Interactions Between the Gut Microbiota and the Host Innate Immune Response Against Pathogens
The mammalian intestine is colonized by over a trillion microbes that comprise the \"gut microbiota,\" a microbial community which has co-evolved with the host to form a mutually beneficial relationship. Accumulating evidence indicates that the gut microbiota participates in immune system maturation and also plays a central role in host defense against pathogens. Here we review some of the mechanisms employed by the gut microbiota to boost the innate immune response against pathogens present on epithelial mucosal surfaces. Antimicrobial peptide secretion, inflammasome activation and induction of host IL-22, IL-17, and IL-10 production are the most commonly observed strategies employed by the gut microbiota for host anti-pathogen defense. Taken together, the body of evidence suggests that the host gut microbiota can elicit innate immunity against pathogens.
Network of job demands-resources and depressive symptoms in critical care nurses: a nationwide cross-sectional study
Background Critical care nurses are vulnerable to depression, which not only lead to poor well-being and increased turnover intention, but also affect their working performances and organizational productivity as well. Work related factors are important drivers of depressive symptoms. However, the non-liner and multi-directional relationships between job demands-resources and depressive symptoms in critical care nurses has not been adequately analyzed. Understanding these relationships would be helpful for reducing depression, increasing nurses’ well-being and retain healthcare forces. Methods This was a cross-sectional study using baseline data from the Nurses’ mental health study (NMHS), a prospective cohort study on nurses from 67 tertiary hospitals in 31 province-level administrative regions in China. Only clinical nurses working in the ICU were included ( n  = 13,745). Data were collected using online questionnaires, and analyzed using network analysis and structural equation model. Job demands (average working hours per week, average number of night shifts per month, paperwork burden and work-life balance), job resources (supervisor support, co-worker support, leader justice, organizational climate satisfaction, work meaning, and career prospect), personal resource (resilience) and depressive symptoms were main variables in the networks, while demographic data and social health (social-emotional support and loneliness) were covariates. Results The prevalence of severe, moderately severe, moderate, mild, and none or minimum depressive symptoms in critical care nurses of this study were 1.21, 3.42, 9.76, 42.88, and 42.07% respectively. In the final network, 132 of 210 possible edges (62.8%) were not zero. “Fatigue” had the highest expected influence, followed by “Motor”, and “Appetite”. Meanwhile, in terms of job demands-resources and personal resources, the node with the highest expected influence was “Supervisor support”, followed by “Work meaning” and “Co-worker support”. Three bridge variables were identified: “Resilience-adaptation”, “Average working hours per week”, and “Co-worker support”. The final structural equation model basically supported the results of network analysis with an acceptable model-fit (GFI = 0.918, AGFI = 0.896, PCFI = 0.789, PNFI = 0.788, NFI = 0.909, IFI = 0.911, CFI = 0.911, SRMR = 0.040, and RMSEA = 0.064). Conclusions There was a rather strong interconnectedness between depressive symptoms and job demands-resources. Fatigue, motor, and appetite were core depressive symptoms of critical care nurses. Close attention to those symptoms could help recognize depression in critical care nurses. Supervisor support, work meaning, and co-worker support played vital roles as job resources in reducing depression, while negative impact of long average working hours per week were more contagious. Resilience, as personal resources, could help mediate the associations between job demands-resources and depression. In clinical practice, it’s recommended for nursing managers to (1) encourage critical care nurses to find their “meaning in work”, (2) implement resilience enhancing programs for nurse, (3) build and maintain meaningful relationships with nurses and support them in daily work, and (4) create a harmonious and dedicated working environment where co-workers are willing to help and support each other. Improvements in those modifiable aspects could help reduce risk and prevent exacerbations of depressive symptoms in critical care nurses.
Comparative Analysis of Volatile Constituents in Root Tuber and Rhizome of Curcuma longa L. Using Fingerprints and Chemometrics Approaches on Gas Chromatography–Mass Spectrometry
The root tuber and rhizome of Curcuma longa L., abbreviated, respectively, as RCL and RHCL, are used as different medicines in China. In this work, volatile oils were extracted from RCL and RHCL. Then, gas chromatography–mass spectrometry (GC–MS) was used for RCL and RHCL volatile oils analysis, and 45 compounds were identified. The dominant constituents both in volatile oils of RCL and RHCL were turmerone, (−)-zingiberene, and β-turmerone, which covered more than 60% of the total area. The chromatographic fingerprint similarities between RCL and RHCL were not less than 0.943, indicating that their main chemical compositions were similar. However, there were also some compounds that were varied in RCL and RHCL. Based on the peak area ratio of 45 compounds, the RCL and RHCL samples were separated into principal component analysis (PCA) and partial least squares discriminant analysis (PLS-DA). Then, 20 compounds with a variable importance for the projection (VIP) value of more than 1 were the high potential contributors for RCL and RHCL differences. Furthermore, ferric ion-reducing antioxidant power (FRAP) assay results demonstrated that the volatile oils of RCL and RHCL had antioxidant activities. This study provided the material basis for the research of volatile components in RCL and RHCL and contributed to their further pharmacological research and quality control.
