Hyodeoxycholic acid ameliorates nonalcoholic fatty liver disease by inhibiting RAN-mediated PPARα nucleus-cytoplasm shuttling
2023
Nonalcoholic fatty liver disease (NAFLD) is usually characterized with disrupted bile acid (BA) homeostasis. However, the exact role of certain BA in NAFLD is poorly understood. Here we show levels of serum hyodeoxycholic acid (HDCA) decrease in both NAFLD patients and mice, as well as in liver and intestinal contents of NAFLD mice compared to their healthy counterparts. Serum HDCA is also inversely correlated with NAFLD severity. Dietary HDCA supplementation ameliorates diet-induced NAFLD in male wild type mice by activating fatty acid oxidation in hepatic peroxisome proliferator-activated receptor α (PPARα)-dependent way because the anti-NAFLD effect of HDCA is abolished in hepatocyte-specific
Pparα
knockout mice. Mechanistically, HDCA facilitates nuclear localization of PPARα by directly interacting with RAN protein. This interaction disrupts the formation of RAN/CRM1/PPARα nucleus-cytoplasm shuttling heterotrimer. Our results demonstrate the therapeutic potential of HDCA for NAFLD and provide new insights of BAs on regulating fatty acid metabolism.
Nonalcoholic fatty liver disease (NAFLD) is often linked to disrupted bile acid homeostasis. Here, the authors show hyodeoxycholic acid (HDCA) ameliorates nonalcoholic fatty liver disease by inhibiting the formation of RAN/CRM1/PPARα nuclear export heterotrimer, resulting in increased nuclear localization of PPARα and activated fatty acid oxidation.

Green Pak Choi is better in suitable environment but the purple ones more resist to drought and shading
2025
Background
Studying how economic vegetable adapt to stressful environment is important not only for plant biology application but also to agronomy. In this study, we selected two commonly used genotypes of pak choi, i.e., larger green pak choi (
Brassica rapa
ssp.
chinensis
) and smaller purple pak choi (
Brassica rapa
var.
chinensis
, ‘Rubi F1’) to examine the divergent response of the two genotypes to drought and shading in the semi-arid region of Xinjiang. We compared the differences in biomass accumulation and plant morphological traits of the two pak choi in response to the interaction effects of drought (55-70% of field water capacity) and shading (24% reduction of canopy light radiation).
Results
The results showed drought and shading significantly reduced the aboveground and belowground biomass of the two pak choi, with a particularly pronounced decrease in shoot biomass under the combined effect of shading + drought. The decline in shoot biomass was mostly resulted from decreasing in the number of leaves rather than in plant height and crown width in response to drought and shading. In terms of morphological traits, green pak choi sensitively responded to increased drought and shading, with aboveground biomass mostly determined by leaf number and root mass. In contrast, purple pak choi likely more resistant to the stressful environment, as its aboveground biomass was also influenced by plant height and crown width.
Conclusions
Hence it is important to consider not only the effects of drought but also the role of adequate light, which plays a key part in promoting the cultivation and growth of pak choi in stressful environments. The research and application of plant biology and agronomy in the region also need to consider the diversity of key economic plants to promote sustainability of vegetable farming in adapting to changing environmental stresses.

