Up until the first half of the 20th century, silver found significant employment in medical applications, particularly in the healing of open wounds, thanks to its antibacterial and antifungal properties. Wound repair is a complex and dynamic biological process regulated by several pathways that cooperate to restore tissue integrity and homeostasis. To facilitate healing, injuries need to be promptly treated. Recently, the interest in alternatives to antibiotics has been raised given the widespread phenomenon of antibiotic resistance. Among these alternatives, the use of silver appears to be a valid option, so a resurgence in its use has been recently observed. In particular, in contrast to ionic silver, colloidal silver, a suspension of metallic silver particles, shows antibacterial activity displaying less or no toxicity. However, the human health risks associated with exposure to silver nanoparticles (NP) appear to be conflicted, and some studies have suggested that it could be toxic in different cellular contexts. These potentially harmful effects of silver NP depend on various parameters including NP size, which commonly range from 1 to 100 nm. In this study, we analyzed the effect of a colloidal silver preparation composed of very small and homogeneous nanoparticles of 0.62 nm size, smaller than those previously tested. We found no adverse effect on the cell proliferation of HaCaT cells, even at high NP concentration. Time-lapse microscopy and indirect immunofluorescence experiments demonstrated that this preparation of colloidal silver strongly increased cell migration, re-modeled the cytoskeleton, and caused recruitment of E-cadherin at cell-cell junctions of human cultured keratinocytes.

Pancreatic Progenitor Commitment Is Marked by an Increase in Ink4a/Arf Expression

The identification of the molecular mechanisms controlling early cell fate decisions in mammals is of paramount importance as the ability to determine specific lineage differentiation represents a significant opportunity for new therapies. Pancreatic Progenitor Cells (PPCs) constitute a regenerative reserve essential for the maintenance and regeneration of the pancreas. Besides, PPCs represent an excellent model for understanding pathological pancreatic cellular remodeling. Given the lack of valid markers of early endoderm, the identification of new ones is of fundamental importance. Both products of the Ink4a/Arf locus, in addition to being critical cell-cycle regulators, appear to be involved in several disease pathologies. Moreover, the locus’ expression is epigenetically regulated in ES reprogramming processes, thus constituting the ideal candidates to modulate PPCs homeostasis. In this study, starting from mouse embryonic stem cells (mESCs), we analyzed the early stages of pancreatic commitment. By inducing mESCs commitment to the pancreatic lineage, we observed that both products of the Cdkn2a locus, Ink4a and Arf, mark a naïve pancreatic cellular state that resembled PPC-like specification. Treatment with epi-drugs suggests a role for chromatin remodeling in the CDKN2a (Cycline Dependent Kinase Inhibitor 2A) locus regulation in line with previous observations in other cellular systems. Our data considerably improve the comprehension of pancreatic cellular ontogeny, which could be critical for implementing pluripotent stem cells programming and reprogramming toward pancreatic lineage commitment.

I17 Curcumin-supplemented diet preserves body weight and ameliorates intestinal functionality in R6/2 mice

BackgroundHuntington Disease (HD) has traditionally been described as a disorder purely of the brain, however emerging evidence indicates that peripheral abnormalities are also commonly seen. Among others, unintended body weight loss represents a hallmark of peripheral HD pathology. It correlates with disease progression and significantly affects the quality of life of HD patients. Although the underlying molecular mechanism is still unknown, evidence suggests that body weight loss seems not to be secondary to inadequate nutrition or to chorea, but rather attributable to changes in the metabolism and defective nutrients absorption along the intestinal tract. Curcumin, a naturally occurring polyphenol, has been validated to exert important beneficial effects in a multitude of gastrointestinal dysfunction and neurodegenerative processes similar to those occurring in HD. Although its therapeutic effect in HD is still questionable, recent evidence indicates that curcumin significantly improves neuropathology as well neurochemical and neurobehavioral defects in a mouse model of the disease, however whether it may be suited to be developed to treat gastrointestinal dysfunction in the disease is not clear yet.AimIn this study, we aimed to investigate the potential beneficial effects that the chronic administration of curcumin may have on gastrointestinal dysfunction commonly occurring in R6/2 mice as the disease progresses.MethodCurcumin along with bioperine (bioavailability enhancing-agent) was orally administered to females 10 days before pregnancy and during the entire gestation period up to the end of lactation. Starting from the third week of age, pups were daily treated with 40 mg/kg curcumin for 7 weeks. KCl-induced contraction was evaluated in isolated intestinal tract from both curcumin and vehicle-treated mice.ResultsOur preliminary data demonstrate that chronic administration of curcumin is safe and well tolerated in R6/2 mice. It prevents the gradual weight loss despite no changes in food intake were observed. From the functional point of view, curcumin considerably ameliorates the intestinal phenotype and restores the normal intestinal reactivity compared to untreated R6/2 mice.

