Background
Chinese cabbage (Brassica rapa ssp. pekinensis) experienced a whole-genome triplication event and thus has three subgenomes: least fractioned, medium fractioned, and most fractioned subgenome. Environmental changes affect leaf development, which in turn influence the yield. To improve the yield and resistance to different climate scenarios, a comprehensive understanding of leaf development is required including insights into the full diversity of cell types and transcriptional networks underlying their specificity.
Results
Here, we generate the transcriptional landscape of Chinese cabbage leaf at single-cell resolution by performing single-cell RNA sequencing of 30,000 individual cells. We characterize seven major cell types with 19 transcriptionally distinct cell clusters based on the expression of the reported marker genes. We find that genes in the least fractioned subgenome are predominantly expressed compared with those in the medium and most fractioned subgenomes in different cell types. Moreover, we generate a single-cell transcriptional map of leaves in response to high temperature. We find that heat stress not only affects gene expression in a cell type-specific manner but also impacts subgenome dominance.
Conclusions
Our study highlights the transcriptional networks in different cell types and provides a better understanding of transcriptional regulation during leaf development and transcriptional response to heat stress in Chinese cabbage.

Pseudopregnant mice generated from Piwil1 deficiency sterile mice

Vasectomized mice play a key role in the production of transgenic mice. However, vasectomy can cause great physical and psychological suffering to mice. Therefore, there is an urgent need to find a suitable replacement for vasectomized mice in the production of transgenic mice. In this study, we generated C57BL/6J mice ( Piwil1 D633A-INS99, Piwil1 mt/mt ) with a 99-base insertion in the Miwi ( Piwil1 ) gene using CRISPR/Cas9 technology and showed that Piwil1 mt/+ heterozygous mice were normally fertile and that homozygous Piwil1 mt/mt males were sterile and females were fertile. Transplantation of normal fertilized eggs into wild pseudopregnant females following mating with Piwil1 mt/mt males produced no Piwil1 mt/mt genotype offspring, and the number of offspring did not differ significantly from that of pseudopregnant mice following mating and breeding with ligated males. The CRISPR‒Cas9 system is available for generating Miwi -modified mice, and provides a powerful resource to replace ligated males in assisted reproduction research.

Effects of Dietary Supplementation with α-Mangostin on Oviduct Inflammation and Eggshell Quality in Aging Laying Hens

This study investigated the effects of dietary supplementation with α-mangostin (α-Ma), a bioactive xanthone derived from mangosteen pericarp, on production performance and egg quality in late-phase laying hens. The experiment was conducted using a completely randomized design. In total, 576 healthy 51-week-old Beinong No. 2 laying hens were randomly assigned to 4 dietary treatments (n = 12): a basal diet (CON) or the basal diet supplemented with 80, 120, or 160 mg/kg α-Ma. The experiment lasted for 4 weeks, after which production performance, egg quality, serum biochemical and antioxidant parameters, inflammatory markers, and uterine gene expression were evaluated. Dietary supplementation with α-mangostin, particularly at 120 mg/kg, significantly improved feed efficiency (p < 0.05), as evidenced by a reduced feed-to-egg ratio from week 2 onward, without affecting average daily feed intake or egg production rate. After 4 weeks, hens receiving 120 mg/kg α-Ma exhibited significantly greater egg weight and eggshell strength (p < 0.05). Serum and hepatic antioxidant capacities were significantly enhanced, with increased glutathione peroxidase and catalase activities, elevated total antioxidant capacity, and decreased malondialdehyde levels (p < 0.05). Moreover, α-Ma at 120 mg/kg specifically lowered the concentration of the pro-inflammatory cytokine interleukin-1β in both serum and uterine tissue (p < 0.05). At the molecular level, this dosage significantly upregulated uterine genes essential for eggshell formation (p < 0.05), including calcium transporters (TRPV6, ATP2B2), the matrix protein gene OC-116, and other key genes (LYZ, CA2, SLC4A9, and ATP6V0D2). In conclusion, dietary supplementation with 120 mg/kg α-Ma effectively enhances feed efficiency, strengthens antioxidant and anti-inflammatory defenses, and upregulates uterine genes involved in biomineralization, thereby improving eggshell quality in aging laying hens. These findings support α-Ma as a promising plant-based feed additive for maintaining productivity and egg quality in antibiotic-free layer production systems.

The non-redundant functions of PIWI family proteins in gametogenesis in golden hamsters

The piRNA pathway is essential for female fertility in golden hamsters and likely humans, but not in mice. However, the role of individual PIWIs in mammalian reproduction remains poorly understood outside of mice. Here, we describe the expression profiles, subcellular localization, and knockout-associated reproductive defects for all four PIWIs in golden hamsters. In female golden hamsters, PIWIL1 and PIWIL3 are highly expressed throughout oogenesis and early embryogenesis, while knockout of PIWIL1 leads to sterility, and PIWIL3 deficiency results in subfertility with lagging zygotic development. PIWIL1 can partially compensate for TE silencing in PIWIL3 knockout females, but not vice versa. PIWIL1 and PIWIL4 are the predominant PIWIs expressed in adult and postnatal testes, respectively, while PIWIL2 is present at both stages. Loss of any PIWI expressed in testes leads to sterility and severe but distinct spermatogenesis disorders. These findings illustrate the non-redundant regulatory functions of PIWI-piRNAs in gametogenesis and early embryogenesis in golden hamsters, facilitating study of their role in human fertility.
The role of PIWI proteins in mammalian reproduction remains poorly understood. Here, using golden hamsters, the authors establish expression profiles for all four PIWIs during gametogenesis and characterize their associated reproductive defects.

