Paroxysmal means PAVB is more dangerous than persistent third-degree AVB, as patients may experience prolonged ventricular asystole without warning, increasing the risk of syncope or sudden death.4 Electrophysiology tests often have a low diagnostic yield in such cases, making prolonged monitoring crucial for diagnosis. Extended ECG monitoring, including implantable loop recorders, can improve the detection rate of PAVB.5 Besides, timely pacemaker implantation is essential to prevent sudden death. The [-2]proPSA (p2PSA) and its derivatives, the p2PSA-to-free PSA ratio (%p2PSA), and the Prostate Health Index (PHI) have greatly improved discrimination between men with and without prostate cancer (PCa) in prostate biopsies. However, little is known about their performance in cases where a digital rectal examination (DRE) and transrectal ultrasonography (TRUS) are negative. A prospective cohort of 261 consecutive patients in China with negative DRE and TRUS were recruited and underwent prostate biopsies. A serum sample had collected before the biopsy was used to measure various PSA derivatives, including total prostate-specific antigen (tPSA), free PSA, and p2PSA. For each patient, the free-to-total PSA ratio (%fPSA), PSA density (PSAD), p2PSA-to-free PSA ratio (%p2PSA), and PHI were calculated. Discriminative performance was assessed using the area under the receiver operating characteristic curve (AUC) and the biopsy rate at 91% sensitivity. The AUC scores within the entire cohort with respect to age, tPSA, %fPSA, PSAD, p2PSA, %p2PSA, and PHI were 0.598, 0.751, 0.646, 0.789, 0.814, 0.808, and 0.853, respectively. PHI was the best predictor of prostate biopsy results, especially in patients with a tPSA of 10.1-20 ng ml-1. Compared with other markers, at a sensitivity of 91%, PHI was the most useful for determining which men did not need to undergo biopsy, thereby avoiding unnecessary procedures. The use of PHI could improve the accuracy of PCa detection by predicting prostate biopsy outcomes among men with a negative DRE and TRUS in China.

The Huashan risk calculators performed better in prediction of prostate cancer in Chinese population: a training study followed by a validation study

The performances of the Prostate Cancer Prevention Trial (PCPT) risk calculator and other risk calculators for prostate cancer (PCa) prediction in Chinese populations were poorly understood. We performed this study to build risk calculators (Huashan risk calculators) based on Chinese population and validated the performance of prostate-specific antigen (PSA), PCPT risk calculator, and Huashan risk calculators in a validation cohort. We built Huashan risk calculators based on data from 1059 men who underwent initial prostate biopsy from January 2006 to December 2010 in a training cohort. Then, we validated the performance of PSA, PCPT risk calculator, and Huashan risk calculators in an observational validation study from January 2011 to December 2014. All necessary clinical information were collected before the biopsy. The results showed that Huashan risk calculators 1 and 2 outperformed the PCPT risk calculator for predicting PCa in both entire training cohort and stratified population (with PSA from 2.0 ng ml^-1 to 20.0 ng ml^-1). In the validation study, Huashan risk calculator 1 still outperformed the PCPT risk calculator in the entire validation cohort (0.849 vs 0.779 in area under the receiver operating characteristic curve [AUC]) and stratified population. A considerable reduction of unnecessary biopsies (approximately 30%) was also observed when the Huashan risk calculators were used. Thus, we believe that the Huashan risk calculators (especially Huashan risk calculator 1) may have added value for predicting PCa in Chinese population. However, these results still needed further evaluation in larger populations.

