The genome of Cymbidium sinense revealed the evolution of orchid traits

Summary The Orchidaceae is of economic and ecological importance and constitutes ˜10% of all seed plant species. Here, we report a genome physical map for Cymbidium sinense, a well‐known species belonging to genus Cymbidium that has thousands of natural variation varieties of flower organs, flower and leaf colours and also referred as the King of Fragrance, which make it arose into a unique cultural symbol in China. The high‐quality chromosome‐scale genome assembly was 3.52 Gb in size, 29 638 protein‐coding genes were predicted, and evidence for whole‐genome duplication shared with other orchids was provided. Marked amplification of cytochrome‐ and photosystem‐related genes was observed, which was consistent with the shade tolerance and dark green leaves of C. sinense. Extensive duplication of MADS‐box genes, and the resulting subfunctional and expressional differentiation, was associated with regulation of species‐specific flower traits, including wild‐type and mutant‐type floral patterning, seasonal flowering and ecological adaption. CsSEP4 was originally found to positively regulate gynostemium development. The CsSVP genes and their interaction proteins CsAP1 and CsSOC1 were significantly expanded and involved in the regulation of low‐temperature‐dependent flowering. Important genetic clues to the colourful leaf traits, purple‐black flowers and volatile trait in C. sinense were also found. The results provide new insights into the molecular mechanisms of important phenotypic traits of Cymbidium and its evolution and serve as a powerful platform for future evolutionary studies and molecular breeding of orchids.

Revealing the decision-making practices in automated external defibrillator deployment: insights from Shanghai, China

In recent years, the government has promoted the increased deployment of automated external defibrillators (AEDs) in public places with dense crowds, which is of great significance for ensuring that residents enjoy equal health rights. However, it is still unclear what factors decision-makers take into account when formulating deployment plans and whether these factors are related to local characteristics such as population distribution and socioeconomic conditions. Taking Shanghai, China as the research area, we adopted the kernel density estimation and spatial autocorrelation analysis to explore the spatial distribution characteristics of AEDs. We constructed a geographically weighted regression (GWR) model to identify the key factors influencing AED deployment. The results showed that AEDs in Shanghai presented obvious clustering distribution characteristics. The GWR model found that the factors considered by decision-makers in different regions when deploying AEDs followed the guidance of existing policies. It was also found that decision-makers in Shanghai mainly deployed more devices in areas with a high density of the elderly population, dense transportation networks, cultural and educational places, and transportation hubs with large population flows. However, it was observed that the city center might lack sufficient preparation for the elderly group. In order to allocate emergency medical resources more reasonably, it is very important to determine the practices of decision-makers in deploying AEDs. The GWR has shown the potential to evaluate and guide the local implementation of deployment plans.

Target search in the CRISPR/Cas9 system: facilitated diffusion with target cues

We study how Cas9, a central component of the CRISPR/Cas9 system, searches for a target sequence on the DNA. We propose a model that includes as key ingredients 3D diffusion, 1D sliding along the DNA, and the effect of short binding sequences preceding the target (protospacer adjacent motifs—PAMs). This last aspect constitutes the main difference with traditional facilitated diffusion of transcription factors. We solve our model, obtaining an expression for the average search time of Cas9 for its target. We find that experimentally measured kinetic parameters are close to the values yielding an optimal search time. Our results rationalize the role of PAMs in guiding the search process, and show that Cas9 searches for its targets in a nearly optimal way.

