Effect of Incorporating Nanoporous Metal Phosphate Materials on the Compressive Strength of Portland Cement

Nanoporous metal phosphate (NP-MPO) materials are being developed for removal of contaminant oxyanions (As(OH)O32−, CrO42−, and TcO4−), and cations (mercury, cadmium, and lead) from water and waste streams. Following sequestration, incorporation of metal laden NP-MPOs as a portion of cement formulation would provide an efficient and low-cost way to immobilize metal laden NP-MPOs in an easily handled waste form suitable for permanent disposal. There are no known investigations regarding the incorporation of NP-MPOs in concrete and the effects imparted on the physical and mechanical properties of concrete. Results of this investigation demonstrated that incorporating of NP-MPO materials requires additional water in the concrete formulation which decreases the compressive strength. Thus, incorporation of NP-MPOs in concrete may not serve as an efficient means for long-term disposal.

Gravel-Corrected Kd Values

Standard measurements of solute sorption to sediments are typically made on the <2 mm sediment fraction. This fraction is used by researchers to standardize the method and to ease experimental protocol so that large labware is not required to accommodate the gravel fraction (>2 mm particles). Since sorption is a phenomenon directly related to surface area, sorption measurements based on the <2 mm fraction would be expected to overestimate actual whole‐sediment values for sediments containing gravel. This inaccuracy is a problem for ground water contaminant transport modelers who use laboratory‐derived sorption values, typically expressed as a distribution coefficients (Kd), to calculate the retardation factor (Rf), a parameter that accounts for solute‐sediment chemical interactions. The objectives of this laboratory study were to quantify the effect of gravel on strontium Kd and Rf values and to develop an empirical method to calculate gravel‐corrected Kdgc values for the study site (Hanford Site in Richland, Washington). Three gravel corrections, Kd values, were evaluated: a correction based on the assumption that the gravel simply diluted the Kd<2mm and had no sorption capacity (Kdgc,g=0), a correction based on the assumption that the Kd of the intact sediment (Kdtot was a composite of the Kd<2mm and the Kd>2mm (Kdgc,g = x), and a correction based on surface area (Kdgc,surf). On average, Kd<2mm tended to overestimate Kdtot by 28% to 47%; Kdgc,g = x overestimated Kdtot by only 3% to 5%; and Kdgc,g = 0 and Kdgc,surf underestimated Kdtot by 10% to 39%. Kdgc,g = x provided the best estimate of actual values (Kdtot); however, Kdgc,g = 0 was appreciably easier to acquire. Although other contaminants will likely have different gravel‐correction values, these results have important implications regarding the traditional approach to modeling contaminant transport which uses Kd<2mm values. Such calculations may overestimate the tendency of gravel‐containing sediments to retard contaminant migration.

As the transportation industry is the central force in the supply chain, how it reacts to and prepares for the Y2K phenomenon will affect everyone from manufacturers to consumers. Not all reports on transportation preparedness are encouraging. In fact, by some estimates, the transportation industry ranks near the bottom of industries addressing the Y2K problem and it is estimated that approximately 50 percent of these companies will experience mission- critical failures. The Y2K phenomenon may, however, turn out to be a profit-making business opportunity for the transportation industry. For example, at least one large trucking firm already has postponed its annual company Christmas party in anticipation of increased demands on its ability to transport the vast volume of goods being purchased by numerous businesses prior to any possible disruption in the supply chain. This expectation of increased usage is not an isolated view. According to a recent Journal of Commerce article, shippers are protecting themselves from any potential Y2K-associated problems by stocking increased inventories. During August, approximately 600 shippers utilizing Prime's temperature-controlled, flatbed and dry van truckload transportation services were mailed a one-page Y2K Truckload Transportation Survey. Ninety-eight or 16 percent of the shippers returned the survey. In the context of Y2K, each of the shippers was asked four primary questions. The first question was, does your firm anticipate an increase in truckload transportation requirements for the second half of 1999? The second question asked was, if your firm anticipates the need for extra trailers during the second half of 1999, would you be willing to rent or lease the trailers? Next the shippers were asked, does your firm anticipate any other additional truckload related services during the second half of 1999? Finally, each shipper was asked to indicate the percentage increase expected for truckload transportation requirements from the first half of 1999 to the second half of 1999.

