The current consensus is that plant responses to canopy shade involve the perception of low red to far-red ratios (R:FRs) by phytochrome B (phyB), which leads to the direct activation of auxin synthesis genes by PHYTOCHROME INTERACTING FACTORS (PIFs). In addition to its effect on R:FRs, shade also reduces irradiance, but whether shade-induced drops in irradiance affect phyB activity has not been demonstrated. To address this issue, we investigated whether irradiance and R:FRs have similar effects on the nuclear distribution of phyB in petiole cells of light-grown plants. Under high-irradiance white light, phyB formed large nuclear bodies. Lowering irradiance without changing R:FRs or lowering R:FRs by adding far-red light led to the appearance of small nuclear bodies containing phyB. Large nuclear bodies remained but with some concomitant reduction in diameter. The appearance of small nuclear bodies was rapid, stable, and reversible upon the return to high irradiance and high R:FRs. High levels of red light but not of blue light were enough to restrain the formation of small phyB nuclear bodies. Irradiance was effective within the range found in natural canopies and even under relatively low R:FRs. The promotion of leaf hyponasty by lowering irradiance was impaired in phyB and pif mutants, as previously reported for the response to R:FRs. The expression of auxin-related genes showed a similar hierarchy of response to low R:FRs and low irradiance. We propose that phyB is able to perceive not only the low R:FRs, but also the low irradiance of shade.

DISTRIBUCIÓN DE POLIMORFISMOS GENÉTICOS DE INTERLEUQUINA-1 EN INDIVIDUOS DE LA REGIÓN CENTROCCIDENTAL DE VENEZUELA

The cytokines belonging to the family of interleukin-1 (IL-1) are encoded by three different genes: IL-1A, IL-1B, and IL-1RN, which encode for IL-1α, IL-1β and the endogenous receptor antagonist for IL-1 (IL-1Ra), respectively. IL-1α and IL-1β operate as pro-inflammatory cytokines, while the IL-1Ra as anti-inflammatory. It has been reported several biallelic polymorphisms in the genes of IL-1B, including IL-1B-511(C/T) and IL-1B+3954(C/T), while IL-1RN presents in intron 2 a penta-allelic VNTR polymorphism. The functionally relevant polymorphisms of these genes have been correlated with a wide range of chronic inflammatory and autoimmune conditions, as well as cancer. In order to determine the distribution of these polymorphisms in the Central-Western region of Venezuela, 100 unrelated apparently healthy individuals were studied. DNA was extracted from peripheral blood, and proceded to the characterization of polymorphisms IL-1B-511 and IL-1B +3954 by PCR-RFLP and VNTR IL-1RN by PCR. Allelic and genotypic frequencies were determined awith the program Arlequin v. 2.0. There was a predominance of T allele (52%) and the C allele (82%) for IL-1B-511 and IL-1B +3954, respectively. While for IL-1RN the more frequent genotypes were 1/1 (47%) and 1/2 (41%). We compare the results with the population frequencies found in other countries, highlighting differences with significant populations of different ethnic origin. These results could provide a valuable reference for future studies of association with cancer and inflammatory diseases in Venezuela.

BackgroundTritrichomonas foetus is the etiologic agent of the sexually transmitted disease Bovine Trichomonosis (BT). In Argentina, BT is endemic and represents a relevant health problem that causes reproductive inefficiency in cattle and large economic losses. Metronidazole is the drug of choice in the treatment of BT. Treatment has been associated with a temporary resolution of the clinical signs but is not able to control the disease. In recent years, the apparition of in vivo and in vitro aerobic and anaerobic resistance leading to ineffective treatments has been reported.AimsThus, the aim of the present study was to explore the susceptibility of six different isolates of T. foetus under aerobic (AC) and anaerobic (ANC) conditions.Results and discussionSix isolates of T. foetus were obtained from samples of preputial smegma of bovine origin. Values of minimum lethal concentration and minimum inhibitory concentration were higher than those observed in other works and represent current data in Argentina and provide information to establish new treatment protocols.

