A news-based climate policy uncertainty index for China

Climate policies can have a significant impact on the economy. However, these policies have often been associated with uncertainty. Quantitative assessment of the socioeconomic impact of climate policy uncertainty is equally or perhaps more important than looking at the policies themselves. Using a deep learning algorithm—the MacBERT model—this study constructed indices of Chinese climate policy uncertainty (CCPU) at the national, provincial and city levels for the first time. The CCPU indices are based on the text mining of news published by a set of major newspapers in China. A clear upward trend was found in the indices, demonstrating increasing policy uncertainties in China in addressing climate change. There is also evidence of clear regional heterogeneity in subnational indices. The CCPU dataset can provide a useful source of information for government actors, academics and investors in understanding the dynamics of climate policies in China. These indices can also be used to investigate the empirical relationship between climate policy uncertainty and other socioeconomic factors in China.

Effects of Chemical Fertilizer Combined with Organic Fertilizer Application on Soil Properties, Citrus Growth Physiology, and Yield

Chemical fertilizer has been excessively used for high yield of citrus around the world, especially in China; meanwhile, it deteriorates the citrus orchard soil environment. To resolve the conflict, the use of organic fertilizer provides a promising solution. However, the data about organic fertilizer used in citrus orchard is rarely available. Here, four treatments including CK (no fertilizer), CF (chemical fertilizer), OF + CF (chemical fertilizer reduction combined with organic fertilizer; application of N, P2O5, K2O fertilizer and organic fertilizer is 0.564, 0.236, 0.336 and 10 kg/plant), and BF + CF (chemical fertilizer reduction combined with bioorganic fertilizer; application of N, P2O5, K2O fertilizer and bioorganic fertilizer is 0.508, 0.320, 0.310 and 10 kg/plant) were performed in a ‘Ponkan’ (Citrus reticulata Blanco) orchard to evaluate the effect of organic fertilizer on citrus yield, growth, soil properties etc. when nutrients of fertilizer of each treatment were equal except CK. The data obtained in 2019 and 2020 showed that both OF + CF and BF + CF were beneficial to improve soil fertility (soil physicochemical and microbe properties) and citrus growth physiology (growth, nutrient and photosynthesis), alleviate NO3−-N leaching, and promote yields. Comprehensive evaluation indicated that BF + CF was more effective than OF + CF. Together, organic fertilizer has the potential to substitute partial chemical fertilizer with improvement in soil properties, growth physiology, and yield of citrus.

Background
Immune checkpoint inhibitors had a great effect in triple-negative breast cancer (TNBC); however, they benefited only a subset of patients, underscoring the need to co-target alternative pathways and select optimal patients. Herein, we investigated patient subpopulations more likely to benefit from immunotherapy and inform more effective combination regimens for TNBC patients.
Methods
We conducted exploratory analyses in the FUSCC cohort to characterize a novel patient selection method and actionable targets for TNBC immunotherapy. We investigated this in vivo and launched a phase 2 trial to assess the clinical value of such criteria and combination regimen. Furthermore, we collected clinicopathological and next-generation sequencing data to illustrate biomarkers for patient outcomes.
Results
CD8-positivity could identify an immunomodulatory subpopulation of TNBCs with higher possibilities to benefit from immunotherapy, and angiogenesis was an actionable target to facilitate checkpoint blockade. We conducted the phase II FUTURE-C-Plus trial to assess the feasibility of combining famitinib (an angiogenesis inhibitor), camrelizumab (a PD-1 monoclonal antibody) and chemotherapy in advanced immunomodulatory TNBC patients. Within 48 enrolled patients, the objective response rate was 81.3% (95% CI, 70.2–92.3), and the median progression-free survival was 13.6 months (95% CI, 8.4–18.8). No treatment-related deaths were reported. Patients with CD8- and/or PD-L1- positive tumors benefit more from this regimen.

Conclusion
This study confirms the efficacy and safety of the triplet regimen in immunomodulatory TNBC and reveals the potential of combining CD8, PD-L1 and somatic mutations to guide clinical decision-making and treatments.

Specificity models in MAPK cascade signaling

The precise execution of various cellular functions relies on the maintenance of signaling specificity from input detection to cellular outputs. However, diverse signaling pathways share similar or identical intermediate components. A well‐conserved intermediate, the Mitogen‐Activated Protein Kinase (MAPK) cascade, participates in a myriad of signaling pathways, regulating signal transduction from input to output. This typifies the “hourglass conundrum”, where a multitude of inputs and outputs all operate through a limited number of common intermediates. Therefore, understanding how MAPK cascades regulate a variety of outputs with specificity is a fundamental question in biology. This review highlights four major insulating mechanisms that improve signaling specificity: selective activation, compartmentalization, combinatorial signaling, and cross‐pathway inhibition. We focus on plant pathways that share MAPK cascade components and compare mechanisms with those of animals and yeast. We hope this conceptual overview will aid future studies to better understand plant signaling specificity. Diverse signaling pathways share similar or identical intermediate components. Efficient synthesis of primary and secondary amides via reacting esters with alkali metal amidoboranes

Amides are one of the most important organic compounds that are widely applied in medicine, biochemistry, and materials science. To find an efficient synthetic method of amides is a challenge for organic chemistry. We report here a facile synthesis method of primary and secondary amides through a direct amidation of esters with sodium amidoboranes (NaNHRBH
3
, R = H, Me), at room temperature without using catalysts and other reagents. This process is rapid and chemoselective, and features quantitative conversion and wide applicability for esters tolerating different functional groups. The experimental and theoretical studies reveal a reaction mechanism with nucleophilic addition followed by a swift proton transfer-induced elimination reaction.
