Effects on enzyme activity and DNA integrity in rainbow trout Oncorhynchus mykiss exposed to fish farm effluents
Fish farm effluents are known to affect water quality and freshwater ecosystems, potentially harming non-target organisms and ecosystem processes. We studied the effect of fish farm effluents at different concentrations (3.125-100% v/v) on catalase (CAT) and glutathione S -transferase (GST) activity as well as the DNA integrity of Oncorhynchus mykiss fry over 24-120 h. Biochemical responses and DNA damage analysis were conducted to assess the impact. We found that fish farm effluent had higher conductivity, nitrate, nitrite, and total dissolved solids concentrations downstream compared to upstream of the farm. Interestingly, no antibiotics were detected in the effluent. CAT activity significantly increased in the fish liver at concentrations of 12.5, 50, and 100% of the effluent after 72 h. In the gills, a significant increase was observed at concentrations ranging from 6.25 to 100% of the effluent after both 24 and 72 h. GST activity increased significantly in the liver at a concentration of 100% of the effluent after 72 h and in the gills at concentrations of 25, 50, and 100% after 24 h, with a decrease noted at higher concentrations. DNA damage assessment revealed significant DNA strand breaks in blood cells at concentrations of 12.5, 25, 50, and 100% of the effluent after 120 h of exposure. The results demonstrate that fish farm effluents can induce oxidative stress, causing damage to DNA integrity in blood cells. Our findings emphasize the potential ecological risks posed by fish farm effluents to aquatic organisms.
Relationship between salt use in fish farms and drift of macroinvertebrates in a freshwater stream
In Chile, salt (NaCl) use per salmon fish farm ranges between 20−30 t yr⁻¹ and is used to prevent and control fungal infections. An increase in salinity in freshwater can have adverse effects on freshwater biodiversity and ecosystem functions and services. We studied the effects of fish-farm effluents on benthic macroinvertebrate communities in a northern Patagonian stream (Chile). Benthic samples were collected at 3 sites near a land-based salmon aquaculture facility (one located 100 m upstream from the fish-farm outlet for effluent, 2 sites located 200 and 400 m downstream from the effluent source). We found changes in benthic macroinvertebrate communities downstream from the ef fluent, with higher abundances of tolerant taxa and lower abundances of sensitive taxa, which was related to nutrient and salt concentration in the water. We also studied the effects of salinity on macroinvertebrate drift in a mesocosm experiment conducted in recirculating channels, measuring the drift of 2 salt-sensitive macroinvertebrates (Andesiops peruvianus and Smicridea annulicornis), collected from an unpolluted northern Patagonian stream, after exposure to a range of salinity concentration pulses similar to those from fish farms. Our results demonstrate that (1) fish-farm effluent can alter stream macroinvertebrate community composition and dynamics, and (2) such effects are at least partly driven by high salt concentrations in effluent waters.
Effect of housefly maggot meal (magmeal) diets on catalase, and glutathione S-transferase in the liver and gills of carp Cyprinus carpio fingerling
Effect of housefly maggot meal (magmeal) diets on catalase, and glutathione S-transferase (GST) in the liver and gills of carp, Cyprinus carpio fingerling was studied. Eight iso-nitrogenous diets were formulated containing fishmeal and magmeal and in combination with soy meal to yield 41.0% crude protein dry matter. After 56 days feeding trial, weight gain and specific growth rate (SGR) of carp improved as fish meal was replaced in diets with magmeal up to an incorporation level of 45%. Results from GST indicate that experimental feed components did not contain unwanted chemicals, such as pharmaceutics or pesticides to critical concentrations that would adversely affect experimental fish performance. Inclusion of soy meal in diets did not confer any significant anti-oxidative and biotransformation stress effect on the fish but compromised the quantity of magmeal in carp diets able to bring about good performance when used without soy meal combination. Based on values of SGR, food conversion ratio (FCR) and enzyme activities, including magmeal above the level of 45% (diet A2) and below 67% (diet A3) in carp diets would improve optimal growth performance of carp. At such a level, magmeal is able to supply between 50% and 75% crude protein needed in the carp diet.
