Gallbladder Carcinoma in a Eurasian Otter (Lutra lutra)
An adult female Eurasian otter (Lutra lutra), introduced with another subject in the National Park of Gran Paradiso (Aosta Valley Region, Italy), was found dead. The necropsy found a mass involving mainly the gallbladder walls with other multicentric masses in the liver and pancreas. In addition to these, through the histological examination, other nodules were detected in the pancreas, with structure similar to gallbladder neoplasm. Histopathology diagnosed it as neoplasia composed of epithelioid cells, forming lobules of tubules and pseudoacini, with a very low mitotic count, discrete cellular pleomorphism, and prominent fibrous stroma. Neoplastic cells demonstrated positive immunoreactivity for cytokeratin and negative immunoreactivity for S100. Gross and histologic lesions and immunohistochemical findings were consistent with a primary gallbladder carcinoma (GBC) of metastatic type. GBC is a rare neoplasm in both humans and animals, sometimes associated with cholelithiasis and cholecystitis, with few reports in the veterinary literature especially in cattle, pigs, dogs and cats. To the best of our knowledge, this is the first report of GBC in a Eurasian otter.
Detection and Genomic Characterization of Novel Mammarenavirus in European Hedgehogs, Italy
Mammarenaviruses are noteworthy zoonotic pathogens, and the main reservoirs are rodent species. We report the detection of a novel mammarenavirus in 6/183 (3.3%) in necropsied European hedgehogs (Erinaceus europaeus) collected in Italy. The whole-genome sequence obtained for 4 strains revealed a marked genetic diversity but a monophyletic origin.
Different Types of Co-Infection by Contagious Ecthyma Virus, Enteropathogenic Escherichia coli, Mycoplasma conjunctivae, Ecto- and Endo-Parasites in Four Young Alpine Ibex (Capra ibex)
The research aimed to investigate the perinatal pathology of Alpine ibex (Capra ibex) through the study of four young subjects (at the age of 3 to 4 months) found dead in Valle d’Aosta, a region of northwestern Italy. The carcasses were submitted to necropsy followed by an examination of ecto- and endoparasites (ECP and ENP); samples from the gross lesions (in summary, cutaneous papilloma and crusts, ocular discharge, lobular haemorrhagic areas in the lungs, catarrhal–haemorrhagic enterocolitis) were analysed by bacteriological, histopathological, and biomolecular methods to define the etiological agent. The subjects, with various co-infection patterns, were affected by contagious ecthyma virus (ORFV) (agent of a highly diffusive pustular dermatitis transmissible to small ruminants and humans), Enteropathogenic Escherichia coli (EPEC) (major etiological agent of infantile diarrhoea especially in developing countries), Mycoplasma conjunctivae (MC) (cause of an ocular infection common to goats and sheep), various ECP (ticks and keds) and ENP (lung and intestinal nematodes, and coccidia). This study emphasises the potential role of the Alpine ibex in the transmission of infectious diseases to other animals such as to humans and, secondly, the need to apply diversified analytical approaches, with the commitment of various specialistic skills, in order to define, in detail, the various and frequently overlapping causes that led a free-ranging animal to the death.
Dirofilaria immitis in wolves recolonizing northern Italy: are wolves competent hosts?
Background Wild carnivores such as the grey wolf ( Canis lupus ), red fox ( Vulpes vulpes ) and golden jackal ( Canis aureus ) are recognized hosts of Dirofilaria immitis . However, few studies have focused on their actual role in the epidemiology of heartworm infection. This study describes the prevalence and distribution of D. immitis in wolves in a heartworm-endemic area in northern Italy where wolves have recently returned after long-time eradication, and investigates the fertility status of the collected adult nematodes. Methods In the frame of a long-term wolf monitoring programme in northwestern Italy, 210 wolf carcasses from four provinces were inspected for the presence of filarioid nematodes in the right heart and pulmonary arteries. Female heartworms were measured, and their uterine content analyzed according to a previously described “embryogram” technique. Results Three wolves, all originating from a single province (Alessandria), were positive for D. immitis (1.42%, 95% CI: 0.48–4.11%, in the whole study area; 13.6%, 95% CI: 4.7–33.3%, limited to the single province from which infected wolves originated). Mean intensity was 5 worms (range: 3–7) and the female worms measured 21–28 cm in length. Six out of 9 female worms harbored uterine microfilariae: 5 were classified as gravid; 1 showed a “discontinuous gradient”; and 3 were non-gravid. Conclusions The present data show that heartworm infection is already prevalent in wolves that have recolonized the known heartworm-endemic area. Based on “embryogram” results, wolves were shown suitable heartworm hosts. Interestingly, investigated wolves appeared similarly exposed to heartworm infection as sympatric unprotected dogs (owned dogs that have never received any heartworm prevention treatment) sampled at the beginning of the wolf return process.