Contextualización: Guasave es un municipio ubicado en el noroeste de México, forma parte de las 18 municipalidades del Estado de Sinaloa, es considerada como una de las regiones más ricas en la actividad agrícola del país, de acuerdo al volumen de producción y superficie sembrada a nivel nacional. Vacío de conocimiento: El calentamiento global, la contaminación y erosión de suelos y aguas, han afectado gravemente a la agricultura, por ello, es necesario que se instrumenten estrategias sustentables que permitan a este sector seguir desarrollándose. La importancia de este artículo recae en hacerle frente a la escasez de instrumentos que coadyuven a la creación de valor en empresas dedicadas a esta actividad, la implementación de éste permitirá obtener información empírica útil para la identificación de las prácticas sustentables que generan valor a las organizaciones agrícolas. Propósito del estudio: El propósito de este artículo es dar a conocer el proceso de construcción y validación, así como divulgar el instrumento “Prácticas sustentables para la creación de valor en organizaciones agrícolas” del norte de Sinaloa, México. Metodología: Son 38 ítems de escala valorativa, organizados en las tres dimensiones de desarrollo sustentable y creación de valor los que integran el instrumento. El enfoque retomado fue el cuantitativo, la población objetivo fue de 32 organizaciones agrícolas del municipio para la prueba de fiabilidad mediante el coeficiente de consistencia interna Alfa de Cronbach y 10 expertos para el caso de la validación. Resultados y conclusiones: Los resultados obtenidos permiten afirmar que el instrumento es apropiado para medir las prácticas sustentables que crean valor en las organizaciones agrícolas de la parte norte de la entidad, al determinarse un coeficiente de 0.941 y una pertinente validez.

A curriculum case study of sheltered English workshops and their effects on student skills

This study investigated the application of a set of instructional strategies in four English workshops. They were designed for high school students of limited-English proficiency Awaiting Redesignation. There were no more than twenty students per workshop. They met on three consecutive Saturday mornings in four hour academic sessions. Each teacher received a syllabus of readings and a set of instructional strategies. Students received a packet of reference materials. Teachers had the freedom to teach the syllabus using their own style, and order of lessons. The instructional strategies were to be applied in the delivery of the lessons. The researcher and two assistants made observations of instruction, using public school observation forms, and an observation form developed by the researcher. Each observation lasted fifteen-minutes, and there were at least three observations made for each teacher, each Saturday. Additionally, all the students in attendance were asked to respond to the same questionnaire at the end of each workshop session. Observations and questionnaires were qualitatively analyzed. The student questionnaire answers were compared to the lesson planning objectives self-reported by the teacher. The analysis suggested that the teachers did utilize the instructional strategies in their delivery of the workshop curriculum. The quantitative analysis consisted of analyzes of the test results for the Language Assessment Scales in reading and writing for the experimental group. The results for each workshop section were displayed in histograms. A Oneway Analysis of Variance was performed. Results showed no significant differences between group Mean performances in both LAS-Reading and LAS-Writing. A T-test for Independent Sample Means was conducted usi ng the experimental group data and comparable test data for another group: Students who qualified to take the workshops, but who did not attend. Results of the T-tests for the LAS reading and writing performance of the two groups, were not statistically significant. However, there was greater variability in the scores of the comparison group.

DNA oxidative damage is one of the main concerns being implicated in severe cell alterations, promoting different types of human disorders and diseases. For their characteristics, male gametes are the most sensitive cells to the accumulation of damaged DNA. We have recently reported the relevance of arginine residues in the Cu(II)-induced DNA breakage of sperm H1 histones. In this work, we have extended our previous findings investigating the involvement of human sperm nuclear basic proteins on DNA oxidative damage in healthy males presenting copper and chromium excess in their semen. We found in 84% of those males an altered protamines/histones ratio and a different DNA binding mode even for those presenting a canonical protamines/histones ratio. Furthermore, all the sperm nuclear basic proteins from these samples that resulted were involved in DNA oxidative damage, supporting the idea that these proteins could promote the Fenton reaction in DNA proximity by increasing the availability of these metals near the binding surface of DNA. In conclusion, our study reveals a new and unexpected behavior of human sperm nuclear basic proteins in oxidative DNA damage, providing new insights for understanding the mechanisms related to processes in which oxidative DNA damage is implicated.

The autonomic nervous system (ANS) and the immune system are deeply interrelated. The ANS regulates both innate and adaptive immunity through the sympathetic and parasympathetic branches, and an imbalance in this system can determine an altered inflammatory response as typically observed in chronic conditions such as systemic autoimmune diseases. Rheumatoid arthritis, systemic lupus erythematosus, and systemic sclerosis all show a dysfunction of the ANS that is mutually related to the increase in inflammation and cardiovascular risk. Moreover, an interaction between ANS and the gut microbiota has direct effects on inflammation homeostasis. Recently vagal stimulation techniques have emerged as an unprecedented possibility to reduce ANS dysfunction, especially in chronic diseases characterized by pain and a decreased quality of life as well as in chronic inflammation.

Violence as a Barrier for HIV Prevention among Female Sex Workers in Argentina

Violence against female sex workers (FSWs) has been increasingly reported as an important determinant of HIV infection risk. This study explores the frequency of different violent experiences (sexual abuse, rejection, beating and imprisonment) among FSWs in Argentina and its association with condom use and HIV and T. pallidum prevalence.
A convenience sample of 1255 FSWs was included in a cross-sectional study conducted between October 2006 and November 2009.
Sexual abuse was reported by 24.1% (219/907) of women. A total of 34.7% (42/1234) reported rejection experiences, 21.9% (267/1215) reported having been beaten and 45.4% (561/1236) stated having been arrested because of their sex work activity. There was a higher frequency of inconsistent condom use with clients among FSWs who had experienced sexual abuse, rejection, and police detention. A higher frequency of HIV and T. pallidum infection was detected among FSWs who reported having been arrested by the police.
The study shows for the first time the frequency of different violent situations among FSWs in Argentina. The association between violence against sex workers, condom use and STI prevalence demonstrated here calls for measures to reduce stigma and violence against FSWs. Such violent experiences may increase vulnerability to STI through coerced unprotected sex.
