Influence of Lifestyle Habits in the Development of Obesity during Adolescence
Background: The alarming increase in childhood obesity is a global public health problem since it has significant health consequences. This cross-sectional study aimed to identify the potentially modifiable risk factors for developing excess weight and determine the importance of developing certain habits to prevent childhood overweight and obesity. Methods: The sample included 416 students between the ages of 12 and 14 (12.8 ± 0.62) first-year high school students from Murcia in Spain. Data were collected on their lifestyle habits through the ENHASA questionnaire, and the somatometry of the participants was measured. Such data were studied through structural equation modeling and importance-performance map analyses. Results: The modifiable risk factors that presented the greatest potency directly regarding when developing excess weight in adolescents were ‘use of electronic devices’ and ‘not performing physical activity’ (p < 0.001). ‘Social and school environment’ and ‘diet’ showed relationships but no significant differences with overweight or obesity. Globally, the lifestyle habit of the greatest importance for not being overweight was ‘engagement in extracurricular physical activities’. On the other hand, the habits related to using new technologies in a sedentary way showed the best capacity for improvement. Therefore, it would be very efficient to focus on them to control excess weight. Conclusion: Responsible and limited use of screens and engaging in extracurricular physical activities may be the most remarkable and cost-effective strategies for obesity prevention programs.
Occult Hepatitis C Virus Infection in Patients in Whom the Etiology of Persistently Abnormal Results of Liver-Function Tests Is Unknown
BackgroundThere are patients in whom the etiology of long-standing abnormal results of liver-function tests is unknown (ALF-EU) after exclusion of all known causes of liver diseases. We analyzed the presence of hepatitis C virus (HCV) RNA in liver-biopsy specimens from 100 patients who were negative for anti-HCV antibodies and for serum HCV RNA and who had ALF-EU MethodsHCV RNA status was tested by reverse-transcription polymerase chain reaction (RT-PCR) and by in situ hybridization, in liver and peripheral-blood mononuclear cells (PBMCs) ResultsHCV RNA was detected in liver-biopsy specimens from 57 of 100 patients negative for anti-HCV antibodies and for serum HCV RNA (i.e., who had occult HCV infection). HCV RNA of negative polarity was found in the liver of 48 (84.2%) of these 57 patients with occult HCV infection. Nucleotide-sequence analysis confirmed the specificity of detection of HCV RNA and that patients were infected with the HCV 1b genotype. Of these 57 patients with intrahepatic HCV RNA, 40 (70%) had viral RNA in their PBMCs. With regard to liver histology, patients with occult HCV infection were more likely to have necroinflammatory activity (P=.017) and fibrosis (P=.022) than were patients without intrahepatic HCV RNA ConclusionPatients with ALF-EU may have intrahepatic HCV RNA in the absence of anti-HCV antibodies and of serum HCV RNA
Influence of physical fitness and weight status on autonomic cardiac modulation in children
Background This study provides reference values for cardiovascular modulation at rest, during maximal exercise test and recovery after exercise in Caucasian children according to weight status and cardiorespiratory fitness (CRF) level. Additionally, the current study analyzed several correlations between autonomic cardiovascular modulation, cardiorespiratory performance and cardiometabolic risk. The principal goal of this study was to analyze cardiac function at rest, during maximum exercise, and during the recovery phase in children grouped according to weight status and CRF level. Methods One hundred and fifty-two healthy children (78 girls) 10–16 years of age were divided into three groups: soccer and basketball players (SBG), endurance group (EG), and sedentary people with overweight and obesity (OOG). A cardiac RR interval monitor recorded the cardiac data and specific software analyzed the cardiac autonomic response through heart rate (HR) and HR variability. The study analyzed resting HR (RHR), HR peak , and HR recovery (HRR). Results OOG showed significant poorer performance in the Léger test lower V̇ O 2 max and higher values of blood pressure at rest and post-exercise than sport groups. The EG presented the best results in CRF and cardiometabolic risk (CMR) in relation to SBG and OOG. The OOG showed higher percentage of HR values, compatible with an unhealthy cardiovascular autonomic modulation than the sport groups, with significant differences in bradycardia, HR reserve, and HRR 5 min. Conclusions Aerobic performance, vagal activity, blood pressure, chronotropic competence, and HRR have significant associations with CMR parameters. Impact The current study presents reference values of autonomic cardiac function in Caucasian children according to weight status and cardiorespiratory fitness level. Aerobic performance, vagal activity, blood pressure, chronotropic competence, and heart rate during the recovery period after exercise have significant associations with cardiometabolic risk parameters. Children with overweight and obesity show signs of autonomic dysfunction reflected as low cardiac vagal activity and poor chronotropic competence.