Bupleurum chinense is an important medicinal plant in China; however, little is known regarding how this plant transcribes and synthesizes saikosaponins under drought stress. Herein, we investigated how drought stress stimulates the transcriptional changes of B. chinense to synthesize saikosaponins. Short-term drought stress induced the accumulation of saikosaponins, especially from the first re-watering stage (RD_1 stage) to the second re-watering stage (RD_2 stage). Saikosaponin-a and saikosaponin-d increased by 84.60% and 75.13%, respectively, from the RD_1 stage to the RD_2 stage. Drought stress also stimulated a rapid increase in the levels of the hormones abscisic acid, salicylic acid, and jasmonic acid. We screened 49 Unigenes regarding the terpenoid backbone and triterpenoid biosynthesis, of which 33 differential genes were significantly up-regulated during drought stress. Moreover, one P450 and two UGTs are possibly involved in the synthesis of saikosaponins, while some transcription factors may be involved in regulating the expression of key enzyme genes. Our study provides a reference for the cultivation of B. chinense and a practical means to ensure the quality (safety and effectiveness) of B. chinense for medicinal use, as well as insights into the modernization of the China Agriculture Research System.

Pay for Being Responsible: The Effect of Target Firm’s Corporate Social Responsibility on Cross-Border Acquisition Premiums

This study examines the effect of a target firm’s corporate social responsibility (CSR) on its cross-border acquisition premium. Building upon the resource-based view and the institutional theory, we argue that the target firm’s CSR positively affect the cross-border acquisition premium, while institutional distance, cultural distance, and the number of fellow acquisitions moderate the above relationship. Hypotheses are tested in a sample of 252 cross-border acquisitions between 1991 and 2016. Empirical findings show that an acquirer is more likely to pay a higher acquisition premium when acquiring a socially responsible target firm; furthermore, such an effect weakens as institutional distance, cultural distance, and the number of fellow acquisitions increase. This study extends existing research on the importance of CSR as a strategic asset and sheds new light on the role of CSR played in the setting of cross-border acquisitions.

The role of histone post-translational modifications in cancer and cancer immunity: functions, mechanisms and therapeutic implications

Histones play crucial roles in both promoting and repressing gene expression, primarily regulated through post-translational modifications (PTMs) at specific amino acid residues. Histone PTMs, including methylation, acetylation, ubiquitination, phosphorylation, lactylation, butyrylation, and propionylation, act as important epigenetic markers. These modifications influence not only chromatin compaction but also gene expression. Their importance extends to the treatment and prevention of various human diseases, particularly cancer, due to their involvement in key cellular processes. Abnormal histone modifications and the enzymes responsible for these alterations often serve as critical drivers in tumor cell proliferation, invasion, apoptosis, and stemness. This review introduces key histone PTMs and the enzymes responsible for these modifications, examining their impact on tumorigenesis and cancer progression. Furthermore, it explores therapeutic strategies targeting histone PTMs and offers recommendations for identifying new potential therapeutic targets.

The potential application of concentrated growth factor in pulp regeneration: an in vitro and in vivo study

