A guideline proposal for mice preparation and care in 18F-FDG PET imaging
The experimental outcomes of small-animal positron emission tomography (PET) imaging with 18F-labelled fluorodeoxyglucose (18F-FDG) can be particularly compromised by animal preparation and care. Several works intend to improve research reporting and amplify the quality and reliability of published research. Though these works provide valuable information to plan and conduct animal studies, manuscripts describe different methodologies—standardization does not exist. Consequently, the variation in details reported can explain the difference in the experimental results found in the literature. Additionally, the resources and guidelines defining protocols for small-animal imaging are scarce, making it difficult for researchers to obtain and compare accurate and reproducible data. Considering the selection of suitable procedures key to ensure animal welfare and research improvement, this paper aims to prepare the way for a future guideline on mice preparation and care for PET imaging with 18F-FDG. For this purpose, a global standard protocol was created based on recommendations and good practices described in relevant literature.
Leptocybe invasa males and the parasitoid Quadrastichus mendeli (Hym.: Eulophidae): First report in Paraguay
Abstract Leptocybe invasa Fisher & La Salle, 2004 (Hymenoptera: Eulophidae) forms galls in tips, young branches, central veins, and petioles of Eucalyptus spp. leaves. This insect reproduces primarily through thelytokous parthenogenesis, with males being rare. The parasitism of L. invasa by Quadrastichus mendeli Kim & La Salle, 2008 (Hymenoptera: Eulophidae), with short life cycle and rapid dispersion, is high. The objective was to record the first occurrence of L. invasa males and its parasitoid Q. mendeli in galls of this pest in Paraguay. Branches of the grancam clone PL 113 (Eucalyptus grandis × Eucalyptus camaldulensis) with L. invasa galls were collected from February to April 2022 in the Maciel District, Caazapá Department, Paraguay. These branches were put in plastic bags, preserved in 70% ethanol and the insects emerged sent to the Laboratory of Biological Control of Forest Pests (LCBPF) at FCA/UNESP in Botucatu, São Paulo state, Brazil. Morphological characteristics confirmed the presence of L. invasa males at a ratio of 38:1 (females to males). Morphological and molecular analysis identified Q. mendeli and its samples deposited in GenBank (OR248672.1). The parasitism rate of L. invasa by Q. mendeli was 26.8%, marking the first record of this occurrence in Paraguay. Resumo Leptocybe invasa Fisher & La Salle, 2004 (Hymenoptera: Eulophidae) forma galhas nos ponteiros, ramos jovens, nervuras centrais e pecíolos das folhas de Eucalyptus spp. Este inseto se reproduz predominantemente por partenogênese telítoca, sendo os machos raros. O parasitismo de L. invasa por Quadrastichus mendeli Kim & La Salle, 2008 (Hymenoptera: Eulophidae), que apresenta ciclo de vida curto e rápida dispersão, é elevado. O objetivo deste estudo foi registrar a primeira ocorrência de machos de L. invasa e de seu parasitoide Q. mendeli em galhas dessa praga no Paraguai. Ramos do clone grancam PL 113 (Eucalyptus grandis × Eucalyptus camaldulensis), com galhas de L. invasa, foram coletados entre fevereiro e abril de 2022, no Distrito de Maciel, Departamento de Caazapá, Paraguai. Esses ramos foram acondicionados em sacos plásticos, preservados em etanol a 70% e os insetos emergidos foram enviados ao Laboratório de Controle Biológico de Pragas Florestais (LCBPF), da FCA/UNESP, em Botucatu, estado de São Paulo, Brasil. As características morfológicas confirmaram a presença de machos de L. invasa em uma proporção de 38:1 (fêmeas: machos). Análises morfológicas e moleculares identificaram Q. mendeli, com amostras depositadas no GenBank (OR248672.1). A taxa de parasitismo de L. invasa por Q. mendeli foi de 26,8%, constituindo o primeiro registro dessa ocorrência no Paraguai.