DISTRIBUCIÓN DE POLIMORFISMOS GENÉTICOS DE INTERLEUQUINA-1 EN INDIVIDUOS DE LA REGIÓN CENTROCCIDENTAL DE VENEZUELA

RESUMEN Las citoquinas pertenecientes a familia de la interleuquina-1 (IL-1) están codificadas por tres genes diferentes: IL-1A, IL-1B, e IL-1RN, los cuales codifican para IL-1 α, IL-1β, y el antagonista endógeno del receptor de IL-1 (IL-1ra), respectivamente. Las IL-1α e IL-1β actúan como citoquinas pro-inflamatorias, mientras que la IL-1ra se comporta como anti-inflamatoria. Han sido reportados varios polimorfismos bialélicos en los genes de IL-1B, incluyendo IL-1B-511(C/T) e IL-1B+3954(C/T), mientras que IL-1RN presenta en el intrón 2 un polimorfismo VNTR penta-alélico. Los polimorfismos funcionalmente relevantes de estos genes han sido correlacionados con un amplio conjunto de condiciones autoinmunes e inflamatorias crónicas, así como con cáncer. Con el fin de determinar la distribución de estos polimorfismos en la región centroccidental de Venezuela, se estudiaron 100 individuos no relacionados aparentemente sanos. Se extrajo ADN genómico a partir de sangre periférica, y se procedió a la tipificación de los polimorfismos IL-1B-511 e IL-1B+3954 por PCR-RFLP y VNTR de IL-1RN por PCR. Se determinaron las frecuencias alélicas y genotípicas con el programa Arlequín ver. 2.000. Se observó un predominio del alelo T (52%) y del alelo C (82%) en IL-1B-511 y IL-1B+3954, respectivamente. Mientras que para IL-1RN los genotipos más frecuente fueron el 1/1 (47%) y 1/2 (41%). Se compararon los resultados con las frecuencias poblacionales encontradas en otros países, destacándose diferencias significativas con poblaciones de diferente origen étnico. Los resultados podrían proporcionar una referencia valiosa para estudios futuros de asociación con cáncer y enfermedades inflamatorias en Venezuela. Cats in a Cat Café: Individual Cat Behavior and Interactions with Humans

Cat cafés are gaining popularity worldwide, yet little is known about their impact on feline welfare. This study examined a Swedish cat café from an animal perspective by assessing space use, cat–cat interactions, and cat–human interactions. A total of 27 neutered domestic cats (12 males, 15 females), with a median stay of 8 weeks, were directly observed in groups of 8–9 individuals for a total of 227 h over 70 days. Scan sampling and focal observations were conducted without prior knowledge of cat background to minimize bias. Results showed that elevated structures (χ2-goodness-of-fit test, χ2 (2) = 1234.2, p < 0.001, post hoc Bonferroni correction padj = 0.013) were important, particularly during periods of high customer occupancy, when cats preferred vertical levels or cat-only rooms. Cat–cat interactions were infrequent (0.58 interactions/cat/h) and mostly affiliative (51.9%), consistent with avoidance as a conflict-reduction strategy. Cat–human interactions were absent in 44.4% of the observation time. Cats varied in their responses to human presence, from engaging to avoiding. Findings suggest that cat cafés should provide complex vertical environments, hiding places, and private areas to support natural behaviors, reduce stress, and promote group stability. As cats come into contact with large numbers of visitors prior to adoption, further research should assess its long-term effects on cat–human relationships and optimize café design to balance cat welfare and visitor experience.

Twice upon a time: The progression of canine visceral leishmaniasis in an Argentinean city

Canine Visceral Leishmaniasis (CVL) prevalence, spatial distribution and associated factors were assessed in four locations in Iguazú department in 2014 and in Puerto Iguazú city again in 2018. The city areas were divided into a grid of 400x400m cells. All cells were sampled in 2014 and a random subsampling was developed in 2018. In each cell, five dogs clustered in a 'critical scenario' (prone to have vectors) were sampled. A rapid immunochromatographic dipstick was used to detect antibodies against Leishmania infantum, confirming by lymph node smears observation and PCR. For Puerto Iguazú, Generalized Linear Models (GLMs) were constructed considering environmental, dog and clinical variables. Pearson's Chi square and Fisher's exact tests were employed to evaluate the association between CVL, dog clinical signs and infestation with other parasites. Cartographic outputs were made and Moran's I indices were calculated as spatial autocorrelation indicators. CVL prevalence rates were 26.18% in 2014 and 17.50% in 2018. No associations were established in environmental models, but dog age and repellent use were significant when running 2014 dog models. Clinical models showed significant associations between seropositive dogs and ophthalmological, dermal signs and onychogryphosis in 2014. In 2018, only adenomegaly was associated. The results of global Moran´s I were not significant but regarding local analysis, six sites in 2014 and one in 2018 presented autocorrelation with neighboring sites. The decrease in CVL prevalence may be associated to transmission stabilization, which could explain the lack of associations with dog-related variables. Further, spatial distribution of CVL is a poor evidence for design of transmission control measures but could be important in case of intensive parasite circulation or when the first autochthonous cases appear. For control success, sensitivity of diagnostic methods, political will and adequate material resources remain critical. Modeling of multiple variables will be required to identify factors that drive disease stabilization/destabilization.

