Regional variety in quality of life in Poland

Research background: Quality of life is viewed as the degree of people?s satisfaction of various needs, especially including material, spiritual, safety-related needs and life aspirations. The aspects taken into consideration are economic, spatial, environmental and cultural ones. Quality of life is therefore determined by various conditions in which a human lives and the source of which is the hierarchy of needs and a value system. At the same time, quality of life can be viewed in terms of collectivity (objective aspect) and an individual?s evaluation (subjective aspect). The former view shapes the existing living conditions and the latter results from the satisfaction from a specific person?s life, experiences and ambitions. The multidimensionality of the quality of life categories translates into difficulties in defining and measuring it, and constitutes an interesting research object. Purpose of the article: The purpose of the article is to diagnose and evaluate the variety of the quality of life in Polish voivodeships, in objective terms by building a synthetic measure allowing for estimating inequalities in the quality of life of regional communities. It was assumed that quality of life will be characterised by indicators grouped into the following areas: material living conditions, economic activity, health, education and living environment quality. Methods: The main method used for measuring quality of life is the linear ordering based on Z. Hellwig?s taxonomic measure which allows for organising voivodeships in terms of the quality of life of communities. Additionally, the cluster analysis (Ward?s agglomerative grouping method) was used, which allowed for distinguishing voivodeships similar in terms of the analysed phenomenon. Findings & Value added: During the realisation of the research goal, special attention was paid to the complexity of the quality of life issue and its measurement in terms of economy and rather important varieties in voivodeships were emphasized. The conducted analysis allowed for distinguishing groups of regions with the highest, high, low and the lowest quality of life.

Climate and energy policies are the key areas of the European Union’s development aspirations in the international arena. The European Green Deal sets ambitious new goals in this respect, emphasizing clean energy. The development of the energy sector, relying chiefly on renewable sources, is one of the conditions for the EU to transition to energies that do not produce waste that pollutes or is otherwise harmful to the Earth and human health. This paper assesses the development of renewable sources of energy in the EU and identifies challenges to the realization of the EU’s energy priorities. An analysis of renewable sources of energy in the EU in 2012 and 2020 is the prime method. The generation of primary energy from renewable sources, the share of energy from renewable sources in the final energy consumption, the structure of energy generation from renewable sources, and the share of energy from renewable sources in the energy used by the transport sector are discussed. Secondary data are employed, made available by Eurostat and Statistics Poland. To reach the objective of the study, Z. Hellwig’s taxonomic method is applied to assess the development of renewable energy sources (RES). Cluster analysis (Ward’s agglomerative clustering method) is additionally used for separation of objects and member states that are similar with regard to the phenomenon reviewed. Particular attention is paid to the challenges that societies and economies face in view of the assumptions of the EU’s new Green Deal, which envisages a balancing of CO2 emissions and absorption. This analysis discovers varied progress on the development of RES in the member states, posing a threat to the European community’s aspirations.

Passive coupling of membrane tension and cell volume during active response of cells to osmosis

During osmotic changes of their environment, cells actively regulate their volume and plasma membrane tension that can passively change through osmosis. How tension and volume are coupled during osmotic adaptation remains unknown, as their quantitative characterization is lacking. Here, we performed dynamic membrane tension and cell volume measurements during osmotic shocks. During the first few seconds following the shock, cell volume varied to equilibrate osmotic pressures inside and outside the cell, and membrane tension dynamically followed these changes. A theoretical model based on the passive, reversible unfolding of the membrane as it detaches from the actin cortex during volume increase quantitatively describes our data. After the initial response, tension and volume recovered from hypoosmotic shocks but not from hyperosmotic shocks. Using a fluorescent membrane tension probe (fluorescent lipid tension reporter [Flipper-TR]), we investigated the coupling between tension and volume during these asymmetric recoveries. Caveolae depletion and pharmacological inhibition of ion transporters and channels, mTORCs, and the cytoskeleton all affected tension and volume responses. Treatments targeting mTORC2 and specific downstream effectors caused identical changes to both tension and volume responses, their coupling remaining the same. This supports that the coupling of tension and volume responses to osmotic shocks is primarily regulated by mTORC2.