First detection of Gonipterus platensis (Coleoptera: Curculionidae) and its parasitoid Anaphes nitens (Hymenoptera: Mymaridae) in eucalyptus plantations in Minas Gerais, Brazil
Ribeiro et al discuss the first detection of Gonipterus platensis and its parasitoid Anaphes nitens in eucalyptus plantations in Minas Gerais, Brazil. The area with forest plantations in Brazil is one of the largest in the world, 9.93 million hectares with 75.8% (7.53 million ha) of eucalyptus plantations with a mean annual productivity of 38.9 m3/ha. These plantations are used to produce raw materials, mainly for the bioenergy, pulp and paper and laminate flooring sectors. Minas Gerais is the Brazilian state with the largest area of eucalyptus plantations, more than 2 million hectares, followed by the Mato Grosso do Sul and Sao Paulo states. Insect pests, especially exotic ones, damage these plantations.
Yersinia massiliensis (Enterobacteriales: Enterobacteriaceae) in the host Anaphes nitens (Hymenoptera: Mymaridae): first report of association with insects
Abstract Endosymbiont bacteria can affect biological parameters and reduce the effectiveness of natural enemies in controlling the target insect. The objective of this work was to identify endosymbiont bacteria in Anaphes nitens (Girault, 1928) (Hymenoptera: Mymaridae), the main natural enemy used to manage Gonipterus platensis (Marelli, 1926) (Coleoptera: Curculionidae). Genomic DNA from six A. nitens populations was extracted and polymerase chain reactions (PCR) were performed with the primers to detect endosymbiont bacteria in this insect. The PCR products were amplified, sequenced, and compared with sequences deposited in the GenBank for the bacteria identification. All A. nitens populations had the bacterium Yersinia massiliensis (Enterobacteriales: Enterobacteriaceae). This bacterium was originally described as free-living, and it is associated with and composes part of the A. nitens microbiota. This is the first report of Y. massiliensis in an insect host. Resumo As bactérias endossimbiontes podem afetar os parâmetros biológicos e reduzirem a eficácia de inimigos naturais no controle do inseto alvo. O objetivo deste trabalho foi identificar bactérias endossimbiontes em Anaphes nitens (Girault, 1928) (Hymenoptera: Mymaridae), o principal inimigo natural usado no manejo de Gonipterus platensis (Marelli, 1926) (Coleoptera: Curculionidae). O DNA genômico de seis populações de A. nitens foi extraído e as reações em cadeia da polimerase (PCR) realizadas com os primers para detectar bactérias endossimbiontes neste inseto. Os produtos de PCR foram amplificados, sequenciados e comparados com as sequências depositadas no GenBank para identificação das bactérias. Todas as populações de A. nitens tinham a bactéria Yersinia massiliensis (Enterobacteriales: Enterobacteriaceae). Esta bactéria foi originalmente descrita como de vida livre e está associada e compõe parte da microbiota de A. nitens. Este é o primeiro relato de Y. massiliensis em um hospedeiro.
