eHealth Interventions for Fruit and Vegetable Intake
Background. An adequate intake of fruits and vegetables (FVI) has shown benefits for reduced risk of manifesting chronic diseases. Thus, development of effective interventions to promote FVI is paramount. Aim. To assess the effectiveness of eHealth interventions for FVI targeted to healthy population, in comparison with control groups, and identify the moderators contributing to their effectiveness. Method. A database search was conducted in April 2016 and July 2018 using MEDLINE, PsycINFO, ERIC, Google Scholar, SciELO, and RISS, yielding 19 eligible studies. Risk of bias was assessed using Cochrane’s Risk of Bias Tool. Random-effects model was used to calculate effect sizes (ES) by weighted standardized mean differences. Results. This meta-analysis includes 19 studies and 6,894 participants. The most common eHealth tool used was Internet-based interventions (n = 10). Most of studies were conducted in adults (n = 11), followed by children (n = 4), and adolescents (n = 4). The minimum number of behavior change techniques (BCTs) used was one, and the maximum seven. The overall ES was small (0.26, p < .001), favoring eHealth interventions. The between-studies heterogeneity was large (I2 = 62.77%, p < .001). Subgroup analyses showed that the components with larger ES were tailored interventions (0.27, p < .001), computer-based interventions (0.44, p < .001), and use of ≥7 BCTs (0.42, p < .001). Discussion. All studies showed a positive ES favoring interventions. Large heterogeneity could be explained in part by the number of BCTs and type of eHealth tool used. Nonetheless, more concrete evidence regarding other intervention components effectiveness was limited by small sample size. Conclusion. The use of eHealth tools for the improvement of FVI showed to be more effective compared with nonintervention and interventions not using these technologies. Nonetheless, more research is needed to determine the specific combination of intervention components that could translate into larger effectiveness.
Relative Advantage and Compatibility as Drivers of ChatGPT Adoption in Latin American Higher Education: A PLS SEM Study Towards Sustainable Digital Education
As Latin American universities pursue digitally and environmentally sustainable teaching models, understanding why students adopt generative AI is essential. We analyzed data from undergraduate students (n = 792) across five Latin American countries (Peru, Chile, Bolivia, Argentina, and Colombia). Grounded in the diffusion of innovations theory, the study evaluated the effects of relative advantage, compatibility, complexity, trialability, and observability on attitudes towards ChatGPT and examined the effect of attitude on intention to use among higher education students in the region. The reliability and validity of the measurement scale were confirmed, and structural relationships were tested using partial least squares structural equation modeling (PLS-SEM). The model explained 58.1% of the variance in attitude: relative advantage (β = 0.247) and compatibility (β = 0.246) exerted the largest effects, followed by trialability (β = 0.223) and observability (β = 0.167); complexity showed a weaker yet significant effect (β = 0.118). Attitude strongly predicted the intention to use ChatGPT (β = 0.777), accounting for 60.4% of its variance. All paths were significant (p < 0.001), and psychometric indicators exceeded recommended thresholds. These findings indicate that student adoption is driven more by perceived academic benefits and alignment with existing learning routines than by technical ease. Highlighting concrete, ethically delineated use cases and providing guided institutional spaces for experimentation may accelerate the responsible, long-term adoption of generative AI in quality higher education.
Effect of flaxseed (Linum usitatissimum) supplementation on glycemic control and insulin resistance in prediabetes and type 2 diabetes: A systematic review and meta-analysis of randomized controlled trials
Prediabetes and type 2 Diabetes Mellitus (T2DM) are characterized by increased blood sugar concentration and insulin resistance. Although there are only a few reports of potential benefits of flaxseed’s consumption on different metabolic parameters, there is no evidence of its effect among people with these conditions. The present systematic review and meta-analysis aimed to assess the effect of flaxseed supplementation on glycemic control variables and insulin resistance in prediabetes and T2DM. A literature search was conducted through PubMed, Cochrane Central Register of Controlled Trials (CENTRAL), and Web of Science, to identify Randomized Control Trials (RCTs) that evaluated the effect of milled or ground flaxseed supplementation on fasting blood glucose, HbA1c, insulin concentrations, or HOMA-IR. The data were analyzed using Comprehensive Meta-Analysis (CMA) software version 3.3 in a fixed-effect model. Seven studies were included in the systematic review and the meta-analysis, the results showed a significant reduction on fasting blood sugar (SMD: −0.392, 95% CI: −0.596, −0.187, p = <0.001, I2 = 64.81%) insulin concentrations, (SMD: −0.287, 95% CI: −0.534, −0.041, p = 0.022, I2 = 32.53%), HbA1c (SMD: −0.442, 95% CI: −0.770, −0.114, p = 0.008, I2 = 11.058%), and HOMA-IR (SMD: −0.284, 95% CI: −0.530, −0.038, p = 0.024, I2 = 0.00%) after flaxseed supplementation. Flaxseed supplementation seems to improve glycemic control variables and insulin resistance in prediabetes and T2DM; however, more RCTs are needed to have more decisive evidence about doses, method of supplementation, and the possible effect of synergy with the dietetic treatment. •Flaxseed supplementation may improve glycemic control in prediabetes and type 2 diabetes mellitus.•Flaxseed supplementation may improve insulin concentrations and insulin resistance in prediabetes and type 2 diabetes mellitus.•Further studies are needed to address methodological aspects, such as the administration, dosage, and the combined effect with other treatments.