Background
Concentrated growth factor (CGF), as a natural biomaterial, is known to contain platelets, cytokines, and growth factors to facilitate the healing process, but there has been little information acquired in regenerative endodontics. The purpose of this study was to investigate the effects of CGF on proliferation, migration, and differentiation in human dental stem pulp cells (hDPSCs) exposed to lipopolysaccharide (LPS) in vitro and its potential role in pulp regeneration of the immature teeth in vivo
.
Methods
In vitro experiments: CGF-conditioned medium were extracted by freeze-dried method. hDPSCs were isolated and identified. The proliferative potential of hDPSCs with different concentration of CGF and LPS was evaluated by Cell Counting Kit-8. Migration capacity was analyzed by Transwell assays, odonto/osteoblastic differentiation was determined by measuring alkaline phosphatase (ALP) activity using ALP staining, and the extent of mineralization was evaluated by using Alizarin red S staining. The mRNA expression level of DMP-1, DSPP, OPN, Runx2, and OCN were determined by quantitative polymerase chain reaction (qPCR).
In vivo experiments: CGF were used as root canal filling agent of the immature single-rooted teeth in the beagle dogs. The teeth were then radiographed, extracted, fixed, demineralized, and subjected to histologic analyses at 8 weeks. The newly formed dentine-pulp complex and the development of apical foramen were evaluated by the hematoxylin-eosin (HE) and Masson trichrome technique. Soft tissues were analyzed by immunohistochemical staining of vascular endothelial growth factor (VEGF) and Nestin.
Results
In vitro experiments: The cultured cells exhibited the characteristics of mesenchymal stem cell. The treatment of LPS significantly increased the expression of TNF-α, IL-1β, IL-6, and IL-8 in hDPSCs, and CGF inhibited the mRNA expression of IL-8 in LPS-stimulated hDPSCs. The proliferation values of the CGF group in LPS-stimulated hDPSCs were significantly higher than that of the control group from day 3 to day 7 (
P
< 0.05). In addition, the number of migratory cells of the CGF group was greater than that of the control group at 24 h with or without LPS treatment. ALP activities increased gradually in both groups from day 4 to day 7. The mineralized nodules and the expression of odontogenesis-related genes DMP-1 and DSPP, osteogenesis-related genes OPN, Runx2, and OCN were dramatically enhanced by CGF in LPS-stimulated hDPSCs at days 21 and 28.
In vivo experiments: In CGF treated group, the results of radiograph, HE, and Masson trichrome staining showed a continuing developed tooth of the immature teeth in the beagle dogs (i.e., the ingrowth of soft tissues into the root canal, the thickened internal root dentin walls, and the closed apex), which resembled the normal tooth development in the positive control group. The immunohistochemical staining showed that VEGF and Nestin were both moderately expressed in the regenerated pulp-like tissues which indicating the vascularization and innervation.
Conclusions
CGF has a positive effect on the proliferation, migration, and differentiation of hDPSCs exposed to LPS in vitro, and it can also promote the regeneration of dentine-pulp complex of the immature teeth in the beagle dogs in vivo. Therefore, CGF could be a promising alternative biomaterial in regenerative endodontics.

Can “new infrastructure” reverse the “growth with pollution” profit growth pattern? An empirical analysis based on listed companies in China

With the increasing severity of environmental regulations, Chinese enterprises are gradually transforming from the profit growth pattern of “growth with pollution” to green development. However, this paper finds that this transformation is not complete. While catering to the national environmental protection governance requirements, some enterprises still persist the profit growth pattern of “growth with pollution.” How to solve this problem? By attracting government subsidies, reducing tax burdens, and increasing the proportion of technical personnel employed, we theoretically and empirically demonstrate that the positive effect of “new infrastructure” in accelerating enterprises’ transformation away from the profit growth pattern of “growth with pollution” to sustainable development. In the heterogeneity analysis section, we found that in enterprises with higher political correlation, more technical personnel, and lower labor cost, the “new infrastructure” has a stronger restraining effect on the “growth with pollution” pattern.

Gamification in higher education administration: a conceptual model for enhancing faculty and staff engagement

This study investigates the influence of gamification, job satisfaction, and organizational culture on faculty and staff engagement within higher education administration in China. A conceptual model was developed to explore these relationships, employing Confirmatory Factor Analysis (CFA) and Structural Equation Modeling (SEM) to ensure the constructs’ reliability and validity. The sample featured a diverse representation of faculty and administrative staff, encompassing various genders, age groups, and professional experiences. The findings indicate that gamification significantly enhances engagement, demonstrating its effectiveness in making administrative tasks more engaging and motivating. Job satisfaction emerged as a crucial factor, positively impacting engagement and mediating the relationship between gamification and engagement. The results further highlight the essential role of organizational culture, showing that a supportive and collaborative work environment can foster higher levels of engagement and job satisfaction. The study underscores the interconnected nature of these constructs, suggesting that higher education institutions should consider incorporating gamification and promoting a positive organizational culture to improve overall engagement. The research provides important theoretical insights and practical recommendations, emphasizing a comprehensive approach to employee engagement and highlighting directions for future studies, such as exploring demographic influences and long-term impacts.
