The LPS-inducible lncRNA Mirt2 is a negative regulator of inflammation

Toll-like receptors (TLRs) are a family of pattern recognition receptors (PRR) with a crucial function in innate immune responses. Activation of TLR4 signaling at the plasma membrane by lipopolysaccharide (LPS) stimulates proinflammatory signaling pathways dependent on the E3 ubiquitin ligase TRAF6. Here we show the LPS-induced long non-coding RNA (lncRNA) Mirt2 functions as a checkpoint to prevent aberrant activation of inflammation, and is a potential regulator of macrophage polarization. Mirt2 associates with, and attenuates Lys63 (K63)-linked ubiquitination of, TRAF6, thus inhibiting activation of NF-κB and MAPK pathways and limiting production of proinflammatory cytokines. Adenovirus mediated gene transfer of Mirt2 protects mice from endotoxemia induced fatality and multi-organ dysfunction. These findings identify lncRNA Mirt2 as a negative feedback regulator of excessive inflammation.
Excessive inflammation can be tissue destructive and contributes to auotinflammatory diseases and sepsis pathology. Here the authors show that the lncRNA Mirt2 is an endogenous negative feedback regulator of LPS-induced inflammation by limiting ubiquitination of TRAF6 and NF-κB activation.

Effects of Aluminum Powder on Ignition Performance of RDX, HMX, and CL-20 Explosives

As a kind of high explosives, aluminized explosive cannot release the energy maximumly, which is a key problem. Using DTA-TG equipment, the ignition performance of three kinds of aluminized explosives (RDX, HMX, and CL-20) with different mass percentages of aluminum powder (0%, 10 wt.%, 20 wt.%, and 30 wt.%) was investigated. The results showed that the energy release of the HMX/Al composite explosive with 10 wt.%, 20 wt.%, and 30 wt.% aluminum powder was only equivalent to 80%, 65%, and 36% of pure HMX, respectively. It was similar to RDX/Al and CL-20/Al composite explosives, except the CL-20/Al mixture with 10% aluminum powder. Rather than participating in the ignition and combustion, the aluminum powder does effect the complete reaction of RDX, HMX, and CL-20 in the initial stage of ignition or in the lower temperature area of the boundary.

The liver coordinates a series of metabolic adaptations to maintain the energy balance of the system and provide adequate nutrients to key organs, tissues and cells during starvation. However, the mediators and underlying molecular mechanisms that mediate these fasting-induced adaptive responses remain unclear.
Male wild-type C57BL/6J littermates (8-weeks-old) were intraperitoneally injected with MCC950 or vehicle, and then randomly divided into three groups: fed, fasted, and refed. Plasma IL1β and insulin levels were detected by ELISA kits. Plasma and hepatic metabolites were determined using commercial assay kits. HepaRG cell line was applied to verify the regulation of NLRP3 on lipogenesis.
NOD-like receptor protein 3 (NLRP3) and its downstream inflammatory cytokines were significantly suppressed after 24 h fasting and recovered upon 6 h refeeding in plasma and liver tissues of mice. Moreover, fasting-induced hepatic steatosis and accompanied liver injury were ameliorated when mice were intraperitoneally injected with MCC950 (a selective NLRP3 inhibitor). Further study revealed that MCC950 suppressed sterol regulatory element-binding protein-1c (SREBP-1c) expression and transcriptional activity, thus inhibited lipogenesis in the liver, which may explain its role in stabilizing lipid metabolism.
The NLRP3 inhibitor-MCC950 protects against fasting-induced hepatic steatosis. The novel and critical role of NLRP3 in lipogenesis may explain its importance in regulating the adaptive responses of the liver upon starvation stress and may provide therapeutic value.

