Sepia officinalis ink mitigates gastric ulcer via modulation of antioxidant/anti-inflammatory pathways

Background
Sepia officinalis
ink is a bioactive secondary metabolite rich in melanin granules, which has a wide range of nutritional and therapeutic values and also has been used to prevent various gastrointestinal disorders. Gastric ulcer, the most common gastrointestinal disease, is characterized by severe gastric mucosa damage, and its prevention is currently one of the main goals of clinical and experimental studies. Thus, the present study was focused on evaluating the potential gastroprotective efficacy of
Sepia officinalis
ink extract (SOIE) against ethanol-induced gastric ulcer in rats.
Results
The current results revealed that SOIE administration at the two selected doses improved significantly gastric mucosa integrity as indicated by the significant (
P
< 0.05) amelioration in gastric secretion indices (pH and volume) and the marked decrease in the ulcer index. Moreover, SOIE could counteract the gastric oxidative stress induced by ethanol via a marked decline in malondialdehyde content as well as a significant (
P
< 0.05) increment in glutathione content and antioxidant enzymes activities (catalase and glutathione-s-transferase). Additionally, SOIE treatment caused a significant (
P
< 0.05) reduction in gastric nitric oxide content. Respecting morphological and histopathological studies, SOIE treatment at 200 mg/kg body weight caused marked healing of gastric lesions as indicated by no hemorrhagic bands or injuries observed as well as significantly reduced severity score of ulcer.
Conclusions
SOIE could be used as a promising alternative antiulcerogenic compound to treat severe gastric lesions.

Chia seeds and coenzyme Q10 alleviate iron overload induced hepatorenal toxicity in mice via iron chelation and oxidative stress modulation

Iron overload (IOL) can cause hepatorenal damage due to iron-mediated oxidative and mitochondrial damage. Remarkably, combining a natural iron chelator with an antioxidant can exert greater efficacy than monotherapy. Thus, the present study aimed to evaluate the efficacy of Chia and CoQ

to chelate excess iron and prevent hepatorenal oxidative damage in IOL mice. Male Swiss albino mice (

. Computational chemistry indicates that the phytic acid found in the Chia seeds is stable, reactive, and able to bind to up to three iron ions (both Fe
2+
and Fe
3+
). IOL induced a significant (P < 0.05) increase in serum iron, ferritin, transferrin, TIBC, TSI, RBCs, Hb, MCV, MCH, WBCs, AST, ALT, creatinine, and MDA. IOL causes a significant (P < 0.05) decrease in UIBC, platelets, and antioxidant molecules (GSH, SOD, CAT, and GR). Also, IOL elicits mitochondrial membrane change depolarization, and DNA fragmentation and suppresses mitochondrial DNA copies. Furthermore, substantial changes in hepatic and renal tissue, including hepatocellular necrosis and apoptosis, glomerular degeneration, glomerular basement membrane thickening, and tubular degeneration, were observed in the IOL group. induced significant (P < 0.05) amelioration in all the mentioned parameters. They can mostly repair the abnormal architecture of hepatic and renal tissues induced by IOL, as signified by normal sinusoids, normal central veins, and neither glomerular damage nor degenerated tubules. In conclusion, the combined treatment with Chia + CoQ

exerts more pronounced efficacy than monotherapy in hepatorenal protection via chelating excess iron and improved cellular antioxidant status and hepatorenal mitochondrial function in IOL mice.

Background
Coelomic fluid, a pharmacologically active compound in earthworms, exhibits a range of biological activities, including antioxidant, anti-inflammatory, and anticancer. However, the biological activities exerted by the coelomic fluid can be restrained by its low bioavailability and stability. Liposomes are progressively utilized as an entrapment system for natural bioactive compounds with poor bioavailability and stability, which could be appropriate for coelomic fluid. Thus, the present study was designed to fabricate, characterize, and evaluate the stability of liposomal formulation for
Allolobophora caliginosa
coelomic fluid (ACCF) as a natural antioxidant compound.
Methods
The ACCF-liposomes were developed with a subsequent characterization of their physicochemical attributes. The physical stability, ACCF release behavior, and gastrointestinal stability were evaluated in vitro. The biological activities of ACCF and its liposomal formulation were also determined.
Results
The liposomal formulation of ACCF had a steady characteristic absorption band at 201 nm and a transmittance of 99.20 ± 0.10%. Its average hydrodynamic particle size was 98 nm, with a PDI of 0.29 ± 0.04 and a negative zeta potential (-38.66 ± 0.33mV). TEM further confirmed the formation of vesicular, spherical nano-liposomes with unilamellar configuration. Additionally, a remarkable entrapment efficiency percent (77.58 ± 0.82%) with a permeability rate equal to 3.20 ± 0.31% and a high retention rate (54.16 ± 2.20%) for ACCF-liposomes were observed. The Fourier transform infrared spectroscopy (FTIR) result demonstrated that ACCF successfully entrapped inside liposomes. The ACCF-liposomes exhibited a slow and controlled ACCF release in vitro. Regarding stability studies, the liposomal formulation enhanced the stability of ACCF during storage and at different pH. Furthermore, ACCF-liposomes are highly stable in intestinal digestion conditions comparable to gastric digestion. The current study disclosed that liposomal formulation potentiates the biological activities of ACCF, especially antioxidant, anti-inflammatory, and thrombolytic activities.
Conclusion
These promising results offer a novel approach to increasing the bioaccessibility of ACCF, which may be crucial for the development of pharmaceuticals and nutraceutical-enriched functional foods.