Empirical evidence for stability of the 405-kiloyear Jupiter–Venus eccentricity cycle over hundreds of millions of years

The Newark–Hartford astrochronostratigraphic polarity timescale (APTS) was developed using a theoretically constant 405-kiloyear eccentricity cycle linked to gravitational interactions with Jupiter–Venus as a tuning target and provides a major timing calibration for about 30 million years of Late Triassic and earliest Jurassic time. While the 405-ky cycle is both unimodal and the most metronomic of the major orbital cycles thought to pace Earth’s climate in numerical solutions, there has been little empirical confirmation of that behavior, especially back before the limits of orbital solutions at about 50 million years before present. Moreover, the APTS is anchored only at its younger end by U–Pb zircon dates at 201.6 million years before present and could even be missing a number of 405-ky cycles. To test the validity of the dangling APTS and orbital periodicities, we recovered a diagnostic magnetic polarity sequence in the volcaniclastic-bearing Chinle Formation in a scientific drill core fromPetrified Forest National Park (Arizona) that provides an unambiguous correlation to the APTS. New high precision U–Pb detrital zircon dates from the core are indistinguishable from ages predicted by the APTS back to 215 million years before present. The agreement shows that the APTS is continuous and supports a stable 405-kiloyear cycle well beyond theoretical solutions. The validated Newark–Hartford APTS can be used as a robust framework to help differentiate provinciality from global temporal patterns in the ecological rise of early dinosaurs in the Late Triassic, amongst other problems.

Regulation of sodium balance is a critical factor in the maintenance of euvolemia, and dysregulation of renal sodium excretion results in disorders of altered intravascular volume, such as hypertension. The amiloride-sensitive epithelial sodium channel (ENaC) is thought to be the only mechanism for sodium transport in the cortical collecting duct (CCD) of the kidney. However, it has been found that much of the sodium absorption in the CCD is actually amiloride insensitive and sensitive to thiazide diuretics, which also block the Na-Cl cotransporter (NCC) located in the distal convoluted tubule. In this study, we have demonstrated the presence of electroneutral, amiloride-resistant, thiazide-sensitive, transepithelial NaCl absorption in mouse CCDs, which persists even with genetic disruption of ENaC. Furthermore, hydrochlorothiazide (HCTZ) increased excretion of Na+ and Cl- in mice devoid of the thiazide target NCC, suggesting that an additional mechanism might account for this effect. Studies on isolated CCDs suggested that the parallel action of the Na+-driven Cl-/HCO3- exchanger (NDCBE/SLC4A8) and the Na+-independent Cl-/HCO3- exchanger (pendrin/SLC26A4) accounted for the electroneutral thiazide-sensitive sodium transport. Furthermore, genetic ablation of SLC4A8 abolished thiazide-sensitive NaCl transport in the CCD. These studies establish what we believe to be a novel role for NDCBE in mediating substantial Na+ reabsorption in the CCD and suggest a role for this transporter in the regulation of fluid homeostasis in mice.

Probiotics for Gastrointestinal Symptoms and Quality of Life in Autism: A Placebo-Controlled Pilot Trial

