Lactic Acid Bacteria and Yeast Fermentation to Improve the Nutritional Value of Ulva rigida
Aquaculture reliance on fishmeal protein has become a bottleneck due to long-term sustainability concerns and increasing costs. Given its abundance and nutrient-rich profile, the green macroalga Ulva rigida is a promising alternative protein source. However, the bioaccessibility of its proteins is hindered by an embedding matrix of ulvan, a gel-forming polysaccharide. Saccharification of the alga crude fiber followed by microbial fermentation improves protein bioaccessibility and leads to products of higher protein content and quality. Also, upon fermentation, the nutritional and bioactive properties of these feed ingredients are enhanced, since microorganisms synthesize vitamins, new proteins, and essential amino acids. The carbohydrate fraction of Ulva rigida was hydrolyzed into a sugar-rich syrup and subsequently used as a substrate in microbial fermentations. Three types of fermentation were tested, namely, with a consortium of four lactic acid bacteria (LAB), with Saccharomyces cerevisiae, and with a co-culture of lactobacilli and yeast. A functional analysis of lyophilized whole-fermentation broths revealed that the yeast-fermented products had stronger antioxidant properties when compared to the LAB-fermented products. The protein bioaccessibility in the fermented products was 11- to 12-fold higher than that of the raw alga. These findings highlight the potential of utilizing S. cerevisiae and lactobacilli starter cultures in seaweed fermentation to produce Ulva-based feed ingredients.
An Antibody-Based Survey of Toxoplasma gondii and Neospora caninum Infection in Client-Owned Cats from Portugal
Toxoplasma gondii and Neospora caninum are obligate intracellular protozoan parasites infecting a wide range of hosts worldwide. However, information on the epidemiology of toxoplasmosis and neosporosis in cats from Portugal is limited. Thus, this study aims to evaluate anti-T. gondii and anti-N. caninum seroprevalence in client-owned cats from Portugal and to identify risk factors using a panel of well-characterized sera. A total of 183 domestic cats were sampled and screened for antibodies against T. gondii and N. caninum using commercial ELISA assays, and their owners answered an online questionnaire designed to obtain background information. The overall anti-T. gondii and anti-N. caninum seroprevalences were 13.1% (CI: 8.97–18.77) and 3.8% (CI: 1.87–7.68), respectively. Univariate analysis revealed that living strictly indoors was a significant protection factor (cOR: 0.053; CI: 0.005–0.627), and the presence of a chronic disease a significant risk factor (cOR: 3.106; CI: 1.062–9.082) to T. gondii seroprevalence. When performing multivariate analysis, only chronic disease (aOR: 57.527; CI: 1.7–1976.7) and seropositivity to N. caninum (aOR: 7.929; CI:0.8–82.9) were found to be a significant risk factor to anti-T. gondii antibodies. To the best of our knowledge, this is the first report of N. caninum seropositivity in cats from Portugal.
International Latin American Survey on Pediatric Intestinal Failure Team
There is little data on the experience of managing pediatric Intestinal Failure (IF) in Latin America. This study aimed to identify and describe the current organization and practices of the IF teams in Latin America and the Caribbean. An online survey was sent to inquire about the existence of IF teams that managed children on home parenteral nutrition (HPN). Our questionnaire was based on a previously published European study with a similar goal. Twenty-four centers with pediatric IF teams in eight countries completed the survey, representing a total number of 316 children on HPN. The median number of children on parenteral nutrition (PN) at home per team was 5.5 (range 1–50). Teams consisted of the following members: pediatric gastroenterologist and a pediatric surgeon in all teams, dietician (95.8%), nurse (91.7%), social worker (79.2%), pharmacist (70.8%), oral therapist (62.5%), psychologist (58.3%), and physiotherapist (45.8%). The majority of the centers followed international standards of care on vascular access, parenteral and enteral nutrition, and IF medical and surgical management, but a significant percentage reported inability to monitor micronutrients, like vitamins A (37.5%), E (41.7%), B1 (66.7%), B2 (62.5%), B6 (62.5%), active B12 (58.3%); and trace elements—including zinc (29.2%), aluminum (75%), copper (37.5%), chromium (58.3%), selenium (58.3%), and manganese (58.3%). Conclusion: There is wide variation in how IF teams are structured in Latin America—while many countries have well-established Intestinal rehabilitation programs, a few do not follow international standards. Many countries did not report having an IF team managing pediatric patients on HPN.