Based on a targeted analysis performed by LC–ESI–MS/MS, based on multiple reaction monitoring, it was found that the dissipation kinetics of the parent compound and its metabolites are compatible with a first-order reaction mechanism. Moreover, it has been proven that formed NTBC metabolites, such as CHD, cause a decrease in
l
-tyrosine,
l
-tryptophan, and
l
-phenylalanine concentrations by about 34%, 59% and 51%, respectively, compared to the untreated model organism. The overall changes in the metabolism of yeast exposed to NTBC or its derivatives were evaluated by non-targeted analysis via LC–ESI–MS/MS in the ion trap scanning mode. Based on principal components analysis, a statistically significant similarity between metabolic responses of yeast treated with ATFA or NTFA was observed. These findings facilitate further studies investigating the influence of NTBC on the human body and the mechanism of its action.

The association between night shift work and nutrition patterns among nurses: a literature review

The shift work system may affect the temporal distribution of eating and diet quality. The paper aimed at reviewing a body of research examining the associations between night shift work and dietary habits among nurses. Data from the PubMed and Google Schoolar databases, as well as references lists in selected papers were searched. The authors used the following keywords: nurses, shift work, diet, nutrition. Papers published in English or Polish were selected for the review, and as many as 19 papers published in 2000-2017 were eventually identified. The studies varied greatly with respect to the study size, subjects' age and the duration of night shift work. The major problem was the heterogeneity of the tools used for dietary assessment. Self-administered questionnaires were used and analyses were rarely adjusted for confounders. Alcohol consumption was the most frequently analyzed aspect (N = 8 studies), followed by the total energy (N = 7), protein, fat (N = 6), and carbohydrate intake, coffee and fruit consumption (N = 5). The results showed quite a consistent association of night work with higher coffee (caffeine) consumption, as well as lower alcohol, and fruit and vegetables consumption. Few studies also reported more frequent snacks consumption, later time of the last meal, eating at night, meals irregularity, and a poorer diet quality among night shift nurses when compared to the reference. The review showed some poor nutritional habits among nurses working night shifts. Novel coronavirus-like particles targeting cells lining the respiratory tract

Virus like particles (VLPs) produced by the expression of viral structural proteins can serve as versatile nanovectors or potential vaccine candidates. In this study we describe for the first time the generation of HCoV-NL63 VLPs using baculovirus system. Major structural proteins of HCoV-NL63 have been expressed in tagged or native form, and their assembly to form VLPs was evaluated. Additionally, a novel procedure for chromatography purification of HCoV-NL63 VLPs was developed. Interestingly, we show that these nanoparticles may deliver cargo and selectively transduce cells expressing the ACE2 protein such as ciliated cells of the respiratory tract. Production of a specific delivery vector is a major challenge for research concerning targeting molecules. The obtained results show that HCoV-NL63 VLPs may be efficiently produced, purified, modified and serve as a delivery platform. This study constitutes an important basis for further development of a promising viral vector displaying narrow tissue tropism.

The New Test Procedure for Group-Type Composition of Base Oils of Lubricating Oils, Especially Emitted into the Environment

