Homogenization and impoverishment of taxonomic and functional diversity of ants in Eucalyptus plantations
Despite its negative impacts on the environment and biodiversity, tree plantations can contribute to biodiversity conservation in fragmented landscapes, as they harbor many native species. In this study, we investigated the impact of Eucalyptus plantations on the taxonomic and functional diversity of ant communities, comparing ant communities sampled in managed and unmanaged (abandoned for 28 years) Eucalyptus plantations, and native Atlantic rain forests. Eucalyptus plantations, both managed and unmanaged, reduced the functional diversity and increased the similarity between ant communities leading to functional homogenization. While communities in managed plantations had the lowest values of both taxonomic and functional ant diversities, ant communities from unmanaged plantations had similar values of species richness, functional redundancy and Rao’s Q compared to ant communities from forest patches (although functional richness was lower). In addition, communities in unmanaged Eucalyptus plantations were taxonomically and functionally more similar to communities located in managed plantations, indicating that Eucalyptus plantations have a severe long-term impact on ant communities. These results indicate that natural regeneration may mitigate the impact of Eucalyptus management, particularly regarding the functional structure of the community (α diversity), although it does not attenuate the effects of long term homogenization in community composition (β diversity).
Dilated Cardiomyopathy Phenotype With Global (Four‐Chamber) Involvement in a Cat: Echocardiographic, Pathological, Histopathological, and Immunohistochemical Findings
An 8‐year‐old castrated male Exotic Shorthair cat was referred due to the onset of abdominal effusion. Echocardiography revealed a dilated cardiomyopathy phenotype affecting both ventricles, associated with biatrial dilatation, left atrial dysfunction with spontaneous echocontrast, and right‐sided congestive heart failure. Additional diagnostic tests (thoracic radiographs, complete blood cell count, serum biochemistry, assessment of thyroid status, cardiac troponin I and plasma taurine concentrations, and urinalysis) ruled out secondary conditions able to cause a similar echocardiographic phenotype. Despite prompt initiation of cardiac therapy (furosemide, pimobendan, clopidogrel, and rivaroxaban) and subsequent hospitalization for further medical support, the cat′s clinical condition deteriorated, ultimately leading to euthanasia. Postmortem examination revealed extensive areas of myofiber thinning with a wavy appearance in both ventricles, closely resembling the attenuated wavy fiber pattern described in dogs affected by dilated cardiomyopathy. Additionally, multifocal replacement of myofibers by adipose tissue was found in both atria. Immunohistochemical analysis showed abnormal expression patterns of desmin, vimentin, and connexin 43, similar to those previously reported in humans and dogs with dilated cardiomyopathy. Given the thorough antemortem and postmortem investigations and the associated findings, a rare case of feline dilated cardiomyopathy with biventricular and biatrial involvement was diagnosed.
DOG1 Immunohistochemical Expression in Normal and Neoplastic Canine Tissues: Is It Only a Marker for GISTs?
DOG1 is a transmembrane protein highly expressed in human GISTs. In dogs, DOG1 has been studied to a limited extent in GISTs, where its diagnostic value is considered comparable to KIT, while its expression in other canine tissues remains uncharacterized. The aim of this study is to assess the expression of DOG1 in canine normal and neoplastic tissues, with particular emphasis on a large cohort of GISTs to verify their diagnostic role in comparison with KIT. To achieve this, we analyzed a total of 143 FFPE samples of normal (n = 55) and neoplastic canine tissues (n = 88) by immunohistochemistry. DOG1 was strongly expressed in several cell types, with distribution and intensity patterns that partially overlap those reported in humans. In canine GISTs, DOG1 showed strong and widespread reactivity with a sensitivity slightly inferior to KIT (92% vs. 96%); however, one intestinal mesenchymal tumor KIT-negative/DOG1-positive highlights the importance of combining both markers to maximize the correct diagnosis. Moderate to strong immunohistochemistry for DOG1 has been found in several other tumors, sometimes comparable to that of GISTs. This suggests that DOG1 should not be associated exclusively with GISTs and may provide a basis for further investigation into the role of DOG1 in canine malignancy.
The Facet of Human Impact: Solenopsis invicta Buren, 1972 Spreading around the Atlantic Forest
The present investigation deals with some aspects of the diversity of fire ants (Hymenoptera: Formicidae) in their native range. The Red Imported Fire Ant Solenopsis invicta is native to the tropical and subtropical inland territories of South America. In Brazil, it mainly occurs around the Pantanal region and across the Paraguay river, a region composed of grasslands which are seasonally flooded. Recent studies have evidenced this fire ant species is gradually spreading to other regions of Brazil. In the present investigation, we surveyed the molecular diversity of S. invicta populations across fragments of Atlantic Forest in São Paulo, Brazil, using mtDNA COI haplotypes. Fire ant nests were sampled along the highways lining the northern and southern slope sides of the mountain range Serra do Mar, SP, Brazil. Four haplotypes were identified (H1–H4), which were assessed for similarity to deposited records by other authors, revealing that the haplotypes H1 and H2 are likely of foreign origin through recent reintroduction via a marine port to the south of the Serra do Mar mountain range. On the other hand, the haplotypes H3 and H4, predominating among the inland samples from the northern side of the mountain range, were most similar to previous records from more central regions of Brazil. Haplotypes clustered into distinct supergroups, further pointing to the occurrence of two separate expansion waves of S. invicta in the region. We suggest the obtained pattern indicates the mountain range may function as a geographical barrier deferring gene flow.
