Sleep problems and risk of all-cause cognitive decline or dementia: an updated systematic review and meta-analysis

ObjectivesTo conduct an updated systematic review and meta-analysis of association between sleep and all-cause cognitive disorders.MethodsPubMed and EMBASE were searched from inception to 18 February 2019. Cohort studies exploring longitudinal associations of sleep with cognitive decline or dementia were included. The multivariable-adjusted effect estimates were pooled by random-effects models, with credibility assessment. The robust error meta-regression model was used to conduct the dose–response meta-analysis for sleep duration.Results11 155 reports were searched and 51 eligible cohorts with 15 sleep problems were included for our meta-analyses. Ten types of sleep conditions or parameters, including six (insomnia, fragmentation, daytime dysfunction, prolonged latency, rapid eye movement sleep behaviour disorder and excessive time in bed) with moderate-to-high levels of evidence, were linked to higher risk of all-cause cognitive disorders. Furthermore, a U-shaped relationship was revealed for the associations with sleep duration.ConclusionsSleep management might serve as a promising target for dementia prevention.

Immunogenic Cell Death-Based Cancer Vaccines

Cancer immunotherapy has achieved great advancement in the past decades. Whereas, its response is largely limited in immunologically cold tumors, in an urgent need to be solve. In recent years, an increasing number of studies have shown that inducing immunogenic cell deaths (ICDs) is an attractive approach to activate antitumor immunity. Upon specific stress, cancer cells undergo ICDs and dying cancer cells release danger associated molecular patterns (DAMPs), produce neoantigens and trigger adaptive immunity. ICDs exert a cancer vaccine-like effect and Inducement of ICDs mimics process of cancer vaccination. In this review, we propose a concept of ICD-based cancer vaccines and summarize sources of ICD-based cancer vaccines and their challenges, which may broaden the understandings of ICD and cancer vaccines in cancer immunotherapy.

ObjectiveInflammation plays a crucial role in the pathogenesis of mild cognitive impairment (MCI) and Alzheimer’s disease (AD). Our study aimed to analyse previous inconsistent results of inflammatory markers in AD and MCI quantitatively.MethodsStudies reporting concentrations of peripheral or cerebrospinal fluid (CSF) markers were included, and eligible data on AD, MCI and control were extracted. Pooled Hedges’s g was adopted to illustrate comparisons, and various confounding factors were used to explore sources of heterogeneity.ResultsA total of 170 studies were included in the meta-analysis and systematic review, which demonstrated increased peripheral levels of high-sensitivity C reactive protein (Hedges’s g 0.281, p<0.05), interleukin-6 (IL-6) (0.429, p<0.005), soluble tumour necrosis factor receptor 1 (sTNFR1) (0.763, p<0.05), soluble tumour necrosis factor receptor 2 (sTNFR2) (0.354, p<0.005), alpha1-antichymotrypsin (α1-ACT) (1.217, p<0.005), IL-1β (0.615, p<0.05) and soluble CD40 ligand (0.868, p<0.005), and CSF levels of IL-10 (0.434, p<0.05), monocyte chemoattractant protein-1 (MCP-1) (0.798, p<0.005), transforming growth factor-beta 1 (1.009, p<0.05), soluble triggering receptor expressed on myeloid cells2 (sTREM2) (0.587, p<0.001), YKL-40 (0.849, p<0.001), α1-ACT (0.638, p<0.001), nerve growth factor (5.475, p<0.005) and visinin-like protein-1 (VILIP-1) (0.677, p<0.005), in AD compared with the control. Higher levels of sTNFR2 (0.265, p<0.05), IL-6 (0.129, p<0.05) and MCP-1 (0.779, p<0.05) and lower levels of IL-8 (−1.293, p<0.05) in the periphery, as well as elevated concentrations of YKL-40 (0.373, p<0.05), VILIP-1 (0.534, p<0.005) and sTREM2 (0.695, p<0.05) in CSF, were shown in MCI compared with the control. Additionally, increased peripheral sTNFR1 (0.582, p<0.05) and sTNFR2 (0.254, p<0.05) levels were observed in AD compared with MCI.ConclusionSignificantly altered levels of inflammatory markers were verified in comparison between AD, MCI and control, supporting the notion that AD and MCI are accompanied by inflammatory responses in both the periphery and CSF.

Models for predicting risk of dementia: a systematic review

BackgroundInformation from well-established dementia risk models can guide targeted intervention to prevent dementia, in addition to the main purpose of quantifying the probability of developing dementia in the future.MethodsWe conducted a systematic review of published studies on existing dementia risk models. The models were assessed by sensitivity, specificity and area under the curve (AUC) from receiver operating characteristic analysis.ResultsOf 8462 studies reviewed, 61 articles describing dementia risk models were identified, with the majority of the articles modelling late life risk (n=39), followed by those modelling prediction of mild cognitive impairment to Alzheimer’s disease (n=15), mid-life risk (n=4) and patients with diabetes (n=3). Age, sex, education, Mini Mental State Examination, the Consortium to Establish a Registry for Alzheimer’s Disease neuropsychological assessment battery, Alzheimer’s Disease Assessment Scale-cognitive subscale, body mass index, alcohol intake and genetic variables are the most common predictors included in the models. Most risk models had moderate-to-high predictive ability (AUC>0.70). The highest AUC value (0.932) was produced from a risk model developed for patients with mild cognitive impairment.ConclusionThe predictive ability of existing dementia risk models is acceptable. Population-specific dementia risk models are necessary for populations and subpopulations with different characteristics.