Effect of Sciatic Nerve Transection on acetylcholinesterase activity in spinal cord and skeletal muscles of the bullfrog Lithobates catesbeianus
Abstract Sciatic nerve transection (SNT), a model for studying neuropathic pain, mimics the clinical symptoms of “phantom limb”, a pain condition that arises in humans after amputation or transverse spinal lesions. In some vertebrate tissues, this condition decreases acetylcholinesterase (AChE) activity, the enzyme responsible for fast hydrolysis of released acetylcholine in cholinergic synapses. In spinal cord of frog Rana pipiens, this enzyme’s activity was not significantly changed in the first days following ventral root transection, another model for studying neuropathic pain. An answerable question is whether SNT decreases AChE activity in spinal cord of frog Lithobates catesbeianus, a species that has been used as a model for studying SNT-induced neuropathic pain. Since each animal model has been created with a specific methodology, and the findings tend to vary widely with slight changes in the method used to induce pain, our study assessed AChE activity 3 and 10 days after complete SNT in lumbosacral spinal cord of adult male bullfrog Lithobates catesbeianus. Because there are time scale differences of motor endplate maturation in rat skeletal muscles, our study also measured the AChE activity in bullfrog tibial posticus (a postural muscle) and gastrocnemius (a typical skeletal muscle that is frequently used to study the motor system) muscles. AChE activity did not show significant changes 3 and 10 days following SNT in spinal cord. Also, no significant change occurred in AChE activity in tibial posticus and gastrocnemius muscles at day 3. However, a significant decrease was found at day 10, with reductions of 18% and 20% in tibial posticus and gastrocnemius, respectively. At present we cannot explain this change in AChE activity. While temporally different, the direction of the change was similar to that described for rats. This similarity indicates that bullfrog is a valid model for investigating AChE activity following SNT. Resumo A transecção do nervo isquiático (SNT), um modelo para estudar dor neuropática, simula os sintomas clínicos do “membro fantasma”, uma condição dolorosa que ocorre nos humanos após amputação ou secção completa da medula espinal. Essa condição muda a atividade da acetilcolinesterase (AChE), a enzima responsável pela rápida hidrólise da acetilcolina liberada nas sinapses colinérgicas, em alguns tecidos de vertebrados. Em medula espinal de rã Rana pipiens, a atividade da AChE não foi significativamente alterada nos primeiros dias após a secção da raiz ventral, outro modelo para o estudo da dor neuropática. Uma questão ainda não respondida é se a SNT diminui a atividade da AChE na medula espinal de rã Lithobates catesbeianus, uma espécie que vem sendo usada como modelo em estudos da dor neuropática induzida por SNT. Como cada modelo animal é criado a partir de metodologia específica, e seus resultados tendem a variar com pequenas mudanças na metodologia de indução da dor, o presente estudo avaliou a atividade da AChE em medula espinal lombossacral de rã-touro Lithobates catesbeianus, adultos, machos, 3 e 10 dias após a completa SNT. Como há diferenças temporais na maturação de placas motoras em músculos esqueléticos de ratos, nosso estudo ainda demonstrou, na rã-touro, os efeitos da SNT sobre a atividade da AChE nos músculos esqueléticos tibial posticus, um músculo postural, e gastrocnêmio, um músculo frequentemente usado em estudos do sistema motor. A atividade da AChE não mudou significativamente na medula espinal aos 3 e 10 dias após a SNT. Nos músculos, a atividade não alterou significativamente aos 3 dias após a lesão, mas reduziu de forma significativa aos 10 dias após a SNT. Aos 10 dias, a diminuição foi 18% no músculo tibial posticus e 20% no gastrocnêmio. No momento, nós não temos explicação para essa mudança na atividade da AChE. Embora temporalmente diferente, o sentido da mudança é similar ao que é descrito em ratos. Esta similaridade torna a rã-touro um modelo válido para se estudar questões ainda não respondidas da SNT sobre a AChE.
Postoperative imaging findings after laparoscopic surgery for deeply infiltrating endometriosis
Deeply infiltrative endometriosis (DIE) is a common gynecologic disease affecting women of reproductive age and often causing chronic pelvic pain and infertility. Clinical treatment options and preventive actions are ineffective due to the lack of knowledge about the etiology of DIE. Surgical treatment is currently the only alternative to eradicate the disease. Diagnostic imaging plays a crucial role for surgical planning and postoperative evaluation. Transvaginal sonography (TVS) with a dedicated protocol and magnetic resonance imaging (MRI) can be used to evaluate recurrent disease. Extensive pelvic surgery may cause anatomical changes and a variable spectrum of postoperative findings. Residual disease and complications can be also evaluated and are of great importance to estimate pain relief and fertility prognosis. The most common imaging findings following radical surgery for DIE are fibrotic scars in the retrocervical space and bowel anastomosis, absence of the posterior vaginal fornix and loculated fluid in the pararectal spaces. Ovaries are the most frequent site of early recurrence. Complications include infection, hemorrhage, urinary/evacuatory voiding dysfunctions as well as bowel and ureteral stenosis. The purpose of this article is to review the surgical techniques currently used to treat endometriosis in the retrocervical space, vagina, bladder, bowel, ureters, and ovaries and to describe the most common imaging findings including normal aspects, residual disease, complications, and recurrence.
