Lactate dehydrogenase activity drives hair follicle stem cell activation
Although normally dormant, hair follicle stem cells (HFSCs) quickly become activated to divide during a new hair cycle. The quiescence of HFSCs is known to be regulated by a number of intrinsic and extrinsic mechanisms. Here we provide several lines of evidence to demonstrate that HFSCs utilize glycolytic metabolism and produce significantly more lactate than other cells in the epidermis. Furthermore, lactate generation appears to be critical for the activation of HFSCs as deletion of lactate dehydrogenase (Ldha) prevented their activation. Conversely, genetically promoting lactate production in HFSCs through mitochondrial pyruvate carrier 1 (Mpc1) deletion accelerated their activation and the hair cycle. Finally, we identify small molecules that increase lactate production by stimulating Myc levels or inhibiting Mpc1 carrier activity and can topically induce the hair cycle. These data suggest that HFSCs maintain a metabolic state that allows them to remain dormant and yet quickly respond to appropriate proliferative stimuli. Flores et al. show that hair follicle stem cells rely on the production of lactate via the LDHA enzyme to become activated. Inducing Ldha through Mpc1 inhibition or Myc activation successfully reactivates the hair cycle in quiescent follicles.
Suicide attempts among children and adolescents: partial or total injury?
This study sought to verify the records on file and the number of cases of attempted suicide among children and adolescents who were attended by Emergency Care health professionals in the municipality of Matozinhos, Minas Gerais, Brazil. Documentary and descriptive research was conducted, the data for which was collected by means of an investigation of Outpatient Records from 2008 to 2010. Of the 73,000 files evaluated, those dealing with cases of attempted suicide among children and adolescents between the age of 3 and 18 years were selected. It was revealed that the health professionals, particularly physicians and nurses, fail to register the cases appropriately, invalidating information about the problem and potential prevention measures. The conclusion reached was that underreporting and the discrepancy of the diagnoses which were not duly referred to the competent agencies require rethinking and reviewing medical practices, and taking a systematic and careful look to address the individual as a complex whole. Neste estudo procurou-se verificar o registro e o número de casos de tentativa de suicídio entre crianças e adolescentes do município de Matozinhos, Minas Gerais, Brasil, que foram atendidos pelos profissionais de saúde do Pronto-Atendimento. Trata-se de uma pesquisa documental e descritiva, cuja coleta dos dados ocorreu por meio de investigação nas Fichas Ambulatoriais, no período de 2008 a 2010. Das 73.000 fichas levantadas, selecionaram-se aquelas que tratavam de casos de tentativa de suicídio entre crianças e adolescentes do município, com idades entre três e 18 anos. Percebeu-se que os profissionais de saúde, mais especificamente os médicos e enfermeiros, não registram os casos de forma adequada, inviabilizando a informação sobre o problema e as medidas de prevenção. Concluiu-se que a subnotificação, a discrepância dos diagnósticos e o não encaminhamento aos órgãos competentes exigem repensar e rever a prática médica e dirigir um olhar sistematizado e cuidadoso para perceber o sujeito como um todo complexo.
Metabolic and Intracellular Signaling Mechanisms of Hair Follicle Stem Cell Activation
The hair follicle is considered a mini-organ which makes it a useful model for studying regenerative processes and cross-tissue interactions due to its unique populations of cell types and specialized pools of adult stem cells. Hair itself is a defining feature of the skin organ and is critical for external protection, thermoregulation, sweat and pheromone relays, and social interactions. In the bulge niche, hair follicle stem cells (HFSCs) oscillate between activation and quiescence to create the hair cycle. This process is cyclically and dynamically maintained throughout a coated animal's lifetime, requiring precise temporal and spatial control of the HFSC niche. However, a complete inventory of the mechanisms underlying hair follicle homeostasis remains unclear. Herein I will briefly explain the current dogma of signaling pathways regulating HFSCs. But, the novelty of my work will go on to further describe the synergistic – yet understudied – roles of metabolic control and canonical G-protein coupled receptors (GPCR) and signaling in modulating downstream genes facilitating HFSC biology. The data out of these projects of course will yield new avenues for the development of metabolic/pharmacological compounds alike for regenerative medicine and equally important molecular management of adult stem cell homeostasis.
Women's temporality after cardiac surgery: contributions to nursing care
to unveil women's existential movement after cardiac surgery. qualitative phenomenological study. The research setting was a hospital in Minas Gerais, in which ten women were interviewed between December 2011 and January 2012. after hospital discharge, the women experienced physical, social and emotional impairments, and expressed the desire to go back to the time before their diagnosis, because they felt as though they still had heart disease. This vague and average understanding led to three units of meaning that, from a Heideggerian hermeneutic point of view, revealed the phenomenon of cardiac surgery as a present circumstance that limited the participants' daily lives. nurses supporting women patients after cardiac surgery should promote health considering existential facets that are expressed during care. The bases for comprehensive care are revealed in singular and whole meetings of subjectivity.
