Rapamycin persistently improves cardiac function in aged, male and female mice, even following cessation of treatment
Even in healthy aging, cardiac morbidity and mortality increase with age in both mice and humans. These effects include a decline in diastolic function, left ventricular hypertrophy, metabolic substrate shifts, and alterations in the cardiac proteome. Previous work from our laboratory indicated that short‐term (10‐week) treatment with rapamycin, an mTORC1 inhibitor, improved measures of these age‐related changes. In this report, we demonstrate that the rapamycin‐dependent improvement of diastolic function is highly persistent, while decreases in both cardiac hypertrophy and passive stiffness are substantially persistent 8 weeks after cessation of an 8‐week treatment of rapamycin in both male and female 22‐ to 24‐month‐old C57BL/6NIA mice. The proteomic and metabolomic abundance changes that occur after 8 weeks of rapamycin treatment have varying persistence after 8 further weeks without the drug. However, rapamycin did lead to a persistent increase in abundance of electron transport chain (ETC) complex components, most of which belonged to Complex I. Although ETC protein abundance and Complex I activity were each differentially affected in males and females, the ratio of Complex I activity to Complex I protein abundance was equally and persistently reduced after rapamycin treatment in both sexes. Thus, rapamycin treatment in the aged mice persistently improved diastolic function and myocardial stiffness, persistently altered the cardiac proteome in the absence of persistent metabolic changes, and led to persistent alterations in mitochondrial respiratory chain activity. These observations suggest that an optimal translational regimen for rapamycin therapy that promotes enhancement of healthspan may involve intermittent short‐term treatments. Cardiac morbidity and mortality increase with age in both mice and humans. Here, we demonstrate that 8‐week treatment with rapamycin is sufficient to persistently improve diastolic function, decrease age‐related cardiac hypertrophy and passive stiffness, and modulate the cardiac proteome, even a further 8 weeks after cessation of treatment. These observations suggest that an optimal translational regimen for rapamycin therapy that promotes enhancement of healthspan may involve intermittent short‐term treatments.
Late-life restoration of mitochondrial function reverses cardiac dysfunction in old mice
Diastolic dysfunction is a prominent feature of cardiac aging in both mice and humans. We show here that 8-week treatment of old mice with the mitochondrial targeted peptide SS-31 (elamipretide) can substantially reverse this deficit. SS-31 normalized the increase in proton leak and reduced mitochondrial ROS in cardiomyocytes from old mice, accompanied by reduced protein oxidation and a shift towards a more reduced protein thiol redox state in old hearts. Improved diastolic function was concordant with increased phosphorylation of cMyBP-C Ser282 but was independent of titin isoform shift. Late-life viral expression of mitochondrial-targeted catalase (mCAT) produced similar functional benefits in old mice and SS-31 did not improve cardiac function of old mCAT mice, implicating normalizing mitochondrial oxidative stress as an overlapping mechanism. These results demonstrate that pre-existing cardiac aging phenotypes can be reversed by targeting mitochondrial dysfunction and implicate mitochondrial energetics and redox signaling as therapeutic targets for cardiac aging.
Effect of tourist satisfaction on revisit intention in Can Tho City, Vietnam
The research goal is to identify the essential components of visitor satisfaction that influence future return intentions. The quantitative method and the deductive approach were implemented with a sample size of 300 tourists in Can Tho city, Vietnam after sending the survey questionnaire to 15 experts and receiving positive feedback within two weeks. The researchers used the main method to evaluate the data, which was structural equation modeling. The findings illustrated those three factors including cultural contact, perceived value, and green practices noticeably impact visitor satisfaction leading to their intention to visit again, with cultural contact being the most significant influence. At the same time, this helps researchers better understand the intention of tourists to return. Service quality and novelty seeking have negligible influence on tourists’ willingness to revisit the location. The valuable data of this study can inform tourism and hotel managers about their customer groups by encouraging visitors to engage in value-creating activities, cultural exposure and promotion of ideas related to green tourism. On the other side, this study contributes hospitality-related material resources for the following research and provides recommendations to create strategies for tourism regulators and hospitality businesses to promote visitor satisfaction and return.This study aims to identify the essential components of tourist satisfaction that encourage repeat visits. The questionnaire was reviewed and received good feedback from more than ten experts after which the survey was conducted with 300 tourists coming to Can Tho city, Vietnam. Through structural equation modelling, the findings illustrated those three factors including cultural contact, perceived value, and green practices noticeably impact visitor satisfaction leading to their intention to visit again, with cultural contact being the most significant influence. This also helps academics understand visitor return intentions. Tourists’ inclination to return is unaffected by service quality and novelty. This study’s findings can help tourism and hotel management understand their customers by encouraging value-creating events, cultural exposure, and green tourism initiatives. This study provides necessary resources for future research and recommends strategies for tourism regulators and hospitality firms to increase tourist happiness and repeat visits.