Preliminary Study of the Antimicrobial Capacity of the Cutaneous Mucus and Smear Cytology of the Epidermis in a Population of European eels (Anguilla anguilla, Linnaeus 1758)
The skin and its products, such as the mucus, represent an important defense mechanism against infection by pathogens. Various environmental conditions can alter mucus composition and/or cells embedded in this matrix. The analysis of skin mucus is considered a useful method to evaluate the biological response of fish to stimuli. The mucus and cells can be considered suitable non-invasive biomarkers. With this preliminary study, an antibacterial capacity method was applied to cutaneous mucus samples of a European eel population. This standardized method highlighted the ability of the mucus to significantly counteract the proliferation of two bacteria, the opportunistic pathogen Aeromonas hydrophila and the primary pathogen Aeromonas salmonicida. The cytological method using skin scraping has proven to be a useful non-invasive tool, having allowed the highlighting of the two most represented cellular populations of the cutaneous covering, the epidermal cells and the goblet cells, in combination with mucous strands. Cytological findings did not disclose pathologic elements in the mucus. A broader use of these two non-invasive microbiological and cytological methods can provide useful information on fish health, indirectly contributing to the conservation of the species.
Cannabinoid and cannabinoid related receptors in fibroblasts, inflammatory and endothelial cells of the equine hoof with and without laminitis: novel pharmacological target
Evidence suggests that the endocannabinoid system (ECS) is crucial for regulating inflammation, cell proliferation and pain. The ECS is composed of cannabinoid receptors such as type 1 (CBR1), type 2 (CBR2) and GPR55, endocannabinoids and enzymes. Proteins of ECS have previously been localized in the epidermal cells of the horse hooves. Given the physio-pathological role and cellular distribution of the ECS across species, the authors hypothesized that cannabinoid receptors are expressed within the inflammatory cells, fibroblasts and endothelial cells of the equine hoof laminae, going beyond the epidermal cells. To preliminary analyze the gene expression of Cn1r, Cn2r and GPR55 in the hoof laminae and test the specificity of the antibody against GPR55. To characterize the distribution and expression of CBRs in the inflammatory cells and fibroblasts of the laminar junction of equine healthy hooves and with laminitis. Animals were divided into 3 groups: healthy, acute laminitis and chronic laminitis. A total of 18 samples were collected and processed from the front limb of animals slaughtered for consumption or euthanized (6 control animals, 4 acute laminitis, 8 chronic laminitis). Analysis of CBR1, CBR2 and GPR55 protein expression was made by fluorescence microscopy with co-localization with antibodies against the macrophages marker IBA1, the T cell marker CD3, the neutrophils marker calprotectin (MAC387), the fibroblasts marker vimentin (Clove V9) and the nerve fibers marker Substance P. Preliminary analysis was performed to evaluate gene expression ( , , and ) using real-time PCR and to verify the specificity of the primary antibody (Gpr55) with Western Blotting (WB). The resident pool of inflammatory cells in the normal laminae and the inflammatory infiltrate cells of the affected equine laminae showed protein expression of CB2R and GPR55; no CB1R staining was seen at the inflammatory cells. Equine dermal fibroblast and endothelial cells exhibited protein expressions of CB1R, CBR2 and GPR55. Substance P positive nerve fibers were positive for CB1R. Cannabinoid receptors are expressed in different immune cell types of the hoof laminae, pointing to the role of the ECS in modulating inflammatory outburst, tissue degeneration and pain. Our results serve as a foundation for the development of new veterinary pharmacotherapies that target the ECS during laminitis.