Underwater image enhancement using multi-task fusion

Underwater images are often scattered due to suspended particles in the water, resulting in light scattering and blocking and reduced visibility and contrast. Color shifts and distortions are also caused by the absorption of different wavelengths of light in the water. This series of problems will make the underwater image quality greatly impaired, resulting in some advanced visual work can not be carried out underwater. In order to solve these problems, this paper proposes an underwater image enhancement method based on multi-task fusion, called MTF. Specifically, we first use linear constraints on the input image to achieve color correction based on the gray world assumption. The corrected image is then used to achieve visibility enhancement using an improved type-II fuzzy set-based algorithm, while the image is contrast enhanced using standard normal distribution probability density function and softplus function. However, in order to obtain more qualitative results, we propose multi-task fusion, in which we solve for similarity, then we obtain fusion weights that guarantee the best features of the image as much as possible from the obtained similarity, and finally we fuse the image with the weights to obtain the output image, and we find that multi-task fusion has excellent image enhancement and restoration capabilities, and also produces visually pleasing results. Extensive qualitative and quantitative evaluations show that MTF method achieves optimal results compared to ten state-of-the-art underwater enhancement algorithms on 2 datasets. Moreover, the method can achieve better results in application tests such as target detection and edge detection.

You Only Look Yourself: Unsupervised and Untrained Single Image Dehazing Neural Network

In this paper, we study two challenging and less-touched problems in single image dehazing, namely, how to make deep learning achieve image dehazing without training on the ground-truth clean image (unsupervised) and an image collection (untrained). An unsupervised model will avoid the intensive labor of collecting hazy-clean image pairs, and an untrained model is a “real” single image dehazing approach which could remove haze based on the observed hazy image only and no extra images are used. Motivated by the layer disentanglement, we propose a novel method, called you only look yourself (YOLY) which could be one of the first unsupervised and untrained neural networks for image dehazing. In brief, YOLY employs three joint subnetworks to separate the observed hazy image into several latent layers, i.e., scene radiance layer, transmission map layer, and atmospheric light layer. After that, three layers are further composed to the hazy image in a self-supervised manner. Thanks to the unsupervised and untrained characteristics of YOLY, our method bypasses the conventional training paradigm of deep models on hazy-clean pairs or a large scale dataset, thus avoids the labor-intensive data collection and the domain shift issue. Besides, our method also provides an effective learning-based haze transfer solution thanks to its layer disentanglement mechanism. Extensive experiments show the promising performance of our method in image dehazing compared with 14 methods on six databases. The code could be accessed at www.pengxi.me.

Graphene saturable absorbers applications in fiber lasers

Two-dimensional materials are widely used in a variety of fields, such as medical treatment, industrial preparation, machining, etc. In this review, we have made a detailed description of the development of fiber lasers as well as the evolution of two-dimensional materials, especially graphene. In addition, we describe the optical properties of graphene and its preparations, for instance, chemical exfoliatio, liquid phase exfoliation, electrochemical technique, chemical vapor deposition, supercritical fluid exfoliation, and thermal exfoliation. Meanwhile, we also summarized several types of graphene saturable absorbers like all fiber, D-shaped, and optical deposition. Furthermore, we summarize the optical applications of fiber lasers based on graphene. Finally, we also take a look at the future perspectives of graphene and discuss the future applications of graphene in the field of optics. It is note worth that future fiber lasers will use more heterostructures or gas-solid mixtures to prepare saturable absorbers.

The Peculiar Precursor of a Gamma-Ray Burst from a Binary Merger Involving a Magnetar

The milestone discovery of GW170817-GRB 170817A-AT 2017gfo has shown that gravitational waves (GWs) could be produced during the merger of a neutron star–neutron star/black hole and that in electromagnetic (EM) waves, a gamma-ray burst (GRB) and a kilonova (KN) are generated in sequence after the merger. Observationally, however, EM properties before the merger phase are still unclear. Here we report a peculiar precursor in a KN-associated long-duration GRB 211211A, providing evidence of the EM before the merger. This precursor lasts ∼0.2 s, and the waiting time between the precursor and the main burst is ∼1 s, comparable to that between GW170817 and GRB 170817A. The spectrum of the precursor could be well fit with a nonthermal cutoff power-law model instead of a blackbody. In particular, a ∼22 Hz quasiperiodic oscillation candidate (∼3σ) is detected in the precursor. These temporal and spectral properties indicate that this precursor is probably produced by a catastrophic flare accompanied with magnetoelastic or crustal oscillations of a magnetar in a binary compact merger. The strong magnetic field of the magnetar can also account for the prolonged duration of GRB 211211A. However, it poses a challenge to reconcile the rather short lifetime of a magnetar with the rather long spiraling time of a binary neutron star system only by the GW radiation before the merger.

Nanoparticles for super-resolution microscopy and single-molecule tracking

We review the use of luminescent nanoparticles in super-resolution imaging and single-molecule tracking, and showcase novel approaches to super-resolution imaging that leverage the brightness, stability, and unique optical-switching properties of these nanoparticles. We also discuss the challenges associated with their use in biological systems, including intracellular delivery and molecular targeting. In doing so, we hope to provide practical guidance for biologists and continue to bridge the fields of super-resolution imaging and nanoparticle engineering to support their mutual advancement.
