Multi-class diagnosis of Alzheimer’s disease using cascaded three dimensional-convolutional neural network

Dementia is a social problem in the aging society of advanced countries. Presently, 46.8 million people affected with dementia worldwide, and it may increase to 130 million by 2050. Alzheimer’s disease (AD) is the most common form of dementia. The cost of care for AD patients in 2015 was 818 billion US dollars and is expected to increase intensely due to the increasing number of patients due to the aging society. It isn’t easy to cure AD, but early detection is crucial. This paper proposes a multi-class classification of AD, mild cognitive impairment (MCI), and normal control (NC) subjects using three dimensional-convolutional neural network with Support Vector Machine classifier. A cross-sectional study on structural MRI data of 465 subjects, including 132 AD patients, 181 MCI, and 152 NC, is performed in this paper. The highly complex and spatial atrophy patterns of the brain related to Alzheimer’s Disease and MCI are extracted from structural MRI images using cascaded layers of the three dimensional convolutional neural network. The hectic process of segmentation and further extraction of handcrafted features is eliminated. The complete image is considered for the processing, thus incorporating every region of the brain for the classification. The features extracted using four cascaded layers of three dimensional-convolutional neural network are fed into the Support Vector Machine classifier. The proposed method achieved 97.77% accuracy which outperforms state of the art, and this algorithm is a promising indicator for the diagnosis of AD.

Coronal anatomic slices of structural MRI images clearly show the topographical structures of the Hippocampus and Amygdala, which are essential for early diagnosis of Alzheimer’s disease (AD). MR coronal sections are best appreciated for studying the complex topographical relationships of the amygdala and the topographical structures of the hippocampus, which helps in the early detection of disease. Early diagnosis helps prevent the disease’s progression to its final stage. It allows the patient to be aware of the severity of the disease and can take the necessary therapeutic medications to prevent its progression. A coronal view study of MR images is proposed in this paper for early diagnosis of disease using a wavelet-pooling-based multi-path and multi-scale convolutional neural network. This work aims to perform a three-way classification of 2D coronal slices of MRI images to diagnose Mild Cognitive Impairment, AD, and Normal Control in a single algorithm and learn the brain-affected regions through Gradcam visualization. wavelet-pooling is utilized to extract the texture details of the image and thus provide spatial attention to the texture features of the image, which is impossible using Max-pooling or Average-pooling. Multi-scale feature learning is incorporated using parallel multiple low-rank convolutional kernels to capture varying scales of atrophy regions. Multi-path mode compensates for the early loss of features and avoids vanishing gradient problems. with ten-fold cross-validation. The multi-scale and multi-path methods significantly reduce the number of learnable parameters.

Alzheimer’s disease is one of the most frequently studied diseases of the nervous system although it has no cure or slowing its progression. There are various options for treating the symptoms of Alzheimer’s disease in different stages and as the disease progresses over time, patients in their various stages need different treatment. Diagnosis of Alzheimer’s in the elderly is quietly difficult and requires representation of a discriminatory factor in isolation due to similar brain patterns and pixel strength. Deep learning strategies are able to learn such representations from the data. In this proposed work we perform multilevel classification of Alzheimer’s disease ie; Mild Demented, Moderate Demented, Non Demented and Very Mild Demented using transfer learning with VGG16 using Fastai. This approach results in 99% predictive accuracy which means a significant increase in accuracy compared to previous studies and clearly demonstrates the effectiveness of the proposed methods.