The lncRNA CASC15 regulates SOX4 expression in RUNX1-rearranged acute leukemia
Background Long non-coding RNAs (lncRNAs) play a variety of cellular roles, including regulation of transcription and translation, leading to alterations in gene expression. Some lncRNAs modulate the expression of chromosomally adjacent genes. Here, we assess the roles of the lncRNA CASC15 in regulation of a chromosomally nearby gene, SOX4, and its function in RUNX1/AML translocated leukemia. Results CASC15 is a conserved lncRNA that was upregulated in pediatric B-acute lymphoblastic leukemia (B-ALL) with t (12; 21) as well as pediatric acute myeloid leukemia (AML) with t (8; 21), both of which are associated with relatively better prognosis. Enforced expression of CASC15 led to a myeloid bias in development, and overall, decreased engraftment and colony formation. At the cellular level, CASC15 regulated cellular survival, proliferation, and the expression of its chromosomally adjacent gene, SOX4 . Differentially regulated genes following CASC15 knockdown were enriched for predicted transcriptional targets of the Yin and Yang-1 (YY1) transcription factor. Interestingly, we found that CASC15 enhances YY1-mediated regulation of the SOX4 promoter. Conclusions Our findings represent the first characterization of this CASC15 in RUNX1-translocated leukemia, and point towards a mechanistic basis for its action.
The Inflammatory Process Modulates the Expression and Localization of WT1 in Podocytes Leading to Kidney Damage
Wilms' tumor 1 (WT1) is involved in the development of the urogenital system and is expressed in podocytes throughout life. Inflammation of renal glomeruli causes renal damage-induced nephrotic syndrome and steroid-resistant nephrotic syndrome have mutations in the WT1 gene. The aim of this work was to determine if the inflammatory process modulates the expression and localization of WT1 in podocytes that cause kidney damage using lipopolysaccharide (LPS)-treated mice as a sepsis model. In investigation of renal damage, proteinuria and histology were analyzed. WT1 modulation was analyzed by indirect immunofluorescence, immunohistochemistry and western blot assays, and proinflammatory cytokines were analyzed by quantitative polymerase chain reaction assay. WT1 expression decreased most at 24 and 36 h after the induction of inflammation and phosphorylated WT1 was mainly localized in the cytoplasm, reduced nephrin mRNA expression and increased mRNA expression of tumor necrosis factor α and interleukin 1β. These results indicate that the immune system plays an important role in the modulation of WT1, leading to kidney damage.
Association of diabetes and tuberculosis: impact on treatment and post-treatment outcomes
Objective To determine the clinical consequences of pulmonary tuberculosis (TB) among patients with diabetes mellitus (DM). Methods We conducted a prospective study of patients with TB in Southern Mexico. From 1995 to 2010, patients with acid-fast bacilli or Mycobacterium tuberculosis in sputum samples underwent epidemiological, clinical and microbiological evaluation. Annual follow-ups were performed to ascertain treatment outcome, recurrence, relapse and reinfection. Results The prevalence of DM among 1262 patients with pulmonary TB was 29.63% (n=374). Patients with DM and pulmonary TB had more severe clinical manifestations (cavities of any size on the chest x-ray, adjusted OR (aOR) 1.80, 95% CI 1.35 to 2.41), delayed sputum conversion (aOR 1.51, 95% CI 1.09 to 2.10), a higher probability of treatment failure (aOR 2.93, 95% CI 1.18 to 7.23), recurrence (adjusted HR (aHR) 1.76, 95% CI 1.11 to 2.79) and relapse (aHR 1.83, 95% CI 1.04 to 3.23). Most of the second episodes among patients with DM were caused by bacteria with the same genotype but, in 5/26 instances (19.23%), reinfection with a different strain occurred. Conclusions Given the growing epidemic of DM worldwide, it is necessary to add DM prevention and control strategies to TB control programmes and vice versa and to evaluate their effectiveness. The concurrence of both diseases potentially carries a risk of global spreading, with serious implications for TB control and the achievement of the United Nations Millennium Development Goals.