Thermal Performance and Comfort Condition Analysis in a Vernacular Building with a Glazed Balcony
The increase in global environmental problems requires more environmentally efficient construction. Vernacular passive strategies can play an important role in helping reducing energy use and CO2 emissions related to buildings. This paper studies the use of glazed balconies in the North of Portugal as a strategy to capture solar gains and reduce heat losses. The purpose is understanding thermal performance and comfort conditions provided by this passive heating strategy. The methodology includes objective (short and long-term monitoring), to evaluate the different parameters affecting thermal comfort and air quality, and subjective assessments to assess occupants’ perception regarding thermal sensation. The results show that the use of glazed balconies as a passive heating strategy in a climate with cold winters is viable. During the mid-seasons, the rooms with balcony have adequate comfort conditions. In the heating season, it is possible to achieve comfort conditions in sunny days while in the cooling season there is a risk of overheating. Regarding indoor air quality, carbon dioxide concentrations were low, but the average radon concentration measured was high when the building was unoccupied, rapidly decreasing to acceptable values, during occupation periods when a minimum ventilation rate was promoted. Occupants’ actions were essential to improving building behavior.
Pulmonary and Gastrointestinal Parasitic Infections in Small Ruminant Autochthonous Breeds from Centre Region of Portugal—A Cross Sectional Study
The production of small ruminant autochthonous breeds in the Centre region of Portugal is practiced in a semi-extensive husbandry system, exposing animals to parasitic infections. The main objective of this study was to estimate the prevalence of lungworm infection and identify risk factors. Fecal samples of 203 goats and 208 sheep from 30 herds were collected per rectum and subjected to the modified Baermann test. The overall prevalence of infection was 57.7%, significantly higher in goats (95.6%) than in sheep (20.7%) (p < 0.001). According to the binary logistic regression model, sheep dewormed with albendazole, mebendazole plus closantel, or ivermectin plus clorsulon presented a risk of Protostrongylidae infection 29.702, 7.426, or 8.720 times higher, respectively, than those dewormed with eprinomectin. Additionally, the presence of gastrointestinal parasites was investigated in 307 fecal samples using Mini-FLOTAC (R). The overall prevalence of infection was 86.3%, also significantly higher in goats (93.2%) than in sheep (79.9%) (p < 0.001). Strongyle-type eggs were the most frequently identified, both in sheep (69.8%) and goats (87.8%), followed by Eimeria oocysts (40.3% in sheep and 68.9% in goats). Considering the high prevalence and the burden of lungworm parasitic infection, it is urgent to determine its economic impact and the repercussions in animal health in the Centre region of Portugal to establish appropriate therapeutic guidelines.
Usability and User Experience of a Digital Platform Prototype (Healthy Bone) to Promote Pharmacological and Nonpharmacological Treatment in Patients With Osteoporosis: Mixed Methods Study
Osteoporosis-related fractures significantly impact older adults, often leading to disability and even premature death. While pharmacological and nonpharmacological interventions are widely recommended for managing osteoporosis, adherence to these interventions remains low. To address this challenge, we developed the Healthy Bone digital platform (desktop, mobile app, and smart TV internet-based) for use in clinical settings to improve disease management and treatment adherence. It integrates a multimedia health-related behavioral change program with a patient monitoring and management system. This study aimed to evaluate the usability and user experience of the desktop version of the Healthy Bone digital platform prototype from the patients' perspective. The findings will provide valuable insights into optimizing the digital platform and enhancing its functionality. A mixed-methods study was conducted. During usability testing, patients completed tasks simulating real-world use of the platform while using a Think-Aloud approach. After each task, participants filled out an After Scenario Questionnaire to assess task satisfaction. Subsequently, participants completed the System Usability Scale (SUS) and the eHealth Usability Benchmarking Instrument (HUBBI) to measure usability quantitatively. Following this, semistructured interviews were conducted to explore participants' experiences with the platform in greater depth. Descriptive statistics were used for quantitative analysis. Qualitative data analysis involved a combined deductive and inductive approach, ensuring a comprehensive evaluation of the platform's usability and user experience. Deductive content analysis was guided by an ontology of eHealth usability components, while thematic analysis adhered to Braun and Clarke's method to identify emerging themes. Seven participants evaluated the digital platform, reporting high usability with a mean overall SUS score of 87.1 (SD 13.3). Similarly, good usability was reported across all categories of the HUBBI, except for the guidance and support category, which presented moderate levels of usability (mean 3.3, SD 1.1). Patients reported high levels of task satisfaction and identified 24 unique usability issues, predominantly related to the basic system performance, interface design, and navigation and structure categories of the eHealth usability ontology. Overall, patients had positive perceptions and acceptability of the digital platform, highlighting its simplicity, accessibility, utility, and potential to empower those with osteoporosis. Barriers to usage included limited skills, lack of suitable equipment, and time, while facilitators included motivation for behavior change, health benefits, and the decrease of potential inequalities. This study provided valuable insights into the usability and user experience of the desktop version of the Healthy Bone digital platform prototype. These findings will play a key role in optimizing the platform to ensure it is effectively tailored to the needs of the target population. This platform adds an understanding of how various information and communication technology tools can support and benefit large numbers of osteoporosis patients in society.
