Acetylation of parental histone H4 segregated to newly replicated DNA and the acetylation of nascent H3 in HeLa cells
This study examined the sites of acetylation of parental H4 segregated to newly replicated DNA, through a series of immunoprecipitations of replicative chromatin with antibodies against H4 acetylated on specific lysines. Experiments established that while acetylation of parental H4 is less than that of nascent H4, parental H4 is preferentially acetylated on lysines 5 and 12. Prior to the studies in this report, there had been no evidence for deposition-related acetylation of nascent H3 in humans. Using an acetyllysine antibody, immunoblots of a cytosolic extract showed that variants H3.2 and H3.1 were acetylated while H3.1 was not. Cytosolic H3 labeled with [3H]lysine, HPLC separated, and analyzed through fluorography indicated that H3.2/H3.3 were acetylated. Microsequencing of nuclear H3 showed low level acetylation occurring.