Distribution of Cannabinoid Receptors in Keratinocytes of Healthy Dogs and Dogs With Atopic Dermatitis
It is commonly accepted that some form of skin barrier dysfunction is present in canine atopic dermatitis (AD), one of the most common cutaneous pruritic inflammatory diseases of dogs. The impaired skin barrier function facilitates the penetration of allergens and subsequently stronger sensitization responses. The role of the endocannabinoid system (ECS) in the physiology and pathology of the skin is becoming increasingly established. It has been demonstrated that cannabinoid receptors are expressed in healthy and diseased skin and, based on current knowledge, it could be stated that cannabinoids are important mediators in the skin. The present study has been designed to immunohistochemically investigate the expression of the cannabinoid receptors type 1 (CB1R) and 2 (CB2R) and the cannabinoid-related receptors G protein-coupled receptor 55 (GPR55), transient receptor potential vanilloid 1 (TRPV1) and ankyrin 1 (TRPA1), peroxisome proliferator-activated receptors alpha (PPARα), and serotoninergic receptor 1a (5-HT1aR) in keratinocytes of healthy dogs and of dogs with AD. Samples of skin tissues were collected from 7 healthy controls (CTRL-dogs) and from 8 dogs with AD (AD-dogs). The tissue samples were processed using an immunofluorescence assay with commercially available antibodies, and the immunolabelling of the receptors studied was quantitatively evaluated. The keratinocytes of the CTRL- and the AD-dogs showed immunoreactivity for all the receptors investigated with a significant upregulation of CB2R, TRPA1, and 5-HT1aR in the epidermis of the AD-dogs. The presence of cannabinoid and cannabinoid-related receptors in healthy keratinocytes suggested the possible role of the ECS in canine epidermal homeostasis while their overexpression in the inflamed tissues of the AD-dogs suggested the involvement of the ECS in the pathogenesis of this disease, having a possible role in the related skin inflammation and itching. Based on the present findings, the ECS could be considered a potential therapeutic target for dogs with AD.
Mutational Analysis of c-KIT and PDGFRA in Canine Gastrointestinal Stromal Tumors (GISTs)
Gastrointestinal stromal tumors (GISTs) are the most common mesenchymal tumors of the canine gastrointestinal tract and are diagnosed by the immunohistochemical expression of the receptor tyrosine kinase (RTK) KIT. Activating mutations of the proto-oncogenes c-KIT and PDGFRA drive GIST oncogenesis and are used to predict the response to RTK-inhibitors in human oncology. Currently, the frequency and significance of these mutations in canine GIST have not been adequately explored. Therefore, we investigated the mutational status of c-KIT (exons 9, 11 and 13) and PDGFRA (exons 12 and 18) genes by PCR followed by fragment analysis for c-KIT deletions and PCR followed by screening with DHPLC and direct sequencing confirmation for single nucleotide variations in 17 formalin-fixed paraffin-embedded canine GISTs confirmed by KIT immunopositivity. c-KIT mutations were detected in 47% of cases, with a mutation detection rate significantly higher (p = 0.0004, Fisher’s exact test) and always involving exon 11. A PDGFRA gene mutation (exon 18) was identified in one case. Even if follow-up data were not available for all cases, four cases with documented abdominal metastases displayed c-KIT mutations. These data confirm that c-KIT exon 11 mutations occur frequently in canine GISTs, and identify the presence of a PDGFRA mutation similar to human GISTs. This study also suggests a potential association of c-KIT mutation with more aggressive biological behavior.
Using Ant Community Data for Monitoring Tropical Forest Restoration in the Seasonal Atlantic Forest in Brazil
Background and Research Aims: Monitoring is essential for assessing the success of ecological restoration efforts. Ants, with their vast diversity and functional roles, serve as a significant group for such analyses. This study focused on evaluating ant communities to identify potential arboreal species that could serve as indicators of the forest restoration process. Methods: The study was carried out in restoration areas within the Seasonal Semideciduous Forest, located in two landscapes within the Atlantic Forest of southeastern Brazil. We examined forest restoration plantations at three different developmental stages—early, middle, and late—alongside a native reference forest. Key parameters such as canopy cover and density of trees and saplings were measured. Ants were actively sampled throughout different seasons. We assessed ant community compositions across restoration gradients and investigated their association with vegetation structure. We also identified ant species that could serve as restoration indicators. Results: We identified a total of 60 species and 19 morphospecies. However, no significant difference in species richness was observed among the areas. Ant communities in restoration areas exhibited more similarity to each other than to those in native forests. Therefore, even in the oldest restored sections, ant communities have not yet converged with those of the reference ecosystem. Although we identified some indicator species within certain areas, none of these were arboreal. Conclusion: The composition of the ant community is more valuable than its species richness for monitoring tropical forest restoration. The historical context of restoration areas and the age of restoration efforts may influence the results. Implications for conservation: Early stages of tropical forest restoration do not recover the biodiversity and composition of arboreal ants found in reference forests. Additional studies are needed to better understand changes in the ant communities over time, providing information to improve monitoring, management and the conservation value.
Unusual Presentation of Aortic Valve Infective Endocarditis in a Dog: Aorto-Cavitary Fistula, Tricuspid Valve Endocarditis, and Third-Degree Atrioventricular Block
A 2-year-old Boxer with a history of subaortic stenosis and immunosuppressant therapy developed aortic valve infective endocarditis. On echocardiographic examination with simultaneous electrocardiographic tracing, multiple uncommon periannular complications of the aortic valve endocarditis were found, including aorto-cavitary fistula with diastolic left-to-right shunt, tricuspid valve endocarditis, and third-degree atrioventricular block. Necropsy confirmed the above echocardiographic findings. Although aortic valve endocarditis represents a well-known disease entity in dogs, the dynamic nature of this condition may allow development of complex and uncommon echocardiographic features.
