A Multi-Year Study of GOES-13 Droplet Effective Radius Retrievals for Warm Clouds over South America and Southeast Pacific

Geostationary satellites can retrieve the cloud droplet effective radius (re) but suffer biases from cloud inhomogeneities, internal retrieval nonlinearities, and 3-D scattering/shadowing from neighboring clouds, among others. A 1-D retrieval method was applied to Geostationary Operational Environmental Satellite 13 (GOES-13) imagery, over large areas in South America (5∘ N–30∘ S; 20∘–70∘ W), the Southeast Pacific (5∘ N–30∘ S; 70∘–120∘ W), and the Amazon (2∘ N–7∘ S; 54∘–73∘ W), for four months in each year from 2014–2017. Results were compared against in situ aircraft measurements and the Moderate Resolution Imaging Spectroradiometer cloud product for Terra and Aqua satellites. Monthly regression parameters approximately followed a seasonal pattern. With up to 108,009 of matchups, slope, intercept, and correlation for Terra (Aqua) ranged from about 0.71 to 1.17, −2.8 to 2.5 μm, and 0.61 to 0.91 (0.54 to 0.78, −1.5 to 1.8 μm, 0.63 to 0.89), respectively. We identified evidence for re overestimation (underestimation) correlated with shadowing (enhanced reflectance) in the forward (backscattering) hemisphere, and limitations to illumination and viewing configurations accessible by GOES-13, depending on the time of day and season. A proposition is hypothesized to ameliorate 3-D biases by studying relative illumination and cloud spatial inhomogeneity.

Plasma Proteome Responses in Zebrafish Following λ-Carrageenan-Induced Inflammation are Mediated by PMN Leukocytes and Correlate Highly with their Human Counterparts

Regulation of inflammation is a critical process for maintaining physiological homeostasis. The λ-carrageenan (λ-CGN) is a mucopolysaccharide extracted from the cell wall of red algae (Chondrus crispus) capable of inducing acute intestinal inflammation, which is translated into the production of acute phase reactants secreted into the blood circulation. However, the associated mechanisms in vertebrates are not well understood. Here, we investigated the crucial factors behind the inflammatory milieu of λ-CGN-mediated inflammation administered at 0, 1.75, and 3.5% (v/w) by i.p. injection into the peritoneal cavity of adult zebrafish (ZF) (Danio rerio). We found that polymorphonuclear leukocytes (neutrophils) and lymphocytes infiltrating the ZF peritoneal cavity had short-term persistence. Nevertheless, they generate a strong pattern of inflammation that affects systemically and is enough to produce edema in the cavity. Consistent with these findings, cell infiltration, which causes notable tissue changes, resulted in the overexpression of several acute inflammatory markers at the protein level. Using reversed-phase high-performance liquid chromatography followed by a hybrid linear ion-trap mass spectrometry shotgun proteomic approach, we identified 2938 plasma proteins among the animals injected with PBS and 3.5% λ-CGN. First, the bioinformatic analysis revealed the composition of the plasma proteome. Interestingly, 72 commonly expressed proteins were recorded among the treated and control groups, but, surprisingly, 2830 novel proteins were differentially expressed exclusively in the λ-CGN-induced group. Furthermore, from the commonly expressed proteins, compared to the control group 62 proteins got a significant (p<0.05) upregulation in the λ-CGN-treated group, while the remaining ten proteins were downregulated. Next, we obtained the major protein-protein interaction networks between hub protein clusters in the blood plasma of the λ-CGN induced group. Moreover, to understand the molecular underpinnings of these effects based on the unveiled protein sets, we performed a bioinformatic structural similarity analysis and generated overlapping 3D reconstructions between ZF and humans during acute inflammation. Biological pathway analysis pointed to the activation and abundance of diverse classical immune and acute phase reactants, several catalytic enzymes, and varied proteins supporting the immune response. Together, this information can be used for testing and finding novel pharmacological targets to treat human intestinal inflammatory diseases.

IntroductionThe aim of this study was to evaluate the clinical, epidemiological and laboratory aspects of SARS-CoV-2 infection during pregnancy and postpartum in 16 maternity hospitals.Methods and analysisA prospective multicentre study, with five axes. First, the prevalence of SARS-CoV-2 infection among women admitted for childbirth will be described in a cross-sectional study. Second, maternal and perinatal outcomes will be assessed in a prospective cohort study including pregnant or postpartum women with suspected COVID-19. Third, a cohort of positive COVID-19 cases with sampling of a variety of biological material. Histopathological and viral analysis of biological maternal and neonatal samples will be performed, and the assessment of nutritional variables to evaluate the association between vitamin D and severity of infection. Fourth, a monitoring and evaluation committee to collect relevant healthcare information and plan actions in centres facing the pandemic. Furthermore, qualitative studies will be performed to study pregnant women, their families and health professionals. Fifth, an ecological study will monitor the number of live births, stillbirths and other outcomes to explore any trend among the periods before, during and after the pandemic. Data will systematically be collected in an electronic platform following standardised operational procedures. For quantitative study components, an appropriate statistical approach will be used for each analysis. For qualitative data, in-depth interviews recorded in audio will be transcribed, checking the text obtained with the recording. Subsequently, thematic analysis with the aid of the NVivo programme will be performed.Ethics and disseminationEthical approval was obtained (letters of approval numbers 4.047.168, 4.179.679 and 4.083.988). All women will be fully informed to sign the consent form before enrolment in the study. Findings will be disseminated through peer-reviewed journals and scientific conferences.

