New Insights into the Pivotal Role of the Amygdala in Inflammation-Related Depression and Anxiety Disorder

Depression and anxiety disorders are the two most prevalent psychiatric diseases that affect hundreds of millions of individuals worldwide. Understanding the etiology and related mechanisms is of great importance and might yield new therapeutic strategies to treat these diseases effectively. During the past decades, a growing number of studies have pointed out the importance of the stress-induced inflammatory response in the amygdala, a kernel region for processing emotional stimuli, as a potentially critical contributor to the pathophysiology of depression and anxiety disorders. In this review, we first summarized the recent progress from both animal and human studies toward understanding the causal link between stress-induced inflammation and depression and anxiety disorders, with particular emphasis on findings showing the effect of inflammation on the functional changes in neurons in the amygdala, at levels ranging from molecular signaling, cellular function, synaptic plasticity, and the neural circuit to behavior, as well as their contributions to the pathology of inflammation-related depression and anxiety disorders. Finally, we concluded by discussing some of the difficulties surrounding the current research and propose some issues worth future study in this field.

Identification of a prefrontal cortex-to-amygdala pathway for chronic stress-induced anxiety

Dysregulated prefrontal control over amygdala is engaged in the pathogenesis of psychiatric diseases including depression and anxiety disorders. Here we show that, in a rodent anxiety model induced by chronic restraint stress (CRS), the dysregulation occurs in basolateral amygdala projection neurons receiving mono-directional inputs from dorsomedial prefrontal cortex (dmPFC→BLA PNs) rather than those reciprocally connected with dmPFC (dmPFC↔BLA PNs). Specifically, CRS shifts the dmPFC-driven excitatory-inhibitory balance towards excitation in the former, but not latter population. Such specificity is preferential to connections made by dmPFC, caused by enhanced presynaptic glutamate release, and highly correlated with the increased anxiety-like behavior in stressed mice. Importantly, low-frequency optogenetic stimulation of dmPFC afferents in BLA normalizes the enhanced prefrontal glutamate release onto dmPFC→BLA PNs and lastingly attenuates CRS-induced increase of anxiety-like behavior. Our findings thus reveal a target cell-based dysregulation of mPFC-to-amygdala transmission for stress-induced anxiety.

Remote Bridge Deflection Measurement Using an Advanced Video Deflectometer and Actively Illuminated LED Targets

An advanced video deflectometer using actively illuminated LED targets is proposed for remote, real-time measurement of bridge deflection. The system configuration, fundamental principles, and measuring procedures of the video deflectometer are first described. To address the challenge of remote and accurate deflection measurement of large engineering structures without being affected by ambient light, the novel idea of active imaging, which combines high-brightness monochromatic LED targets with coupled bandpass filter imaging, is introduced. Then, to examine the measurement accuracy of the proposed advanced video deflectometer in outdoor environments, vertical motions of an LED target with precisely-controlled translations were measured and compared with prescribed values. Finally, by tracking six LED targets mounted on the bridge, the developed video deflectometer was applied for field, remote, and multipoint deflection measurement of the Wuhan Yangtze River Bridge, one of the most prestigious and most publicized constructions in China, during its routine safety evaluation tests. Since the proposed video deflectometer using actively illuminated LED targets offers prominent merits of remote, contactless, real-time, and multipoint deflection measurement with strong robustness against ambient light changes, it has great potential in the routine safety evaluation of various bridges and other large-scale engineering structures.

Practical implications of ICD-11 personality disorder classifications

Personality disorders (PDs) are associated with an inferior quality of life, poor health, and premature mortality, leading to heavy clinical, familial, and societal burdens. The International Classification of Diseases-11 (ICD-11) makes a thorough, dramatic paradigm shift from the categorical to dimensional diagnosis of PD and expands the application into adolescence. We have reviewed the recent literature on practical implications, and severity and trait measures of ICD-11 defined PDs, by comparing with the alternative model of personality disorders in the fifth edition of the Diagnostic and Statistical Manual of Mental Disorders (DSM-5), by mentioning the relevance in forensic and social concerns, and by referencing the developmental implication of life span, especially in adolescence. Study results strongly support the dimensional utility of ICD-11 PD diagnosis and application in adolescence which warrants early detection and intervention. More evidence-based research is needed along the ICD-11 PD application, such as its social relevance, measurement simplification, and longitudinal design of lifespan observation and treatment.

Panoramic/Dual-Surface Digital Image Correlation Measurement Using a Single Camera

We propose a cost-effective and simple-to-implement mirror-assisted single-camera panoramic digital image correlation (DIC) method for panoramic/dual-surface profile and deformation measurement. Specifically, two planar mirrors and a single camera attached with a four-mirror adapter are used to capture stereo images of the front and rear surfaces of a test object. These stereo images can be processed by regular stereo-DIC to retrieve shape and kinematics fields of each surface. Further, with the speckle patterns prefabricated on the mirrors, reflection transformation matrices are obtained and applied to transform all reconstructed surfaces into a common world coordinate system. As such, panoramic/dual-surface shape and deformation measurements can be realized. For validation, a high-resolution smartphone camera and an industrial camera were, respectively, used to construct mirror-assisted single-camera panoramic DIC systems. Real experiments, including panoramic shape measurement of an aluminum cylinder, dual-surface shape measurement of an aluminum plate and uniaxial tensile tests of aluminum sheet specimens, were performed, confirming the feasibility and accuracy of the method. Since only a single camera and a few auxiliary reflective mirrors are required, the proposed method provides a cost-effective and convenient way for taking panoramic/dual-surface shape and deformation measurements of regular-sized cylindrical and bar samples.

