Mapping and Validation of the Major Sex-Determining Region in Nile Tilapia (Oreochromis niloticus L.) Using RAD Sequencing
Sex in Oreochromis niloticus (Nile tilapia) is principally determined by an XX/XY locus but other genetic and environmental factors also influence sex ratio. Restriction Associated DNA (RAD) sequencing was used in two families derived from crossing XY males with females from an isogenic clonal line, in order to identify Single Nucleotide Polymorphisms (SNPs) and map the sex-determining region(s). We constructed a linkage map with 3,802 SNPs, which corresponded to 3,280 informative markers, and identified a major sex-determining region on linkage group 1, explaining nearly 96% of the phenotypic variance. This sex-determining region was mapped in a 2 cM interval, corresponding to approximately 1.2 Mb in the O. niloticus draft genome. In order to validate this, a diverse family (4 families; 96 individuals in total) and population (40 broodstock individuals) test panel were genotyped for five of the SNPs showing the highest association with phenotypic sex. From the expanded data set, SNPs Oni23063 and Oni28137 showed the highest association, which persisted both in the case of family and population data. Across the entire dataset all females were found to be homozygous for these two SNPs. Males were heterozygous, with the exception of five individuals in the population and two in the family dataset. These fish possessed the homozygous genotype expected of females. Progeny sex ratios (over 95% females) from two of the males with the \"female\" genotype indicated that they were neomales (XX males). Sex reversal induced by elevated temperature during sexual differentiation also resulted in phenotypic males with the \"female\" genotype. This study narrows down the region containing the main sex-determining locus, and provides genetic markers tightly linked to this locus, with an association that persisted across the population. These markers will be of use in refining the production of genetically male O. niloticus for aquaculture.

The 2019/2020 mega-fires exposed Australian ecosystems to an unprecedented extent of high-severity fire
Extreme fire seasons characterised by very large ‘mega-fires’ have demonstrably increased area burnt across forested regions globally. However, the effect of extreme fire seasons on fire severity, a measure of fire impacts on ecosystems, remains unclear. Very large wildfires burnt an unprecedented area of temperate forest, woodland and shrubland across south-eastern Australia in 2019/2020, providing an opportunity to examine the impact of extreme fires on fire severity patterns. We developed an atlas of wildfire severity across south-eastern Australia between 1988 and 2020 to test (a) whether the 2019/2020 fire season was more severe than previous fire seasons, and (b) if the proportion of high-severity fire within the burn extent (HSp) increases with wildfire size and annual area burnt. We demonstrate that the 2019/2020 wildfires in south-eastern Australia were generally greater in extent but not proportionally more severe than previous fires, owing to constant scaling between HSp and annual fire extent across the dominant dry-forest communities. However, HSp did increase with increasing annual fire extent across wet-forests and the less-common rainforest and woodland communities. The absolute area of high-severity fire in 2019/2020 (∼1.8 M ha) was larger than previously seen, accounting for ∼44% of the area burnt by high-severity fire over the past 33 years. Our results demonstrate that extreme fire seasons are a rare but defining feature of fire regimes across forested regions, owing to the disproportionate influence of mega-fires on area burnt.
Some Wildfire Ignition Causes Pose More Risk of Destroying Houses than Others
Many houses are at risk of being destroyed by wildfires. While previous studies have improved our understanding of how, when and why houses are destroyed by wildfires, little attention has been given to how these fires started. We compiled a dataset of wildfires that destroyed houses in New South Wales and Victoria and, by comparing against wildfires where no houses were destroyed, investigated the relationship between the distribution of ignition causes for wildfires that did and did not destroy houses. Powerlines, lightning and deliberate ignitions are the main causes of wildfires that destroyed houses. Powerlines were 6 times more common in the wildfires that destroyed houses data than in the wildfires where no houses were destroyed data and lightning was 2 times more common. For deliberate- and powerline-caused wildfires, temperature, wind speed, and forest fire danger index were all significantly higher and relative humidity significantly lower (P < 0.05) on the day of ignition for wildfires that destroyed houses compared with wildfires where no houses were destroyed. For all powerline-caused wildfires the first house destroyed always occurred on the day of ignition. In contrast, the first house destroyed was after the day of ignition for 78% of lightning-caused wildfires. Lightning-caused wildfires that destroyed houses were significantly larger (P < 0.001) in area than human-caused wildfires that destroyed houses. Our results suggest that targeting fire prevention strategies around ignition causes, such as improving powerline safety and targeted arson reduction programmes, and reducing fire spread may decrease the number of wildfires that destroy houses.

Cognitive and Behavioral Changes Arising From Spirituality
This article aims to explore the concept of spiritual transformation and address the question, ‘How does spirituality bring about changes in cognition and behavior?’ It draws on the findings of a larger qualitative study that explored the essence of spirituality and spiritual engagement from the perspective of palliative care clients and caregivers. Four clients and ten caregivers from across regional South Australia participated in the larger study utilizing van Manen’s phenomenology. Secondary analysis of the qualitative data was undertaken to highlight the statements and phrases that portrayed the marked changes in thinking and behavior catalyzed by spirituality. The findings relate to the participants’ spiritual beliefs and practices that helped them journey through the process of death and dying. Spirituality is a plausible explanation of the transformation that occurred, manifested by new thinking and behavior.
