A Bioactive Extract Rich in Triterpenic Acid and Polyphenols from Olea europaea Promotes Systemic Immunity and Protects Atlantic Salmon Smolts Against Furunculosis
In the present study, the modulation of the transcriptional immune response (microarray analysis) in the head kidney (HK) of the anadromous fish Atlantic salmon ( Salmo salar ) fed a diet supplemented with an olive fruit extract (AQUOLIVE ® ) was evaluated. At the end of the trial (133 days), in order to investigate the immunomodulatory properties of the phytogenic tested against a bacterial infection, an in vivo challenge with Aeromonas salmonicida was performed. A total number of 1,027 differentially expressed genes (DEGs) (805 up- and 222 downregulated) were found when comparing the transcriptomic profiling of the HK from fish fed the control and AQUOLIVE ® diets. The HK transcripteractome revealed an expression profile that mainly favored biological processes related to immunity. Particularly, the signaling of i-kappa B kinase/NF-kappa and the activation of leukocytes, such as granulocytes and neutrophils degranulation, were suggested to be the primary actors of the innate immune response promoted by the tested functional feed additive in the HK. Moreover, the bacterial challenge with A . salmonicida that lasted 12 days showed that the cumulative survival was higher in fish fed the AQUOLIVE ® diet (96.9 ± 6.4%) than the control group (60.7 ± 13.5%). These results indicate that the dietary supplementation of AQUOLIVE ® at the level of 0.15% enhanced the systemic immune response and reduced the A . salmonicida cumulative mortality in Atlantic salmon smolts.
Biochar and Energy Production: Valorizing Swine Manure through Coupling Co-Digestion and Pyrolysis
Anaerobic digestion is an established technological option for the treatment of agricultural residues and livestock wastes beneficially producing renewable energy and digestate as biofertilizer. This technology also has significant potential for becoming an essential component of biorefineries for valorizing lignocellulosic biomass due to its great versatility in assimilating a wide spectrum of carbonaceous materials. The integration of anaerobic digestion and pyrolysis of its digestates for enhanced waste treatment was studied. A theoretical analysis was performed for three scenarios based on the thermal needs of the process: The treatment of swine manure (scenario 1), co-digestion with crop wastes (scenario 2), and addition of residual glycerine (scenario 3). The selected plant design basis was to produce biochar and electricity via combined heat and power units. For electricity production, the best performing scenario was scenario 3 (producing three times more electricity than scenario 1), with scenario 2 resulting in the highest production of biochar (double the biochar production and 1.7 times more electricity than scenario 1), but being highly penalized by the great thermal demand associated with digestate dewatering. Sensitivity analysis was performed using a central composite design, predominantly to evaluate the bio-oil yield and its high heating value, as well as digestate dewatering. Results demonstrated the effect of these parameters on electricity production and on the global thermal demand of the plant. The main significant factor was the solid content attained in the dewatering process, which excessively penalized the global process for values lower than 25% TS.
SHOC2 scaffold protein modulates daunorubicin-induced cell death through p53 modulation in lymphoid leukemia cells
SHOC2 scaffold protein has been mainly related to oncogenic ERK signaling through the RAS-SHOC2-PP1 phosphatase complex. In leukemic cells however, SHOC2 upregulation has been previously related to an increased 5-year event-free survival of pediatric pre-B acute lymphoid leukemia, suggesting that SHOC2 could be a potential prognostic marker. To address such paradoxical function, our study investigated how SHOC2 impact leukemic cells drug response. Our transcriptome analysis has shown that SHOC2 can modulate the DNA-damage mediated by p53. Notably, upon genetic inhibition of SHOC2 we observed a significant impairment of p53 expression, which in turn, leads to the blockage of key apoptotic molecules. To confirm the specificity of DNA-damage related modulation, several anti-leukemic drugs has been tested and we did confirm that the proposed mechanism impairs cell death upon daunorubicin-induced DNA damage of human lymphoid cells. In conclusion, our study uncovers new insights into SHOC2 function and reveals that this scaffold protein may be essential to activate a novel mechanism of p53-induced cell death in pre-B lymphoid cells.
The Shallow Structure of the Jalisco Block (Western Trans-Mexican Volcanic Belt) Inferred from Aeromagnetic Data—Implications for Mineral Deposits
The complex geology of southwestern Mexico results from prolonged interaction between the North American and Farallon plates along an active subduction zone. This process led to crustal growth via oceanic lithosphere consumption, island arc accretion and batholith exhumation, forming great geological features like the Guerrero composite terrane. On the other hand, the Zihuatanejo subterrane, evolved into the Jalisco Block is now bounded by major grabens. Aeromagnetic data from the Mexican Geological Service (1962–2016) were used to map geological structures and contribute to the mineral exploration. Advanced magnetic processing and 3D modeling (VOXI Magnetic Vector Inversion) revealed the Jalisco Block’s complex structure, including Triassic basement, Jurassic–Cretaceous volcanics, and plutonic bodies such as the Puerto Vallarta batholith. Magnetic anomalies are related to intrusive bodies and mineralized zones, notably Peña Colorada (Fe), El Barqueño (Au), and La Huerta. Iron deposits are linked to intrusive volcanic–sedimentary contacts, while gold aligns with intrusive zones and observed magnetic maxima. A notable NW–SE magnetic low at 20 km depth suggests a reactivated back-arc basin and crustal fracture zone. These findings underscore aeromagnetic surveys’ value in both mineral exploration and geological interpretation.