Towards new care strategies in a suburban healthcare center in La Matanza, Argentina: performance evaluation of rapid diagnostic tests for Chagas disease in a non-endemic area

Chagas disease, caused by the protozoan parasite
, remains a major public health problem, affecting approximately seven million people worldwide. Although historically restricted to endemic regions, migration has led to its global distribution, increasing the relevance of congenital transmission in non-endemic areas. If not timely diagnosed and treated, infection can progress to chronic Chagas cardiomyopathy in nearly 30 % of patients, generating substantial social and healthcare burdens. Diagnosis requires concordant results from two or three serological assays based on different antigenic principles, limiting timely access to care in peripheral settings.
We aimed to characterize the clinical, parasitological, and sociodemographic profile of individuals with
infection in La Matanza (Buenos Aires Province, Argentina), and to assess the diagnostic performance of two rapid diagnostic tests (RDTs) from serum samples in this at-risk population.
In this cross-sectional prospective study including 103 subjects, epidemiological analysis revealed that most infected individuals originated from endemic areas, with a substantial proportion of infections consistent with congenital transmission. Clinical evaluation identified electrocardiographic abnormalities in 30.9 % of infected patients, predominantly right bundle branch block. Molecular characterization showed parasites belonging to DTU V (Discrete Typing Unit V) in 7 infections (one coinfected with DTU VI). RDT1 showed a sensitivity of 82.5 % and specificity of 100 %, whereas RDT2 exhibited a sensitivity of 87.5 % and specificity of 85.7 %.
Overall, our findings provide a comprehensive characterization of
infected individuals in a non-endemic region with ongoing transmission dynamics shaped by migration and congenital infection, pointing out advantages and limitations of RDT use in peripheral healthcare settings.

Cardiorenal syndrome (CRS) is a pathology where the heart and kidney are involved, and the deterioration of one of them leads to the malfunction of the other. Diabetes mellitus (DM) carries a higher risk of HF and a worse prognosis. Furthermore, almost half of people with DM will have chronic kidney disease (CKD), which means that DM is the main cause of kidney failure. The triad of cardiorenal syndrome and diabetes is known to be associated with increased risk of hospitalization and mortality. Cardiorenal units, with a multidisciplinary team (cardiologist, nephrologist, nursing), multiple tools for diagnosis, as well as new treatments that help to better control cardio-renal-metabolic patients, offer holistic management of patients with CRS. In recent years, the appearance of drugs such as sodium-glucose cotransporter type 2 inhibitors, have shown cardiovascular benefits, initially in patients with type 2 DM and later in CKD and heart failure with and without DM2, offering a new therapeutic opportunity, especially for cardiorenal patients. In addition, glucagon-like peptide-1 receptor agonists have shown CV benefits in patients with DM and CV disease in addition to a reduced risk of CKD progression.

Prevalence of resistance associated substitutions and efficacy of baseline resistance-guided chronic hepatitis C treatment in Spain from the GEHEP-004 cohort

Treatment guidelines differ in their recommendation to determine baseline resistance associated substitutions (RAS) before starting a first-line treatment with direct-acting antivirals (DAAs). Here we analyze the efficacy of DAA treatment with baseline RAS information. We conducted a prospective study involving 23 centers collaborating in the GEHEP-004 DAA resistance cohort. Baseline NS5A and NS3 RASs were studied by Sanger sequencing. After issuing a comprehensive resistance report, the treating physician decided the therapy, duration and ribavirin use. Sustained virological response (SVR12) data are available in 275 patients. Baseline NS5A RAS prevalence was between 4.3% and 26.8% according to genotype, and NS3 RASs prevalence (GT1a) was 6.3%. Overall, SVR12 was 97.8%. Amongst HCV-GT1a patients, 75.0% had >800,000 IU/ml and most of those that started grazoprevir/elbasvir were treated for 12 weeks. In genotype 3, NS5A Y93H was detected in 9 patients. 42.8% of the HCV-GT3 patients that started sofosbuvir/velpatasvir included ribavirin, although only 14.7% carried Y93H. The efficacy of baseline resistance-guided treatment in our cohort has been high across the most prevalent HCV genotypes in Spain. The duration of the grazoprevir/elbasvir treatment adhered mostly to AASLD/IDSA recommendations. In cirrhotic patients infected with GT-3 there has been a high use of ribavirin.

Distinct Roles for RAB10 and RAB29 in Pathogenic LRRK2-Mediated Endolysosomal Trafficking Alterations

Mutations in the gene encoding leucine-rich repeat kinase 2 (LRRK2) cause familial Parkinson’s disease, and sequence variations are associated with the sporadic form of the disease. LRRK2 phosphorylates a subset of RAB proteins implicated in secretory and recycling trafficking pathways, including RAB8A and RAB10. Another RAB protein, RAB29, has been reported to recruit LRRK2 to the Golgi, where it stimulates its kinase activity. Our previous studies revealed that G2019S LRRK2 expression or knockdown of RAB8A deregulate epidermal growth factor receptor (EGFR) trafficking, with a concomitant accumulation of the receptor in a RAB4-positive recycling compartment. Here, we show that the G2019S LRRK2-mediated EGFR deficits are mimicked by knockdown of RAB10 and rescued by expression of active RAB10. By contrast, RAB29 knockdown is without effect, but expression of RAB29 also rescues the pathogenic LRRK2-mediated trafficking deficits independently of Golgi integrity. Our data suggest that G2019S LRRK2 deregulates endolysosomal trafficking by impairing the function of RAB8A and RAB10, while RAB29 positively modulates non-Golgi-related trafficking events impaired by pathogenic LRRK2.
