Re-evaluating the link between brain size and behavioural ecology in primates
Comparative studies have identified a wide range of behavioural and ecological correlates of relative brain size, with results differing between taxonomic groups, and even within them. In primates for example, recent studies contradict one another over whether social or ecological factors are critical. A basic assumption of such studies is that with sufficiently large samples and appropriate analysis, robust correlations indicative of selection pressures on cognition will emerge. We carried out a comprehensive re-examination of correlates of primate brain size using two large comparative datasets and phylogenetic comparative methods. We found evidence in both datasets for associations between brain size and ecological variables (home range size, diet and activity period), but little evidence for an effect of social group size, a correlation which has previously formed the empirical basis of the Social Brain Hypothesis. However, reflecting divergent results in the literature, our results exhibited instability across datasets, even when they were matched for species composition and predictor variables. We identify several potential empirical and theoretical difficulties underlying this instability and suggest that these issues raise doubts about inferring cognitive selection pressures from behavioural correlates of brain size.
Protein disulphide isomerase inhibition as a potential cancer therapeutic strategy
The protein disulphide isomerase (PDI) gene family is a large, diverse group of enzymes recognised for their roles in disulphide bond formation within the endoplasmic reticulum (ER). PDI therefore plays an important role in ER proteostasis, however, it also shows involvement in ER stress, a characteristic recognised in multiple disease states, including cancer. While the exact mechanisms by which PDI contributes to tumorigenesis are still not fully understood, PDI exhibits clear involvement in the unfolded protein response (UPR) pathway. The UPR acts to alleviate ER stress through the activation of ER chaperones, such as PDI, which act to refold misfolded proteins, promoting cell survival. PDI also acts as an upstream regulator of the UPR pathway, through redox regulation of UPR stress receptors. This demonstrates the pro‐protective roles of PDI and highlights PDI as a potential therapeutic target for cancer treatment. Recent research has explored the use of PDI inhibitors with PACMA 31 in particular, demonstrating promising anti‐cancer effects in ovarian cancer. This review discusses the properties and functions of PDI family members and focuses on their potential as a therapeutic target for cancer treatment. Protein disulphide isomerases (PDIs) play a vital role in folding proteins into their correct comformation. Inhibition of PDIs can cause ER‐stress leading to cellular apoptosis. In many cancers, PDI expression and activity is increased suggesting they are potential targets for novel therapies. Here we review the latest on PDI in cancer and highlight key PDI inhibitors that may be in the future used as cancer treatments.
Readability Analysis of Online Breast Cancer Surgery Patient Education Materials from National Cancer Institute-Designated Cancer Centers Compared with Top Internet Search Results
BackgroundThe National Institutes of Health (NIH) recommends patient education materials reflect the average reading grade level of the US population. Due to the importance of shared decision-making in breast cancer surgery, this study evaluates the reading level of patient education materials from National Cancer Institute-designated cancer centers (NCI-DCC) compared with top Internet search results.MethodsOnline materials from NCI-DCC and top Internet search results on breast cancer, staging, surgical options, and pre- and postoperative expectations were analyzed using three validated readability algorithms: Simplified Measure of Gobbledygook Readability Formula, Coleman–Liau index, and Flesch–Kincaid grade level. Mean readability was compared across source groups and information subcategories using an unpaired t-test with statistical significance set at p < 0.05. Mean readability was compared using a one-way analysis of variance.ResultsMean readability scores from NCI-DCC and Internet groups ranged from a 9th–12th grade level, significantly above the NIH recommended reading level of 6th–7th grade. There was no significant difference between reading levels from the two sources. The discrepancy between actual and recommended reading level was most pronounced for “surgical options” at a 10th–12th grade level from both sources.ConclusionsPatient education materials on breast cancer from both NCI-DCC and top Internet search results were written several reading grade levels higher than the NIH recommendation. Materials should be revised to enhance patient comprehension of breast cancer surgical treatment and guide patients in this important decision-making process to ultimately improve health outcomes.