Systematic investigation of the high-spin structures in the odd-odd nuclei \\(^{166, 168, 170, 172}\\)Re by a particle-number conserving method

The recently observed two and four-quasiparticle high-spin rotational bands in the odd-odd nuclei \\(^{166, 168, 170, 172}\\)Re are investigated using the cranked shell model with pairing correlations treated by a particle-number conserving method. The experimental moments of inertia and alignments can be reproduced well by the present calculation if appropriate bandhead spins and configurations are assigned for these bands, which in turn confirms their spin and configuration assignments. It is found that the bandhead spins of those two rotational bands observed in \\(^{166}\\)Re~[Li {\\it et al.}, Phys. Rev. C 92 014310 (2015)] should be both increased by \\(2\\hbar\\) to get in consistent with the systematics of the experimental and calculated moments of inertia for the same configurations in \\(^{168, 170, 172}\\)Re. The variations of the backbendings/upbendings with increasing neutron number in these nuclei are investigated. The level crossing mechanism is well understood by analysing the variations of the occupation probabilities of the single-particle states close to the Fermi surface and their contributions to the angular momentum alignment with rotational frequency. In addition, the influence of the deformation driving effects of the proton \\(1/2^-[541]\\) (\\(h_{9/2}\\)) orbtial on the level crossing in \\(^{172}\\)Re is also discussed.

Dear Editor We report here the performance of prostate cancer antigen 3 (PCA3) and genetic risk score (GRS) in predicting prostate cancer (PCa) from the prostate biopsy. To the best of our knowledge, this is the first report of simultaneously evaluating these two biomarkers in the same study.

Configuration and bandhead spin assignments for the 2-quasiparticle rotational bands in the neutron-rich nuclei \\(^{154, 156}\\)Pm

The rotational bands in the neutron-rich nuclei \\(^{153-157}\\)Pm are investigated by a particle-number conserving method. The kinematic moments of inertia for the 1-quasiparticle bands in odd-\\(A\\) Pm isotopes \\(^{153, 155, 157}\\)Pm are reproduced quite well by the present calculation. By comparison between the experimental and calculated moments of inertia for the three 2-quasiparticle bands in the odd-odd nuclei \\(^{154, 156}\\)Pm, their configurations and bandhead spins have been assigned properly. For the 2-quasiparticle band in \\(^{154}\\)Pm, the configuration is assigned as \\(\\pi5/2^-[532]\\otimes \\nu3/2^-[521]\\) (\\(K^\\pi=4^+\\)) with the bandhead spin \\(I_0=4\\hbar\\). In \\(^{156}\\)Pm, the same configuration and bandhead spin assignments have been made for the 2-quasiparticle band with lower excitation energy. The configuration \\(\\pi5/2^+[413]\\otimes \\nu5/2^+[642]\\) (\\(K^\\pi=5^+\\)) with the bandhead spin \\(I_0=5\\hbar\\) is assigned for that with higher excitation energy.
TRIM25 activates AKT/mTOR by inhibiting PTEN via K63-linked polyubiquitination in non-small cell lung cancer

The PTEN/AKT/mTOR signaling pathway is frequently dysregulated in non-small cell lung cancer (NSCLC), but the mechanisms are not well-understood. The present study found that the ubiquitin ligase TRIM25 is highly expressed in NSCLC tissues and promotes NSCLC cell survival and tumor growth. Mechanistic studies revealed that TRIM25 binds to PTEN and mediates its K63-linked ubiquitination at K266. This modification prevents the plasma membrane translocation of PTEN and reduces its phosphatase activity therefore accumulating PI(3,4,5)P3. TRIM25 thus activates the AKT/mTOR signaling. Moreover, we found that the antibacterial nitroxoline can activate PTEN by reducing its K63-linked polyubiquitination and sensitizes NSCLC to cisplatin-induced apoptosis. This study thus identified a novel modulation on the PTEN signaling pathway by TRIM25 and provides a potential target for NSCLC treatment.

A novel S-scheme heterojunction photocatalyst, Yb6Te5O19.2/g-C3N4: synthesis, characterization, photocatalytic activity, and mechanism

