Radiation-induced lung injury (RILI) remains a dose-limiting and life-threatening complication of thoracic radiotherapy. The present study aimed to evaluate the therapeutic efficacy and mechanism of the naturally extracted flavonoid, 5,7,8-trimethoxyflavone (HY-N7656), in inhibiting RILI. Lung injury in mice was evaluated using micro-computed tomography, histopathological analysis, enzyme-linked immunosorbent assay and western blotting. Network pharmacology was conducted to predict the potential therapeutic targets and signaling pathways of HY-N7656 in RILI. Cell Counting Kit-8, wound healing, immunofluorescence, reverse transcription-quantitative (RT-q) PCR and protein expression analyses were carried out in vitro using TGF-β-stimulated A549 cells to evaluate epithelial-mesenchymal transition (EMT) and signaling activity. Results of the present study revealed that HY-N7656 markedly alleviated pulmonary inflammation and fibrosis in irradiated mice, leading to a reduction in α-smooth muscle actin expression. In addition, EMT was effectively reversed following treatment with HY-N7656 in A549 alveolar epithelial cells treated with TGF-β, accompanied by restoration of E-cadherin expression and downregulation of mesenchymal markers, such as N-cadherin and vimentin. Network pharmacology analysis and molecular docking validation identified the PI3K/Akt pathway as a central target, which was subsequently confirmed via western blot analysis. Moreover, results of the present study demonstrated that HY-N7656 inhibited radiation-induced activation of PI3K and Akt. To the best of the authors' knowledge, the present study was the first to demonstrate that HY-N7656 modulates the PI3K/Akt signaling pathway to suppress the progression of EMT in RILI, establishing HY-N7656 as a multi-target inhibitor of RILI. These findings present a potential strategy to enhance the safety of radiotherapy, warranting further preclinical and clinical evaluation.

Behaviors of Polyurethane Filled Double Skin Steel Tubular Members

Based on the nonlinear finite element software, the mechanical properties of polyurethane filled double skin steel tubular (PFDSST) members with different hoop coefficients and slenderness ratios under axial compression have been studied by nonlinear finite element software. On the basis of the researches on the numerical simulation results, it was summarized that the influences on stability, ductility and properties of the PFDSST members under axial compression, and then the work processes of the PFDSST members have been revealed deeply, the theoretical basis for the designs of the PFDSST members was established.

Development of a Real-Time PCR Method (Taqman) for Rapid Identification and Quantification of Prorocentrum donghaiense

Prorocentrum donghaiense is a dinoflagellate that is widely distributed in the East China Sea and has become increasingly involved in Harmful Algal Blooms (HABs). Therefore, it is necessary to study this dinoflagellate to monitor HABs. In this study, 13 pairs of primers specific to P. donghaiense (within its internal transcribed spacer (ITS) regions) were designed for SYBR Green I real-time PCR. As the SYBR Green I real-time PCR could not identify P. donghaiense in a specific manner, a Taqman real-time PCR method was developed by designing a set of specific primers and a Taqman probe. A 10-fold serial dilution of recombinant plasmid containing ITS regions of P. donghaiense was prepared as standard samples and the standard curve was established. Additionally, we quantified the genomic DNA in P. donghaiense cells and utilized this DNA to prepare another 10-fold serial dilution of standard sample and accordingly set up the standard curve. The mathematic correlation between the cell number and its corresponding plasmid copy number was also established. In order to test the efficiency of the real-time PCR method, laboratory samples and P. donghaiense HAB field samples were employed for identification and quantitative analysis. As to laboratory samples, as few as 102 cells of P. donghaiense could be quantified precisely utilizing both centrifugation and filtration techniques. The quantification results from field samples by real-time PCR were highly similar to those by light microscopy. In conclusion, the real-time PCR could be applied to identify and quantify P. donghaiense in HABs.

Formability of Fe-Mn-C Twinning Induced Plasticity Steel

A comparative analysis of formability was investigated between F~Mn-C twinning induced plasticity steel with different Mn contents and interstitial free steel. Tensile test combing with the morphology of fracture reveals that element Mn is helpful for the forming of inclusion or particles with film or rod shapes inducing the crack initia- tion and propagation. During stamping process, twinning induced plasticity steel without earing shows better anisot- ropy than interstitial-free steel because a typical 〈111〉 fiber texture forms accompanied by a weaker 〈100〉 fiber texture. The difference between the two steels is not evident during Erichsen cone cupping test, but the result of cone cupping test indicates that the twinning induced plasticity steel has superior drawing ability compared with intersti tial-free steel. The different performances can be attributed to the different deformation mechanism during cupping test. FED (forming limit diagram) of tested steels further suggests twinning induced plasticity steel has slightly su- perior deep drawability but low stretchability than that of IF steel, whose FLD0 value can reach 30%.
