A Comparison of the Acute Effects of Different Forms of Yoga on Physiological and Psychological Stress: A Pilot Study

Yoga is a frequently recommended stress management strategy; however, the acute stress response to varying types of yoga are not fully clear. Thus, the purpose of this study was to compare the acute effects of meditative and power yoga on indices of physiological and psychological stress. In a crossover counterbalanced design, physically active females (n = 13; age = 20.8 yrs ± 0.8, height = 164.5 cm ± 6.1, body mass = 65.0 kg ± 13.8) who did not regularly participate in yoga or mindful training enrolled in this study. Participants completed two visits each, with a standardized instructional-video 30-min yoga session with either A) meditative (Hatha style) yoga or B) power (Vinyasa style) yoga. Prior to and immediately after each yoga bout, psychological stress was assessed using the State–Trait Anxiety Inventory (STAI) questionnaire, and salivary cortisol samples were obtained to measure indices of physiological stress. State anxiety scores were significantly lower following meditative yoga (p = 0.047) but were not different following power yoga (p = 0.625). Salivary cortisol levels were significantly lower following meditative yoga (p = 0.020) but not following power yoga (p = 0.242). Results indicate that acute engagement in meditative yoga decreases markers of psychological and physiological stress, while power yoga does not impart a significant stress-relieving benefit. Findings indicate that differing types of yoga may have various stress-relieving capabilities and should be considered by individuals seeking anxiolytic benefits.

EphA3-targeted chimeric antigen receptor T cells are effective in glioma and generate curative memory T cell responses

BackgroundHigh-grade gliomas including glioblastoma (GBM) and diffuse midline gliomas (DMG) represent the most lethal and aggressive brain cancers where current treatment modalities offer limited efficacy. Chimeric antigen receptor (CAR) T cell therapies have emerged as a promising strategy, boasting tumor-specific targeting and the unique ability to penetrate the blood-brain barrier. However, the effective clinical application hinges on the optimal choice of antigen, with a limited number, currently under investigation.MethodsWe employed cell surface proteomic analysis of primary human high-grade glioma samples from both adult and pediatric patients. This led to the identification of Ephrin type-A receptor 3 (EphA3) as a prevalently expressed target. We engineered a second-generation EphA3-targeted CAR T cell and assessed function using in vitro and in vivo models of GBM and DMG.ResultsEphA3-targeted CAR T cells demonstrated robust antigen-specific killing of human GBM and DMG cell lines in vitro. In an orthotopic xenograft NSG mouse model, EphA3-targeted CAR T cells not only effectively eradicated tumors but also established a functional T cell population protective on rechallenge. Remarkably, mice rechallenged with a second contralateral orthotopic tumor implantation achieved complete tumor clearance and maintained a sustained complete response 6 months following initial treatment.ConclusionBuilding on the proven safety profile of EphA3 antibodies in clinical settings, our study provides compelling preclinical evidence supporting the efficacy of EphA3-targeted CAR T cells against high-grade gliomas. These findings underscore the potential for transitioning this innovative therapy into clinical trials, aiming to revolutionize the treatment landscape for patients afflicted with these formidable brain cancers.

Inter-annual variation in American redstart (Setophaga ruticilla) plumage colour is associated with rainfall and temperature during moult: an 11-year study

Carotenoid-based colouration plays an important role in sexual signaling in animals as an honest indicator of individual quality during mate choice and competitive interactions. However, few studies have examined how natural variation in weather conditions influences interannual variation in the expression of ornamentation, potentially through affecting the dietary availability of carotenoids. In this study, we examine variation in the expression of carotenoid-based plumage colouration in relation to temperature and rainfall during the pre-moulting and moulting period over 11 years in a population of American redstarts, Setophaga ruticilla, breeding in eastern Canada. We used reflectance spectrometry of tail feathers collected from male and female redstarts to relate feather colour with weather conditions the previous breeding season during the months over which redstarts are likely to moult (June-September). At a population level, birds expressed feathers with higher red chroma and lower brightness in years following high July rainfall and low August temperature. The pattern was stronger in males, but was generally consistent across ages and sexes. Analyses of feathers from repeatedly captured birds indicated that the above patterns could be explained by individual change in feather colour. We suggest that higher rainfall during the moulting period may increase insect abundance and the availability of dietary carotenoids. This is among the first studies to show effects of weather conditions on a sexual signalling trait, which may have important consequences for sexual selection, mate choice, and the reliability of putative signals.

“Everything had stopped, no meeting, no gathering”: Social interactions during the COVID-19 pandemic in the Central African Republic, the Democratic Republic of Congo, and Bangladesh

Understanding the spread of COVID-19 in humanitarian and fragile settings is challenging for many reasons, including the lack of data on social dynamics and preventive behaviors during an epidemic. We investigate social interactions in three such settings - Democratic Republic of the Congo (DRC), Central African Republic (CAR), Cox’s Bazar (CXB), Bangladesh – and how they changed during the first year of the pandemic. This comparative mixed-methods study uses a representative household survey and focus group discussions or key informant interviews in each site. Descriptive weighted analysis of survey responses was conducted; multivariate logistic regression identified factors associated with changes in social interactions. Thematic analysis was conducted on qualitative data. Nearly all participants had social interactions the day before the survey, although the average number of daily interactions was low. Interactions primarily occurred indoors, at home and without masks. We saw a discrepancy between knowledge about and practice of preventive behaviors. Most respondents reported interacting less often (77.3% CXB, 86.7% CAR, 58.8% DRC) and having shorter meetings (80% CXB, 77.8% CAR, and 47.8% DRC). Reluctance towards the COVID-19 vaccine was a risk factor for non-compliant behaviors in CAR (OR increased frequency = 3.51, 95%CI = 1.41–8.75; OR increased duration = 2.47, 95%CI = 1.15–5.29) and DRC (OR increased duration = 3.06, 95%CI = 1.71–5.49), likely pointing to distrust towards institutional policies. Respondents from IDP communities in DRC were less likely to reduce the frequency of interaction, likely because living conditions did not facilitate physical distancing. Increased knowledge in CXB was associated with compliant behavior (for 1pt-increase: OR increased frequency = 0.47, 95%CI = 0.32–0.68; OR increased duration = 0.46, 95%CI = 0.31–0.69). Understanding social dynamics is fundamental to predict infectious disease spread, particularly in humanitarian settings. More evidence is needed to understand behaviors influencing disease dynamics and drivers of behaviors, including trust in authorities, social, and economic factors. Peace, community engagement, and reduction of misinformation remain critical for epidemic responses in humanitarian settings.