Synergism between two BLA-to-BNST pathways for appropriate expression of anxiety-like behaviors in male mice

Understanding how distinct functional circuits are coordinated to fine-tune mood and behavior is of fundamental importance. Here, we observe that within the dense projections from basolateral amygdala (BLA) to bed nucleus of stria terminalis (BNST), there are two functionally opposing pathways orchestrated to enable contextually appropriate expression of anxiety-like behaviors in male mice. Specifically, the anterior BLA neurons predominantly innervate the anterodorsal BNST (adBNST), while their posterior counterparts send massive fibers to oval BNST (ovBNST) with moderate to adBNST. Optogenetic activation of the anterior and posterior BLA inputs oppositely regulated the activity of adBNST neurons and anxiety-like behaviors, via disengaging and engaging the inhibitory ovBNST-to-adBNST microcircuit, respectively. Importantly, the two pathways exhibited synchronized but opposite responses to both anxiolytic and anxiogenic stimuli, partially due to their mutual inhibition within BLA and the different inputs they receive. These findings reveal synergistic interactions between two BLA-to-BNST pathways for appropriate anxiety expression with ongoing environmental demands.
How distinct circuits get coordinated to allow individuals to express appropriate level of anxiety is unclear. Here, authors show there are two functionally opposing BLA-BNST pathways interacting via the local inhibitory networks to enable anxiety expression with environmental needs.

Background
The development of antinociceptive tolerance following repetitive administration of opioid analgesics significantly hinders their clinical use. Evidence has accumulated indicating that microglia within the spinal cord plays a critical role in morphine tolerance. The inhibitor of microglia is effective to attenuate the tolerance; however, the mechanism is not fully understood. Our present study investigated the effects and possible mechanism of a natural product procyanidins in improving morphine tolerance via its specific inhibition on NOD-like receptor protein3 (NLRP3) inflammasome in microglia.
Methods
CD-1 mice were used for tail-flick test to evaluate the degree of pain. The microglial cell line BV-2 was used to investigate the effects and the mechanism of procyanidins. Reactive oxygen species (ROS) produced from BV-2 cells was evaluated by flow cytometry. Cell signaling was measured by western blot assay and immunofluorescence assay.
Results
Co-administration of procyanidins with morphine potentiated its antinociception effect and attenuated the development of acute and chronic morphine tolerance. Procyanidins also inhibited morphine-induced increase of interleukin-1β and activation of NOD-like receptor protein3 (NLRP3) inflammasome. Furthermore, procyanidins decreased the phosphorylation of p38 mitogen-activated protein kinase, inhibited the translocation of nuclear factor-κB (NF-κB), and suppressed the level of reactive oxygen species in microglia.
Conclusions
Procyanidins suppresses morphine-induced activation of NLRP3 inflammasome and inflammatory responses in microglia, and thus resulting in significant attenuation of morphine antinociceptive tolerance.

Post-synaptic specialization of the neuromuscular junction: junctional folds formation, function, and disorders

Post-synaptic specialization is critical to the neurotransmitter release and action potential conduction. The neuromuscular junctions (NMJs) are the synapses between the motor neurons and muscle cells and have a more specialized post-synaptic membrane than synapses in the central nervous system (CNS). The sarcolemma within NMJ folded to form some invagination portions called junctional folds (JFs), and they have important roles in maintaining the post-synaptic membrane structure. The NMJ formation and the acetylcholine receptor (AChR) clustering signal pathway have been extensively studied and reviewed. Although it has been suggested that JFs are related to maintaining the safety factor of neurotransmitter release, the formation mechanism and function of JFs are still unclear. This review will focus on the JFs about evolution, formation, function, and disorders. Anticipate understanding of where they are coming from and where we will study in the future.

Full-Field Bridge Deflection Monitoring with Off-Axis Digital Image Correlation

Video deflectometer based on using off-axis digital image correlation (DIC) has emerged as a robust non-contact optical tool for deflection measurements of bridges. In practice, a video deflectometer often needs to measure the deflections at multiple positions of the bridge. The existing 2D-DIC-based measurement methods usually use a laser rangefinder to measure the distance from each point to the camera to obtain the scale factor for the point. It is only suitable for the deflection measurements of a few points since manually measuring distances for a large number of points is time consuming and impractical. In this paper, a novel method for full-field bridge deflection measurement based on off-axis DIC is proposed. Because the bridge is usually a slender structure and the region of interest on the bridge is often a narrow band, the new approach can determine the scale factors of all the points of interest with a spatial straight-line fitting scheme. Moreover, the proposed technique employs reliability-guided processing and a fast initial parameter estimation strategy for real-time and accurate image-matching analysis. An indoor cantilever beam experiment verified the accuracy of the proposed approach, and a field test of a high-speed railway bridge demonstrated the robustness and practicability of the technique.
