Effect of the vaginal microbiome on the pregnancy rate in women receiving assisted reproductive treatment
PurposeTo investigate if the vaginal microbiome influences the IVF outcome.MethodsThirty-one patients undergoing assisted reproductive treatment (ART) with own or donated gametes and with cryotransfer of a single euploid blastocyst were recruited for this cohort study. Two vaginal samples were taken during the embryo transfer procedure, just before transferring the embryo. The V3 V4 region of 16S rRNA was used to analyze the vaginal microbiome, and the bioinformatic analysis was performed using QIIME2, Bioconductor Phyloseq, and MicrobiomeAnalyst packages. Alpha diversity was compared between groups according to the result of the pregnancy test.ResultsFourteen (45.2%) patients did not and seventeen (54.8 %) did achieve pregnancy under ART. A greater index of alpha diversity was found in patients who did not achieve pregnancy comparing to those who did, although this difference was not significant (p = 0.088). In the analysis of beta diversity, no statistically significant differences were observed between groups established as per the pregnancy status. Samples from women who achieved pregnancy showed a greater presence of Lactobacillus spp. The cluster analysis identified two main clusters: the first encompassed the genera Lactobacillus, Gardnerella, Clostridium, Staphylococcus, and Dialister, and the second included all other genera. Women who achieved pregnancy were mainly detected microorganisms from the first cluster.ConclusionsThe vaginal microbiome can influence the results of ART. The profiles dominated by Lactobacillus were associated with the achievement of pregnancy, and there was a relationship between the stability of the vaginal microbiome and the achievement of pregnancy.
INCURVATA11 and CUPULIFORMIS2 Are Redundant Genes That Encode Epigenetic Machinery Components in Arabidopsis
All critical developmental and physiological events in a plant’s life cycle depend on the proper activation and repression of specific gene sets, and this often involves epigenetic mechanisms. Some Arabidopsis thaliana mutants with disorders of the epigenetic machinery exhibit pleiotropic defects, including incurved leaves and early flowering, due to the ectopic and heterochronic derepression of developmental regulators. Here, we studied one such mutant class, the incurvata11 (icu11) loss-of-function mutants. We have identified ICU11 as the founding member of a small gene family that we have named CUPULIFORMIS (CP). This family is part of the 2-oxoglutarate/Fe(II)-dependent dioxygenase superfamily. ICU11 and its closest paralog, CP2, have unequally redundant functions: although cp2 mutants are phenotypically wild type, icu11 cp2 double mutants skip vegetative development and flower upon germination. This phenotype is reminiscent of loss-of-function mutants of the Polycomb-group genes EMBRYONIC FLOWER1 (EMF1) and EMF2. Double mutants harboring icu11 alleles and loss-of-function alleles of genes encoding components of the epigenetic machinery exhibit synergistic, severe phenotypes, and some are similar to those of emf mutants. Hundreds of genes are misexpressed in icu11 plants, including SEPALLATA3 (SEP3), and derepression of SEP3 causes the leaf phenotype of icu11. ICU11 and CP2 are nucleoplasmic proteins that act as epigenetic repressors through an unknown mechanism involving histone modification, but not DNA methylation.
Impact of the Vaginal and Endometrial Microbiome Pattern on Assisted Reproduction Outcomes
Uterine microbiota may be involved in reproductive health and disease. This study aims to describe and compare the vaginal and endometrial microbiome patterns between women who became pregnant and women who did not after in vitro fertilization. We also compared the vaginal and endometrial microbiome patterns between women with and without a history of repeated implantation failures (RIF). This pilot prospective cohort study included 48 women presenting to the fertility clinic for IVF from May 2017 to May 2019. Women who achieved clinical pregnancy presented a greater relative abundance of Lactobacillus spp. in their vaginal samples than those who did not (97.69% versus 94.63%; p = 0.027. The alpha and beta diversity of vaginal and endometrial samples were not statistically different between pregnant and non-pregnant women. The Faith alpha diversity index in vaginal samples was lower in women with RIF than those without RIF (p = 0.027). The alpha diversity of the endometrial microbiome was significantly higher in women without RIF (p = 0.021). There were no significant differences in the vaginal and endometrial microbiomes between pregnant and non-pregnant women. The relative abundance of the genera in women with RIF was different from those without RIF. Statistically significant differences in the endometrial microbiome were found between women with and without RIF.
Roles of the Arabidopsis KEULE Gene in Postembryonic Development
Cytokinesis in plant cells begins with the fusion of vesicles that transport cell wall materials to the center of the cell division plane, where the cell plate forms and expands radially until it fuses with the parental cell wall. Vesicle fusion is facilitated by trans-SNARE complexes, with assistance from Sec1/Munc18 (SM) proteins. The SNARE protein KNOLLE and the SM protein KEULE are required for membrane fusion at the cell plate. Due to the crucial function of KEULE, all Arabidopsis (Arabidopsis thaliana) keule mutants identified to date are seedling lethal. Here, we identified the Arabidopsis serrata4-1 (sea4-1) and sea4-2 mutants, which carry recessive, hypomorphic alleles of KEULE. Homozygous sea4-1 and sea4-2 plants are viable and fertile but have smaller rosettes and fewer leaves at bolting than the wild type. Their leaves are serrated, small, and wavy, with a complex venation pattern. The mutant leaves also develop necrotic patches and undergo premature senescence. RNA-seq revealed transcriptome changes likely leading to reduced cell wall integrity and an increase in the unfolded protein response. These findings shed light on the roles of KEULE in postembryonic development, particularly in the patterning of rosette leaves and leaf margins.