Seasonality of food groups and total energy intake: a systematic review and meta-analysis
Background/Objectives: The aim of this systematic review and meta-analysis was to assess the effect of season on food intake from selected food groups and on energy intake in adults. Subjects/Methods: The search process was based on selecting publications listed in the following: Medline, Scopus, Web of Science, Embase and Agris. Food frequency questionnaires, 24-h dietary recalls and food records as methods for assessment of dietary intake were used to assess changes in the consumption of 11 food groups and of energy intake across seasons. A meta-analysis was performed. Results: Twenty-six studies were included. Articles were divided into those reporting data on four seasons (winter, spring, summer and autumn) or on two seasons (pre-and post-harvest). Four of the studies could be utilized for meta-analysis describing changes in food consumption across four season scheme: from winter to spring fruits decreased, whereas vegetables, eggs and alcoholic beverages increased; from spring to summer vegetable consumption further increased and cereals decreased; from summer to autumn fruits and cereals increased and vegetables, meat, eggs and alcoholic beverages decreased; from autumn to winter cereals decreased. A significant association was also found between energy intake and season, for 13 studies reporting energy intake across four seasons (favors winter) and for eight studies across pre- and post-harvest seasons (favors post-harvest). Conclusions: The winter or the post-harvest season is associated with increased energy intake. The intake of fruits, vegetables, eggs, meat, cereals and alcoholic beverages is following a seasonal consumption pattern and at least for these foods season is determinant of intake.
Circulating vitamin D concentration and risk of seven cancers: Mendelian randomisation study
Objective To determine if circulating concentrations of vitamin D are causally associated with risk of cancer.Design Mendelian randomisation study.Setting Large genetic epidemiology networks (the Genetic Associations and Mechanisms in Oncology (GAME-ON), the Genetic and Epidemiology of Colorectal Cancer Consortium (GECCO), and the Prostate Cancer Association Group to Investigate Cancer Associated Alterations in the Genome (PRACTICAL) consortiums, and the MR-Base platform).Participants 70 563 cases of cancer (22 898 prostate cancer, 15 748 breast cancer, 12 537 lung cancer, 11 488 colorectal cancer, 4369 ovarian cancer, 1896 pancreatic cancer, and 1627 neuroblastoma) and 84 418 controls.Exposures Four single nucleotide polymorphisms (rs2282679, rs10741657, rs12785878 and rs6013897) associated with vitamin D were used to define a multi-polymorphism score for circulating 25-hydroxyvitamin D (25(OH)D) concentrations.Main outcomes measures The primary outcomes were the risk of incident colorectal, breast, prostate, ovarian, lung, and pancreatic cancer and neuroblastoma, which was evaluated with an inverse variance weighted average of the associations with specific polymorphisms and a likelihood based approach. Secondary outcomes based on cancer subtypes by sex, anatomic location, stage, and histology were also examined.Results There was little evidence that the multi-polymorphism score of 25(OH)D was associated with risk of any of the seven cancers or their subtypes. Specifically, the odds ratios per 25 nmol/L increase in genetically determined 25(OH)D concentrations were 0.92 (95% confidence interval 0.76 to 1.10) for colorectal cancer, 1.05 (0.89 to 1.24) for breast cancer, 0.89 (0.77 to 1.02) for prostate cancer, and 1.03 (0.87 to 1.23) for lung cancer. The results were consistent with the two different analytical approaches, and the study was powered to detect relative effect sizes of moderate magnitude (for example, 1.20-1.50 per 25 nmol/L decrease in 25(OH)D for most primary cancer outcomes. The Mendelian randomisation assumptions did not seem to be violated.Conclusions There is little evidence for a linear causal association between circulating vitamin D concentration and risk of various types of cancer, though the existence of causal clinically relevant effects of low magnitude cannot be ruled out. These results, in combination with previous literature, provide evidence that population-wide screening for vitamin D deficiency and subsequent widespread vitamin D supplementation should not currently be recommended as a strategy for primary cancer prevention.