The Effect of Microsized Aluminum Powder on Thermal Decomposition of HNIW (CL-20)

DSC experiments were conducted on the 2,4,6,8,10,12-hexanitro-2,4,6,8,10,12-hexaaza-tetracyclo-[5.5.0.05,9.03,11]-dodecane (also known as HNIW or CL-20) and CL-20 containing 10 wt.% microsized aluminum (Al) powders. The kinetic parameters of CL-20 and CL-20/Al were obtained by ASTM685 and Friedman methods, respectively, indicating that Al powder decreases the activation energy of CL-20 slightly and has a catalytic effect on the thermal decomposition of CL-20. By the method of nonlinear multivariate regression, kinetic models of CL-20 and CL-20/Al were derived as fα=1−αn1+kcat⋅α, where the lgkcat of CL-20 and CL-20/Al is 1.97 and 2.12, respectively, showing that the autocatalytic ability of CL-20 had been increased by adding Al powder. From the SEM images of CL-20/Al and the XRD pattern of the decomposition residues of CL-20/Al, it can be inferred that partial combustion of Al particles happened in the microscale view and led to the release of heat.

Absence of Interferon Regulatory Factor 1 Protects Against Atherosclerosis in Apolipoprotein E-Deficient Mice

Deciphering the molecular and cellular processes involved in foam cell formation is critical to understanding the pathogenesis of atherosclerosis. Interferon regulatory factor 1 (IRF1) was first identified as a transcriptional regulator of type-I interferons (IFNs) and IFN inducible genes. Our study aims to explore the role of IRF1 in atherosclerotic foam cell formation and understand the functional diversity of IRF1 in various cell types contributing to atherosclerosis.

mice and evaluated the effect of IRF1 on disease progression and foam cell formation.
: IRF1 expression was increased in human and mouse atherosclerotic lesions. IRF1 deficiency inhibited modified lipoprotein uptake and promoted cholesterol efflux, along with altered expression of genes implicated in lipid metabolism. Gene expression analysis identified scavenger receptor (SR)-AI as a regulated target of IRF1, and SR-AI silencing completely abrogated the increased uptake of modified lipoprotein induced by IRF1. Our data also explain a mechanism underlying endotoxemia-complicated atherogenesis as follows: two likely pro-inflammatory agents, oxidized low-density lipoprotein (ox-LDL) and bacterial lipopolysaccharide (LPS), exert cooperative effects on foam cell formation, which is partly attributable to a shift of IRF1-Ubc9 complex to IRF1- myeloid differentiation primary response protein 88 (Myd88) complex and subsequent IRF1 nuclear translocation. Additionally, it seems that improved function of vascular smooth muscle cells (VSMCs) also accounts for the diminished and more stable atherosclerotic plaques observed in ApoE

mice.
: Our findings demonstrate an unanticipated role of IRF1 in the regulation of gene expression implicated in foam cell formation and identify IRF1 activation as a new risk factor in the development, progression and instability of atherosclerotic lesions.

Dynamic global tracker for online multi camera multi vehicle tracking

Multi-camera multi-target Tracking (MCMT) is often regarded as a downstream task of Multi-Object Tracking (MOT). Traditional methods typically follow an offline pipeline involving detection, re-identification, single-camera tracking, and post-hoc clustering, which leads to poor real-time performance, high computational cost, and weak adaptability in dynamic environments. Moreover, trackers tailored for specific locations overly rely on manually crafted information like road topology and camera calibration, reducing their effectiveness in varied scenarios. We propose Dynamic Global Tracking (DGT), an innovative online framework for Multi-Camera Multi-Target (MCMT) vehicle tracking. Unlike traditional methods that rely on full trajectory extraction and then clustering, the DGT integrates cross-camera associations directly into the tracking process. This transformation reduces the computational burden and enhances real-time performance. Especially, our framework includes a Hybrid Fusion Module (HFM) to address resolution disparities and a Stable Trajectory Manager (STM) to improve stability and robustness. Extensive experiments demonstrate that DGT significantly improves tracking accuracy and adaptability in various environments, achieving an IDF1 score of 61.19 on the HST dataset (speed version) and 70.49 (performance version) with FPS of 90.

