A hypomyelinating leukodystrophy in German Shepherd dogs
Abstract Background Shaking puppy syndrome is commonly attributed to abnormal myelination of the central nervous system. Hypothesis/Objectives To report the long-term clinical course and the imaging characteristics of hypomyelinating leukodystrophy in German Shepherd dogs. Animals and Methods Three related litters with 11 affected dogs. Results The 11 affected dogs experienced coarse, side-to-side tremors of the head and trunk, which interfered with normal goal-oriented movements and disappeared at rest. Signs were noticed shortly after birth. Nine dogs were euthanized, 3 dogs underwent pathological examination, and 2 littermates were raised by their breeder. Tremors improved gradually until 6 to 7 months of age. Adult dogs walked with severe residual pelvic limb ataxia. One dog developed epilepsy with tonic-clonic seizures at 15 months of age. Conventional magnetic resonance imaging (MRI) disclosed homogenous hyperintense signal of the entire subcortical white matter in 3 affected 7-week-old dogs and a hypointense signal in a presumably unaffected littermate. Subcortical white matter appeared isointense to gray matter at 15 and 27 weeks of age on repeated MRI. Abnormal white matter signal with failure to display normal gray-white matter contrast persisted into adulthood. Cerebellar arbor vitae was not visible at any time point. Clinical signs, MRI findings, and pathological examinations were indicative of a hypomyelinating leukodystrophy. All parents of the affected litters shared a common ancestor and relatedness of the puppies suggested an autosomal recessive mode of inheritance. Conclusion We describe a novel hypomyelinating leukodystrophy in German Shepherd dogs with a suspected inherited origin.
The cat as a small dog?—Comparison of trabecular and cortical bone microarchitecture of radius and ulna in cats and small dogs using microcomputed tomography
The forearms of dogs and cats do not only differ anatomically from each other, but there are also differences in prevalence of radius and ulna fractures between the two species. The prevalence of antebrachial fractures is 18.0% in dogs and 2.0–8.0% in cats. Many studies focus solely on the trabecular and cortical bone structure of dogs and the characteristics of the cat are often disregarded. The aim of this study was to evaluate the trabecular structure parameters [bone volume fraction per total volume (BV/TV), bone surface per total volume (BS/BV), trabecular number (Tb.N), trabecular thickness (Tb.Th), trabecular separation (Tb.Sp), connectivity density (Conn. D), degree of anisotropy (DA)] and the diaphyseal cortical bone density (Mean Density) of the antebrachium in cats and small dogs to visualise their differences. For this purpose, a total of 32 forearms of cats (n = 8) and small dogs (n = 8) were evaluated using microcomputed tomography and the findings were compared. The results of the study showed that cats had higher values for BV/TV, Tb.Th, Tb.Sp, DA and Mean Density and lower values for BS/BV, Tb.N and Conn.D at radius and ulna compared to dogs. According to the results of this study, the higher bone volume fraction (BV/TV), thicker trabeculae (Tb.Th), increased anisotropy (DA) and significantly higher diaphyseal cortical density (Mean Density) could contribute to the lower fracture risk of the antebrachium in cats compared to small dogs. In the present study trabecular and cortical bone structure of the antebrachium of cats and small dogs were investigated and the differences had been visualized. Comparative studies of feline and canine structural bone architecture are not found at all in the current literature. Furthermore the aim of this study was to detect reasons for the decreased antebrachial fracture incidence of cats comparing to dogs .
