Long known as digestive organelles, lysosomes have now emerged as multifaceted centers responsible for degradation, nutrient sensing, and immunity. Growing evidence also implicates role of lysosome-related mechanisms in pathologic process. In this review, we discuss physiological function of lysosomes and, more importantly, how the homeostasis of lysosomes is disrupted in several diseases, including atherosclerosis, neurodegenerative diseases, autoimmune disorders, pancreatitis, lysosomal storage disorders, and malignant tumors. In atherosclerosis and Gaucher disease, dysfunction of lysosomes changes cytokine secretion from macrophages, partially through inflammasome activation. In neurodegenerative diseases, defect autophagy facilitates accumulation of toxic protein and dysfunctional organelles leading to neuron death. Lysosomal dysfunction has been demonstrated in pathology of pancreatitis. Abnormal autophagy activation or inhibition has been revealed in autoimmune disorders. In tumor microenvironment, malignant phenotypes, including tumorigenesis, growth regulation, invasion, drug resistance, and radiotherapy resistance, of tumor cells and behaviors of tumor-associated macrophages, fibroblasts, dendritic cells, and T cells are also mediated by lysosomes. Based on these findings, a series of therapeutic methods targeting lysosomal proteins and processes have been developed from bench to bedside. In a word, present researches corroborate lysosomes to be pivotal organelles for understanding pathology of atherosclerosis, neurodegenerative diseases, autoimmune disorders, pancreatitis, and lysosomal storage disorders, and malignant tumors and developing novel therapeutic strategies.

Observation of negative capacitance in antiferroelectric PbZrO3 Films

Negative capacitance effect in ferroelectric materials provides a solution to the energy dissipation problem induced by Boltzmann distribution of electrons in conventional electronics. Here, we discover that besides ferroelectrics, the antiferroelectrics based on Landau switches also have intrinsic negative capacitance effect. We report both the static and transient negative capacitance effect in antiferroelectric PbZrO
3
films and reveal its possible physical origin. The capacitance of the capacitor of the PbZrO
3
and paraelectric heterostructure is demonstrated to be larger than that of the isolated paraelectric capacitor at room temperature, indicating the existence of the static negative capacitance. The opposite variation trends of the voltage and charge transients in a circuit of the PbZrO
3
capacitor in series with an external resistor demonstrate the existence of transient negative capacitance effect. Strikingly, four negative capacitance effects are observed in the antiferroelectric system during one cycle scan of voltage pulses, different from the ferroelectric counterpart with two negative capacitance effects. The Role of the p21-Activated Kinase Family in Tumor Immunity

The p21-activated kinases (PAKs) are a group of evolutionarily conserved serine/threonine protein kinases and serve as a downstream target of the small GTPases Rac and Cdc42, both of which belong to the Rho family. PAKs play pivotal roles in various physiological processes, including cytoskeletal rearrangement and cellular signal transduction. Group II PAKs (PAK4-6) are particularly closely linked to human tumors, such as breast and pancreatic cancers, while Group I PAKs (PAK1-3) are indispensable for normal physiological functions such as cardiovascular development and neurogenesis. In recent years, the association of PAKs with diseases like cancer and the rise of small-molecule inhibitors targeting PAKs have attracted significant attention. This article focuses on the analysis of PAKs’ role in tumor progression and immune infiltration, as well as the current small-molecule inhibitors of PAKs and their mechanisms.