2023
Background
Nucleoside analogues are currently applied as a first-line treatment for chronic hepatitis B (CHB) patients. However, the long-term effects of this type of treatment on kidney and bone tissue need to be further investigated.
Methods
We conducted a search of entecavir (ETV), tenofovir disoproxil fumarate (TDF), and tenofovir alafenamide fumarate (TAF) for treatment of CHB patients through October 29, 2023. Side effects of the three drugs were compared. Standardized mean difference (SMD), 95% confidence interval (95%CI), and surface under the cumulative ranking curve (SUCRA) were reported for each outcome. Further subgroup analysis was conducted according to duration of administration.
Results
ETV and TAF exhibited less effect on estimated glomerular filtration rate (eGFR) than TDF (SMD = -3.60 (95%CI: -1.94 ~ -5.26) and SMD = -4.27 (95%CI: -2.62 ~ -5.93)). ETV also exhibited less effect on creatinine rise than TAF and TDF (SMD = -0.55 (95%CI: -0.09 ~ -1.01) and SMD = -0.61 (95%CI: -0.15 ~ -1.06)). Moreover, the effect of TAF on bone mineral density (BMD) was less than that of TDF (SMD = -0.02 (95%CI: -0.01 ~ -0.02)). The probabilities of the three drugs changing relevant indicators exhibited similar patterns: eGFR (TDF (100.0%) > ETV (41.2%) > TAF (8.8%)), creatinine (TDF (94.7%) > TAF (54.7%) > ETV (0.6%)), BMD (TDF (79.7%) > ETV (50.6%) > TAF (19.6%)), and blood phosphorus (TDF (90.6%) > TAF (49.8%) > ETV (9.7%)). After 6 and 24 months of treatment, no statistically significant difference in renal function or bone tissue was observed between ETV and TDF. However, greater adverse effects on renal function were observed for TDF than ETV at 60 months compared to 12 months. TDF also exhibited greater adverse effects on bone tissue than ETV at 36 months than at 12 months.
Conclusions
Long-term administration of TDF has resulted in stronger adverse effects than TAF and ETV in regard to both renal function and bone tissue in CHB patients. The effect of TAF on creatinine increase was greater than ETV. The difference in side effects between ETV and TDF was independent of treatment duration.

Dynamic Analysis of a Cross-Lingual Coupled Rumor Propagation Model with Response Delay in Online Social Networks
2026
As online social networks (OSNs) evolve into multilingual ecosystems, rumors can cross language boundaries through translation and bilingual re-expression, increasing governance difficulty. To characterize cross-lingual coupling and response delay, this study proposes a time-delay S2LCHR dynamical model for bilingual OSNs, in which the coupled spreader state
describes cross-lingual coupled rumor transmission and a fixed response delay represents cross-lingual comprehension, judgment, and re-expression. The basic reproduction number R0 is derived using the next-generation matrix method. Lyapunov analysis, the Routh-Hurwitz criterion, characteristic-equation analysis, and numerical simulations are combined to examine equilibrium stability, delay-induced Hopf bifurcation, parameter sensitivity, and social-impact indicators. A real-world aggregate trend-fitting case study using English-Spanish COVID-19-related tweet data is further conducted to assess empirical plausibility. The results show that R0 determines the threshold between rumor extinction and persistence in the delay-free system, while an excessive response delay can destabilize the rumor-prevailing equilibrium and induce bounded oscillatory behavior. Sensitivity and social-impact analyses indicate that α and β promote rumor persistence, whereas σ and φ associated with state
are key inhibitory factors. These findings suggest that coupled spreaders should be prioritized in cross-lingual rumor governance.

Liquid biopsies to track trastuzumab resistance in metastatic HER2-positive gastric cancer
2019
ObjectiveTo monitor trastuzumab resistance and determine the underlying mechanisms for the limited response rate and rapid emergence of resistance of HER2+ metastatic gastric cancer (mGC).DesignTargeted sequencing of 416 clinically relevant genes was performed in 78 paired plasma and tissue biopsy samples to determine plasma-tissue concordance. Then, we performed longitudinal analyses of 97 serial plasma samples collected from 24 patients who were HER2+ to track the resistance during trastuzumab treatment and validated the identified candidate resistance genes.ResultsThe results from targeted sequencing-based detection of somatic copy number alterations (SCNA) of HER2 gene were highly consistent with fluorescence in situ hybridisation data, and the detected HER2 SCNA was better than plasma carcinoembryonic antigen levels at predicting tumour shrinkage and progression. Furthermore, most patients with innate trastuzumab resistance presented high HER2 SCNA during progression compared with baseline, while HER2 SCNA decreased in patients with acquired resistance. PIK3CA mutations were significantly enriched in patients with innate resistance, and ERBB2/4 genes were the most mutated genes, accounting for trastuzumab resistance in six (35.3%) and five (29.4%) patients in baseline and progression plasma, respectively. Patients with PIK3CA/R1/C3 or ERBB2/4 mutations in the baseline plasma had significantly worse progression-free survival. Additionally, mutations in NF1 contributed to trastuzumab resistance, which was further confirmed through in vitro and in vivo studies, while combined HER2 and MEK/ERK blockade overcame trastuzumab resistance.ConclusionLongitudinal circulating tumour DNA sequencing provides novel insights into gene alterations underlying trastuzumab resistance in HER2+mGC.