Effects of Fear of Crime and Financial Scarcity on Wellbeing and Prison Sentences
In the present article we will examine whether the fear felt towards crime is related to the perception of financial scarcity when satisfying needs, thus attempting to determine whether such a fear influences considerations of whether the transgressor must serve a prison sentence. In addition, we through a correlational design, we will analyse through a correlational design, how fear of crime and perceived financial scarcity is linked to mood and wellbeing. The results from among a large sample of Spanish participants (1,474) have demonstrated that no relation exists between both concerns (fear of crime and perceived financial scarcity); that a not at all insignificant percentage (1.1%) wants the offender to serve a prison sentence; that fear does not correlate with general wellbeing and that financial scarcity does not provoke significant differences on positive mood. These findings allow us to advance in the study of the issues here analysed and thus propose solutions in order to address their consequences.
Characterization of the Endometrial Microbiome in Patients with Recurrent Implantation Failure
An abnormal endometrial microbiota has been associated with implantation failure; therefore, it may be important to evaluate it in order to improve reproductive outcomes in infertile patients. The main objective of our study was to compare the endometrial microbiome of patients with recurrent implantation failure (RIF) and control patients undergoing assisted reproduction treatment (ART). A prospective cohort study including forty-five patients with their own or donated gametes. The endometrial microbiome was analysed by massive sequencing of the bacterial 16S rRNA gene. Different bacterial communities were detected in RIF and control patients. Lactobacillus stands out as the most frequent genus, with 92.27% in RIF patients and 97.96% in control patients, and significant differences were reported between the two groups (p = 0.002). No significant differences were found regarding alpha diversity index. In beta diversity analysis, a significant trend was observed in the separation of the bacterial community between established groups (p < 0.07). Relative abundance analysis identified genera Prevotella (p < 0.001), Streptococcus (p < 0.001), Bifidobacterium (p = 0.002), Lactobacillus (p = 0.002) and Dialister (p = 0.003). Our results demonstrated the existence of an endometrial microbiota characteristic of RIF patients and showed that there might be a relationship between population of the endometrial microbiome and embryo implantation failure, providing us the possibility to improve clinical results in this patients.
Justicia restaurativa y su relación con la empatía y los valores Sociales
Con el propósito de conocer cómo se relacionan los indicadores psicosociales, caracterizados por la empatía y los valores sociales, se realizó un estudio cuasiexperimental de Justicia Restaurativa (JR) en el que participaron 187 sujetos pertenecientes a la población general, con una media de edad de 40.27 (DE = 12.32) años. Éstos fueron asignados al azar a un supuesto sobre JR que contenía 3 condiciones (desafiante, arrepentimiento e impunidad) y cuestiones sobre la gestión del delito, como son: el castigo, el diálogo, la responsabilización y la reparación. Además, los participantes cumplimentaron escalas sobre emociones negativas (α = 0.81), empatía (α = 0.72) y valores sociales (α = 0.76). A través de análisis correlacionales (r de Pearson) y de comparación de medias (prueba t y ANOVA) se constató que la emoción que se siente con más intensidad, ante el delito que sufre otra persona, es el enfado, existiendo diferencias en función de las actitudes desafiante y de arrepentimiento mostradas por el agresor, para el conjunto de emociones negativas, así como para la posibilidad de aceptar el diálogo entre las partes. El hecho de poseer alta amabilidad empática hace que se faciliten los procesos de JR caracterizados por la responsabilización y la reparación, mientras que los valores de conservadurismo (tradición y seguridad) se vincularon con el castigo. Todos estos hallazgos permiten avanzar en el estudio de la JR, la empatía y los valores sociales.
Experiencias innovadoras e integradoras de justicia restaurativa en ejecución de condena: proyecto restauravidas. Reintegración en el país de origen
La presente publicación sintetiza un estudio basado en datos descriptivos provenientes del desarrollo de un programa de intervención con personas españolas condenadas a privación de libertad en elextranjero por el tráfico de drogas. El doble objetivo de este trabajo radica en el análisis de determinados procesos psicológicos, criminológicos y restaurativos presentes en estas personas en su proceso de  Reintegración tras la experiencia traumática, así como la detección del perfil mayoritario en este tipo de delitos, para la aplicación de este conocimiento decara a futuras políticas y acciones sensibilizadoras y preventivas.

Sex differences in social support perceived by polymedicated older adults with multimorbidity. MULTIPAP study
The beneficial effects of social support on morbidity, mortality, and quality of life are well known. Using the baseline data of the MULTIPAP study (n = 593), an observational, descriptive, cross-sectional study was carried out that analyzed the sex differences in the social support perceived by polymedicated adults aged 65 to 74 years with multimorbidity. The main outcome variable was social support measured through the Duke–UNC-11 Functional Social Support (DUFSS) questionnaire in its two dimensions (confident support and affective support). For both sexes, the perception of functional social support was correlated with being married or partnered and having a higher health-related quality of life utility index. In women, it was correlated with a higher level of education, living alone, and treatment adherence, and in men with higher monthly income, prescribed drugs and fewer diagnosed diseases.
