Gender, Candidate Emotional Expression, and Voter Reactions During Televised Debates
Voters evaluate politicians not just by what they say, but also how they say it, via facial displays of emotions and vocal pitch. Candidate characteristics can shape how leaders use—and how voters react to—nonverbal cues. Drawing on role congruity expectations, we study how the use of and reactions to facial, vocal, and textual communication in political debates varies by candidate gender. Relying on full-length videos of four German federal election debates (2005–2017) and a minor party debate, we use video, audio, and text data to measure candidate facial displays of emotion, vocal pitch, and speech sentiment. Consistent with our expectations, Angela Merkel expresses less anger than her male opponents, but she is just as emotive in other respects. Combining these measures of emotional expression with continuous responses recorded by live audiences, we find that voters punish Merkel for anger displays and reward her happiness and general emotional displays.
Revealing centrality in the spatial structure of cities from human activity patterns
Identifying changes in the spatial structure of cities is a prerequisite for the development and validation of adequate planning strategies. Nevertheless, current methods of measurement are becoming ever more challenged by the highly diverse and intertwined ways of how people actually make use of urban space. Here, we propose a new quantitative measure for the centrality of locations, taking into account not only the numbers of people attracted to different locations, but also the diversity of the activities they are engaged in. This 'centrality index' allows for the identification of functional urban centres and for a systematic tracking of their relative importance over time, thus contributing to our understanding of polycentricity. We demonstrate the proposed index using travel survey data in Singapore for different years between 1997 and 2012. It is shown that, on the one hand, the city-state has been developing rapidly towards a polycentric urban form that compares rather closely with the official urban development plan. On the other hand, however, the downtown core has strongly gained in its importance, and this can be partly attributed to the recent extension of the public transit system.
Leader of the Pack? Changes in “Wolf Warrior Diplomacy” after a Politburo Collective Study Session
This research report measures changes in China's public diplomacy after a May 2021 collective study session of the Chinese Communist Party Politburo. The session examined the country's global communications strategy and fuelled speculation about what might change in China's external communications, particularly with regard to its “wolf warrior” diplomats. Combining hand-coding and quantitative text analysis, we develop and validate a measure of “wolf warrior diplomacy” rhetoric and apply it to over 200,000 tweets from nearly 200 institutional, media and diplomatic Twitter accounts. Using a difference-in-difference research design, we evaluate if the session led to a noticeable change in the tweets of diplomats based in OECD countries. After the announcement, PRC diplomats in the OECD moderated their tweets in comparison to non-OECD diplomats, but we do not detect a major re-orientation of PRC communication strategies. These findings have relevance for scholars of Chinese foreign policy, nationalism and public diplomacy.
Nostalgia in European Party Politics: A Text-Based Measurement Approach
Traditional research on political parties pays little attention to the temporal focus of communication. It usually concentrates on promises, issue attention, and policy positions. This lack of scholarly attention is surprising, given that voters respond to nostalgic rhetoric and may even adjust issue positions when policy is framed in nostalgic terms. This article presents a novel dataset, PolNos, which contains six text-based measures of nostalgic rhetoric in 1,648 party manifestos across 24 European democracies from 1946 to 2018. The measures combine dictionaries, word embeddings, sentiment approaches, and supervised machine learning. Our analysis yields a consistent result: nostalgia is most prevalent in manifestos of culturally conservative parties, notably Christian democratic, nationalist, and radical right parties. However, substantial variation remains regarding regional differences and whether nostalgia concerns the economy or culture. We discuss the implications and use of our dataset for studying political parties, party competition, and elections.