Anaerobic digestion of fourth range fruit and vegetable products: comparison of three different scenarios for its valorisation by life cycle assessment and life cycle costing
Changes in consumer demand due to preferences for a healthier lifestyle have led to a new market offering fruit and salad products ready to eat. This affects the agro-industrial sector and the characteristic of waste streams generated having the organic fraction higher quality and representing a new opportunity of valorisation. This study experimentally evaluated the digestion of wastes derived from the fourth range product sector. It was also proposed the use of this digestate as a fermentation medium for producing plant growth-promoting cultures. Three digestion scenarios were studied: Scenario 1 considered biogas valorisation using a combined heat and power (CHP) unit. Scenario 2 featured biogas upgrading to be used as vehicle fuel. Finally, scenario 3 evaluated the transport of waste materials to the digestion plant by a network of pipes and pumps directly from the production chain. All three scenarios included the land application of a biostimulator based on the production of a plant growth-promoting culture derived from digestate. Life cycle analysis and life cycle costing were used to determine potential environmental impacts and costs over a lifetime of 25 years. The study showed that scenario 1 was the most favourable option for valorising this type of waste, although the economic assessment resulted in negative values for all three alternatives.
iHandU: A Novel Quantitative Wrist Rigidity Evaluation Device for Deep Brain Stimulation Surgery
Deep brain stimulation (DBS) surgery is the gold standard therapeutic intervention in Parkinson’s disease (PD) with motor complications, notwithstanding drug therapy. In the intraoperative evaluation of DBS’s efficacy, neurologists impose a passive wrist flexion movement and qualitatively describe the perceived decrease in rigidity under different stimulation parameters and electrode positions. To tackle this subjectivity, we designed a wearable device to quantitatively evaluate the wrist rigidity changes during the neurosurgery procedure, supporting physicians in decision-making when setting the stimulation parameters and reducing surgery time. This system comprises a gyroscope sensor embedded in a textile band for patient’s hand, communicating to a smartphone via Bluetooth and has been evaluated on three datasets, showing an average accuracy of 80%. In this work, we present a system that has seen four iterations since 2015, improving on accuracy, usability and reliability. We aim to review the work done so far, outlining the iHandU system evolution, as well as the main challenges, lessons learned, and future steps to improve it. We also introduce the last version (iHandU 4.0), currently used in DBS surgeries at São João Hospital in Portugal.
Study of the Spatial Distribution of the Bark Beetle in the Ejido Tixtlancingo
The spatial distribution of Dendroctonus frontalis Zimmermann, 1868, and Dendroctonus mexicanus Hopkins, 1905, was determined from 2020 to 2021 in the Tixtlancingo ejido. The information came from two surveys because, despite the abundant forested areas in this geographical area of the state of Guerrero, Mexico, economic resources for pest biomonitoring are limited. However, it was possible to identify the presence of 76 outbreaks affecting 1117.697 hectares and 95,078 trees, totaling 14,223.8 m3 of standing timber volume. In 2020, 28 outbreaks were reported, with the eastern spatial distribution showing the most damage from bark beetles, particularly in outbreaks 7 and 14 of the surveyed area. The most affected conifers were Pinus maximinoi H. E. Moore (44.71%), Pinus oocarpa Schiede ex Schltdl. (39.93%), and Pinus pseudostrobus Lindl. (15.36%). The affected timber volume was observed in diameter categories of 30 cm for the three pine species, with infestation of 90,549 trees (13,497.6 m3 t.t.v. (total tree volume)) across 1057.64 hectares. In 2021, 48 outbreaks were recorded, with the northeastern and southern parts of the surveyed area showing the most damage from the bark beetle. The trees most affected by the bark beetle were P. oocarpa (59.17%), P. maximinoi (33.94%), and P. pseudostrobus (6.89%). It was observed that the affected volume occurred in trees with diameter categories of 50 cm for the three pine species, affecting 4529 trees (726.214 m3 t.t.v.) distributed over 60.06 hectares. The contribution of this work lies in establishing a baseline for monitoring damage caused by this beetle, which affects forest resources and diminishes the possibility of maintaining carbon capture areas in the medium and long term, thus impacting the Sustainable Development Goals (SDGs) of the 2030 agenda, specifically Goals 11, 13, and 15.