Changes in physical activity and sedentary behavior during the first COVID-19 pandemic- restrictions in Germany: a nationwide survey
The COVID-19 pandemic restrictions posed challenges to maintaining healthy lifestyles and physical well-being. During the first mobility restrictions from March to mid-July 2020, the German population was advised to stay home, except for work, exercise, and essential shopping. Our objective was to comprehensively assess the impact of these restrictions on changes in physical activity and sedentary behavior to identify the most affected groups. Between April 30, 2020, and May 12, 2020, we distributed a COVID-19-specific questionnaire to participants of the German National Cohort (NAKO). This questionnaire gathered information about participants' physical activity and sedentary behavior currently compared to the time before the restrictions. We integrated this new data with existing information on anxiety, depressive symptoms, and physical activity. The analyses focused on sociodemographic factors, social relationships, physical health, and working conditions. Out of 152,421 respondents, a significant proportion reported altered physical activity and sedentary behavioral patterns due to COVID-19 restrictions. Over a third of the participants initially meeting the WHO's physical activity recommendation could no longer meet the guidelines during the restrictions. Participants reported substantial declines in sports activities (mean change (M) = -0.38; 95% CI: -.390; -.378; range from -2 to + 2) and reduced active transportation (M = -0.12; 95% CI: -.126; -.117). However, they also increased recreational physical activities (M = 0.12; 95% CI: .117; .126) while engaging in more sedentary behavior (M = 0.24; 95% CI: .240; .247) compared to pre-restriction levels. Multivariable linear and log-binomial regression models indicated that younger adults were more affected by the restrictions than older adults. The shift to remote work, self-rated health, and depressive symptoms were the factors most strongly associated with changes in all physical activity domains, including sedentary behavior, and the likelihood to continue following the physical activity guidelines. Mobility patterns shifted towards inactivity or low-intensity activities during the nationwide restrictions in the spring of 2020, potentially leading to considerable and lasting health risks.
Use of dietary supplements in the European Prospective Investigation into Cancer and Nutrition calibration study
Background: Dietary supplement use is increasing, but there are few comparable data on supplement intakes and how they affect the nutrition and health of European consumers. The aim of this study was to describe the use of dietary supplements in subsamples of the 10 countries participating in the European Prospective Investigation into Cancer and Nutrition (EPIC). Methods: Specific questions on dietary supplement use were asked as a part of single 24-h recalls performed on 36 034 men and women aged 35–74 years from 1995 to 2000. Results: Between countries, the mean percentage of dietary supplement use varied almost 10-fold among women and even more among men. There was a clear north–south gradient in use, with a higher consumption in northern countries. The lowest crude mean percentage of use was found in Greece (2.0% among men, 6.7% among women), and the highest was in Denmark (51.0% among men, 65.8% among women). Use was higher in women than in men. Vitamins, minerals or combinations of them were the predominant types of supplements reported, but there were striking differences between countries. Conclusions: This study indicates that there are wide variations in supplement use in Europe, which may affect individual and population nutrient intakes. The results underline the need to monitor consumption of dietary supplements in Europe, as well as to evaluate the risks and benefits.