Developing a Chitosan/polyvinyl alcohol hydrogel for gastro-retentive release of ranitidine and enhanced anti-ulcerative properties

Ranitidine is widely used to treat gastrointestinal conditions, but recent studies have revealed severe potential side effects, including a link to cancer. Therefore, this study aims to develop a new gastro-retentive formulation of ranitidine by utilizing the biocompatibility and biodegradability of Chitosan, along with the strength and hydrophilicity of polyvinyl alcohol (PVA). A chitosan/PVA/ranitidine hydrogel was created using the freeze-thaw method and evaluated for stability, ranitidine release behavior, and efficacy in treating ulcers in rats compared to a commercial formulation. The hydrogel demonstrates an average particle size of 69 nm, a polydispersity index of 0.344, and a zeta potential of + 38 mV. Transmission Electron Microscopy confirmed the spherical shape of the formulation, while X-ray diffraction verified its crystalline structure. Additionally, the study observed an impressive encapsulation efficiency of 98.66% ± 1.01 and a high drug content of 49.82% ± 1.29, as confirmed by Fourier transform infrared analysis. The prepared hydrogel controls the release of ranitidine over 12 h, with an average release of 87.98% ± 4.01%. The hydrogel exhibits minimal degradation over 15 days, greater thermal stability than ranitidine, and adequate stability in acidic gastric conditions. Furthermore, the cytotoxicity assay demonstrated that the hydrogel is biocompatible and promotes cell growth. The study discovered that the hydrogel formulation enhances the effects of ranitidine, particularly its antioxidant and anti-inflammatory properties. In vivo studies illustrated the hydrogel’s promising ulcer-healing properties, suggesting potential use in treating peptic ulcers. Hence, the chitosan/PVA hydrogel can be used as a possible drug delivery system for the sustained release of ranitidine.

mono-fatty acyl amino acid glucosyl esters were synthesized, and their physicochemical properties and biological activities were evaluated. The design and preparation of these esters were chemically performed via the reaction of glucose with different fatty acyl amino acids as renewable starting materials, with the suggestion that they would acquire functional characteristics superior and competitive to certain conventional surfactants. Clinical and molecular characterization of 14 Egyptian children with fructose-1,6-bisphosphatase deficiency

Background
Fructose-1,6-bisphosphatase (FBP1) deficiency is a rare inherited disease characterized by recurrent episodes of lactic acidosis and ketotic hypoglycemia. To date, no cases have been reported in the Egyptian population. This study aimed to elucidate the phenotypic and molecular spectrum of FBP1 deficiency in Egypt.
Methods
This observational study included children with FBP1 deficiency diagnosed and managed at an Egyptian medical center between 2022 and 2024. Clinical and laboratory data of acute metabolic episodes were thoroughly reviewed. All patients underwent blood acylcarnitine assay, urinary organic acids analysis, and whole-exome sequencing. Patients’ outcomes were classified into favorable, neurodevelopmental impairment, and death.
Results
This cohort included 14 Egyptian children (from 11 families) with FBP1 deficiency. The median age at disease onset was 13 months, ranging from the first week of life to 36 months. All patients exhibited acute lactic acidosis, and most (13/14) had hypoglycemia. Four
FBP1
variants were identified: c.88G > T (p.Glu30Ter), c.652_661delinsTCACGAGGGCT (p.Arg218SerfsTer9), c.960delinsGG (p.Ser321ValfsTer13), and c.902_904del (Glu301del). The c.960delinsGG variant was detected in nine cases, suggesting a founder effect. The c.652_661delinsTCACGAGGGCT is a novel variant. One case had a coexisting partial biotinidase deficiency. Regarding outcome, two patients died during the neonatal period, while the remainder achieved normal neurodevelopment.
Conclusion
This is the first study of FBP1 deficiency in Egypt, which expands the demographic, clinical, and genetic spectrum of this rare disease.