COVID-19 and Its Effects on the Management of the Basic Quality Conditions in Universities of Peru, 2022
The purpose of this study was to determine the influences of variables, crisis management, distance education, the organizational image, and student satisfaction on the basic quality conditions in Peruvian universities during the COVID-19 pandemic. A quantitative, nonexperimental explanatory approach with stratified random sampling was used. A questionnaire was applied to 513 students from public and private universities in southern Peru who received distance education during the months of September to November 2022. For the analysis of the results, a structural equation model (SEM). A hierarchical linear regression was carried out to test the hypotheses according to the dimensions studied on the student satisfaction scale. The study findings showed that distance education and student satisfaction had positive impacts on the basic quality conditions, while crisis management and the organizational image had a positive relationship but a smaller effect on the quality of the conditions. We concluded that the applied model allows the causal relationships between variables to be explored and that the results will allow university authorities to generate policies that improve their organizational image and crisis management processes and, at the same time, allow them to better plan their crisis management strategies to achieve better satisfaction within the framework of a sustainable university.
Using the optimal seed germination temperature approach to determine the potential distribution of Inga jinicuil in Mexico under climate change scenarios
Inga jinicuil is used extensively in shade coffee farms in Mexico, a diversified agroforestry system providing important environmental goods and services. However, its recalcitrant seeds represent an important barrier to its propagation. Given the climate change scenarios, it will be necessary to generate information on the effect of temperature on germination, a key stage for the establishment and conservation of the species. The objective of the study was to determine the optimal germination temperatures for I. jinicuil using linear and non-linear models, as well as the species’ potential distribution under contrasting climate change scenarios using the GISS-E2-1-G model. Seeds were placed in germination chambers at constant temperatures of 5 ± 0.5 to 40 ± 0.5 °C, and their thermal responses were then modelled using a thermal timing approach. Results indicated a good fit of models of I. jinicuil germination in response to temperature. Seeds germinated across a wide temperature range; the base temperature for germination was in the range of 4.8 to 9.45 °C (average T b : 6.21 ± 2.23 °C). and the ceiling temperature in the range of 44.51 to 49.20 °C (average T c : 47.6 ± 2.73 °C). While the optimal temperature was found in the range of 29.58 to 33.02 °C (average T o : 31.52 ± 1.43 °C). The suboptimal thermal time ( 1 (50)) for germination of 50% of the seed lot was 117.164 ± 0.636°Cd, which under current climatic conditions is reached in 6.6 days. According to climate modeling, the distribution of I. jinicuil populations will decrease by up to 23% in the future relative to the current distribution. Results indicate that high temperatures have a negative effect on germination, which may be related to seed physiology. More research on seed germination and growth is needed to improve the management and conservation of this species and its continued use as a shade tree in coffee agroforestry systems.
Curcumin Sensitizes 4T1 Murine Breast Cancer Cells to Cisplatin Through PAR4 Secretion
Prostate apoptosis response 4 (PAR4), a tumour-suppressor protein, selectively induces apoptosis of cancer cells without affecting normal cells. Its soluble form is induced by secretagogues (e.g., chloroquine), and it induces apoptosis by interacting with the receptor of glucose-regulated protein 78, which is overexpressed in cancer cells. In this study, curcumin was analyzed as an inducer of PAR4 expression in 4T1 murine breast cancer cell. and its ability to induce PAR4 secretion in Balb/c mice. In addition, the cisplatin sensitizing effect of soluble PAR4 was analyzed. The 4T1 cell line was treated in vitro using different concentrations of curcumin; cell viability was analyzed using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay and PAR4 expression by western blotting. The expression of soluble PAR4 in the serum of mice treated with intraperitoneal curcumin was analyzed using the dot-blot method. Moreover, MTT assay was used to analyze the effects of serum from curcumin-treated mice on cell viability. Tumor size was analyzed in mice treated with curcumin alone and in combination with cisplatin. Curcumin showed a dose- and time-dependent effects on cell viability on 4T1 cells, as well as increasing PAR4 expression. Compared with the control group (phosphate-buffered saline), mice treated with curcumin showed an increase in plasma PAR4. In the Balb/C tumor model, mice treated with curcumin and cisplatin showed greater tumor shrinkage than the control group. These results indicate that curcumin induces expression of soluble PAR4 and sensitizes tumor cells to cisplatin.
MicroRNAs in B cell development and malignancy
MicroRNAs are small RNA molecules that regulate gene expression and play critical roles in B cell development and malignancy. miRNA expression is important globally, as B cell specific knockouts of Dicer show profound defects in B cell development; and is also critical at the level of specific miRNAs. In this review, we discuss miRNAs that are involved in normal B cell development in the bone marrow and during B cell activation and terminal differentiation in the periphery. Next, we turn to miRNAs that are dysregulated during diseases of B cells, including malignant diseases and autoimmunity. Further study of miRNAs and their targets will lead to a better understanding of B cell development, and should also lead to the development of novel therapeutic strategies against B cell diseases.