Transcribed Ultraconserved Regions Are Associated with Clinicopathological Features in Breast Cancer
Ultraconserved regions (UCRs) are 481 genome segments, with length longer than 200 bp, that are 100% conserved among humans, mice, and rats. The majority of UCRs are transcriptionally active (T-UCRs) as many of them produce non-coding RNAs. In a previous study, we evaluated the expression level of T-UCRs in breast cancer (BC) patients and found that 63% of transcripts correlated with some clinical and/or molecular parameter of BC. In this study, we delved into the expression levels of 12 T-UCRs and correlated them with clinicopathological parameters, immunohistochemical markers, and overall survival in two breast cancer cohorts: TCGA and Brazilian patients. We found that uc.268 is more expressed in TCGA patients under 40 years of age, associated with progesterone receptor (PR) and estrogen receptor (ER), and its high expression is found in luminal A. Lower uc.84 and uc.376 were respectively observed in metastatic and stage IV tumors associated with good prognostic in luminal B. Moreover, uc.84 was only related to the HER2+, while uc.376 was related to ER+ and PR+, and HER2+. A panel composed of uc.147, uc.271, and uc.427 distinguished luminal A from triple negative patients with an AUC of 0.9531 (sensitivity 92.19% and specificity 86.76%). These results highlight the potential role of T-UCRs in BC and provide insights into the potential application of T-UCRs as biomarkers.
Renal transplantation in lupus nephritis: a Brazilian cohort
Objective: To determine the epidemiological profile and outcome of patients with lupus nephritis (LN) undergoing renal transplantation. Methods: The archival records of 50 patients with LN and end-stage renal disease (ESRD) treated by kidney transplantation from March 1992 to December 2010 were reviewed. All patients met the American College of Rheumatology criteria for systemic lupus erythematosus (SLE). Results: Fourteen patients were included in the study. The majority were women (85.7%) and non-Caucasian (85.7%); the mean age at diagnosis of SLE and LN was 24 ± 8 and 25 ± 8 years, respectively. Renal biopsy was performed in 12 patients, with 75% of them showing proliferative lesions (class III and IV according to the World Health Organization and International Society of Nephrology/Renal Pathology Society classification). Thirteen patients (93%) underwent intermittent hemodialysis or peritoneal dialysis before transplantation. The median time between the start of dialysis and transplantation was 30 months (range 3–103 months); 67% of the procedures involved deceased donors and 33% involved living-related donors. The graft survival rates were 93.3%, 90.9%, and 85.7% at 1, 5 and 10 years, respectively. Post-transplant immunosuppressive agents were mycophenolate mofetil (84%), azathioprine (17%), tacrolimus (25%), sirolimus (58%) and cyclosporine (8%). Eight episodes of acute rejection were noted in six patients. There was a graft loss due to renal vein thrombosis in the one patient with secondary antiphospholipid syndrome. The mean SLICC by the time of kidney transplantation was 5 ± 2. In total, 13 patients (92.8%) developed at least one infectious event during the follow-up, with one dying in the immediate post-transplant period because of sepsis. Two patients (14%) had a lupus flare. There was no clinical or histological evidence of LN recurrence. Conclusion: LN is the major cause of morbidity in SLE, with progression to ESRD in 10–22% of cases. Despite concerns about LN recurrence after renal transplantation, the data obtained in our sample indicate this procedure as a safe alternative therapy for ESRD in this population.