Studies on the association of malondialdehyde as a biomarker for oxidative stress and degree of malignancy in dogs with mammary adenocarcinomas
Background Mammary adenocarcinomas are one of the most common tumour diseases in bitches. The relationship between oxidative stress and the degree of malignancy of the tumour has not been sufficiently researched in veterinary medicine. Objectives The main objective was to investigate the potential role of MDA as a practice‐relevant biomarker for the assessment of systemic oxidative stress and to determine whether this parameter can indicate the malignancy grade of a mammary adenocarcinoma. Methods In the present pilot study, MDA plasma concentrations were analysed in 55 bitches with (n = 28) and without (n027) malignant adenocarcinomas of the mammary gland using two different measurement methods and the relationship to tumour size was investigated. Results The mean MDA concentration measured by enzyme‐linked immunosorbent assay (ELISA) was 289 ng/mL (range 365–634 ng/mL) in dogs with grade 1 adenocarcinoma (n = 13), 288.5 ng/mL (range 85–752 ng/mL) in dogs with grade 2 adenocarcinoma (n = 10), 332 ng/mL (range 239–947 ng/mL) in dogs with grade 3 (n = 5) adenocarcinoma and 293 ng/mL (range 175–549 ng/mL) in dogs without a mammary tumour (n = 27). When MDA was measured by HPLC, the average MDA concentration in the study group (n = 11) was 0.24 µmol/L (range 0.16–0.37) and that of the control group (n = 15) was 0.27 µmol/L (range 0.16–1.62). Thus, there were no significant differences between the study group with malignant adenocarcinomas and the control group in both examination methods (p > 0.05). Furthermore, there was no correlation between the MDA concentrations and the approximate volume of the mammary tumour. Conclusion The results highlight the challenges of providing a prognosis for the malignancy of a mammary adenocarcinoma based on MDA concentrations in plasma using ELISA or HPLC. As a result, histopathological examination remains the gold standard for diagnosing and differentiating adenocarcinomas of the mammary gland. In the present pilot study, malondialdehyde (MDA), a parameter for oxidative stress, was measured in plasma of bitches with and without malignant mammary adenocarcinomas. The results showed no correlation between MDA concentrations in the blood and malignancy or approximative volume of the mammary adenocarcinomas. As a result, histopathological examination remains the gold standard for diagnosing and differentiating adenocarcinomas of the mammary gland.
Performance and overview of clinically relevant areas of application of saliva testing in the cat
The cat represents an important model in order to investigate basic physiological knowledge of salivary secretion as well as pharmacokinetics of active substances. The aim of the study was to review in which diagnostic application areas saliva testing is routinely used and in which areas it could be further explored in the future. Literature relevant to the research question was collected in March 2022 using the Pubmed database. The diagnosis of infectious diseases in cat saliva is one of the most important fields of application. Saliva diagnostics may also indicate dental diseases, allergies or kidney and other metabolic diseases. Sexual and stress hormones can also be measured in cat saliva. A number of clinically relevant allergens in cat saliva that may cause allergies in humans has been investigated and described, in addition to infectious agents that can be transmitted from cats to humans. Saliva testing in cats can be useful in many areas, including the detection of infectious diseases, allergies and dental disease. However, it is far from being used to its full potential within veterinary medicine.
Evaluation of the soft tissue biocompatibility of MgCa0.8 and surgical steel 316L in vivo: a comparative study in rabbits
Background Recent studies have shown the potential suitability of magnesium alloys as biodegradable implants. The aim of the present study was to compare the soft tissue biocompatibility of MgCa0.8 and commonly used surgical steel in vivo . Methods A biodegradable magnesium calcium alloy (MgCa0.8) and surgical steel (S316L), as a control, were investigated. Screws of identical geometrical conformation were implanted into the tibiae of 40 rabbits for a postoperative follow up of two, four, six and eight weeks. The tibialis cranialis muscle was in direct vicinity of the screw head and thus embedded in paraffin and histologically and immunohistochemically assessed. Haematoxylin and eosin staining was performed to identify macrophages, giant cells and heterophil granulocytes as well as the extent of tissue fibrosis and necrosis. Mouse anti-CD79α and rat anti-CD3 monoclonal primary antibodies were used for B- and T-lymphocyte detection. Evaluation of all sections was performed by applying a semi-quantitative score. Results Clinically, both implant materials were tolerated well. Histology revealed that a layer of fibrous tissue had formed between implant and overlying muscle in MgCa0.8 and S316L, which was demarcated by a layer of synoviocyte-like cells at its interface to the implant. In MgCa0.8 implants cavities were detected within the fibrous tissue, which were surrounded by the same kind of cell type. The thickness of the fibrous layer and the amount of tissue necrosis and cellular infiltrations gradually decreased in S316L. In contrast, a decrease could only be noted in the first weeks of implantation in MgCa0.8, whereas parameters were increasing again at the end of the observation period. B-lymphocytes were found more often in MgCa0.8 indicating humoral immunity and the presence of soluble antigens. Conversely, S316L displayed a higher quantity of T-lymphocytes. Conclusions Moderate inflammation was detected in both implant materials and resolved to a minimum during the first weeks indicating comparable biocompatibility for MgCa0.8 and S316L. Thus, the application of MgCa0.8 as biodegradable implant material seems conceivable. Since the inflammatory parameters were re-increasing at the end of the observation period in MgCa0.8 it is important to observe the development of inflammation over a longer time period in addition to the present study.