Molecular Survey for Major Canine Enteric Viral Pathogens in Wild Carnivores, Northwestern Italy
Wild carnivores can harbor pathogens affecting wildlife conservation and domestic animal health. This study surveyed major viral pathogens in free-ranging wolves, red foxes, stone martens, and Eurasian badgers in Northwestern Italy. Duodenal samples from 140 carcasses were screened by consensus PCR for members of the species Protoparvovirus carnivoran1 and for canine adenoviruses (CAdV-1/2). PCR-positive samples underwent sequence-independent amplification and Oxford Nanopore sequencing. Canine parvovirus type 2 (CPV-2) and feline panleukopenia virus (FPV) DNAs were identified in three wolves (6.4%) and one badger (4.3%), whereas CAdV-1 was detected in one red fox (1.8%). Nanopore sequencing yielded near-complete genomes of two CPV-new 2a, one CPV-2c, and one FPV strains, along with partial CAdV-1 sequences. Furthermore, the complete genome of a canine circovirus (CaCV) strain in co-infection with a CPV-2c-positive wolf and partial sequences of a canine kobuvirus (CaKoV) strain were also obtained. Phylogenetic analysis placed these viruses within known European lineages and linked them to domestic and wild hosts. These findings revealed the circulation of multiple viral pathogens among wild carnivores, reflecting ongoing cross-species spillover. Continuing molecular surveillance at the wildlife–domestic interface is recommended.
Current Knowledge of Hepatitis E Virus (HEV) Epidemiology in Ruminants
Hepatitis E virus (HEV) infection represents an emerging public health concern worldwide. In industrialized countries, increasing numbers of autochthonous cases of human HEV infection are caused by zoonotic transmission of genotypes 3 and 4, mainly through the consumption of contaminated raw or undercooked meat of infected pigs and wild boars, which are considered the main reservoirs of HEV. However, in the last few years, accumulating evidence seems to indicate that several other animals, including different ruminant species, may harbor HEV. Understanding the impact of HEV infection in ruminants and identifying the risk factors affecting transmission among animals and to humans is critical in order to determine their role in the epidemiological cycle of HEV. In this review, we provide a summary of current knowledge on HEV ecology in ruminants. A growing body of evidence has revealed that these animal species may be potential important hosts of HEV, raising concerns about the possible implications for public health.
Detection and Characterization of Zoonotic Pathogens in Game Meat Hunted in Northwestern Italy
Wildlife can represent a reservoir of zoonotic pathogens and a public health problem. In the present study, we investigated the spread of zoonotic pathogens (Salmonella spp., Yersinia enterocolitica, Listeria monocytogenes, Shiga-toxin-producing Escherichia coli (STEC), and hepatitis E virus (HEV)) considering the presence of virulence and antibiotic resistance genes in game meat from animals hunted in northwest Italy. During two hunting seasons (2020 to 2022), samples of liver and/or muscle tissue were collected from chamois (n = 48), roe deer (n = 26), deer (n = 39), and wild boar (n = 35). Conventional microbiology and biomolecular methods were used for the detection, isolation, and characterization of the investigated pathogens. Two L. monocytogenes serotype IIa strains were isolated from wild boar liver; both presented fosfomycin resistance gene and a total of 22 virulence genes were detected and specified in the text. Eight Y. enterocolitica biotype 1A strains were isolated from chamois (2), wild boar (5), and deer (1) liver samples; all showed streptogramin and beta-lactam resistance genes; the virulence genes found were myfA (8/8 strains), ymoA (8/8), invA (8/8), ystB (8/8), and ail (4/8). Our data underscore the potential role of wildlife as a carrier of zoonotic and antibiotic-resistant pathogens in northwest Italy and a food safety risk for game meat consumers.

Characterisation of Yersinia enterocolitica strains isolated from wildlife in the northwestern Italian Alps
Yersiniosis is a zoonosis causing gastroenteritis, diarrhoea, and occasionally reactive arthritis and septicaemia. Cases are often linked to meat consumption and the most common aetiological agent is the Gram-negative bacilliform Yersinia enterocolitica bacterium. The occurrence of Yersinia spp. among wild animals has mostly been studied in wild boar, but it has seldom been in other species.A total of 1,868 faecal samples from animals found dead or hunted were collected between 2015 and 2018 in the Valle d’Aosta region of the northwestern Italian Alps. Alpine ibex faecal samples were collected during a health monitoring program in 2018. Bacteria were isolated via PCR and confirmed as Y. enterocolitica biochemically. Strain antimicrobial susceptibility was tested by Kirby–Bauer disc diffusion, and the presence of virulence factors and antimicrobial resistance genes was investigated using whole-genome sequencing.Yersinia enterocolitica strains of biotype 1A were detected in six faecal samples from red deer (0.93%), roe deer (0.49%) and red foxes (0.7%). Strains found in beech martens (3.57%) and Alpine ibex (2.77%) belonged to biotypes 1B and 5, respectively and harboured the pYPTS01 plasmid that had only been detected in Y. pseudotuberculosis PB1/+. All the isolates were resistant to ampicillin and erythromycin.The biovar 1A strains exhibited different virulence factors and behaved like non-pathogenic commensals. The strain from an Alpine ibex also harboured the self-transmissible pYE854 plasmid that can mobilise itself and the pYPTS01 plasmid to other strains. The beech marten could be considered a sentinel animal for Y. enterocolitica. Phenotypic resistance may account for the ability of all the strains to resist β-lactams.