Identification of EMT-associated prognostic features among grade II/III gliomas

Grade II/III gliomas have a highly heterogeneous clinical course. Identifying prognostic biomarkers in grade II/III gliomas is essential to guide clinical management. We explored epithelial-mesenchymal transition (EMT)-related genes to uncover prognostic features in grade II/III gliomas. Consensus cluster analysis of 200 EMT-related genes classified 512 grade II/III glioma samples into two molecular subtypes, C1 and C2. The C1 subtype had significantly worse overall survival compared to the C2 subtype. Pathway analysis revealed C1 tumors were highly associated with tumor progression pathways and demonstrated higher immune cell infiltration scores. Differential expression analysis identified four genes (ACTN1, AQP1, LAMC3, NRM) that discriminated the two subtypes. Validation in external datasets confirmed that high expression of this four-gene signature predicted poor prognosis in grade II/III gliomas. Cellular experiments showed ACTN1, AQP1 and NRM promoted glioma cell proliferation, migration and invasion. We examined correlations of the signature genes with T cell exhaustion markers and found ACTN1 expression had the strongest association. Immunohistochemistry analysis further demonstrated that ACTN1 protein expression in grade II/III gliomas was negatively correlated with patient overall survival. In summary, our study identified a concise four-gene signature that robustly predicts grade II/III gliomas prognosis across multiple datasets. The signature provides clinical relevance in distinguishing more aggressive grade II/III glioma tumors. Targeting the ACTN1, AQP1 and NRM genes may offer new therapeutic opportunities to improve grade II/III gliomas patient outcomes.

Crop Mapping Using the Historical Crop Data Layer and Deep Neural Networks: A Case Study in Jilin Province, China

Machine learning combined with satellite image time series can quickly, and reliably be implemented to map crop distribution and growth monitoring necessary for food security. However, obtaining a large number of field survey samples for classifier training is often time-consuming and costly, which results in the very slow production of crop distribution maps. To overcome this challenge, we propose an ensemble learning approach from the existing historical crop data layer (CDL) to automatically create multitudes of samples according to the rules of spatiotemporal sample selection. Sentinel-2 monthly composite images from 2017 to 2019 for crop distribution mapping in Jilin Province were mosaicked and classified. Classification accuracies of four machine learning algorithms for a single-month and multi-month time series were compared. The results show that deep neural network (DNN) performed the best, followed by random forest (RF), then decision tree (DT), and support vector machine (SVM) the least. Compared with other months, July and August have higher classification accuracy, and the kappa coefficients of 0.78 and 0.79, respectively. Compared with a single phase, the kappa coefficient gradually increases with the growth of the time series, reaching 0.94 in August at the earliest, and then the increase is not obvious, and the highest in the whole growth cycle is 0.95. During the mapping process, time series of different lengths produced different classification results. Wetland types were misclassified as rice. In such cases, authors combined time series of two lengths to correct the misclassified rice types. By comparing with existing products and field points, rice has the highest consistency, followed by corn, whereas soybeans have the least consistency. This shows that the generated sample data set and trained model in this research can meet the crop mapping accuracy and simultaneously reduce the cost of field surveys. For further research, more years and types of crops should be considered for mapping and validation.

An adaptive hierarchical hybrid kernel ELM optimized by aquila optimizer algorithm for bearing fault diagnosis

As a critical component of rotating machinery, the operating status of rolling bearings is not only related to significant economic interests but also has a far-reaching impact on social security. Hence, ensuring an effective diagnosis of faults in rolling bearings is paramount in maintaining operational integrity. This paper proposes an intelligent bearing fault diagnosis method that improves classification accuracy using a stacked denoising autoencoder (SDAE) and adaptive hierarchical hybrid kernel extreme learning machine (AHHKELM). First, a hybrid kernel extreme learning machine (HKELM) is initially constructed, leveraging SDAE’s deep network architecture for automatic feature extraction. The hybrid kernel functions address the limitations of single kernel functions by effectively capturing both linear and nonlinear patterns in the data. Subsequently, the hierarchical hybrid kernel extreme learning machine (HHKELM) is refined through an enhanced Aquila Optimizer (AO) algorithm, which iteratively optimizes the kernel hyperparameter combination. The AO algorithm is further enhanced by incorporating chaos mapping, implementing a refined balanced search strategy, and fine-tuning parameter
, which collectively improve its ability to escape local optima and conduct global searches, thus strengthening the robustness of the model during parameter optimization. Experimental results on the CWRU , MFPT and JNU datasets demonstrate that stacked denoising autoencoder-adaptive hierarchical hybrid kernel extreme learning machine (SDAE-AHHKELM) has better fault classification accuracy, robustness, and generalization than KELM and other methods.