Microcomputed tomographic comparison of the bony structure of the canine and feline carpus
Background Feline carpal joint injuries pose a significant clinical challenge due to their complex anatomy and the limited evidence available to guide treatment decisions. A detailed understanding of the osseous microarchitecture is essential for improving surgical planning and therapeutic outcomes. A total of 80 carpal joints from 20 cats and 20 toy breed dogs were examined and compared using microcomputed tomography. The parameters bone volume (BV/TV), bone surface (BS/BV), trabecular thickness (Tb.Th), number of trabeculae (Tb.N), trabecular spacing (Tb.Sp), degree of anisotropy (DA) and connectivity density (Conn. D) were measured and compared. To classify the cortical structure, a three-staged scoring system was applied. Results All carpal bones exhibited a cancellous structure. Overall, cats had fewer but thicker trabeculae than the toy-dogs in the analysed areas of the carpus, which resulted in a greater trabecular distance in cats. Cats also had a higher bone volume (BV/TV) and thicker cortex. Conclusions There were significant differences in structure between dogs and cats which may be important when assessing carpal bone injuries.
Physiological Anti-Müllerian Hormone Concentrations in Male and Female Dogs and Cats before and around Puberty
In recent years several studies established the diagnostic value of anti-Müllerian hormone (AMH) in companion animals. However, less is known about physiological AMH concentrations in young individuals highlighting the necessity to apply the diagnostic findings to this group. The aim of this study was to determine the AMH values of healthy male and female dogs between the age of 8 and 48 weeks, tomcats under 8 weeks and up to 48 weeks of age and queens between 2 to 12 weeks of age. In total, 96 blood samples were collected. Anti-Müllerian hormone was measured in all samples and testosterone was measured in the oldest age group of the males in both species. The hormones were analyzed using a human based chemiluminescence immune assay. Overall, AMH concentrations were higher in males than in females (p < 0.001). According to the AMH concentration there was no difference in males, but queens had significant higher AMH concentrations than bitches (p < 0.001). AMH remained high in males up to week 24 and decreased significantly thereafter (tomcats: p = 0.015; male dogs: p = 0.013), which correlated with an increase in testosterone levels for male dogs only. In bitches, AMH remained below the detection limit until the week 16 and slightly increased subsequently. In queens, AMH was detectable from the beginning with a significant increase in the older age group (p = 0.003). Half of the cats in the older age group even approached the chemiluminescence immune assay’s upper limit. The results show that female cats secrete AMH much earlier than female dogs in which AMH secretion begins just shortly before the start of the puberty. In the male animals, the decrease in AMH concentration around puberty was similar in dogs and cats, but a correlation with the increase of testosterone was only observed in dogs. Further research is required to determine the origin of the high AMH concentrations in female kittens and the lack of correlation between testosterone and AMH concentrations in male kittens.
Diagnostic value of contrast-enhanced CT for elbow joint disorders in dogs
Computed tomography (CT) with intravenous contrast agents can provide additional diagnostic information. To our knowledge, no previous study has evaluated the role of contrast agents in CT imaging of the canine elbow joint. This study aimed to determine the diagnostic value of post-contrast imaging. A total of 326 elbow joints from 163 dogs with unilateral or bilateral lameness caused by elbow joint pathology were examined using radiography and CT, both with and without contrast agents. Diagnoses assessed from radiographs and CT scans in the bone window included primary diseases and periarticular osteophytes. Possible primary diseases included medial coronoid process disease (MCPD), ununited anconeal process (UAP), osteochondrosis dissecans (OCD), humeral intracondylar fissure/incomplete ossification of the humeral condyle (HIF/IOHC), epicondylar spur, caudal or medial calcified bodies. Contrast enhancement of the joint capsule and flexor muscles was evaluated in the soft tissue window. Elbow joints without pathological findings, lameness, or contrast enhancement served as controls (  = 21). Among the 137 elbow joints showing contrast enhancement, 94 demonstrated enhancement limited to the joint capsule, 16 to the flexor muscles, and 27 to both joint structures. Epicondylar spurs were most strongly associated with flexor muscle enhancement, whereas medially located calcified bodies were not. Joint capsule enhancement was most frequently associated with periarticular osteophytes. Notably, elbows with flexor muscle involvement but no epicondylar spur or calcified bodies (  = 14) could only be detected through contrast-enhanced imaging. A contrast agent could be administered during most CT scans of the elbow joint to ensure that no pathological changes are overlooked. Nevertheless, it is important to weigh the benefits for treatment and prognosis against the risks of administering a contrast agent to the patient.
