First detection of Fusarium oxysporum f. sp. cubense tropical race 4 (TR4) on Cavendish banana in India

Fusarium wilt, caused by the soil-borne fungus Fusarium oxysporum f. sp. cubense (Foc), is regarded as one of the most devastating diseases of bananas globally. Foc consists of three races, of which Foc races 1 and 2 occur in India. Foc race 4 has not been found in the country despite the importance and extensive cultivation of Cavendish cultivars. In 2015, a banana grower from Barari village in the Bihar State of India reported the wilting of Cavendish banana cvs. Robusta and Grand Naine. The incidence of the disease ranged from 2 to 26.6%. Symptoms included yellowing and browning of older leaves, which progressed from the oldest to the youngest leaves. Samples were collected from pseudostems and the responsible fungus isolated. The cultures were then identified using fungal morphology, vegetative compatibility group (VCG) analysis, volatile production and PCR with Foc tropical race (TR) 4-specific primers. Pathogenicity tests were conducted using Grand Naine plantlets produced by micropropagation. The cultures were identified as volatile-producing F. oxysporum strains, and pathogenicity tests confirmed their status as the banana pathogen Foc. VCG analysis, and the use of race-specific PCR markers, identified them as members of VCG 01213/16 and Foc TR4, respectively. This indicated that the fungus responsible for Fusarium wilt in the Bihar state of India is Foc TR4. Most Indian farmers use suckers as planting material, and are not aware of the impact of the disease. If the disease spreads to other states in India it can lead to an unemployment problem for farm labourers and the change in production to non-remunerative crops that may reduce profits.

The development of a multiplex PCR assay for the detection of Fusarium oxysporum f. sp. cubense lineage VI strains in East and Central Africa

Banana is a staple food and cash crop grown in East and Central Africa (ECA). The main banana varieties grown in ECA are the East African highland bananas (EAHB), although dessert/beer bananas such as Sukari Ndizi, Kayinja (Pisang Awak) and Gros Michel are also produced due to their high value at local markets. The Fusarium wilt fungus Fusarium oxysporum f. sp. cubense (Foc) causes disease of susceptible dessert/beer bananas, which significantly reduces yields. Banana Fusarium wilt is managed by excluding the pathogen from disease-free areas and by planting disease-resistant varieties in infested fields. Six phylogenetically closely-related vegetative compatibility groups (VCGs) of Foc, VCGs 0124, 0125, 0128, 01212, 01220 and 01222 are present in ECA, which all group together in Foc Lineage VI. Rapid and accurate detection of Foc Lineage VI strains is thus important to prevent its spread to disease-free areas. In this study, molecular markers specific to Foc Lineage VI were therefore developed. Primer sets were then combined in a multiplex PCR assay, and validated on a worldwide population of 623 known Foc isolates, other formae speciales and non-pathogenic Fusarium oxysporum isolates. The Foc Lineage VI multiplex PCR was used to identify Foc isolates collected in banana fields at five locations in Uganda and Tanzania. Foc Lineage VI DNA was detected at a concentration as low as 0.1 ng/μl, both in the absence and presence of banana DNA, and can therefore be used as an accurate diagnostic tool for Foc Lineage VI strains.

The distribution and host range of the banana Fusarium wilt fungus, Fusarium oxysporum f. sp. cubense, in Asia

Fusarium oxysporum formae specialis cubense (Foc) is a soil-borne fungus that causes Fusarium wilt, which is considered to be the most destructive disease of bananas. The fungus is believed to have evolved with its host in the Indo-Malayan region, and from there it was spread to other banana-growing areas with infected planting material. The diversity and distribution of Foc in Asia was investigated. A total of 594 F. oxysporum isolates collected in ten Asian countries were identified by vegetative compatibility groups (VCGs) analysis. To simplify the identification process, the isolates were first divided into DNA lineages using PCR-RFLP analysis. Six lineages and 14 VCGs, representing three Foc races, were identified in this study. The VCG complex 0124/5 was most common in the Indian subcontinent, Vietnam and Cambodia; whereas the VCG complex 01213/16 dominated in the rest of Asia. Sixty-nine F. oxysporum isolates in this study did not match any of the known VCG tester strains. In this study, Foc VCG diversity in Bangladesh, Cambodia and Sri Lanka was determined for the first time and VCGs 01221 and 01222 were first reported from Cambodia and Vietnam. New associations of Foc VCGs and banana cultivars were recorded in all the countries where the fungus was collected. Information obtained in this study could help Asian countries to develop and implement regulatory measures to prevent the incursion of Foc into areas where it does not yet occur. It could also facilitate the deployment of disease resistant banana varieties in infested areas.

Long-term outcome after severe traumatic brain injury: a systematic literature review

BackgroundExpectation of long-term outcome is an important factor in treatment decision-making after severe traumatic brain injury (sTBI). Conclusive long-term outcome data substantiating these decisions is nowadays lacking. This systematic review aimed to provide an overview of the scientific literature on long-term outcome after sTBI.MethodsA systematic search was conducted using PubMed from 2008 to 2020. Studies were included when reporting long-term outcome ≥ 2 years after sTBI (GCS 3–8 or AIS head score ≥ 4), using standardized outcome measures. Study quality and risk of bias were assessed using the QUIPS tool.ResultsTwenty observational studies were included. Studies showed substantial variation in study objectives and study methodology. GOS-E (n = 12) and GOS (n = 8) were the most frequently used outcome measures. Mortality was reported in 46% of patients (range 18–75%). Unfavourable outcome rates ranged from 29 to 100% and full recovery was seen in 21–27% of patients. Most surviving patients reported SF-36 scores lower than the general population.ConclusionLiterature on long-term outcome after sTBI was limited and heterogeneous. Mortality and unfavourable outcome rates were high and persisting sequelae on multiple domains common. Nonetheless, a considerable proportion of survivors achieved favourable outcome. Future studies should incorporate standardized multidimensional and temporal long-term outcome measures to strengthen the evidence-base for acute and subacute decision-making.Highlights1. Expectation of long-term outcome is an important factor in treatment decision-making for patients with severe traumatic brain injury (sTBI).2. Favourable outcome and full recovery after sTBI are possible, but mortality and unfavourable outcome rates are high.3. sTBI survivors are likely to suffer from a wide range of long-term consequences, underscoring the need for long-term and multi-modality outcome assessment in future studies.4. The quality of the scientific literature on long-term outcome after sTBI can and should be improved to advance treatment decision-making.