Mineral base oil, a product of multistep fine refining of the relevant fractions from vacuum distillation of crude oil, is a main component of so-called mineral lubricating oils containing aliphatic and alicyclic substituted aromatic hydrocarbons, i.e., derivatives of benzene and biphenyl. Mineral lubricating oil is composed mostly of mineral base oil and a low amount of enriching additives, most often products of advanced organic chemical technology. The application of mineral lubricating oils in open cutting systems has a very negative impact on environment and on the operator’s health. This work presents a simple, cheap and fast methodology allows identification of the group-type composition of base oil in lubricating oil and to estimate the content or total absence of base oil of mineral and vegetable origin in lubricating oil. The first step of the test is an in-situ screening for fluorescence of petroleum fraction under the 365 nm light. The next is the performance of infrared spectra with Fourier transformation (FT-MIR) to identify and estimate the content of vegetable oil and its derivatives and the performance of UV-Vis spectra to identify and determine the content of aromatic hydrocarbons, as well as dyes present in the lubricating oil. The last stage is normal phase thin layer chromatography (NP-TLC) using different visualization methods to evaluate the group-type composition of lubricating oil. Effectivity of the developed procedure has been confirmed during control of group-type composition evaluation of lubricating oils in cutting systems. The procedure can be also applied with respect to different oil matrices.

Over the past two decades, regulatory B cells (Breg cells or Bregs) have emerged as an immunosuppressive subset of B lymphocytes playing a key role in inflammation, infection, allergy, transplantation, and cancer. However, the involvement of Bregs in various pathological conditions of the gastrointestinal tract is not fully understood and is the subject of much recent research. In this review, we aimed to summarize the current state of knowledge about the origin, phenotype, and suppressive mechanisms of Bregs. The relationship between the host gut microbiota and the function of Bregs in the context of the disturbance of mucosal immune homeostasis is also discussed. Moreover, we focused our attention on the role of Bregs in certain diseases and pathological conditions related to the digestive tract, especially Helicobacter pylori infection, parasitic diseases (leishmaniasis and schistosomiasis), and gastrointestinal neoplasms. Increasing evidence points to a relationship between the presence and number of Bregs and the severity and progression of these pathologies. As the number of cases is increasing year by year, also among young people, it is extremely important to understand the role of these cells in the digestive tract.

Synthetic lethality between VPS4A and VPS4B triggers an inflammatory response in colorectal cancer

Somatic copy number alterations play a critical role in oncogenesis. Loss of chromosomal regions containing tumor suppressors can lead to collateral deletion of passenger genes. This can be exploited therapeutically if synthetic lethal partners of such passenger genes are known and represent druggable targets. Here, we report that
VPS4B
gene, encoding an ATPase involved in ESCRT‐dependent membrane remodeling, is such a passenger gene frequently deleted in many cancer types, notably in colorectal cancer (CRC). We observed downregulation of
VPS4B
mRNA and protein levels from CRC patient samples. We identified
VPS4A
paralog as a synthetic lethal interactor for
VPS4B in vitro
and in mouse xenografts. Depleting both proteins profoundly altered the cellular transcriptome and induced cell death accompanied by the release of immunomodulatory molecules that mediate inflammatory and anti‐tumor responses. Our results identify a pair of novel druggable targets for personalized oncology and provide a rationale to develop VPS4 inhibitors for precision therapy of VPS4B‐deficient cancers.

Comparison and Optimization of Saccharification Conditions of Alkaline Pre-Treated Triticale Straw for Acid and Enzymatic Hydrolysis Followed by Ethanol Fermentation

This paper concerns the comparison of the efficiency of two-stage hydrolysis processes, i.e., alkaline pre-treatment and acid hydrolysis, as well as alkaline pre-treatment followed by enzymatic hydrolysis, carried out in order to obtain reducing sugars from triticale straw. For each of the analyzed systems, the optimization of the processing conditions was carried out with respect to the glucose yield. For the alkaline pre-treatment, an optimal catalyst concentration was selected for constant values of temperature and pre-treatment time. For enzymatic hydrolysis, optimal process time and concentration of the enzyme preparation were determined. For the acidic hydrolysis, performed with 85% phosphoric acid, the optimum temperature and hydrolysis time were determined. In the hydrolysates obtained after the two-stage treatment, the concentration of reducing sugars was determined using HPLC. The obtained hydrolysates were subjected to ethanol fermentation. The concentrations of fermentation inhibitors are given and their effects on the alcoholic fermentation efficiency are discussed.