Comparison of self-reported and biomedical data on hypertension and diabetes: findings from the China Health and Retirement Longitudinal Study (CHARLS)
ObjectivesWe examined the level of agreement between biomedical and self-reported measurements of hypertension and diabetes in a Chinese national community sample, and explored associations of the agreement and possible contextual effects among provinces and geographic regions in China.DesignSecondary analysis of a cohort sample.Setting and participantsCommunity samples were drawn from the national baseline survey of the China Health and Retirement Longitudinal Study (CHARLS, 2011–2012) through multistage probability sampling, which included households with members 45 years of age or above with a total sample size of 17 708 individuals.Outcome measuresSensitivity, specificity and κ were used as measurements of agreements or validity; variance of validity measures among provinces and communities was estimated using random-effects models.ResultsSelf-reports for hypertension and diabetes showed high specificity (96.3% and 98.3%, respectively) but low sensitivity (56.3% and 61.5%, respectively). Agreement between self-reported data and biomedical measurements was moderate for both hypertension (κ 0.57) and diabetes (κ 0.65), with respondents who were older, of higher socioeconomic status, better educated and who had hospital admissions in the past 12 months showing stronger agreements than their counterparts. Large and significant variations in the sensitivity among provinces for hypertension, and among communities for both hypertension and diabetes, could neither be attributed to the effects of respondents’ characteristics nor to the contextual effects of city–village differences.ConclusionsAs a considerable number of people in the overall sample were unaware of their conditions, self-reports will lead to an underestimation of the prevalence of hypertension and diabetes. However, in more developed communities or provinces, self-reported data can be a reliable estimate of the prevalence of the two conditions. Further investigations of contextual effects at provincial and community levels could highlight public health strategies to improve awareness of the two conditions.
Design and optimization of a bird’s nest foreign object removal robot
To enhance the automation level of foreign object removal in bird’s nest processing and reduce the costs and misjudgment rates associated with manual inspection, this paper proposes and develops a parallel system for automatic foreign object removal in bird’s nest based on machine vision. A two-stage foreign object detection algorithm is proposed: the first stage introduces an attention mechanism to improve the U-Net++ model, enhancing segmentation accuracy under texture interference; the second stage combines binary mask processing to achieve precise classification of foreign object and non-foreign object regions. Experimental results show that the algorithm achieves an F1 score of 94.80% and a recall rate of 97.90%, outperforming traditional methods in overall performance. An evaluation system for bird’s nest foreign object removal is established, incorporating criteria such as cleanliness, loss rate, structural integrity, and removal time. A set of automated equipment with identification, localization, and removal functions is designed and implemented. The algorithm and equipment were deeply integrated, and through prototype construction and performance testing, the system’s ability to accurately identify and stably remove various types of foreign objects under complex operating conditions was validated. This study provides a feasible intelligent solution for the bird’s nest impurity removal field, with good application prospects and industrial promotion value.
Dipeptidyl peptidase 4 inhibitor sitagliptin protected against dextran sulfate sodium-induced experimental colitis by potentiating the action of GLP-2
Dipeptidyl peptidase 4 (DPP4), a ubiquitously expressed protease that cleaves off the N-terminal dipeptide from proline and alanine on the penultimate position, has important roles in many physiological processes. In the present study, experimental colitis was induced in mice receiving 3% dextran sulfate sodium (DSS) in drinking water. We found that mice with DSS-induced colitis had significantly increased intestinal DPP activity and decreased serum DPP activity, suggesting a probable correlation of DPP4 with experimental colitis. Then, we investigated whether sitagliptin, a specific DPP4 inhibitor could protect against DSS-induced colitis. We showed that oral administration of single dose of sitagliptin (30 mg/kg) on D7 remarkably inhibited DPP enzyme activity in both serum and intestine of DSS-induced colitic mice. Repeated administration of sitagliptin (10, 30 mg/kg, bid, from D0 to D8) significantly ameliorated DSS-induced colitis, including reduction of disease activity index (DAI) and body weight loss, improvement of histological score and colon length. Sitagliptin administration dose-dependently increased plasma concentrations of active form of GLP-1 and colonic expression of GLP-2R. Co-administration of GLP-2R antagonist GLP-2 3-33 (500 μg/kg, bid, sc) abolished the protective effects of sitagliptin in DSS-induced colitic mice. Moreover, sitagliptin administration significantly decreased the ratio of apoptotic cells and increased the ratio of proliferative cells in colon epithelium of DSS-induced colitic mice, and this effect was also blocked by GLP-2 3-33 . Taken together, our results demonstrate that sitagliptin could attenuate DSS-induced experimental colitis and the effects can be attributed to the enhancement of GLP-2 action and the subsequent protective effects on intestinal barrier by inhibiting epithelial cells apoptosis and promoting their proliferation. These findings suggest sitagliptin as a novel therapeutic approach for the treatment of ulcerative colitis.
