In recent decades, advances in lighting technology have precipitated exponential increases in night sky brightness worldwide, raising concerns in the scientific community about the impact of artificial light at night (ALAN) on crepuscular and nocturnal biodiversity. Long‐term records show that insect abundance has declined significantly over this time, with worrying implications for terrestrial ecosystems. The majority of investigations into the vulnerability of nocturnal insects to artificial light have focused on the flight‐to‐light behavior exhibited by select insect families. However, ALAN can affect insects in other ways as well. This review proposes five categories of ALAN impact on nocturnal insects, highlighting past research and identifying key knowledge gaps. We conclude with a summary of relevant literature on bioluminescent fireflies, which emphasizes the unique vulnerability of terrestrial light‐based communication systems to artificial illumination. Comprehensive understanding of the ecological impacts of ALAN on diverse nocturnal insect taxa will enable researchers to seek out methods whereby fireflies, moths, and other essential members of the nocturnal ecosystem can coexist with humans on an increasingly urbanized planet. Psychoanalysing ambivalence with Freud and Lacan : on and off the couch
\"Taking a deep dive into contemporary Western culture, this book suggests we are all fundamentally ambivalent beings. A great deal has been written about how to love-to be kinder, more empathic, a better person, and so on. But trying to love without dealing with our ambivalence, with our hatred, is often a recipe for failure. Any attempt, therefore, to love our neighbour as ourselves-or even, for that matter, to love ourselves-must recognize that we love where we hate and we hate where we love. Psychoanalysis, beginning with Freud, has claimed that to be in two minds about something or someone is characteristic of human subjectivity. Owens and Swales trace the concept of ambivalence through its various iterations in Freud and Lacan in order to question how the contemporary subject deals with its ambivalence. They argue that experiences of ambivalence are, in present-day cultural life, increasingly excised or foreclosed, and that this foreclosure has symptomatic effects at the individual as well as social levels. Owens and Swales examine ambivalence as it is at work in mourning, in matters of sexuality, in our enjoyment under neo-liberalism and capitalism. Above all, the authors consider how today's ambivalent subject relates to the racially, religiously, culturally, or sexually different neighbour as a result of the current societal dictate of complete tolerance of the other. International criteria for electrocardiographic interpretation in athletes: Consensus statement

Sudden cardiac death (SCD) is the leading cause of mortality in athletes during sport. A variety of mostly hereditary, structural or electrical cardiac disorders are associated with SCD in young athletes, the majority of which can be identified or suggested by abnormalities on a resting 12-lead electrocardiogram (ECG). Whether used for diagnostic or screening purposes, physicians responsible for the cardiovascular care of athletes should be knowledgeable and competent in ECG interpretation in athletes. However, in most countries a shortage of physician expertise limits wider application of the ECG in the care of the athlete. A critical need exists for physician education in modern ECG interpretation that distinguishes normal physiological adaptations in athletes from distinctly abnormal findings suggestive of underlying pathology. Since the original 2010 European Society of Cardiology recommendations for ECG interpretation in athletes, ECG standards have evolved quickly, advanced by a growing body of scientific data and investigations that both examine proposed criteria sets and establish new evidence to guide refinements. On 26–27 February 2015, an international group of experts in sports cardiology, inherited cardiac disease, and sports medicine convened in Seattle, Washington (USA), to update contemporary standards for ECG interpretation in athletes. The objective of the meeting was to define and revise ECG interpretation standards based on new and emerging research and to develop a clear guide to the proper evaluation of ECG abnormalities in athletes. This statement represents an international consensus for ECG interpretation in athletes and provides expert opinion-based recommendations linking specific ECG abnormalities and the secondary evaluation for conditions associated with SCD.