Therapeutic success in fragmented coronoid process disease and other canine medial elbow compartment pathology: a systematic review with meta-analyses
IntroductionThe correct treatment of elbow dysplasia is controversial in modern small animal orthopedics. The aim of this study was to compile all relevant literature of the therapy of fragmented coronoid process and other hereditary disorders of the medial elbow compartment and to statistically evaluate the therapeutic results in three meta-analyses.MethodsThe basis for the systematic literature review was a comprehensive database search of Web of Science, PubMed and Medline. Studies on living patients with above mentioned degenerative joint disease were included in the initial literature search. The data from the final studies, selected according to the PRISMA guidelines, was subsequently extracted. Finally, the success of the different therapies was compared and analyzed by three meta-analyses: success rate, mean difference and standardized mean difference.ResultsFourteen of 494 publications covered by the systematic literature search remained. Their overall truth was: In studies where surgery outcomes was determined by clinical examination and owner questionnaires, it was found that surgical intervention had a significant positive outcome in the presence of fragmented coronoid process and medial compartment disease. Surgical outcomes were also good in three cross-over studies that investigated treatment success using computerized gait analysis. In contrast, comparative studies between surgical and conservative management yielded controversial results. The meta-analysis found no significant difference between medical and surgical therapy.DiscussionThe positive results of studies investigating owner satisfaction and veterinary clinical examination of surgical therapy for medial compartment disease were confirmed by two meta-analyses. However, their study designs were susceptible to observer biases. A third meta-analysis of standardized mean difference differentiating computerized gait analysis results of surgical and conservative management found no evidence of significant superiority of each treatment modality. It however had a limited number of subjects. More comparative studies of high evidence are needed to better understand medial compartment disease and provide the clinician with more accurate diagnostics to separate pathology that should be treated surgically from pathology that can benefit from conservative therapy similarly. Given the invasiveness a more cautious approach might be warranted regarding generally recommending surgery for pathology of the medial elbow compartment.
Salivary cortisol measurements in brachycephalic dog breeds as part of a standardized stress test
Brachycephalic obstructive airway syndrome (BOAS) is a common condition in brachycephalic dogs, with Pugs (PG) and French Bulldogs (FB) appearing to be particularly typically affected. Objective and easy-to-perform tests are necessary to detect the disease at an early stage and to exclude dogs affected by BOAS from breeding. The present study investigated the extent to which vital signs and salivary cortisol concentrations can be used to distinguish between healthy and BOAS-affected dogs in a standardized fitness test. A total of 57 PG, 56 FB and 27 meso- and dolichocephalic dogs were studied as control group (CG). In addition to vital signs, salivary cortisol concentrations were measured before and after the exercise test. It emerged that non-brachycephalic dogs showed a higher fitness level than brachycephalic dogs. The PG recovered significantly slower than the FB after the exercise test. In unaffected PG, cortisol levels rose significantly after the test and then fell again, in unaffected FB they fell significantly during the test. Unexpectedly, cortisol levels remained constant in BOAS affected dogs of both breeds. A possible explanation could be a disturbance of the pituitary-hypothalamic-adrenal axis, which could be due to the chronic stress of affected animals. This would have to be clarified in further studies. In conclusion, a submaximal fitness test may be a useful method to identify dogs suffering from BOAS as it is imperative to prevent the breeding and reproduction of affected dogs.