O conhecimento dos adolescentes escolarizados sobre o papiloma vírus humano: revisão integrativa
Objective: To analyze the evidences found in the scientific literature regarding the human papilloma virus for the adolescent sexual and reproductive life. Method: literature integrative review through the search of publications in journals indexed in SciELO, MEDLINE and LILACS databases. Results/Discussion: The final study sample consisted of 27 articles with a predominance of studies addressing adolescence and human papillomavirus (37%), followed by studies on adolescence and sexuality (33%). Most of the studies were carried out in the American continent (56%) followed by the European continent (22%), evidencing the lack of knowledge about the transmission, prevention, screening and oncogenicity of human papilloma virus in different regions, with more knowledge among girls, homosexual boys and the vaccinated population, which revealed the importance of education. Conclusion: This review revealed that actions that make possible changes in the current scenario are fundamental to improve education, awareness, reflection about the risks and health promotion of adolescents, building a network of new meanings and behaviors for their lives. Objetivo: Analizar las evidencias encontradas en la literatura científica acerca del virus papiloma humano para la vida sexual y reproductiva del adolescente. Método: Revisión integradora de la literatura mediante la búsqueda de publicaciones en los periódicos indexados en las bases de datos de SciELO, MEDLINE y LILACS. Resultados / Discusión: La muestra final del estudio está constituida de 27 artículos con predominio de los estudios que abordaban adolescencia y virus papiloma humano (37%), seguido de los estudios sobre adolescentes y sexualidad (33%). La mayoría de los estudios ha sido realizada en el continente americano (56%) seguido del continente europeo (22%), evidenciando el nivel deficitario de conocimiento de los adolescentes sobre la transmisión, prevención, rastreo y oncogenicidad del virus papiloma humano en distintas regiones, siendo mayor el conocimiento entre chicas, chicos homosexuales y entre la población vacunada, lo que ha revelado la importancia de la educación. Conclusión: Esta revisión ha mostrado que las acciones que posibilitan el cambio en el escenario actual son fundamentales para mejorar la educación, concienciación, reflexión sobre los riesgos y la promoción de la salud de los adolescentes, construyendo una red de nuevos significados y comportamientos para sus vidas. Temporalidade da mulher após cirurgia cardíaca: contribuições para o cuidado de enfermagem
RESUMO Objetivo: desvelar o movimento existencial da mulher após a intervenção cirúrgica cardíaca. Método: pesquisa qualitativa de abordagem fenomenológica. Teve como cenário uma instituição hospitalar em Minas Gerais, na qual dez mulheres foram entrevistadas entre dezembro de 2011 e janeiro de 2012. Resultados: após a alta hospitalar, as mulheres vivenciam comprometimentos físicos, sociais e emocionais, desejando a volta do tempo anterior ao diagnóstico, uma vez que ainda se sentem cardiopatas. Essa compreensão vaga e mediana emerge de três unidades de significação, as quais, na perspectiva da hermenêutica heideggeriana, permitiram desvelar o fenômeno da cirurgia cardíaca como um agora que limita o dia a dia das mulheres. Conclusão: o enfermeiro, na disposição de ser-com-o-ser-aí-mulher-após-a-cirurgia-cardíaca, deve promover a saúde, considerando as facetas existenciais que se mostram nos momentos de cuidado. Nessa oportunidade de encontros singulares e plenos de subjetividade se revelam as bases para a integralidade da assistência. RESUMEN Objetivo: desvelar la existencia de la mujer tras la cirugía cardíaca. Método: investigación cualitativa de enfoque fenomenológico. La escena fue un hospital en Minas Gerais, en el cual se entrevistaron diez mujeres entre diciembre de 2011 y enero de 2012. Resultados: tras el alta hospitalaria que todavía sienten las enfermedades del corazón a través de experiencias físicas, sociales y emocionales, las mujeres anhelan regresar al tiempo anterior al diagnóstico. Esa vaga y mediana comprensión viene de tres unidades de significado, las cuales, teniendo en vista la hermenéutica de Heidegger, ayudaron a descubrir que el fenómeno de la cirugía cardiaca limita la vida cotidiana de las mujeres. Conclusión: el enfermero ayudando después de la cirugía cardiaca debe promover la salud de la paciente, teniendo en cuenta sus aspectos existenciales en los momentos de atención.