AimTo investigate the prevalence of alcohol and tobacco consumption as well as behavioral and socio-demographic aspects among undergraduate students in the healthcare area of a Brazilian private university.Subjects and methodsA cross-sectional study was conducted with undergraduate students in health sciences courses, with randomized probabilistic sampling proportional to the number of students in each course. Two trained researchers, by means of a socio-demographic and behavioral questionnaire, carried out the data collection. One questionnaire related to the smoking habit and the other the Alcohol Use Disorder Identification Test. The data were entered and organized in SPSS software, version 20.0 for Windows (Statistical Package for the Social Sciences), presented through descriptive (absolute and percentage distribution) and analytic statistics.ResultsThe total sample was composed of 317 students (29.7% male and 70.3% female). The prevalence rate of nicotine consumption was 3.2% and hazardous alcohol consumption 31.9% among university students. It was verified that the habit of smoking was more prevalent among male students (p = 0.007). The hazardous use of alcohol was proportionally greater among male undergraduates (p < 0.001) in the physical education course (p = 0.003) who were smokers (p < 0.001).ConclusionThe prevalence of smoking was low, with the majority of smokers being male, while the hazardous use of alcohol among university students was high, also predominantly among males.

Are remote sensing evapotranspiration models reliable across South American climates and ecosystems?

Many remote sensing-based evapotranspiration (RSBET) algorithms have been proposed in the past decades and evaluated using flux tower data, mainly over North America and Europe. Model evaluation across South America has been done locally or using only a single algorithm at a time. Here, we provide the first evaluation of multiple RSBET models, at a daily scale, across a wide variety of biomes, climate zones, and land uses in South America. We used meteorological data from 25 flux towers to force four remote sensing based ET models: Priestley & Taylor Jet Propulsion Laboratory (PT-JPL), Global Land Evaporation Amsterdam Model (GLEAM), Penman-Monteith Mu model (PM-MOD), and Penman-Monteith Nagler model (PM-VI). ET was predicted satisfactorily by all four models, with correlations consistently higher (R²>0.6) for GLEAM and PT-JPL, and PM-MOD and PM-VI presenting overall better responses in terms of PBIAS (-10

The renin-angiotensin system (RAS) comprises a biochemical cascade that hydrolyzes angiotensinogen into several different bioactive peptides, which can activate different receptors promoting plenty of specific effects. The aim of this study was to evaluate the presence of the putative product of alamandine, the pentapeptide alamandine-(1-5) in the circulation and its biological activity. To accomplish this we have used mass spectrometry (MALDI/TOF/TOF, LC-MS/MS) and several methodologies including isolated blood vessels, isolated perfused hearts, isolated cardiomyocytes, blood pressure recording in freely-moving normotensive and hypertensive rats (SHR), high resolution echocardiography (VEVO 2100), central administration (ICV infusion and microinjection in the insular cortex), cell culture (endothelial cells and GPCR-transfected CHO cells) and wild type and Mas, MrgD or AT2R deficient mice. Our results show that alamandine-(1-5) circulates in the human and rodent blood and promotes many biological central and peripheral actions. More importantly, its plasma concentration is increased in pediatric nephropathic patients. A major role for plasma ACE activity in the formation of alamandine-(1-5) from alamandine was observed using plasma samples from Angiotensinogen-KO mice. Alamandine-(1-5) increased Baroreflex sensitivity and produced a long-lasting (∼6 hours) anti-hypertensive effect in SHR, associated with a significant reduction in cardiac output. A particularly important effect of this pentapeptide was observed in isolated perfused heart and cardiomyocyte contractility (reduced inotropism). It was capable of stimulating NO production through all receptors from the renin-angiotensin protective arm, (MAS, MrgD and AT2R) in CHO-transfected cells. Our data shows that Alamandine-(1-5) exhibits selective actions that set it apart from traditional concepts of the vasodilatory axis of the RAS and that are possibly intricately linked to a complex interplay between Mas, MrgD and AT2 receptors. This novel finding suggests that RAS may possess a complexity that surpasses our current understanding.

Summary Establishing new experimental animal models to assess the safety and immune response to the antigen used in the development of COVID-19 vaccine is an imperative issue. Based on the advantages of using zebrafish as a model in research, herein we suggest doing this to test the safety of the putative vaccine candidates and to study immune response against the virus. We produced a recombinant N-terminal fraction of the Spike SARS-CoV-2 protein and injected it into adult female zebrafish. The specimens generated humoral immunity and passed the antibodies to the eggs. However, they presented adverse reactions and inflammatory responses similar to severe cases of human COVID-19. The analysis of the structure and function of zebrafish and human Angiotensin-converting enzyme 2, the main human receptor for virus infection, presented remarkable sequence similarities. Moreover, bioinformatic analysis predicted protein-protein interaction of the Spike SARS-CoV-2 fragment and the Toll-like receptor pathway. It might help in the choice of future therapeutic pharmaceutical drugs to be studied. Based on the in vivo and in silico results presented here, we propose the zebrafish as a model for translational research into the safety of the vaccine and the immune response of the vertebrate organism to the SARS-CoV-2 virus.