MicroRNAs in Renal Cell Carcinoma: Diagnostic Implications of Serum miR-1233 Levels
MicroRNA expression is altered in cancer cells, and microRNAs could serve as diagnostic/prognostic biomarker for cancer patients. Our study was designed to analyze circulating serum microRNAs in patients with renal cell carcinoma (RCC). We first explored microrna expression profiles in tissue and serum using taqman low density arrays in each six malignant and benign samples: Although 109 microRNAs were circulating at higher levels in cancer patients' serum, we identified only 36 microRNAs with up-regulation in RCC tissue and serum of RCC patients. Seven candidate microRNAs were selected for verification based on the finding of up-regulation in serum and tissue of RCC patients: miR-7-1*, miR-93, miR-106b*, miR-210, miR-320b, miR-1233 and miR-1290 levels in serum of healthy controls (n = 30) and RCC (n = 33) patients were determined using quantitative real-time PCR (TaqMan MicroRNA Assays). miR-1233 was increased in RCC patients, and thus validated in a multicentre cohort of 84 RCC patients and 93 healthy controls using quantitative real-time PCR (sensitivity 77.4%, specificity 37.6%, AUC 0.588). We also studied 13 samples of patients with angiomyolipoma or oncocytoma, whose serum miR-1233 levels were similar to RCC patients. Circulating microRNAs were not correlated with clinical-pathological parameters. MicroRNA levels are distinctly increased in cancer patients, although only a small subset of circulating microRNAs has a tumor-specific origin. We identify circulating miR-1233 as a potential biomarker for RCC patients. Larger-scaled studies are warranted to fully explore the role of circulating microRNAs in RCC.
Microbial-induced meprin β cleavage in MUC2 mucin and a functional CFTR channel are required to release anchored small intestinal mucus
The mucus that covers and protects the epithelium of the intestine is built around its major structural component, the gel-forming MUC2 mucin. The gel-forming mucins have traditionally been assumed to be secreted as nonattached. The colon has a two-layered mucus system where the inner mucus is attached to the epithelium, whereas the small intestine normally has a nonattached mucus. However, the mucus of the small intestine of meprin β-deficient mice was now found to be attached. Meprin β is an endogenous zinc-dependent metalloprotease now shown to cleave the N-terminal region of the MUC2 mucin at two specific sites. When recombinant meprin β was added to the attached mucus of meprin β-deficient mice, the mucus was detached from the epithelium. Similar to meprin β-deficient mice, germ-free mice have attached mucus as they did not shed the membrane-anchored meprin β into the luminal mucus. The ileal mucus of cystic fibrosis (CF) mice with a nonfunctional cystic fibrosis transmembrane conductance regulator (CFTR) channel was recently shown to be attached to the epithelium. Addition of recombinant meprin β to CF mucus did not release the mucus, but further addition of bicarbonate rendered the CF mucus normal, suggesting that MUC2 unfolding exposed the meprin β cleavage sites. Mucus is thus secreted attached to the goblet cells and requires an enzyme, meprin β in the small intestine, to be detached and released into the intestinal lumen. This process regulates mucus properties, can be triggered by bacterial contact, and is nonfunctional in CF due to poor mucin unfolding.
Activated SUMOylation restricts MHC class I antigen presentation to confer immune evasion in cancer
Activated SUMOylation is a hallmark of cancer. Starting from a targeted screening for SUMO-regulated immune evasion mechanisms, we identified an evolutionarily conserved function of activated SUMOylation, which attenuated the immunogenicity of tumor cells. Activated SUMOylation allowed cancer cells to evade CD8+ T cell-mediated immunosurveillance by suppressing the MHC class I (MHC-I) antigen-processing and presentation machinery (APM). Loss of the MHC-I APM is a frequent cause of resistance to cancer immunotherapies, and the pharmacological inhibition of SUMOylation (SUMOi) resulted in reduced activity of the transcriptional repressor scaffold attachment factor B (SAFB) and induction of the MHC-I APM. Consequently, SUMOi enhanced the presentation of antigens and the susceptibility of tumor cells to CD8+ T cell-mediated killing. Importantly, SUMOi also triggered the activation of CD8+ T cells and thereby drove a feed-forward loop amplifying the specific antitumor immune response. In summary, we showed that activated SUMOylation allowed tumor cells to evade antitumor immunosurveillance, and we have expanded the understanding of SUMOi as a rational therapeutic strategy for enhancing the efficacy of cancer immunotherapies.