Direct tissue-sensing reprograms TLR4+ Tfh-like cells inflammatory profile in the joints of rheumatoid arthritis patients
CD4+ T cells mediate rheumatoid arthritis (RA) pathogenesis through both antibody-dependent and independent mechanisms. It remains unclear how synovial microenvironment impinges on CD4+ T cells pathogenic functions. Here, we identified a TLR4+ follicular helper T (Tfh) cell-like population present in the blood and expanded in synovial fluid. TLR4+ T cells possess a two-pronged pathogenic activity whereby direct TLR4+ engagement by endogenous ligands in the arthritic joint reprograms them from an IL-21 response, known to sponsor antibody production towards an IL-17 inflammatory program recognized to fuel tissue damage. Ex vivo, synovial fluid TLR4+ T cells produced IL-17, but not IL-21. Blocking TLR4 signaling with a specific inhibitor impaired IL-17 production in response to synovial fluid recognition. Mechanistically, we unveiled that T-cell HLA-DR regulates their TLR4 expression. TLR4+ T cells appear to uniquely reconcile an ability to promote systemic antibody production with a local synovial driven tissue damage program.In order to identify how the synovial microenvironment impinges on CD4+ T cells pathogenic functions in Rheumatoid Arthritis (RA), Amaral-Silva examined RA patient blood and synovial fluif and identified the presence of a TLR4+ follicular helper T (Tfh) cell-like population. They provided mechanistic insight into how TLR4+ T cells uniquely reconcile an ability to promote systemic antibody production with a local synovial driven-tissue damage program.
Seroprevalence of SARS-CoV-2 in Client-Owned Cats from Portugal
The close contact between humans and domestic cats raises concerns about the potential risks of SARS-CoV-2 transmission. Thus, this study aims to investigate anti-SARS-CoV-2 seroprevalence in client-owned cats from Portugal and evaluate the infection risk of cats that maintain contact with human COVID-19 cases. A total of 176 cats, belonging to 94 households, were sampled. Cat owners answered an online questionnaire, and cats were screened for antibodies against SARS-CoV-2 using a commercial ELISA. Twenty (21.3%) households reported at least one confirmed human COVID-19 case. Forty cats (22.7%) belonged to a COVID-19-positive and 136 (77.3%) to a COVID-19-negative household. The seroprevalences of cats from COVID-19-positive and -negative households were 5.0% (2/40) and 0.7% (1/136). The two SARS-CoV-2-seropositive cats from COVID-19-positive households had an indoor lifestyle, and their owners stated that they maintained a close and frequent contact with them, even after being diagnosed with COVID-19, pointing towards human-to-cat transmission. The SARS-CoV-2-seropositive cat from the COVID-19-negative household had a mixed indoor/outdoor lifestyle and chronic diseases. Owners of the three SARS-CoV-2-seropositive cats did not notice clinical signs or behavior changes. This study highlights the low risk of SARS-CoV-2 transmission from COVID-19-positive human household members to domestic cats, even in a context of close and frequent human–animal contact.
Characteristics associated with poor COVID-19 outcomes in people with psoriasis, psoriatic arthritis and axial spondyloarthritis: data from the COVID-19 PsoProtect and Global Rheumatology Alliance physician-reported registries
ObjectivesTo investigate factors associated with severe COVID-19 in people with psoriasis (PsO), psoriatic arthritis (PsA) and axial spondyloarthritis (axSpA).MethodsDemographic data, clinical characteristics and COVID-19 outcome severity of adults with PsO, PsA and axSpA were obtained from two international physician-reported registries. A three-point ordinal COVID-19 severity scale was defined: no hospitalisation, hospitalisation (and no death) and death. ORs were estimated using multivariable ordinal logistic regression.ResultsOf 5045 cases, 18.3% had PsO, 45.5% PsA and 36.3% axSpA. Most (83.6%) were not hospitalised, 14.6% were hospitalised and 1.8% died. Older age was non-linearly associated with COVID-19 severity. Male sex (OR 1.54, 95% CI 1.30 to 1.83), cardiovascular, respiratory, renal, metabolic and cancer comorbidities (ORs 1.25–2.89), moderate/high disease activity and/or glucocorticoid use (ORs 1.39–2.23, vs remission/low disease activity and no glucocorticoids) were associated with increased odds of severe COVID-19. Later pandemic time periods (ORs 0.42–0.52, vs until 15 June 2020), PsO (OR 0.49, 95% CI 0.37 to 0.65, vs PsA) and baseline exposure to TNFi, IL17i and IL-23i/IL-12+23i (OR 0.57, 95% CI 0.44 to 0.73; OR 0.62, 95% CI 0.45 to 0.87; OR 0.67, 95% CI 0.45 to 0.98; respectively; vs no disease-modifying antirheumatic drug) were associated with reduced odds of severe COVID-19.ConclusionOlder age, male sex, comorbidity burden, higher disease activity and glucocorticoid intake were associated with more severe COVID-19. Later pandemic time periods, PsO and exposure to TNFi, IL17i and IL-23i/IL-12+23i were associated with less severe COVID-19. These findings will enable risk stratification and inform management decisions for patients with PsO, PsA and axSpA during COVID-19 waves or similar future respiratory pandemics.