Objective:
A randomized pilot trial of gastrointestinal (GI) symptoms targeting probiotic for quality of life in autism spectrum disorder (ASD).
Methods:
Thirteen children, 3–12 years of age with ASD, anxiety, and GI symptoms, were randomized into a probiotic crossover trial of 8 weeks each on VISBIOME and placebo separated by a 3-week washout. VISBIOME contains eight probiotic species, mostly Lactobacillus and Bifidobacterium. Primary outcome was the Pediatric Quality of Life Inventory (PedsQL) GI module. Secondary outcomes included gut microbiota analysis, the Parent-Rated Anxiety Scale for ASD (PRAS-ASD), and parent-selected target symptoms. A mixed analysis model was applied.
Results:
Thirteen children were randomized, with 10 completing the study (77% retention): 6 in probiotic/placebo sequence, 4 in placebo/probiotic sequence. Adherence to study treatment was 96%. There were no serious adverse events (AEs), and more nonserious AEs occurred with placebo than with probiotic, including those attributable to treatment. Only 6 of the 10 guessed the correct treatment at the end of week 8. Over the 19-week trial, each outcome improved from baseline and PedsQL correlated significantly with abundance of Lactobacillus without discernable changes to microbiota composition/diversity. Although probiotic showed more improvement than placebo, PedsQL and PRAS-ASD were not statistically significant, as expected at this sample size. PedsQL effect size was d = 0.49 by the general model and d = 0.79 by simple comparison of week 8 changes. A parent-selected target symptom showed significant improvement in GI complaints on probiotic compared with placebo (p = 0.02, d = 0.79). Probiotic effects carried over through the 3-week washout.
Conclusion:
The VISBIOME formulation was safe and suggested a health benefit in children with ASD and GI symptoms who retained Lactobacillus. The moderate effect size compared with placebo warrants a larger trial using a parallel-group design.

A CALL FOR NEW APPROACHES TO QUANTIFYING BIASES IN OBSERVATIONS OF SEA SURFACE TEMPERATURE

Global surface temperature changes are a fundamental expression of climate change. Recent, much-debated variations in the observed rate of surface temperature change have highlighted the importance of uncertainty in adjustments applied to sea surface temperature (SST) measurements. These adjustments are applied to compensate for systematic biases and changes in observing protocol. Better quantification of the adjustments and their uncertainties would increase confidence in estimated surface temperature change and provide higher-quality gridded SST fields for use in many applications. Bias adjustments have been based on either physical models of the observing processes or the assumption of an unchanging relationship between SST and a reference dataset, such as night marine air temperature. These approaches produce similar estimates of SST bias on the largest space and time scales, but regional differences can exceed the estimated uncertainty. We describe challenges to improving our understanding of SST biases. Overcoming these will require clarification of past observational methods, improved modeling of biases associated with each observing method, and the development of statistical bias estimates that are less sensitive to the absence of metadata regarding the observing method. New approaches are required that embed bias models, specific to each type of observation, within a robust statistical framework. Mobile platforms and rapid changes in observation type require biases to be assessed for individual historic and present-day platforms (i.e., ships or buoys) or groups of platforms. Lack of observational metadata and high-quality observations for validation and bias model development are likely to remain major challenges.

Angiotensin II-dependent TGF-Beta signaling contributes to Loeys-Dietz syndrome vascular pathogenesis

Loeys-Dietz syndrome (LDS) is a connective tissue disorder that is characterized by a high risk for aneurysm and dissection throughout the arterial tree and phenotypically resembles Marfan syndrome. LDS is caused by heterozygous missense mutations in either TGF-β receptor gene (TGFBR1 or TGFBR2), which are predicted to result in diminished TGF-bgr; signaling. In this paper, the authors generated 2 knockin mouse strains with LDS mutations in either Tgfbr1 or Tgfbr2 and a transgenic mouse overexpressing mutant Tgfbr2. Knockin and transgenic mice, but not haploinsufficient animals, recapitulated the LDS phenotype. The analysis of TGF-β signaling in the aortic wall in vivo revealed progressive upregulation of Smad2 phosphorylation and TGF-β target gene output, which paralleled worsening of aneurysm pathology and coincided with upregulation of TGF-β1 ligand expression. Importantly, the suppression of Smad2 phosphorylation and TGF-β1 expression correlated with the therapeutic efficacy of the angiotensin II type 1 receptor antagonist losartan.