Metastatic and locally-advanced neuroendocrine neoplasms (aNEN) form clinically and genetically heterogeneous malignancies, characterized by distinct prognoses based upon primary tumor localization, functionality, grade, proliferation index and diverse outcomes to treatment. Oral Supplementation with Lactobacillus casei Subspecies rhamnosus Prevents Enteric Colonization by Candida Species in Preterm Neonates: A Randomized Study

Background. Colonization by Candida species is the most important predictor of the development of invasive fungal disease in preterm neonates, and the enteric reservoir is a major site of colonization. We evaluated the effectiveness of an orally supplemented probiotic (Lactobacillus casei subspecies rhamnosus; Dicoflor [Dicofarm spa]; 6 × 109 cfu/day) in the prevention of gastrointestinal colonization by Candida species in preterm, very low birth weight (i.e., <1500-g) neonates during their stay in a neonatal intensive care unit. Methods. Over a 12-month period, a prospective, randomized, blind, clinical trial that involved 80 preterm neonates with a very low birth weight was conducted in a large tertiary neonatal intensive care unit. During the first 3 days of life, the neonates were randomly assigned to receive either an oral probiotic added to human (maternal or pooled donors') milk (group A) or human milk alone (group B) for 6 weeks or until discharge from the NICU, if the neonate was discharged before 6 weeks. On a weekly basis, specimens obtained from various sites (i.e., oropharyngeal, stool, gastric aspirate, and rectal specimens) were collected from all patients for surveillance culture, to assess the occurrence and intensity of fungal colonization in the gastrointestinal tract. Results. The incidence of fungal enteric colonization (with colonization defined as at least 1 positive culture result for specimens obtained from at least 1 site) was significantly lower in group A than in group B (23.1% vs. 48.8%; relative risk, 0.315 [95% confidence interval, 0.120–0.826]; P = .01). The numbers of fungal isolates obtained from each neonate (P = .005) and from each colonized patient (P = .005) were also lower in group A than in group B. L. casei subspecies rhamnosus was more effective in the subgroup of neonates with a birth weight of 1001–1500 g. There were no changes in the relative proportions of the different Candida strains. No adverse effects potentially associated with the probiotic were recorded. Conclusions. Orally administered L. casei subspecies rhamnosus significantly reduces the incidence and the intensity of enteric colonization by Candida species among very low birth weight neonates.

This article assesses the institutional prescriptions of adaptive (co-)management based on a literature review of the (water) governance literature. The adaptive (co-)management literature contains four institutional prescriptions: collaboration in a polycentric governance system, public participation, an experimental approach to resource management, and management at the bioregional scale. These prescriptions largely resonate with the theoretical and empirical insights embedded in the (water) governance literature. However, this literature also predicts various problems. In particular, attention is called to the complexities associated with participation and collaboration, the difficulty of experimenting in a real-world setting, and the politicized nature of discussion on governance at the bioregional scale. We conclude this article by outlining a common research agenda that invites the collaborative efforts of adaptive (co-)management and governance scholars.

Motor Disturbance in ASD: A Pilot Study Showing Hypokinetic Behavior?

Data supporting theoretical models linking autism spectrum disorders (ASD) to motor disturbance are inconclusive. In the present study, children and adolescents with ASD (n = 44) were compared with a matched group of typically developing individuals (n = 49) on both instrumental and observational assessments of motor abnormalities. No group differences were found in the instrumental data. However, more bradykinetic motor behavior was found using an observational scale in the ASD groups. More rigid motor behavior was found in the adolescents with ASD but not in the children. Individuals with ASD show significantly more hypokinetic behavior, which may not be strictly dopaminergic in origin, but may reflect a weak central coherency in neuronal networks related to the motor system in which developmental changes are present.

There are several hypotheses that could explain territory size in mammals, including the resource dispersion hypothesis (RDH), the intruder pressure hypothesis (IPH), and the intraguild predation hypothesis (IGPH). In this study, we tested predictions of these three hypotheses regarding territories of 19 packs of endangered African wild dogs (Lycaon pictus) over 2 years in the Kruger National Park, South Africa. If territory size was supported by the RDH, then we would observe (1) wild dog territories would be larger when resource patches were more dispersed, (2) pack sizes would be larger when resource patches were rich, and (3) pack size would not affect territory size. If supported by the IPH, then we would observe (4) larger territories would experience less intrusions, and (5) there would be an increase in territory overlap in areas of low resource dispersion. Finally, if supported by the IGPH, we would observe (6) territories would be larger in areas of higher lion (Panthera leo) density, as evidence of a spatial avoidance strategy. We found that the IGPH was fully supported (6), the IPH half supported (5), and the RDH partially supported (1 and 3), where we found spatial partitioning of wild dogs with lions, potentially mediated by resources and territory overlap with conspecifics. Ultimately, our results show that subordinate carnivores must balance a trade-off between dominant interspecific competitors and conspecifics to successfully coexist in areas with dominant carnivores.