The Impact of Cognitive Testing on the Welfare of Group Housed Primates
Providing cognitive challenges to zoo-housed animals may provide enriching effects and subsequently enhance their welfare. Primates may benefit most from such challenges as they often face complex problems in their natural environment and can be observed to seek problem solving opportunities in captivity. However, the extent to which welfare benefits can be achieved through programmes developed primarily for cognitive research is unknown. We tested the impact of voluntary participation cognitive testing on the welfare of a socially housed group of crested macaques (Macaca nigra) at the Macaque Study Centre (Marwell Zoo). First, we compared the rate of self-directed and social behaviours on testing and non-testing days, and between conditions within testing days. Minimal differences in behaviour were found when comparing testing and non-testing days, suggesting that there was no negative impact on welfare as a result of cognitive testing. Lipsmacking behaviours were found to increase and aggressive interaction was found to decrease in the group as a result of testing. Second, social network analysis was used to assess the effect of testing on associations and interactions between individuals. The social networks showed that testing subjects increased their association with others during testing days. One interpretation of this finding could be that providing socially housed primates with an opportunity for individuals to separate from the group for short periods could help mimic natural patterns of sub-group formation and reunion in captivity. The findings suggest, therefore, that the welfare of captive primates can be improved through the use of cognitive testing in zoo environments.
Characterizing unsuccessful animal adoptions: age and breed predict the likelihood of return, reasons for return and post-return outcomes
A considerable number of adopted animals are returned to animal shelters post-adoption which can be stressful for both the animal and the owner. In this retrospective analysis of 23,932 animal records from a US shelter, we identified animal characteristics associated with the likelihood of return, key return reasons, and outcomes post-return for dogs and cats. Binary logistic regression models were used to describe the likelihood of return, return reason and outcome based on intake age, intake type, sex, breed and return frequency. Behavioral issues and incompatibility with existing pets were the most common return reasons. Age and breed group (dogs only) predicted the likelihood of return, return reason and post-adoption return outcome. Adult dogs had the greatest odds of post-adoption return (OR 3.40, 95% CI 2.88–4.01) and post-return euthanasia (OR 3.94, 95% CI 2.04–7.59). Toy and terrier breeds were 65% and 35% less likely to be returned compared with herding breeds. Pit bull-type breeds were more likely to be returned multiple times ( X 2  = 18.11, p  = 0.01) and euthanized post-return (OR 2.60, 95% CI 1.47–4.61). Our findings highlight the importance of animal behavior in the retention of newly adopted animals and provide useful direction for allocation of resources and future adoption counselling and post-adoption support services.
A prospective study of mental wellbeing, quality of life, human-animal attachment, and grief among foster caregivers at animal shelters
Foster care programs at animal shelters have emerged as an important tool for promoting animal welfare and supporting shelter life-saving efforts. Preliminary evidence suggests that foster caregiving may also be health-promoting for humans. The goals of this study were to investigate the experiences of foster caregivers at animal shelters based on measures of positive and negative affect, quality of life, and grief, and to describe human-animal attachment among foster populations. Between March 2022 and 2023, 131 foster caregivers from five shelters in the United States completed three online surveys before, during and after providing foster care to a shelter animal. Positive affect decreased significantly from baseline to post-foster (F = 5.71, p<0.01), particularly among dog caregivers (F = 6.17, p<0.01). Negative affect remained unchanged (F = 0.47, p = 0.63). Foster caregivers perceived their foster animal provided companionship, affection and emotional support, although dog foster caregivers reported significantly higher emotional (U = 313.50, p<0.01) and social/physical quality of life (t = 4.42, p<0.01) than cat foster caregivers. Caregivers reported low mean avoidant and anxious attachment, suggesting they were able to develop secure bonds with their foster animals. Retention of fosters was also strong, with 86% of caregivers reporting they were likely to provide foster care in the future. Our findings suggest that fostering at animal shelters may serve as a One Health intervention to offer companionship, affection and emotional support to human caregivers while promoting animal welfare. However, these benefits did not translate to improvements in caregiver mental wellbeing, so caution should be applied when considering foster caregiving as a potential mental health promotion tool.