SIRT7 facilitates endometrial cancer progression by regulating PTEN stability in an estrogen-dependent manner

The prognosis of metastatic endometrial carcinoma (EC), one of the most common gynecological malignancies worldwide, remains poor, and the underlying driver of metastases is poorly understood. Dysregulation in estrogen-related signaling and inactivation of tumor suppressor PTEN are two essential risk factors of EC. However, whether and how they are interconnected during EC development remains unclear. Here, we demonstrate that the deacetylase SIRT7 is upregulated in EC patients and mouse models, facilitating EC progression in vitro and in vivo. Mechanistically, in an estrogen-dependent fashion, SIRT7 mediates PTEN deacetylation at K260, promoting PTEN ubiquitination by the E3 ligase NEDD4L, accelerating PTEN degradation and, consequently, expediting EC metastasis. Additionally, SIRT7 expression strongly correlates with poor survival in EC patients with wild-type PTEN, though no significant correlation is observed in PTEN mutation patients. These results lay the foundation for the study of targeting estrogen-SIRT7-PTEN axis, to restore PTEN abundance, offering potential avenues for EC therapy.

Farrerol directly activates the deubiqutinase UCHL3 to promote DNA repair and reprogramming when mediated by somatic cell nuclear transfer

Farrerol, a natural flavanone, promotes homologous recombination (HR) repair to improve genome-editing efficiency, but the specific protein that farrerol directly targets to regulate HR repair and the underlying molecular mechanisms have not been determined. Here, we find that the deubiquitinase UCHL3 is the direct target of farrerol. Mechanistically, farrerol enhanced the deubiquitinase activity of UCHL3 to promote RAD51 deubiquitination, thereby improving HR repair. Importantly, we find that embryos of somatic cell nuclear transfer (SCNT) exhibited defective HR repair, increased genomic instability and aneuploidy, and that the farrerol treatment post nuclear transfer enhances HR repair, restores transcriptional and epigenetic network, and promotes SCNT embryo development. Ablating UCHL3 significantly attenuates farrerol-mediated stimulation in HR and SCNT embryo development. In summary, we identify farrerol as an activator of the deubiquitinase UCHL3, highlighted the importance of HR and epigenetic changes in SCNT reprogramming and provide a feasible method to promote SCNT efficiency.
Here the authors find that farrerol directly binds to UCHL3 and activates its deubiquitinase activity, and that farrerol promotes development of SCNT embryos by enhancing HR repair and restoring epigenetic networks in a UCHL3-dependent manner.

In this paper, a broadband reflective multi-polarization converter based on single-layer double-L-shaped metasurface is proposed. The proposed metasurface can effectively convert linear-polarized (TE/TM) incident wave into the reflected wave with three different polarizations within the frequency bands of 5.5–22.75 GHz. Based on the electric and magnetic resonant features of the double-L-shaped structure, the proposed metasurface can convert linearly polarized waves into cross-polarized waves at three resonant frequency bands. Furthermore, the incident linearly polarized waves can be effectively converted into left/right handed circular-polarized (LHCP and RHCP) waves at other four non-resonance frequency bands. Thus, the proposed metasurface can be regarded as a seven-band multi-polarization converter. The prototype of the proposed polarization converter is analyzed and measured. Both simulated and measured results show the 3-dB axis ratio bandwidth of circular polarization bands and the high polarization conversion efficiency of cross-polarization bands when the incident wave changes from 0° to 30° at both TE and TM modes.

A two-step deep learning method for 3DCT-2DUS kidney registration during breathing

This work proposed KidneyRegNet, a novel deep registration pipeline for 3D CT and 2D U/S kidney scans of free breathing, which comprises a feature network, and a 3D–2D CNN-based registration network. The feature network has handcrafted texture feature layers to reduce the semantic gap. The registration network is an encoder-decoder structure with loss of feature-image-motion (FIM), which enables hierarchical regression at decoder layers and avoids multiple network concatenation. It was first pretrained with a retrospective dataset cum training data generation strategy and then adapted to specific patient data under unsupervised one-cycle transfer learning in onsite applications. The experiment was performed on 132 U/S sequences, 39 multiple-phase CT and 210 public single-phase CT images, and 25 pairs of CT and U/S sequences. This resulted in a mean contour distance (MCD) of 0.94 mm between kidneys on CT and U/S images and MCD of 1.15 mm on CT and reference CT images. Datasets with small transformations resulted in MCDs of 0.82 and 1.02 mm, respectively. Large transformations resulted in MCDs of 1.10 and 1.28 mm, respectively. This work addressed difficulties in 3DCT-2DUS kidney registration during free breathing via novel network structures and training strategies.