Alginate oligosaccharides can maintain activities of lysosomes under low pH condition
The objective of this study was to report that lysosome extracted from egg white could be used as a drug through oral administration for treating diseases by using pH sensitive alginate oligosaccharides. Lysosome-alginate oligosaccharides composite were formulated for oral administration of lysosomes. The dissolution test confirmed the availability of the oral dosage form. When lysosome were used as an independent drug, the activity of protein was lost due to influence of low pH. Its antibacterial activity was also remarkably reduced. However, when lysosome-alginate oligosaccharides composite form was used, antimicrobial activity of lysozyme was maintained. At low pH, a gel-like matrix was formed by alginate oligosaccharides to protect the lysosome. When the pH was increased, alginate oligosaccharides were dissolved and the lysosome was released. SDS–polyacrylamide gel electrophoresis analysis of released lysosomes revealed that alginate oligosaccharide could effectively protect the lysosome from degradation or hydrolysis under acidic conditions for at least 2 h. The results of this study are important for application of lysosomes as therapeutic agents, and also it was confirmed that alginate oligosaccharides have potential as direct delivery system for the oral application of protein derived therapies.
Determinants of green consumer behavior: A case study from Vietnam
The rapid upward thrust of the economy has harmed the environment, which has been compounded by pollution. More and more people are becoming concerned about their surroundings and looking for strategies to mitigate their unfavorable consequences. This is seen in governments' and enterprises' determination to expand and produce environmentally friendly products, as well as raise demand for green products. The purpose of this research is to uncover characteristics influencing green consumption behavior in Vietnam and to determine the most powerful motivator that drives customers to buy green products. The quantitative technique was applied in this investigation, with 231 samples obtained via an online survey. To analyze and assess the acquired data, several methods such as Cronbach's Alpha, EFA, SEM, One-way ANOVA, and Independent Sample T-test were used using SPSS and Amos software. According to the findings, three elements, including Attitudes, Social Norms, and Environmental Concerns, have a beneficial effect on green consumption behavior in Vietnam, with environmental concerns playing a particularly important role. The latest findings offered meaningful data about buyer purchasing characteristics, allowing businesses to expand green goods and devise marketing practices to get more customers to make use of them.
Fixed-term contracts and firm productivity: Do workers' skills and firm conversion rates from fixed-term to permanent contracts matter?
PurposeWe aim to elucidate the relationship between fixed-term employment and firm productivity by examining workers’ skills and considering how firm-level conversion rates influence this relationship.Design/methodology/approachWe use longitudinal employer-employee data between 2011 and 2017 in the Netherlands to estimate a nonlinear regression derived from a production function proposed by Addessi (2014) and Castellani et al. (2020).FindingsThe contribution of fixed-term contracts to firm-level productivity is less than that of permanent contracts. However, this contribution is greater when firms exhibit a high conversion rate from fixed-term to permanent positions. The effect of the conversion rate is more substantial for high-skilled fixed-term workers than for low-skilled ones.Originality/valueOur results suggest the extent to which firms benefit from fixed-term contracts when these are used for screening high-skilled workers for permanent employment.