Short-term storage conditions do not affect canine fecal cortisol and secretory immunoglobulin A concentrations
Fecal cortisol and secretory immunoglobulin A (sIgA) are frequently used biomarkers in animal welfare, wildlife conservation, and other fields. However, these research settings often lack immediate access to a -80 °C freezer or other ultra-low temperature storage equipment. This study aims to determine whether canine fecal cortisol and sIgA concentrations remain stable over 24 h at either ambient temperature or on wet ice. Fecal samples were collected from 10 adult dogs housed in two Philadelphia area animal shelters in May and June of 2024. After aliquoting, baseline samples were frozen on dry ice within 15 minutes of defecation. Remaining aliquots were stored at ambient temperature (15–21°C) or on wet ice (4°C) for 1, 2, 4, 8, 12, or 24 hours before freezing on dry ice. Cortisol and sIgA concentrations were quantified using ELISAs and analyzed with a linear mixed effects model. No significant differences were found between time points or storage conditions. These data show that concentrations of fecal canine cortisol and sIgA remain stable if stored unfrozen for 24 hours. Immediate freezing at -80°C does not appear to be necessary for accurate quantification of these analytes. This knowledge removes logistical and regulatory barriers to conducting quality noninvasive research in animal shelter, clinical, and field settings.
Maternal investment, life histories and the evolution of brain structure in primates
Life history is a robust correlate of relative brain size: larger-brained mammals and birds have slower life histories and longer lifespans than smaller-brained species. The cognitive buffer hypothesis (CBH) proposes an adaptive explanation for this relationship: large brains may permit greater behavioural flexibility and thereby buffer the animal from unpredictable environmental challenges, allowing for reduced mortality and increased lifespan. By contrast, the developmental costs hypothesis (DCH) suggests that life-history correlates of brain size reflect the extension of maturational processes needed to accommodate the evolution of large brains, predicting correlations with pre-adult life-history phases. Here, we test novel predictions of the hypotheses in primates applied to the neocortex and cerebellum, two major brain structures with distinct developmental trajectories. While neocortical growth is allocated primarily to pre-natal development, the cerebellum exhibits relatively substantial post-natal growth. Consistent with the DCH, neocortical expansion is related primarily to extended gestation while cerebellar expansion to extended post-natal development, particularly the juvenile period. Contrary to the CBH, adult lifespan explains relatively little variance in the whole brain or neocortex volume once pre-adult life-history phases are accounted for. Only the cerebellum shows a relationship with lifespan after accounting for developmental periods. Our results substantiate and elaborate on the role of maternal investment and offspring development in brain evolution, suggest that brain components can evolve partly independently through modifications of distinct developmental phases, and imply that environmental input during post-natal maturation may be particularly crucial for the development of cerebellar function. They also suggest that relatively extended post-natal maturation times provide a developmental mechanism for the marked expansion of the cerebellum in the apes.
Companion dog acquisition and mental well-being: a community-based three-arm controlled study
Background Dog ownership is suggested to improve mental well-being, although empirical evidence among community dog owners is limited. This study examined changes in human mental well-being following dog acquisition, including four measures: loneliness, positive and negative affect, and psychological distress. Methods We conducted an eight-month controlled study involving three groups ( n  = 71): 17 acquired a dog within 1 month of baseline (dog acquisition); 29 delayed dog acquisition until study completion (lagged control); and 25 had no intentions of acquiring a dog (community control). All participants completed the UCLA Loneliness Scale (possible scores 0–60), Positive and Negative Affect Schedule and Kessler10 at baseline, three-months and eight-months. We used repeated measures ANCOVAs to analyse data with owner age and sex included as covariates. Post-hoc tests were performed for significant effects ( p  < 0.05). Results There was a statistically significant group by time interaction for loneliness ( p  = 0.03), with an estimated reduction of 8.41 units (95% CI -16.57, − 0.26) from baseline to three-months and 7.12 (95% CI -12.55, − 1.69) from baseline to eight-months in the dog acquisition group. The group by time interaction for positive affect was also significant ( p  = 0.03), although there was no change in the dog acquisition group. Conclusions Companion dog acquisition may reduce loneliness among community dog owners. Our study provides useful direction for future larger trials on the effects of dog ownership on human mental well-being. Trial registration This trial was retrospectively registered on 5th July 2017 with the Australian New Zealand Clinical Trials Registry ( ACTRN12617000967381 ).