Optimizing the Growth, Health, Reproductive Performance, and Gonadal Histology of Broodstock Fantail Goldfish (Carassius auratus, L.) by Dietary Cacao Bean Meal

The potential effects of cacao bean meal, Theobroma cacao L., (CBM) on the growth, health status, blood biochemical parameters, antioxidant, immune status, physiological parameters, female reproductive performance, and gonadal histological features of fantail goldfish (Carassius auratus, L.) were evaluated using a complete randomized block design with sex as a block. The trial lasted for 60 days. A total of 54 healthy fantail goldfish (36 broodstock females and 18 broodstock males) were randomly allocated into three treatments with supplementation of three levels of cocoa powder 0, 5, and 10 g kg−1 diet, CBM0, CBM5, and CBM10, respectively, with the sex ratio being four females:two males per replicate. The body weight gain and feed conversion ratio of males were increased in the CBM10 treatment (p < 0.05). The CBM10 diet improved relative feed costs (p < 0.05). Females fed on the CBM10 diet had an increase in the serum level of total protein (p = 0.001). Females fed on a diet supplemented with CBM5 showed a decrease in the serum level of triglyceride compared to females fed on CBM0 and CBM10 diets (p = 0.03). CBM10 diet increased the serum superoxide dismutase (SOD) activity of fish compared to CBM0 and CBM5 diets (p = 0.004). Serum levels of testosterone and estradiol were significantly increased in males fed on the CBM10 diet. The female reproductive performance was improved by CBM supplementation (p < 0.05). Ovarian histology exhibited increased granulation and follicle numbers after dietary CBM supplementation compared to the control treatment. Therefore, cacao bean meal can be used as a feed supplement in the diets of fantail goldfish for improving the growth, health status, and female reproductive performance, economic efficiency, and gonadal histological structure.

Novel Features of Interactive Augmented Reality Advertisements and its Effect on Stimulating User Engagement

Modernity features, technological advances and economic conditions have added new challenges to the world of advertising and tourism promotion. In accordance with the current intelligent era and its special features, it requires technical development in the presentation of attractive tourist advertising ideas so that Egyptian tourism advertising campaigns are unique and different. It became necessary to find an effective interactive way to establish a close relationship with potential users for the purpose of motivating the potential user and to make it an authentic party that shares his senses in the advertising process to consolidate his relationship with advertising and thus the tourist service and achieve the elements of attraction and engagement. According to the development of the properties of interactive augmented reality technology and its unique features in creating a built-in environment that penetrates the spatial and temporal boundaries where the advertising designer was able to embody the worlds of tourism accurately in addition to many distinctive qualities such as interactive quality and dazzle and others, which helps to dipping the user with the advertising experience. The research used this technology as one of the dazzling tourism advertising solutions that help in effectively embodying the Egyptian tourism reality for users. This study follows an experimental approach using two ads implemented with AR technology and two traditional ones for testing two aspects: firstly, the capability to interactively integrate AR into tourism advertisements (ads) using a smartphone to increase the rates of user engagement over those of traditional ones; and secondly, the effect of an interactive tourism advertising experience on enhancing the novelty principle, This study proposed that, H1: the interactive integration of AR in tourism advertising using smartphones leads to higher rates of user engagement. H2: the impact of an augmented tourism advertising experience enhances the novelty principle. The results indicated the effectiveness of such interactive AR technology for increasing the user engagement rates and enhance the novelty principle.

The Political Interests of the Live Stream Egyptian Global Ceremonies on YouTube Channel as a Novel form of Video Digital Advertising

Egypt has recently embraced a modern approach to challenge traditional digital advertisements, capitalizing on political trends to bolster its global standing, particularly in conjunction with internationally announced presidential development processes. To achieve this, the country leveraged Live Stream Egyptian Global Ceremonies on YouTube channel, broadcasting them live via the Internet to audiences worldwide. The research problem focused on examining the role of political interest in organizing Global Ceremonies in Egypt, and their contribution to the development of a contemporary form of video digital advertising. This research aims to provide valuable insights into the multifaceted effects of such practices. This study investigates the role of these ceremonies as an innovative video digital advertising platform for conveying political messages and shaping public opinion in the realm of political communication. By analyzing their impact, and shed light on how these events strengthen Egypt's international standing and influence global public opinion, encompassing political, economic, and touristic aspects. This study specifically centers on notable events such as the Pharaoh's Golden Parade and the Way of the Rams ceremony. It explores how these events influence users' expectations for the opening of the Grand Egyptian Museum. The study adopts an analytical approach, examining worldwide reactions to these events and transforming them into digital advertisements. The statistical analysis reveals a strong positive response from global users of the YouTube channel.