Incidence and Etiology of Sudden Cardiac Arrest and Death in High School Athletes in the United States

To determine the incidence and etiology of sudden cardiac arrest and death (SCA/D) in US high school athletes.
A prospective media database of SCA/D was queried for cases aged 14 to 18 years from 7 states over 6 school years (September 1, 2007, to August 30, 2013). Event details were investigated to determine participation on a high school athletic team, sex, sport, and occurrence during school-sponsored activity or exertion. National sports participation numbers were used and a conversion factor was applied to account for multisport athletes. Autopsy reports were reviewed and cause of death was adjudicated by an expert panel.
A total of 16,390,409 million athlete-seasons representing 6,974,640 athlete-years (AY) were examined, encompassing 36% of the total US high school athlete population. A total of 104 cases of SCA/D were identified (35 SCA with survival and 69 sudden cardiac deaths [SCDs]). The rate of SCD was 1:101,082 AY and of SCA/D 1:67,064 AY. Eighty-eight percent (92) of events occurred in male athletes. The rate of SCA/D in male athletes was 1:44,832 AY and in female athletes 1:237,510 AY (incidence rate ratio, 5.3; 95% CI, 2.9-10.6; P<.001). Men's basketball was the highest risk sport with an SCA/D incidence of 1:37,087 AY followed by men's football at 1:86,494 AY. Men's basketball and football athletes accounted for 57% (39) of deaths. Eighty percent of SCDs (55 of 69) were exertional and 55% (38 of 69) occurred while playing for a school-sponsored team. Autopsy reports were obtained in 73% (50) of cases. The most common findings of autopsy were idiopathic left ventricular hypertrophy or possible cardiomyopathy (13 of 50 [26%]), autopsy-negative sudden unexplained death (9 of 50 [18%]), hypertrophic cardiomyopathy (7 of 50 [14%]), and myocarditis (7 of 50 [14%]).
The rate of SCA/D in male high school athletes was 1:44,832 AY, with almost half due to possible or confirmed cardiomyopathy disease. It is likely that many cases were not identified because of reliance on media reports, and these numbers represent a minimum estimate.

A massively parallel reporter assay library to screen short synthetic promoters in mammalian cells

Cellular responses to stimuli underpin discoveries in drug development, synthetic biology, and general life sciences. We introduce a library comprising 6144 synthetic promoters, each shorter than 250 bp, designed as transcriptional readouts of cellular stimulus responses in massively parallel reporter assay format. This library facilitates precise detection and amplification of transcriptional activity from our promoters, enabling the systematic development of tunable reporters with dynamic ranges of 50−100 fold. Our library proved functional in numerous cell lines and responsive to a variety of stimuli, including metabolites, mitogens, toxins, and pharmaceutical agents, generating robust and scalable reporters effective in screening assays, biomarkers, and synthetic circuits attuned to endogenous cellular activities. Particularly valuable in therapeutic development, our library excels in capturing candidate reporters to signals mediated by drug targets, a feature we illustrate across nine diverse G-protein coupled receptors (GPCRs), critical targets in drug development. We detail how this tool isolates and defines discrete signaling pathways associated with specific GPCRs, elucidating their transcriptional signatures. With its ease of implementation, broad utility, publicly available data, and comprehensive documentation, our library will be beneficial in synthetic biology, cellular engineering, ligand exploration, and drug development.
Context-dependent, responsive synthetic promoters are crucial for a wide range of applications, yet currently available options are limited. Here, authors develop a library of thousands of candidate promoters based on binding motifs of hundreds transcription factors for use in mammalian cells.

iLAM: Imaging Locomotor Activity Monitor for circadian phenotyping of large‐bodied flying insects