A meta-analysis of unilateral axillary approach for robotic surgery compared with open surgery for differentiated thyroid carcinoma
The Da Vinci Robot is the most advanced micro-control system in endoscopic surgical instruments and has gained a lot of valuable experience today. However, the technical feasibility and oncological safety of the robot over open surgery are still uncertain. This work is to systematically evaluate the efficacy of the unilateral axillary approach for robotic surgery compared to open surgery for differentiated thyroid carcinoma. PubMed, Embase, Cochrane Library, and Web of Science databases were utilized to search for relevant literatures of robotic thyroid surgery using unilateral axillary approach compared to open thyroid surgery, and a meta-analysis was performed using RevMan software version 5.3. Statistical analysis was performed through Mantle-Haenszel and inverse variance methods. Twelve studies with a total of 2660 patients were included in the meta-analysis. The results showed that compared with the open group, the robotic group had a longer total thyroidectomy time, shorter hospital stay, less intraoperative bleeding, more postoperative drainage, fewer retrieved central lymph nodes, and higher cosmetic satisfaction (all P < 0.05). In contrast, temporary and permanent laryngeal recurrent nerve injury, temporary and permanent hypoparathyroidism or hypocalcemia, brachial plexus nerve injury, number of retrieved central lymph nodes, number of retrieved lymph nodes in the lateral cervical region, number of lymph node metastases in the lateral cervical region, hematoma, seroma, lymphatic leak, stimulated thyroglobulin (sTg) and unstimulated thyroglobulin (uTg), and the number and recurrence rate of patients with sTg <1ng/ml were not statistically different between the two groups (P > 0.05). The unilateral axillary approach for robotic thyroid surgery may achieve outcomes similar to those of open surgery. Further validation is required in a prospective randomized controlled trial.
Elemental Mapping Reveals Selective Dolomitization in Tonian Stromatolites: Implications for Early Diagenesis and Paleoenvironmental Proxies
Stromatolites archive critical information on Precambrian marine environments, but their geochemical signals are often obscured using complex diagenetic processes. Tonian stromatolites from the Weiji Formation, North China, show selective dolomitization in dark stromatolitic laminae, forming zoned dolomites. The zoned dolomite crystals with well‐preserved growth zoning record the evolution of diagenetic fluid, yet their genesis remains controversial. To constrain selective dolomitization processes and zoned dolomite formation in stromatolites, and further assess their reliability as paleoceanographic proxies, we use LA‐ICP‐TOF‐MS elemental mapping to reveal crystallization mechanisms and early diagenetic controls in stromatolitic carbonates. Results show distinct geochemical distribution patterns between dark and light stromatolitic laminae, reflecting complex microbial‐induced diagenetic reactions during early diagenesis. Fe is preferentially concentrated in dark stromatolitic layers, while Mn anomalously accumulates in light layers (with Mn/Fe ratio up to ∼2). Meanwhile, high terrigenous‐indicative elemental contents (Al‐Si‐K) are observed in dark laminae and the adjacent clay‐rich matrix. The anomalous Fe‐Mn distribution is attributed to the redox oscillations and terrigenous pulses (ROTP) model. Selective dolomitization proceeds through an Ion‐Exchange Motor mechanism during penecontemporaneous to early diagenesis, where Mg2+ derived from seawater undergoes dehydration via clay mineral adsorption and/or microbial mediation. Within ferroan zoned dolomite, elemental zoning exhibits alternating Mn‐enriched bands and Fe‐rich zones, indicating redox‐controlled diagenesis with preferential Mn(IV) reduction prior to Fe(II) mobilization. While Precambrian stromatolites remain valuable proxies for paleo‐ocean chemistry, our results emphasize the critical need for in situ analytical approaches to distinguish primary signals from diagenetic overprints in Precambrian carbonate systems.
Active methanogenesis during the melting of Marinoan snowball Earth
Geological evidence indicates that the deglaciation of Marinoan snowball Earth ice age (~635 Myr ago) was associated with intense continental weathering, recovery of primary productivity, transient marine euxinia, and potentially extensive CH 4 emission. It is proposed that the deglacial CH 4 emissions may have provided positive feedbacks for ice melting and global warming. However, the origin of CH 4 remains unclear. Here we report Ni isotopes (δ 60 Ni) and Yttrium-rare earth element (YREE) compositions of syndepositional pyrites from the upper most Nantuo Formation (equivalent deposits of the Marinoan glaciation), South China. The Nantuo pyrite displays anti-correlations between Ni concentration and δ 60 Ni, and between Ni concentration and Sm/Yb ratio, suggesting mixing between Ni in seawater and Ni from methanogens. Our study indicates active methanogenesis during the termination of Marinoan snowball Earth. This suggests that methanogenesis was fueled by methyl sulfides produced in sulfidic seawater during the deglacial recovery of marine primary productivity. The deglaciation of Marinoan snowball Earth (~635 Myr ago) has been associated with potentially extensive CH 4 emissions in relation to transient marine euxinia. Here, the authors find that active methanogenesis occurred during the termination of Marinoan snowball Earth, fueled by methyl sulfide production in sulfidic seawater.