The increasing reliance on fossil fuels has led to a surge in atmospheric CO₂, intensifying global warming, whereas water contamination by organic pollutants presents another pressing global issue. In response, we have developed Yb₆Te₅O₁₉.₂/g-C₃N₄ (YTO/GCN) composites as novel S-scheme photocatalysts that efficiently target the degradation of pollutants and CO₂ reduction. These composites were synthesized through a straightforward hydrothermal method and comprehensively characterized via XRD, SEM, TEM‒EDS, DRS, BET, PL, and XPS analyses. Compared with the individual YTO and GCN semiconductors, the YTO/GCN heterojunction demonstrated superior photocatalytic efficiency in both crystal violet (CV) degradation under visible-light irradiation and CO₂ photoreduction. The YTO-90%GCN composite achieved the highest CV degradation rate constant (0.043 h⁻¹), which surpassed those of YTO and GCN by 2.97 and 1.11 times, respectively. Furthermore, the YTO-90%GCN composite produced 0.449 µmol/g CH₄ from CO₂ reduction over 216 h, which was 1.07 and 9.78 times greater than that of YTO and GCN alone, respectively. Scavenger and ESR experiments were conducted to identify the active species involved in CV degradation and to elucidate the photocatalytic mechanism of the YTO/GCN composite.

Prostate volume does not provide additional predictive value to prostate health index for prostate cancer or clinically significant prostate cancer: results from a multicenter study in China

To evaluate whether prostate volume (PV) would provide additional predictive utility to the prostate health index (phi) for predicting prostate cancer (PCa) or clinically significant prostate cancer, we designed a prospective, observational multicenter study in two prostate biopsy cohorts. Cohort 1 included 595 patients from three medical centers from 2012 to 2013, and Cohort 2 included 1025 patients from four medical centers from 2013 to 2014. Area under the receiver operating characteristic curves (AUC) and logistic regression models were used to evaluate the predictive performance of PV-based derivatives and models. Linear regression analysis showed that both total prostate-specific antigen (tPSA) and free PSA (fPSA) were significantly correlated with PV (all P < 0.05). [-2]proPSA (p2PSA) was significantly correlated with PV in Cohort 2 (P< 0.001) but not in Cohort 1 (P= 0.309), while no significant association was observed between phi and PV. When combining phi with PV, phi density (PHID) and another phi derivative (PHIV, calculated as phi/PV0.5) did not outperform phi for predicting PCa or clinically significant PCa in either Cohort 1 or Cohort 2. Logistic regression analysis also showed that phi and PV were independent predictors for both PCa and clinically significant PCa (all P < 0.05); however, PV did not provide additional predictive value to phi when combining these derivatives in a regression model (all models vs phi were not statistically significant, all P > 0.05). In conclusion, PV-based derivatives (both PHIV and PHID) and models incorporating PV did not improve the predictive abilities of phi for either PCa or clinically significant PCa.

Lactate Decreases Bortezomib Sensitivity and Predicts Poor Clinical Outcomes of Multiple Myeloma

ObjectiveMetabolic disorders are regarded as hallmarks of multiple myeloma (MM) and are responsible for rapid cancer cell proliferation and tumor growth. However, the exact biological roles of metabolites in MM cells have not been fully explored. This study aimed to explore the feasibility and clinical significance of lactate for MM and investigate the molecular mechanism of lactic acid (Lac) in the proliferation of myeloma cells and cell sensitivity to bortezomib (BTZ).MethodsMetabolomic analysis of the serum was carried out to obtain metabolites expression and clinical characteristics in MM patients. The CCK8 assay and flow cytometry were used to detect cell proliferation, apoptosis, and cell cycle changes. Western blotting was used to detect the potential mechanism and apoptosis- and cycle-related protein changes.ResultsLactate was highly expressed in both the peripheral blood and bone marrow of MM patients. It was significantly correlated with Durie-Salmon Staging (DS Staging) and the International Staging System (ISS Staging) and the serum and urinary involved/uninvolved free light chain ratios. Patients with relatively high lactate levels had a poor treatment response. Moreover, in vitro experiments showed that Lac could promote the proliferation of tumor cells and decrease the proportion of G0/G1-phase cells, which was accompanied by an increased proportion of S-phase cells. In addition, Lac could decrease tumor sensitivity to BTZ by disrupting the expression of nuclear factor kappa B subunit 2 (NFkB2) and RelB.ConclusionMetabolic changes are important in MM cell proliferation and treatment response; lactate could be used as a biomarker in MM and as a therapeutic target to overcome cell resistance to BTZ.