Effect of Carboxymethyl chitosan-sodium alginate hydrogel loaded with Astragalus membranaceus-Panax notoginseng on wound healing in rats

Skin wound healing is a complex physiological process that involves many different cell types and signaling molecules. In traditional Chinese medicine, A. membranaceus and Panax notoginseng are commonly used together for the treatment of wound injury for their significant efficacy. The application of new materials may make A. membranaceus -P. notoginseng (AP) play a better curative effect. In this study, we fabricated a Carboxymethyl chitosan-Sodium alginate hydrogel loaded with the extract of Astragalus membranaceus - P. notoginseng (APCS), which showed favorable stability, biocompatibility, and the ability to rapidly release drugs. Cell culture experiments demonstrated that the APCS hydrogel dramatically increased fibroblast proliferation, migration, and differentiation into myofibroblast. In vivo experiments of wound healing on SD rats showed that APCS hydrogels significantly accelerated skin wound healing, reduced inflammatory response, enhanced the formation of blood vessels, granulation tissues, and collagen fibers, and promoted re-epithelialization at the wound site. Increased expression of catalase, VEGF, and PGP9.5 of wound tissue indicated that APCS hydrogels inhibited oxidative stress and promoted vascular and neuronal regeneration. In summary, the APCS hydrogel displayed great potential as a dressing for achieving satisfactory healing of full-thickness wounds.

Demystifying and Assessing Code Understandability in Java Decompilation

Decompilation, the process of converting machine-level code into readable source code, plays a critical role in reverse engineering. Given that the main purpose of decompilation is to facilitate code comprehension in scenarios where the source code is unavailable, the understandability of decompiled code is of great importance. In this paper, we propose the first empirical study on the understandability of Java decompiled code and obtained the following findings: (1) Understandability of Java decompilation is considered as important as its correctness, and decompilation understandability issues are even more commonly encountered than decompilation failures. (2) A notable percentage of code snippets decompiled by Java decompilers exhibit significantly lower or higher levels of understandability in comparison to their original source code. (3) Unfortunately, Cognitive Complexity demonstrates relatively acceptable precision while low recall in recognizing these code snippets exhibiting diverse understandability during decompilation. (4) Even worse, perplexity demonstrates lower levels of precision and recall in recognizing such code snippets. Inspired by the four findings, we further proposed six code patterns and the first metric for the assessment of decompiled code understandability. This metric was extended from Cognitive Complexity, with six more rules harvested from an exhaustive manual analysis into 1287 pairs of source code snippets and corresponding decompiled code. This metric was also validated using the original and updated dataset, yielding an impressive macro F1-score of 0.88 on the original dataset, and 0.86 on the test set.
Exosome: A Novel Approach to Stimulate Bone Regeneration through Regulation of Osteogenesis and Angiogenesis

The clinical need for effective bone regeneration therapy remains in huge demands. However, the current “gold standard” treatments of autologous and allogeneic bone grafts may result in various complications. Furthermore, safety considerations of biomaterials and cell-based treatment require further clarification. Therefore, developing new therapies with stronger osteogenic potential and a lower incidence of complications is worthwhile. Recently, exosomes, small vesicles of endocytic origin, have attracted attention in bone regeneration field. The vesicles travel between cells and deliver functional cargoes, such as proteins and RNAs, thereby regulating targeted cells differentiation, commitment, function, and proliferation. Much evidence has demonstrated the important roles of exosomes in osteogenesis both in vitro and in vivo. In this review, we summarize the properties, origins and biogenesis of exosomes, and the recent reports using exosomes to regulate osteogenesis and promote bone regeneration.

Astragalus Polysaccharide Improves Insulin Sensitivity via AMPK Activation in 3T3-L1 Adipocytes

Astragalus polysaccharide (APS) is an important bioactive component of Astragalus membranaceus which is used as an anti-diabetes herb in traditional Chinese medicine. The objective of this study was to investigate the effects and mechanisms of APS on insulin-sensitizing of adipocytes. Mouse 3T3-L1 preadipocytes were used as a model. The results showed that APS increased preadipocytes proliferation in a dose dependent manner, and 0.1 μg/mL APS sufficiently increased Proliferating Cell Nuclear Antigen (PCNA) content (p < 0.01). Moreover, APS enhanced intracellular lipid accumulation and mRNA expression of proliferator-activated receptor γ (PPARγ, p < 0.01), CCAAT/enhancer binding protein α (C/EBPα, p < 0.01) and fatty acid binding protein (aP2, p < 0.01). As expected, corresponding protein contents were elevated. Importantly, APS increased 2-(N-(7-Nitrobenz-2-oxa-1,3-diazol-4-yl)Amino)-2-Deoxyglucose (2-NBDG) uptake (p < 0.01). Meanwhile, both mRNA and protein content of glucose transporter 4 (Glut4) were elevated by APS (p < 0.01). The APS treatment enhanced tyrosine phosphorylation of insulin receptor substrate 1 (IRS1, p < 0.05) and phosphor-Akt content (p < 0.01). Besides, phosphorylated AMP-activated protein kinase (AMPK) content was increased in the APS treated cells (p < 0.01). Taken together, APS improved insulin sensitivity by enhancing glucose uptake, possibly through AMPK activation. These results suggested that APS might be a therapeutic candidate for insulin resistance.