Childhood and adolescence factors and multiple sclerosis: results from the German National Cohort (NAKO)
Background Multiple Sclerosis (MS) represents the most common inflammatory neurological disease causing disability in early adulthood. Childhood and adolescence factors might be of relevance in the development of MS. We aimed to investigate the association between various factors (e.g., prematurity, breastfeeding, daycare attendance, weight history) and MS risk. Methods Data from the baseline assessment of the German National Cohort (NAKO) were used to calculate adjusted hazard ratios (HR) and 95% confidence intervals (CI) for the association between childhood and adolescence factors and risk of MS. Analyses stratified by sex were conducted. Results Among a total of 204,273 participants, 858 reported an MS diagnosis. Male sex was associated with a decreased MS risk (HR 0.48; 95% CI 0.41–0.56), while overweight (HR 2.03; 95% CI 1.41–2.94) and obesity (HR 1.89; 95% CI 1.02–3.48) at 18 years of age compared to normal weight were associated with increased MS risk. Having been breastfed for ≤ 4 months was associated with a decreased MS risk in men (HR 0.59; 95% CI 0.40–0.86) compared to no breastfeeding. No association with MS risk was observed for the remaining factors. Conclusions Apart from overweight and obesity at the age of 18 years, we did not observe considerable associations with MS risk. The proportion of cases that can be explained by childhood and adolescence factors examined in this study was low. Further investigations of the association between the onset of overweight and obesity in childhood and adolescence and its interaction with physical activity and MS risk seem worthwhile.
Association between alcohol consumption and serum paraoxonase and arylesterase activities: a cross-sectional study within the Bavarian population
High alcohol consumption is an important risk factor for chronic disease and liver degeneration. Paraoxonase (PON1) and arylesterase (AE) are functions of the enzyme paraoxonase, which is synthesised by the liver. Paraoxonase circulates in plasma bound to HDL and hydrolyses lipid peroxides, protecting lipoproteins against oxidative modification. It has been shown that excessive alcohol consumption leads to a reduction of serum PON1 and AE activities; however, studies investigating the association with low and moderate alcohol consumption are scarce. We investigated the cross-sectional association between alcohol consumption and serum activities of PON1 and AE using data from the population-based Bavarian Food Consumption Survey II survey. PON1 and AE activities were quantified in serum samples of 566 male and female study participants (aged 18–80 years), and dietary intake including alcohol consumption was estimated from three 24-h dietary recalls. The association between alcohol consumption and PON1 and AE activities was analysed using linear regression, adjusted for age, sex and socio-economic status. There was no strong association between alcohol consumption and enzymatic activities of PON1 and AE in the Bavarian population. PON1 activity was seen to be lowest in non-drinkers (0 g/d) and highest in people who consumed 15·1–30 g of alcohol/d. AE activity increased across alcohol consumption categories, with a mean maximum difference of 14 U/ml (P for linear trend 0·04). These associations were attenuated after adjustment for blood concentrations of HDL. The results of this study do not support the hypothesis that alcohol consumption is related to important alterations in PON1 and AE activities.
Development and evaluation of a short 24-h food list as part of a blended dietary assessment strategy in large-scale cohort studies
Background/Objectives: The validity of dietary assessment in large-scale cohort studies has been questioned. Combining data sources for the estimation of usual intake in a blended approach may enhance the validity of dietary measurement. Our objective was to develop a web-based 24-h food list for Germany to identify foods consumed during the previous 24 h and to evaluate the performance of the new questionnaire in a feasibility study. Subjects/Methods: Available data from the German National Nutrition Survey II were used to develop a finite list of food items. A total of 508 individuals were invited to fill in the 24-h food list via the Internet up to three times during a 3–6-month time period. In addition, participants were asked to evaluate the questionnaire using a brief online evaluation form. Results: In total, 246 food items were identified for the 24-h food list, reflecting >75% variation in intake of 27 nutrients and four major food groups. Among the individuals invited, 64% participated in the feasibility study. Of these, 100%, 85% and 68% of participants completed the 24-h food list one, two or three times, respectively. The average time needed to complete the questionnaire was 9 min, and its acceptability by participants was rated as high. Conclusions: The 24-h food list represents a promising new dietary assessment tool that can be used as part of a blended approach combining multiple data sources for valid estimation of usual dietary intake in large-scale cohort studies.