Phytogenics From Sage and Lemon Verbena Promote Growth, Systemic Immunity and Disease Resistance in Atlantic Salmon (Salmo salar)
The transcriptomic response of the head kidney, the main lymphohematopoietic tissue of the body, was evaluated in Atlantic salmon ( Salmo salar ) smolts fed a functional feed containing a phytogenic rich in verbascoside and triterpenic compounds like ursolic acid. Fish (initial body weight = 55.0 ± 0.1 g) were fed two experimental diets (40% crude protein, 22% crude fat; 21.6 MJ/kg gross energy) that only differed in the phytogenic content (0.1% inclusion). Each diet has six replicates and was tested over a period of 133 days. The tested zootechnical feed additive a medicinal plant leaf extract (MPLE) obtained from sage ( Salvia officinalis ) and lemon verbena ( Lippia citriodora ). At the end of the trial, smolts fed the MPLE diet were heavier than their congeners from the control group (271.5 ± 7.9 g vs. 240.2 ± 19.3 g, respectively; P < 0.05). Feed conversion ratio (FCR) values in fish fed the control diet were higher than those in fish fed the MPLE diet (FCR control diet = 1.27 ± 0.08 vs. FCR 0 . 1% MPLE diet = 1.08 ± 0.05; P < 0.05). The immunomodulatory properties of the functional diet were evaluated by means of an in vivo challenge with Aeromonas salmonicida subsp. salmonicida (1 × 10 7 CFU mL –1 ). The microarray analysis of head kidney samples from both dietary groups revealed 1,178 differentially expressed genes (802 upregulated and 376 downregulated). Among them, several biological processes related to immunity were identified in fish fed the MPLE diet ( i.e ., interferon-gamma-mediated signaling pathway, antigen processing and presentation of peptide antigen via MHC class II, autophagy, regulation of i-kappaB kinase/NF-kappaB signaling, and leukocyte activation). Results from the bacterial challenge showed that survival rates were higher in smolts from the MPLE group (90.6 ± 6.4%) in comparison to the control group (60.7 ± 13.5%), confirming the functional benefits of the phytogenic in terms of host’s immunity and disease resistance. Biological processes such as cytoskeleton organization and regulation of cellular protein metabolic process detected in fish fed the MPLE diet supported the metabolic changes related to increased somatic growth promoted. The present findings showed that the inclusion at 0.1% of the tested MPLE obtained from sage and lemon verbena in diets for Atlantic salmon smolts promoted somatic growth, and enhanced their systemic immune response and reduced mortality when fish were challenged with A . salmonicida cumulative, the causative agent of furunculosis in salmonids.
Beyond Institutional Racism? An analysis of the content of the Black Population Health Policy
The concept of Institutional Racism (IR) gained emphasis in the late 1990s and the 2000s. It became one of the central terms in the discussion on racism and the Black Population Health. In Brazil, this category hegemonically influences the understanding of racism in health studies and debates. This qualitative case study paper analyzes the incorporation of the IR concept in Brazil in implementing the National Comprehensive Health Policy for the Black Population. Government documents from the Ministry of Health and the Institute of Applied Economic Research were analyzed. Although the formal inclusion of the discussion about IR in the public policy agenda in the early 21st century is a great achievement, apprehending this fact in isolation disregards the dialectical interaction between individual, institutional, and structural forms in the racialization dehumanizing process. Thus, combating IR depends on facing the reasons that lead institutions to reproduce racism and its consequences on access to housing, work, education, and health services and actions.The concept of Institutional Racism (IR) gained emphasis in the late 1990s and the 2000s. Importance of local and systemic factors in preventing implant displacement in the mandibular body: a scoping review of existing literature
Background The aim of this research was to analyse the current literature on displaced dental implants in the mandibular body, including local and systemic variables related to their cause, and to identify the most frequent location. Methods The study conducted a search of three databases (Pubmed, Scopus, and Web of Science) using specific index terms such as ‘dental implant’, ‘displacement’, ‘dislocation’, ‘displaced’, and ‘mandible’. The analysis focused on the direction of displacement and the characteristics of the bone tissue (bone quality, density, and quantity) in cases where dental implants were displaced. Results A total of 371 articles were obtained. Thirteen of these articles were selected and read in full. To define bone quality, the Lekholm and Zarb classification, modified by Rosas et al., was used. The type II-B bone, which is characterized by thick cortical bone surrounding cancellous bone with extremely wide medullary spaces, presented the largest number of complications. Twenty-two cases were found in which the displacement direction was horizontal. Of these, four were displaced vestibularly, fourteen lingually, and four remained in the center. Additionally, 24 cases presented vertical displacement, with 12 displaced towards the inferior border of the mandible, 9 towards the middle or adjacent to the inferior dental nerve canal, and 3 above the inferior dental nerve canal. Conclusion The accidental displacement of implants within the mandibular body is associated with various risk factors, including the characteristics of the bony trabeculum and the size of the medullary spaces. It is reasonable to suggest that only an adequate pre-surgical diagnostic evaluation, with the help of high-resolution tomographic images that allow a previous evaluation of these structures, will help to have better control over the other factors, thus minimizing the risk of displacement.