Historically, most insect chronoecological research has used direct observations, cameras or infrared beam‐based monitors to quantify movement across timed intervals. Although some alternative DIY systems are cheaper than the current standard locomotor activity monitor, these options remain complicated to build and/or computationally intensive. We developed the imaging Locomotor Activity Monitor (iLAM), an affordable (~ $75 USD/unit) system for activity quantification. The iLAM utilizes a Raspberry Pi Zero W computer and night‐vision camera inside a flight cage to photograph a population of insects at user‐defined intervals. Open‐source, modular R‐scripts process the images and output a file containing the number, size, coordinate location and timestamp of all movements (blobs) identified between consecutive images. Output can be analysed directly or converted into the standard TriKinetics DAM format. We demonstrated the flexibility and power of the iLAM system by comparing diel and circadian activity of different insect species (fireflies: Photinus marginellus, P. greeni, P. obscurellus), ecotypes (moths: Ostrinia nubilalis) and sexes (moths: O. nubilalis). Data captured by only six iLAMs ($ 450) identified that peak activity of O. nubilalis females (AZT: 19.2 h) occurs significantly earlier than males (22.0 h). Additionally, male moths from a univoltine population exhibited a significantly shorter endogenous period length (AZT: 21.3 h) than males from a bivoltine genetic background (22.7 h). The iLAM will serve as a valuable tool for researchers seeking to measure locomotor activity across diverse species, sexes and populations in constant and changing environments.

In subjects with aortic regurgitation (AR) or mitral regurgitation (MR), transthoracic echocardiography (TTE) is recommended for surveillance. Few prospective studies have directly compared the ability of TTE and cardiac magnetic resonance (CMR) to predict clinical outcomes in AR and MR. We hypothesized that, given its higher reproducibility, CMR would predict the need for valve surgery or heart failure (HF) hospitalization better than TTE. Quantitative TTE and CMR were performed on the same day for 51 subjects: 29 with chronic AR and 22 with chronic, primary MR for quantification of valve regurgitation. Baseline measurements of valve regurgitation were compared to the combined primary end point of new HF and valve surgery using receiver operating characteristics, simple logistic regression, and Kaplan–Meier survival analyses. The primary end point occurred in 5 AR subjects (all surgery) and 8 MR subjects (7 surgery, 1 HF) after a mean follow-up of 4.4 ± 1.5 years. For AR, CMR-derived regurgitant volume >50 ml identified those at high risk with 50% undergoing valve surgery versus 0% for those with regurgitant volume ≤50 ml and was more strongly associated with outcomes than regurgitant volume by TTE (p <0.05). For MR, 6.8% of those with regurgitant volume by TTE ≤30 ml developed the primary end point versus 70% in those with regurgitant volume >30 ml. Regurgitant volume by CMR showed no significant separation of survival curves for MR. In conclusion, regurgitant volume by CMR was more predictive of outcomes than by TTE in subjects with AR. In MR, the 2 methods performed similarly.

Effects of cyanobacterial-driven pH increases on sediment nutrient fluxes and coupled nitrification-denitrification in a shallow fresh water estuary

Summer cyanobacterial blooms caused an elevation in pH (9 to ~10.5) that lasted for weeks in the shallow and tidal-fresh region of the Sassafras River, a tributary of Chesapeake Bay (USA). Elevated pH promoted desorption of sedimentary inorganic phosphorus and facilitated conversion of ammonium (NH4+) to ammonia (NH3). In this study, we investigated pH effects on exchangeable NH4+ desorption, pore water diffusion and the flux rates of NH4+, soluble reactive phosphorus (SRP) and nitrate (NO3−), nitrification, denitrification, and oxygen consumption. Elevated pH enhanced desorption of exchangeable NH4+ through NH3 formation from both pore water and adsorbed NH4+ pools. Progressive penetration of high pH from the overlying water into sediment promoted the mobility of SRP and the release of total ammonium (NH4+ and NH3) into the pore water. At elevated pH levels, high sediment-water effluxes of SRP and total ammonium were associated with reduction of nitrification, denitrification and oxygen consumption rates. Alkaline pH and the toxicity of NH3 may inhibit nitrification in the thin aerobic zone, simultaneously constraining coupled nitrification–denitrification with limited NO3− supply and high pH penetration into the anaerobic zone. Geochemical feedbacks to pH elevation, such as enhancement of dissolved nutrient effluxes and reduction in N2 loss via denitrification, may enhance the persistence of cyanobacterial blooms in shallow water ecosystems.