Statistical validation of Aeolus L2A particle backscatter coefficient retrievals over ACTRIS/EARLINET stations on the Iberian Peninsula
The Global Observing System (GOS) has encountered some limitations due to a lack of worldwide real-time wind measurements. In this context, the European Space Agency (ESA) has developed the Aeolus satellite mission, based on the ALADIN (Atmospheric Laser Doppler Instrument) Doppler wind lidar; this mission aims to obtain near-real-time wind retrievals at the global scale. As spin-off products, the instrument retrieves aerosol optical properties such as particle backscatter and extinction coefficients. In this work, a validation of Aeolus reprocessed (baseline 10) co-polar backscatter coefficients (βAeoluspart) is presented through an intercomparison with analogous ground-based measurements taken at the ACTRIS (Aerosols, Clouds and Trace gases Research InfraStructure Network)/EARLINET (European Aerosol Research Lidar Network) stations of Granada (Spain), Évora (Portugal) and Barcelona (Spain) over the period from July 2019 until October 2020. Case studies are first presented, followed by a statistical analysis. The stations are located in a hot spot between Africa and the rest of Europe, which guarantees a variety of aerosol types, from mineral dust layers to continental/anthropogenic aerosol, and allows us to test Aeolus performance under different scenarios. The so called Aeolus-like profiles (βAeoluslike,355part) are obtained from total particle backscatter coefficient and linear particle depolarization ratio (δlinearpart) profiles at 355 and 532 nm measured from the surface, through a thorough bibliographic review of dual-polarization measurements for relevant aerosol types. Finally, the study proposes a relation for the spectral conversion of δlinearpart, which is implemented in the Aeolus-like profile calculation. The statistical results show the ability of the satellite to detect and characterize significant aerosol layers under cloud-free conditions, along with the surface effect on the lowermost measurements, which causes the satellite to largely overestimate co-polar backscatter coefficients. Finally, the Aeolus standard correct algorithm middle bin (SCAmb) shows a better agreement with ground-based measurements than the standard correct algorithm (SCA), which tends to retrieve negative and meaningless coefficients in the clear troposphere. The implementation of Aeolus quality flags entails a vast reduction in the number of measurements available for comparison, which affects the statistical significance of the results.
Real-World Evaluation of Once-Weekly Subcutaneous Semaglutide in Patients with Type 2 Diabetes Mellitus in Spain (SEMA-RW Study)
Although, in randomized clinical trials, once-weekly subcutaneous semaglutide (OW s.c.) has demonstrated superior efficacy in comparison with placebo and active controls in terms of glycemic control and body weight reduction in patients with type 2 diabetes mellitus (T2DM), these results need to be confirmed in a real-world (RW) setting. An RW ambispective study (6 months retrospective and 6 months prospective) was conducted in 10 tertiary hospitals in Spain. We evaluated changes in HbA1c and body weight in patients with T2DM treated with semaglutide OW s.c. Additionally, we analyzed different subgroups of patients treated with semaglutide OW s.c. as an add-on to glucose-lowering therapy. A total of 752 patients with a mean age of 60.2 years, a mean HbA1c level of 8.5%, a mean body weight of 101.6 kg, and a mean T2DM duration of 10 years were included. At 12 months, compared with baseline, there was a mean difference of −2.1% in HbA1c levels (p < 0.001) and a mean difference of 9.2 kg in body weight (p < 0.001). Moreover, there were statistically significant differences (p < 0.001) between baseline and month 12 in both HbA1c and body weight in the four subgroups receiving semaglutide OW s.c. as an add-on to glucose-lowering therapy. Semaglutide OW s.c. was well tolerated, with gastrointestinal disorders being the most commonly reported side effects. In this RW study, 12 months of treatment with semaglutide OW s.c. in patients with T2DM was associated with significant and clinically relevant improvements in glycemic control and weight loss, regardless of the glucose-lowering therapy received, and the overall safety profile was positive.
SARS-CoV-2 accessory proteins involvement in inflammatory and profibrotic processes through IL11 signaling
SARS-CoV-2, the cause of the COVID-19 pandemic, possesses eleven accessory proteins encoded in its genome. Their roles during infection are still not completely understood. In this study, transcriptomics analysis revealed that both WNT5A and IL11 were significantly up-regulated in A549 cells expressing individual accessory proteins ORF6, ORF8, ORF9b or ORF9c from SARS-CoV-2 (Wuhan-Hu-1 isolate). IL11 is a member of the IL6 family of cytokines. IL11 signaling-related genes were also differentially expressed. Bioinformatics analysis disclosed that both WNT5A and IL11 were involved in pulmonary fibrosis idiopathic disease and functional assays confirmed their association with profibrotic cell responses. Subsequently, data comparison with lung cell lines infected with SARS-CoV-2 or lung biopsies from patients with COVID-19, evidenced altered profibrotic gene expression that matched those obtained in this study. Our results show ORF6, ORF8, ORF9b and ORF9c involvement in inflammatory and profibrotic responses. Thus, these accessory proteins could be targeted by new therapies against COVID-19 disease.
Applications of non-intrusive methods to study the sand cat: a field study in the Sahara Desert
Surveys based on indirect signs and camera trapping are two non-invasive methods extensively used for monitoring elusive mammals. Both approaches can be useful to obtain key information on wildlife in remote areas, since they may allow for the logistically viable design of optimal field frameworks. The sand cat (Felis margarita) is a feline that inhabits the Sahara Desert, the Arabian Peninsula, and western Asian deserts. Its basic ecology is poorly known and the status and impacts of threats are difficult to assess. Some local population declines have been detected, and more research is needed. Based on field surveys carried out in the Atlantic Sahara, we have evaluated the applications of both methods to study this species. Our results show that (a) camera trapping provided reliable data on several key aspects of its ecology, (b) walking surveys to collect feces for molecular data failed completely, and (c) for footprints, identification problems and the marked effects of the absence of optimal substrates and the prevalence of wind are relevant handicaps. Beyond this evaluation, we provide for the first time some key aspects of the ecology of sand cats in the Sahara Desert, including habitat selection, density, diel activity, and predator–prey relationships.
