Liangzhu culture : society, belief, and art in Neolithic China
\"Liangzhu culture (5,300-4,300 cal B.P.) represented the peak of prehistoric cultural and social development in the Yangtze Delta. Its centre is located near nowadays Hangzhou city and is considered one of the earliest urban centres in prehistoric China, called by archaeologists the Liangzhu Site Complex. Although it remains a mystery for many in the west, Liangzhu is well known in China for its fine jade crafting industry; its enormous, well-structured earthen compound and recently discovered hydraulic system; and its far-flung impact on contemporary and succeeding cultures. With six chapters contributed by frontline archaeologists, Liangzhu Culture contextualises Liangzhu in broad socioeconomic and cultural backgrounds and provides new, first-hand data to help explain the development and structure of this early urban centre. Exploring the Association Between Overactive Bladder (OAB) and Cognitive decline: mediation by depression in elderly adults, a NHANES weighted analysis

Overactive bladder (OAB) is a prevalent condition among older adults and may be linked to cognitive health. This study explored a relationship between OAB and cognitive health among adults aged 60 ≥ years in the United States, using NHANES 2011–2014. A cross-sectional analysis was conducted using a nationally representative sample of 2,324 (45.95% male, 54.05% female), within 755 individuals have OAB. Cognitive health was assessed using the Consortium to Establish a Registry for Alzheimer’s Disease-Word Learning (CERAD W-L), Animal Fluency Test (AFT), and Digit Symbol Substitution Test (DSST). OAB was defined by a score more than 3 on the Overactive Bladder Syndrome Symptom Score (OABSS). Depression incidence was also recorded. Weighted linear regression models adjusted for demographic, behavioral factors, and comorbidities (e.g., diabetes, kidney disease etc.), were employed to examine the independent association between cognitive health and OAB. The results indicated that cognitive scores were significantly lower in the OAB group compared to the non-OAB group across all cognitive tests (p<0.001). The incidence of depression was significantly higher in the OAB group (15%) compared to the non-OAB group (6.05%) (p<0.001). Regression analysis revealed that a 14% decrease in cognitive health z-scores in patients with OAB (β=-0.14, CI (95%) [-0.22, -0.06]). Furthermore, depression was found to mediate 34.56% of the association between OAB and cognitive health relationship [CI (95%): 0.20, 0.69]. This negative association was robust in subgroups by demographics, behavioral factors, and comorbidities. This study’s findings, supported by mediation analysis, smoothed curve analysis, and sensitivity analysis, suggest that lower cognitive health is significantly associated with OAB, and depression plays an important mediating role in this association.

To describe the clinical manifestation, immunotherapy, and long-term outcomes of anti-leucine-rich glioma-inactivated 1 (LGI1) encephalitis.
This study was a retrospective analysis of 117 patients with a diagnosis of anti-LGI1 encephalitis identified from the databases of multiple clinical centers between September 2014 and December 2019. The clinical features, ancillary test results, and details of long-term outcomes were evaluated.
Among the 117 patients with anti-LGI1 encephalitis, 69.2% (81/117) were male and 30.8% (36/117) were female. The median age of all patients at the onset of the disease was 57 years (interquartile range [IQR], 52-67). The median time from symptom onset to diagnosis was 8.7 weeks (IQR, 4.2-25). The main clinical features identified were seizures, cognitive impairment, and mental and behavioral abnormalities. Of the 117 patients, 109 were treated with immunotherapy. Symptoms including memory, mental ability, and behavior improved in all 109 patients after 3-5 days of treatment. The median time of follow-up for the treated patients was 33 months (IQR, 17-42). Of the treated patients, 16.2% (19/117) experienced a relapse, with a median delay of 5 months (IQR, 2.1-17) between onset and the first relapse. There were no mortalities over the follow-up period.
The long-term outcome of patients with anti-LGI1 encephalitis was mostly favorable, although some patients continued to experience cognitive dysfunction. Early recognition is important for prompt initiation of immunotherapy that can improve clinical symptoms of anti-LGI1 encephalitis.

Immunotherapy is a revolutionized therapeutic strategy for tumor treatment attributing to the rapid development of genomics and immunology, and immune checkpoint inhibitors have successfully achieved responses in numbers of tumor types, including hematopoietic malignancy. However, acute myeloid leukemia (AML) is a heterogeneous disease and there is still a lack of systematic demonstration to apply immunotherapy in AML based on PD-1/PD-L1 blockage. Thus, the identification of molecules that drive tumor immunosuppression and stratify patients according to the benefit from immune checkpoint inhibitors is urgently needed. Here, we reported that STAT5 was highly expressed in the AML cohort and activated the promoter of glycolytic genes to promote glycolysis in AML cells. As a result, the increased-lactate accumulation promoted E3BP nuclear translocation and facilitated histone lactylation, ultimately inducing PD-L1 transcription. T cells in the microenvironment when co-culture with STAT5 constitutively activated AML cells. Clinically, lactate accumulation in bone marrow was positively correlated with STAT5 as well as PD-L1 expression in newly diagnosed AML patients. Therefore, we have illustrated a STAT5-lactate-PD-L1 network in AML progression, which demonstrates that AML patients with STAT5 induced-exuberant glycolysis and lactate accumulation may be benefited from PD-1/PD-L-1-based immunotherapy.

Oncogene-induced cellular senescence (OIS) is a complex program that is triggered in response to aberrant activation of oncogenic signaling. Initially, OIS was thought to be a barrier to malignant transformation because of its suppression on cell proliferation. Later studies showed that senescence induced by oncogenes can also promote the initiation and development of cancer. The opposing effects of OIS occur through different combinations of downstream effectors as well as the interplay of senescent cells and the microenvironment, such as senescence-associated inflammation. Here, we review the common features and molecular mechanisms underlying OIS and the interaction between senescent cells and the microenvironment. We propose that targeting senescent cells may have a beneficial therapeutic effect during the treatment of cancer.
