Microbial-induced calcite precipitation (MICP), which leverages ureolytic microorganisms, has received significant attention during the past decade as a promising method for sustainable building and geoenvironmental applications. However, transitioning from lab-scale experimentation to volumes suitable for practical use poses challenges. species have been identified in the natural environment, and their ureolytic potential has been compared with that of other strains. A reference, banked microorganism yielded the highest ureolysis rate. , was subsequently used to induce the biocementation of a 650 kg column of 0–1 mm sand. The results reveal the successful stabilization of the whole mass, with undrained Tresca strength values ranging from 90 to 140 kPa. This research lays the groundwork for scalable MICP production, which is capable of meeting the demands of real-world building and geoenvironmental projects.

Shewanella oneidensis and Methanosarcina barkerii augmentation and conductive material effects on long-term anaerobic digestion performance

In short-term tests with acetate the syntrophic co-culture with RVC resulted in 86% higher maximum velocity of methane production, while in long term with real feed 13% increased rate was observed: the addition of 1.77 (S/m)*m

was among the dominant taxa at the end of the experiment, while
Shewanella oneidensis
was outcompeted. Among the methanogenesis production pathways, an increase of hydrogenotrophic methanogenesis has been observed in presence of conductive material. Further research is needed to understand the role of RVC in sulfur compounds production. Utilization of RVC to augment methane production yielded interesting results for real-scale application. As an added carrier, RVC remains unaltered and can be readily recuperated and reused multiple times.

The quagga mussel Dreissena rostriformis bugensis is an invasive species that has recently spread to Western Europe, causing major impacts on aquatic ecosystems and human activities. We considered lakes Lugano and Maggiore (Ticino River basin, Italy and Switzerland) to be at high risk of colonization due to leisure boat traffic from already-colonized lakes. We performed an environmental DNA based survey to detect any quagga mussel invasions in these lakes as early as possible. The results of the survey confirmed the presence of this species in the lakes and support the use of molecular approaches for the early detection of invasive mussels.

Fungal communities on alpine cheese rinds in Southern Switzerland

BackgroundThe biodiversity of the mycobiota of soft cheese rinds such as Brie or Camembert has been extensively studied, but scant information is available on the fungi colonizing the rinds of cheese produced in the Southern Switzerland Alps. This study aimed at exploring the fungal communities present on rinds of cheese matured in five cellars in Southern Switzerland and to evaluate their composition with regards to temperature, relative humidity, type of cheese, as well as microenvironmental and geographic factors. We used macro- and microscopical morphology, matrix-assisted laser desorption/ionization–time of flight (MALDI-TOF) mass spectrometry, and sequencing to characterize the fungal communities of the cheeses, and compared them with metabarcoding targeting the ITS region.ResultsIsolation by serial dilution yielded 201 isolates (39 yeasts and 162 filamentous fungi) belonging to 9 fungal species. Mucor and Penicillium were dominant, with Mucor racemosus, M. lanceolatus, P. biforme, and P. chrysogenum/rubens being the most frequent species. All but two yeast isolates were identified as Debaryomyces hansenii. Metabarcoding detected 80 fungal species. Culture work and metabarcoding produced comparable results in terms of similarity of the fungal cheese rind communities in the five cellars.ConclusionsOur study has shown that the mycobiota on the rinds of the cheeses studied is a comparatively species-poor community influenced by temperature, relative humidity, type of cheese, and manufacturing steps, as well as microenvironmental and possibly geographic factors.

Addition of Conductive Materials to Support Syntrophic Microorganisms in Anaerobic Digestion

Syntrophy and interspecies electron transfer among different microbial groups occurs in anaerobic digestion, and many papers recently reported their positive effect on biogas and methane production. In this paper, we present the results on the effect of conductive material, i.e., graphene, PAC and biochar addition in 3.5 L batch experiments, analyzing the biogas production curve. A peculiar curve pattern occurred in the presence of conductive materials. Compared to the respective controls, the addition of graphene produced a biogas surplus of 33%, PAC 20% and biochar 8%. Microbial community molecular analysis showed that syntrophic microorganisms present in the inoculum were stimulated by the conductive material addition. Graphene also appears to promote an interspecies electron transfer between Geobacter sp. and ca. Methanofastidiosum. This paper contributes to the understanding of the DIET-related microbial community dynamic in the presence of graphene and PAC, which could be exploited to optimize biogas and methane production in real-scale applications.

Molecular analysis of a human PAX6 homeobox mutant

Pax6 controls eye, pancreas and brain morphogenesis. In humans, heterozygous PAX6 mutations cause aniridia and various other congenital eye abnormalities. Most frequent PAX6 missense mutations are located in the paired domain (PD), while very few missense mutations have been identified in the homeodomain (HD). In the present report, we describe a molecular analysis of the human PAX6 R242T missense mutation, which is located in the second helix of the HD. It was identified in a male child with partial aniridia in the left eye, presenting as a pseudo-coloboma. Gel-retardation assays revealed that the mutant HD binds DNA as well as the wild-type HD. In addition, the mutation does not modify the DNA-binding properties of the PD. Cell transfection assays indicated that the steady-state levels of the full length mutant protein are higher than those of the wild-type one. In cotransfection assays a PAX6 responsive promoter is activated to a higher extent by the mutant protein than by the wild-type protein.
In vitro
limited proteolysis assays indicated that the presence of the mutation reduces the sensitivity to trypsin digestion. Thus, we suggest that the R242T human phenotype could be due to abnormal increase of PAX6 protein, in keeping with the reported sensitivity of the eye phenotype to increased PAX6 dosage.

Vinegar and Its Active Component Acetic Acid Inhibit SARS-CoV-2 Infection In Vitro and Ex Vivo

Effective and economical measures are needed to either prevent or inhibit the replication of SARS-CoV-2, the causative agent of COVID-19, in the upper respiratory tract. As fumigation of vinegar at low concentration (0.34%) ameliorated the symptoms of mild SARS-CoV-2 infection, we tested in vitro the potential antiviral activity of vinegar and of its active component, acetic acid. We here demonstrate that both vinegar and acetic acid indeed strongly inactivate SARS-CoV-2 infectivity in Vero cells. Furthermore, vinegar treatment caused a 90% inhibition of the infectious titer when directly applied to a nasopharyngeal swab transfer medium of a COVID-19 patient. These effects were potentiated if conduced at a temperature of 45 vs. 37 Celsius degrees, a condition that is transiently generated in the upper respiratory tract during fumigation. Our findings are consistent and extend the results of studies performed in the early and mid-20th century on the disinfectant capacity of organic acids and can provide an affordable home-made aid to prevent or contain SARS-CoV-2 infection of the upper respiratory tract.
