion precipitation products is slower than that of satellite-derived products. This study demonstrates the performance of eight precipitation products over the hinterland of the TP, which is expected to provide valuable information for hydrometeorology applications.

Transcriptomic, biochemical and physio-anatomical investigations shed more light on responses to drought stress in two contrasting sesame genotypes

Sesame is an important oilseed crop with a high oil quality. It is prone to drought stress in the arid and semi-arid areas where it is widely grown. This study aims to decipher the response of tolerant (DT) and sensitive (DS) genotypes to progressive drought based on transcriptome, biochemical and physio-anatomical characterizations. Results indicated that under severe stress, DT relied on a well-functioning taproot while DS displayed a disintegrated root due to collapsed cortical cells. This was attributed to a higher accumulation of osmoprotectants and strong activity of antioxidant enzymes especially peroxidases in DT. From roots, DT could supply water to the aboveground tissues to ensure photosynthetic activities and improve endurance under stress. Temporal transcriptome sequencing under drought further confirmed that DT strongly activated genes related to antioxidant activity, osmoprotection and hormonal signaling pathways including abscisic acid and Ethylene. Furthermore, DT displayed unique differentially expressed genes in root functioning as peroxidases, interleukin receptor-associated kinase, heat shock proteins, APETALA2/ethylene-responsive element-binding protein and mitogen activated protein kinase, to effectively scavenge reactive oxygen species and preserve root cell integrity. Finally, 61 candidate genes conferring higher drought tolerance in DT were discovered and may constitute useful resources for drought tolerance improvement in sesame.

Altered theta oscillations in basolateral amygdala and ventral hippocampus related to social defeat

Background
Depression is a prevalent mental disorder, and prolonged exposure to social defeat is a major contributing factor in the onset of depression. Repeated social defeat stress (RSDS) is a commonly used animal model for depression, significantly impacting on the pathogenesis of depression-related to social disorders. The basolateral amygdala (BLA) and the ventral hippocampus (vHPC) are critical brain regions involved in RSDS-induced social behavioral disorders, but the specific neural oscillations occurring in these regions following social defeat remain unclear.
Methods
Using simultaneous multi-electrode recordings, we captured local field potentials (LFPs) from BLA and vHPC while the stressed mice underwent a social interaction test. Power spectral analysis and Amplitude transform entropy were respectively applied to assess social defeat–induced alterations in neural oscillatory activity and directional inter-regional communication.
Results
Our study demonstrated that repeated social defeat induces social avoidance and depression-like behaviors. Notably, the power spectral analysis within the BLA and vHPC revealed statistically differences in the theta band (4–12 Hz) between control and RSDS groups, particularly during the With CD1 phase in the 0–3 s stage, when mice entered the social interaction zone, compared to the − 3 –0 s stage prior to enter the zone. Moreover, machine learning analysis successfully classified control and RSDS groups based on neural oscillatory activity in the BLA and vHPC. Finally, ketamine treatment was found to reduce social avoidance and depressive-like behaviors, as well as enhance theta oscillation in the BLA and vHPC.
Conclusion
These results suggest that social defeat alters theta oscillations in the BLA and vHPC, highlighting potential therapeutic avenues for addressing depression-related social dysfunction.

Unlike most of the important food crops, sesame can survive drought but severe and repeated drought episodes, especially occurring during the reproductive stage, significantly curtail the productivity of this high oil crop. Genome‐wide association study was conducted for traits related to drought tolerance using 400 diverse sesame accessions, including landraces and modern cultivars. Ten stable QTLs explaining more than 40% of the phenotypic variation and located on four linkage groups were significantly associated with drought tolerance related traits. Accessions from the tropical area harboured higher numbers of drought tolerance alleles at the peak loci and were found to be more tolerant than those from the northern area, indicating a long‐term genetic adaptation to drought‐prone environments. We found that sesame has already fixed important alleles conferring survival to drought which may explain its relative high drought tolerance. However, most of the alleles crucial for productivity and yield maintenance under drought conditions are far from been fixed. This study also revealed that pyramiding the favourable alleles observed at the peak loci is of high potential for enhancing drought tolerance in sesame. In addition, our results highlighted two important pleiotropic QTLs harbouring known and unreported drought tolerance genes such as SiABI4, SiTTM3, SiGOLS1, SiNIMIN1 and SiSAM. By integrating candidate gene association study, gene expression and transgenic experiments, we demonstrated that SiSAM confers drought tolerance by modulating polyamine levels and ROS homeostasis, and a missense mutation in the coding region partly contributes to the natural variation of drought tolerance in sesame.

GWAS Uncovers Differential Genetic Bases for Drought and Salt Tolerances in Sesame at the Germination Stage

Sesame has great potential as an industrial crop but its production is challenged by drought and salt stresses. To unravel the genetic variants leading to salinity and drought tolerances at the germination stage, genome-wide association studies of stress tolerance indexes related to NaCl-salt and polyethylene glycol-drought induced stresses were performed with a diversity panel of 490 sesame accessions. An extensive variation was observed for drought and salt responses in the population and most of the accessions were moderately tolerant to both stresses. A total of 132 and 120 significant Single Nucleotide Polymorphisms (SNPs) resolved to nine and 15 Quantitative trait loci (QTLs) were detected for drought and salt stresses, respectively. Only two common QTLs for drought and salt responses were found located on linkage groups 5 and 7, respectively. This indicates that the genetic bases for drought and salt responses in sesame are different. A total of 13 and 27 potential candidate genes were uncovered for drought and salt tolerance indexes, respectively, encoding transcription factors, antioxidative enzymes, osmoprotectants and involved in hormonal biosynthesis, signal transduction or ion sequestration. The identified SNPs and potential candidate genes represent valuable resources for future functional characterization towards the enhancement of sesame cultivars for drought and salt tolerances.

ECG R-wave peaks marking with simultaneously recorded continuous blood pressure

ECG signal is relatively weak and vulnerable to various noise interferences, such as electromyography. There will be robustness problems when detecting the instantaneous heart rate independently. In some cases, multiple human physiologic parameters are monitored to help in heart rate detection.
In this paper, an algorithm that marks the R-wave peaks with the help of simultaneously recorded continuous blood pressure is proposed and tested on two databases. One database, called the challenge database, is provided by the PhysioNet/Computing in Cardiology Challenge 2014, and the other is the MGH/MF waveform database.
The final scores of the proposed algorithm are 97.3% for the challenge database and 96.6% for the MGH/MF waveform database.
The experimental results show that this algorithm has high detection accuracy and a relatively strong robustness.
