Exploring the Mechanism of Indigo Naturalis in the Treatment of Ulcerative Colitis Based on TLR4/MyD88/NF-κB Signaling Pathway and Gut Microbiota

Background: Clinical trials have proven that indigo naturalis is a candidate drug for treating ulcerative colitis (UC), but its therapeutic mechanism is still unclear. Purpose: This study aimed to evaluate the protective effect and mechanism of indigo naturalis to treat mice with dextran sulfate sodium (DSS)-induced UC. Methods: DSS-induced UC mice were treated with indigo naturalis (200 mg/kg), indigo (4.76 mg/kg), and indirubin (0.78 mg/kg) for 1 week. The anti-UC mechanism of indigo naturalis was studied by pathological section, inflammatory factor, western blot, and 16S rRNA sequencing. Results: According to body weight change, disease activity index, and colon length, indigo naturalis had the strongest anti DSS-induced UC effect, followed by indirubin and indigo. Pathological section showed that indigo naturalis, indigo, and indirubin could reduce the infiltration of inflammatory cells, increase the secretion of intestinal mucus, and repair the intestinal mucosa. Indigo naturalis, indigo, and indirubin could reduce IL-1β,IL-6, and TNF-α by inhibiting TLR4/MyD88/NF-κB signal transduction. Indigo naturalis and indigo could also reduce IgA and IgG both in serum and colon tissue. In addition, indigo naturalis, indigo, and indirubin could adjust the gut microbiota structure of DSS-induced UC mice, reducing the ratio of Firmicutes/Bacteroidetes and increasing the abundance of probiotics. Conclusion: Indigo and indirubin are one of the main anti-UC components of indigo naturalis. INN could regulate intestinal flora, reduce inflammation, repair intestinal mucosa, and improve the physiological status of DSS-induced UC mice and its anti-UC mechanism may be involved in inhibiting TLR4/MyD88/NF-κB signal transduction.

Application of MobileNetV2 to waste classification

Today, the topic of waste separation has been raised for a long time, and some waste separation devices have been installed in large communities. However, the vast majority of domestic waste is still not properly sorted and put out, and the disposal of domestic waste still relies mostly on manual classification. The research in this paper applies deep learning to this persistent problem, which has important significance and impact. The domestic waste is classified into four categories: recyclable waste, kitchen waste, hazardous waste and other waste. The garbage classification model trained based on MobileNetV2 deep neural network can classify domestic garbage quickly and accurately, which can save a lot of labor, material and time costs. The absolute accuracy of the trained network model is 82.92%. In comparison with CNN network model, the classification accuracy of MobileNetV2 model is 15.42% higher than that of CNN model. In addition, the trained model is light enough to be better applied to mobile.

A fault diagnosis method for rotating machinery components based on enhanced YOLO v8 and integrated attention mechanism

Accurate fault diagnosis of rotating machinery components is the key to ensuring the safe operation of the mechanical system. Aiming at problems such as inaccurate detection of small target fault features and loss of fault information in the process of advanced feature extraction that exist in conventional machine learning and traditional deep learning methods in the fault diagnosis of rotating machinery, this paper improves the YOLO v8. It proposes a YOLO v8-C-OD fault diagnosis method. Firstly, the original vibration signal is transformed into a time-frequency image with enhanced fault features using continuous wavelet transform (CWT) based on Morlet wavelet as the optimal wavelet basis (WBF) to form an experimental sample. Secondly, the C2F module in YOLO v8 Backbone is improved by introducing Omni-dimensional Dynamic Convolution (ODConv) into C2F to dynamically adjust the weight of each convolution kernel to enhance the fault feature extraction capability. Finally, the Convolutional Block Attention Module (CBAM) is fused into YOLO v8 to suppress unnecessary features and significantly improve the model fault diagnosis rate. Finally, experiments conducted on datasets such as those from Case Western Reserve University (CWRU) and Jiangsu Qianpeng Diagnostic Engineering Co. show that the fault classification diagnosis accuracy of this method reaches 100% and 99.75%, respectively, which outperforms existing state-of-the-art models. This study introduces a novel fault diagnosis method and provides a valuable reference for the fault diagnosis of aircraft and industrial rotating machinery components.

Computational Insights into Cyclodextrin Inclusion Complexes with the Organophosphorus Flame Retardant DOPO

Cyclodextrins (CDs) were used as green char promoters in the formulation of organophosphorus flame retardants (OPFRs) for polymeric materials, and they could reduce the amount of usage of OPFRs and their release into the environment by forming [host:guest] inclusion complexes with them. Here, we report a systematic study on the inclusion complexes of natural CDs (α-, β-, and γ-CD) with a representative OPFR of DOPO using computational methods of molecular docking, molecular dynamics (MD) simulations, and quantum mechanical (QM) calculations. The binding modes and energetics of [host:guest] inclusion complexes were analyzed in details. α-CD was not able to form a complete inclusion complex with DOPO, and the center of mass distance [host:guest] distance amounted to 4–5 Å. β-CD and γ-CD allowed for a deep insertion of DOPO into their hydrophobic cavities, and DOPO was able to frequently change its orientation within the γ-CD cavity. The energy decomposition analysis based on the dispersion-corrected density functional theory (sobEDAw) indicated that electrostatic, orbital, and dispersion contributions favored [host:guest] complexation, while the exchange–repulsion term showed the opposite. This work provides an in-depth understanding of using CD inclusion complexes in OPFRs formulations.

Research Progress on Main Symptoms of Novel Coronavirus Pneumonia Improved by Traditional Chinese Medicine

Novel coronavirus (COVID-19) pneumonia has become a major threat to worldwide public health, having rapidly spread to more than 180 countries and infecting over 1.6 billion people. Fever, cough, and fatigue are the most common initial symptoms of COVID-19, while some patients experience diarrhea rather than fever in the early stage. Many herbal medicine and Chinese patent medicine can significantly improve these symptoms, cure the patients experiencing a mild 22form of the illness, reduce the rate of transition from mild to severe disease, and reduce mortality. Therefore, this paper summarizes the physiopathological mechanisms of fever, cough, fatigue and diarrhea, and introduces Chinese herbal medicines (Ephedrae Herba, Gypsum Fibrosum, Glycyrrhizae Radix et Rhizoma, Asteris Radix et Rhizoma, Ginseng Radix et Rhizoma, Codonopsis Radix, Atractylodis Rhizoma,
) and Chinese patent medicines (Shuang-huang-lian, Ma-xing-gan-shi-tang,
) with their corresponding therapeutic effects. Emphasis was placed on their material basis, mechanism of action, and clinical research. Most of these medicines possess the pharmacological activities of anti-inflammatory, antioxidant, antiviral, and immunity-enhancement, and may be promising medicines for the treatment or adjuvant treatment of COVID-19 patients.