Parents’ Intentions of Human Papillomavirus Vaccination for Students in Vietnam: A Cross-Sectional Study
Background Human papillomavirus (HPV) is known as a common agent of sexually transmitted infections and cervical cancer. One of the most effective ways for parents to protect their children from HPV is by ensuring they receive vaccinations. Aim To determine the percentage of parents who intend to vaccinate their children against HPV and associated factors. Method A cross-sectional study was conducted on 365 parents who had children attending high school in Ha Tinh province, Vietnam, from April to May 2023, using stratified and random sampling methods. Data were collected by a self-administered questionnaire designed based on previous studies and the domains of the Theory of Planned Behavior and Health Belief Model. A multivariable logistic regression was performed to determine the association between several factors and vaccination status. Result A total of 365 participants took part in the study. The rate of parents intending to vaccinate their children against HPV was 55.9%. Knowledge about the HPV disease and vaccine (all P < .05) and the attitude of parents (P < .001) were determined as the motivation factors that affect the intention to vaccinate children against HPV. Conclusion Many parents still do not have the intention to vaccinate children against HPV. Health education communication should focus on the motivation factors, not only to improve the parents’ knowledge and perspective but also to increase the coverage of the vaccine to prevent cancers caused by HPV. Knowledge, Attitude, and Practices Regarding COVID-19 Among Chronic Illness Patients at Outpatient Departments in Ho Chi Minh City, Vietnam
The novel coronavirus disease (COVID-19) has become a major threat to human life around the world. This study aims to assess the knowledge, attitude, and practices regarding COVID-19 among people with chronic diseases at the outpatient departments in Ho Chi Minh City. A cross-sectional study was carried out between February and March 2020 using a convenience sampling strategy in three hospitals in Ho Chi Minh City (HCMC) via the use of a structured self-administered questionnaire. Factors relating to practices, prevalence ratio (PR), and 95% confidence interval were estimated by using the Poisson regression with robust options. P-value <0.05 was considered as statistically different. A total of 522 participants had a mean age of 51.5 ± 10.6 years. Most of them reported seeing information regarding the COVID-19 pandemic (93.7%) via television and social media (72.8% and 62.1%, respectively). Just over two-thirds of the participants (68.4%) answered with sufficient knowledge of COVID-19. Most respondents had a positive attitude toward COVID-19 (90.8%), although some misconceptions existed. Almost over three-fourths of them (77.2%) maintained good practices for prevention. The rate of good practices in those who had sufficient knowledge was 1.24 times greater than that among those who had insufficient knowledge (PR 1.24, 95% CI: 1.10-1.41, P<0.05). Also, the rate of good practices in males was lower than that of females (PR: 0.91, 95% CI: 0.83-0.99, P<0.05). There still exists an amount of insufficient knowledge and negative attitude regarding COVID-19, which may be barriers to good prevention practices among chronic illness patients. Education programs need to continue via television and social media and emphasize that people with chronic diseases are more likely to experience severe symptoms, including death from COVID-19. Additionally, management authorities should prolong specific policies to protect the more vulnerable in our community.
Predictors of Intention to Get a COVID-19 Vaccine of Health Science Students: A Cross-Sectional Study
Vaccination is one of the interventions that can be employed to control coronavirus disease (COVID-19). The aim of this study is to identify factors that affect vaccination intention among health science students, by using the health belief model (HBM) as a theoretical framework. This study was undertaken in April 2021 using a convenience sample strategy among health science students in Tra Vinh University, which is a university in South Vietnam. Participants were required to complete the self-report questionnaire. All data were calculated by using STATA 14 software, to determine the factors associated with COVID-19 vaccine acceptance, a multivariable logistic regression analysis was performed, along with odds ratio (OR) and 95% confidence interval (95% CI). A total of 412 eligible participants were taken part in the study, with a questionnaire response rate of 48.2%, reporting that 77.7% of participants intended to receive a COVID-19 vaccination when available. An overall score of knowledge was 7.8 ± 1.5 (range 0-10), a positive attitude toward vaccination including perceived susceptibility and severity (3.1 ± 0.7), perceived benefits (3.1 ± 0.7) and cues to action (4.0 ± 0.6). Determinants of willingness to get a COVID-19 vaccination included their field of education, knowledge and factors in HBM model including perceived susceptibility and severity, perceived benefits and cues to action (all p < 0.05). In contrast, perceived barriers had a negative association with the probability of vaccine acceptance (p < 0.05). The study did indicate a number of participants not willing to receive a vaccination due to, mainly, safety (67.4%), development issues (58.7%) and side effects (79.3%). The theoretical framework recognized factors that influence vaccine acceptance. These are considered evidence for the development of planning strategies to optimize the vaccination uptake and offer success in vaccination campaigns for both health science students and the general population.
Multinational firms and the quest for global talent: Employing (skilled) foreign workers at home and abroad
Multinational firms can access global talent in two ways: by employing migrants in their home country, or by employing foreign workers in their overseas affiliates. Taking a knowledge-based perspective, we conceptualize these employment decisions as simultaneous and subject to management coordination. Substitution effects are greater when there is a larger wage cost differential between home and host countries, leading to a cost-reduction motivation for foreign expansion and the offshoring of employment. Substitution also occurs when R&D intensive firms employ highly skilled and internationally mobile foreign workers and employ these where the worker’s knowledge and skills can be most productively put to use. In contrast, a complementary relationship occurs when the migrant country exhibits a high contextual distance with the home country of the firm, leading to knowledge (diversity) benefits of migrant employment at home when expanding abroad. Analyzing employee–employer and foreign affiliate data for multinational firms in the Netherlands (2008–2016) and estimating simultaneous equation models, we find support for these hypotheses. Our findings suggest that policies that restrict immigration may have a negative impact on the competitiveness of home-country multinational firms by limiting their ability to engage in value enhancing coordination of domestic and foreign employment growth.
